The actin cytoskeleton plays crucial roles in many cellular processes, including regulated secretion. However, the mechanisms controlling F-actin dynamics in this process are largely unknown. Through 3D time-lapse imaging in a secreting organ, we show that F-actin is actively disassembled along the apical plasma membrane at the site of secretory vesicle fusion and re-assembled directionally on vesicle membranes. Moreover, we show that fusion pore formation and PIP2 redistribution precedes actin and myosin recruitment to secretory vesicle membranes. Finally, we show essential roles for the bran

Candida albicans, a commensal fungal pathogen, is a leading cause of opportunistic infections in immunocompromised individuals. High-resolution imaging of its cellular architecture is essential for uncovering pathogenic mechanisms and identifying antifungal targets. Here, we applied live-cell stimulated emission depletion (STED) microscopy to visualize subcellular structures in C. albicans with nanoscale precision. We found that fluorophores commonly used for STED imaging in mammalian cells were ineffective in C. albicans. In contrast, Nile Red enabled robust labelling of dynamic lipid-rich st

Store-operated Ca2+ channel (SOC)-regulated Ca2+ entry is involved in inflammation and colorectal cancer (CRC) progression, but clinically applicable treatments targeting this mechanism are lacking. Recent studies have shown that nonsteroidal anti-inflammatory drugs (NSAIDs) not only inhibit inflammation but they also suppress Ca2+ entry via SOC (SOCE). Therefore, delineating the mechanisms of SOCE inhibition by NSAIDs may lead to new CRC treatments. In this study, we tested eight candidate NSAIDs in Ca2+ imaging experiments and found that Aspirin and Sulindac were the most effective at suppre

Type I collagen is the most abundant extracellular matrix protein in bone and other connective tissues and plays key roles in normal and pathological bone formation as well as in connective tissue disorders and fibrosis. Although much is known about the collagen biosynthetic pathway and its regulatory steps, the mechanisms by which it is assembled extracellularly are less clear. We have generated GFPtpz and mCherry-tagged collagen fusion constructs for live imaging of type I collagen assembly by replacing the α2(I)-procollagen N-terminal propeptide with GFPtpz or mCherry. These novel imaging p

Leader and follower cells drive collective cancer cell migration through distinct and interactive roles, but their specific metabolic signatures remain largely unknown. Here, we developed and applied a workflow that integrates time-lapse imaging, single-cell tracking, microsampling and direct infusion high-resolution mass spectrometry to profile the lipidomes of individually sampled leader and follower cells from highly migratory TNBC lines HCC1143 and HCC38. Approximately 120 metabolites were detected per cell at the MS1 level, and 70 compounds were assigned based on matched MS2 fragments, in

Over the years, the utilization of in vitro fertilization (IVF) in bovine embryo production has increased globally to accelerate the selection of cows with high genetic values. The selection of embryos with high implantation potential is a critical factor in establishing pregnancy. Time-lapse monitoring (TLM) has emerged as a new technique that allows frequent and non-invasive imaging of developing embryos. TLM is considered to have several advantages over the conventional morphological evaluation of embryos, which has been widely used in bovine embryo production. Establishing a novel embryo s

Colobomata, persistent optic fissures, frequently cause congenital blindness. Here, we focused on optic fissure fusion using in vivo time-lapse imaging in zebrafish. We identified the fusion initiating cells, which we termed "pioneer cells." Based on morphology, localization, and downregulation of the neuroretinal (NR) precursor marker rx2, these cells could be considered as retinal pigment epithelial (RPE) progenitors. Notably, pioneer cells regain rx2 expression and integrate into the NR after fusion, indicating that they do not belong to the pool of RPE progenitors, supported by the lack of

Cell sheet morphogenesis characterizes key developmental transitions and homeostasis, in vertebrates and throughout phylogeny, including gastrulation, neural tube formation and wound healing. Dorsal closure, a process during Drosophila embryogenesis, has emerged as a model for cell sheet morphogenesis. ∼140 genes are currently known to affect dorsal closure and new genes are identified each year. Many of these genes were identified in screens that resulted in arrested development. Dorsal closure is remarkably robust and many questions regarding the molecular mechanisms involved in this complex

Intravital microscopy is a powerful tool that enables imaging various biological processes in live animals. Here, we describe a series of procedures designed to image subcellular structures, such as endosomes and secretory vesicles in the salivary glands (SGs) of live rats. To this aim, we used fluorescently labeled molecules and/or fluorescently tagged proteins that were transiently transfected in the live animal.

Chromium and nickel cause allergic contact dermatitis, a common biological skin response to sensitizing agents. This study used a conventional in vitro wounding model to study the impact of sensitizing agents on the innate immune response of human keratinocytes. Experiments were designed to evaluate the involvement of specific Toll-like receptors and metalloproteinases as effectors molecules downstream, at a molecular level. Further, keratinocytes were co-cultured with monocytes (THP-1 cells) to reproduce an inductive stimulus on monocytes made by metals. Human keratinocytes (HaCat) were grown

The Lyme disease spirochete Borrelia burgdorferi exists in nature in an enzootic cycle that involves the arthropod vector Ixodes scapularis and mammalian reservoirs. To disseminate within and between these hosts, spirochetes must migrate through complex, polymeric environments such as the basement membrane of the tick midgut and the dermis of the mammal. To date, most research on the motility of B. burgdorferi has been done in media that do not resemble the tissue milieus that B. burgdorferi encounter in vivo. Here we show that the motility of Borrelia in gelatin matrices in vitro resembles th

Cd(2+) is carcinogenic to both humans and experimental animals. We present quantitative time-course imaging of Cd(2+)-induced variation in the membrane permeability of single live human bladder cancer cells (T24) to ferrocenemethanol using scanning electrochemical microscopy (SECM). High temporal resolution combined with non-invasive nature renders a time-lapse SECM depth scan, a promising method to quantitatively investigate the effectiveness, kinetics, and mechanism of metal ions based on the responses of single live cells in real time. Under unstimulated conditions, T24 cells have constant

An automated technique for the identification, tracking and analysis of biological cells is presented. It is based on the use of nanoparticles, enclosed within intra-cellular vesicles, to produce clusters of discrete, point-like fluorescent, light sources within the cells. Computational analysis of these light ensembles in successive time frames of a movie sequence, using k-means clustering and particle tracking algorithms, provides robust and automated discrimination of live cells and their motion and a quantitative measure of their proliferation. This approach is a cytometric version of the

Pseudomyxoma peritonei (PMP) is a rare condition that results from the dissemination of a mucinous primary tumor and the resultant accumulation of mucin-secreting tumor cells in the peritoneal cavity. PMP can arise from various types of cancers, including appendiceal, ovarian, and colorectal, though appendiceal neoplasms are by far the most common etiology. PMP is challenging to study due to its (1) rarity, (2) limited murine models, and (3) mucinous, acellular histology. The method presented here allows real-time visualization and interrogation of these tumor types using patient-derived ex vi

The high kidney uptake of prostate-specific membrane antigen (PSMA) radiotracers remains a significant challenge in the clinical diagnosis and treatment of prostate cancer (PCa). The aim of this study was to reduce the absolute kidney uptake of PSMA tracers by modifying targeting groups, thereby increasing the tumor-to-kidney (T/K) ratio. [99mTc]Tc-AuK-NapLPRO-HYNIC-EDDA demonstrated the most prominent imaging potential. It bound stably within the active cavity of the PSMA protein with nanomolar affinity (Kd = 14.73 nM). Density functional theory (DFT) and molecular dynamics provide a theoreti

The plasma membrane of cells is an ordered environment, giving rise to anisotropic orientation and restricted motion of molecules and proteins residing in the membrane. At the same time as being an organized matrix of defined structure, the cell membrane is heterogeneous and dynamic. Here we present a method where we use fluorescence imaging of linear dichroism to measure the orientation of molecules relative to the cell membrane. By detecting linear dichroism as well as fluorescence anisotropy, the orientation parameters are separated from dynamic properties such as rotational diffusion and h

The transformation of epithelial cells into Non-Professional Phagocytes (NPPs) is a conserved and versatile adaptation that occurs in response to immune challenges, tissue remodeling, and apoptotic debris clearance. In the Drosophila melanogaster ovary, follicle cells (FCs) acquire phagocytic capabilities under stress, providing a powerful model to study this process. Using time-lapse live imaging, we captured dynamic behaviors associated with FC-to-NPP differentiation that are too transient to detect by static imaging. Our approach confirmed established features, including germline cell death

Conventional multi-step warming protocols (MWP) for vitrified blastocysts are labor-intensive and susceptible to handling variability. This study evaluated whether a shortened warming protocol (SWP) could support post-warming recovery and developmental competence comparable to those of conventional MWP. A retrospective propensity score-matched cohort study was conducted using 546 single vitrified-warmed blastocyst transfer cycles (271 SWP and 275 MWP). Post-warming developmental behavior was assessed using time-lapse imaging, including the start of re-expansion (tSRE), completion of re-expansi

Currently, incubators with a time-lapse system are widely used for in vitro embryo production in several species, however, their effect on alpaca embryo development compared to conventional incubators remains unknown. The aim of this study was to compare early in vitro embryo development in alpacas using a time-lapse incubator system versus a conventional incubator. Ovaries were obtained from a slaughterhouse and 1048 cumulus-oocyte complexes (COCs) were collected and in vitro matured for 26 h in either a time-lapse system (n = 542) or a conventional incubator (n = 542). All the matured COCs w
