Enzymic evaluation of thiamin, riboflavin and pyridoxine status of parturient women and their newborn infants.

1. Thiamin, riboflavin and pyridoxine status of 'low-income-group' mothers and their newborn infants was assessed by analysing paired samples of maternal and umbilical cord blood for erythrocyte transketolase (EC 2.2.1.1) (ETK), erythrocyte glutathione reductase (EC 1.6.4.2) (EGR), and erythrocyte aspartate aminotransferase (EC 2.6.1.1) (EAA) activities. 2. The vitamin status of the infants was better than that of the mothers. 3. Most of the mothers and some of the infants had biochemical evidence of thiamin and riboflavin deficiency. 4. The pregnant women had a higher EAA activity and also hi…

Thiamin and Alzheimer's disease.

Because of clinical and neuropathological overlap between the characteristics of dementia of the Alzheimer type (DAT) and of a human thiamin deficiency syndrome (Wernicke-Korsakoff syndrome), thiamin pyrophosphate (TPP) dependent processes have been studied in DAT brain and other tissues. The activities of 3 TPP-dependent enzymes are reduced in DAT brain: transketolase (TK), the pyruvate dehydrogenase complex (PDHC), and the alpha-ketoglutarate dehydrogenase complex (KGDHC). Quantitatively, the most marked reductions are in KGDHC (to less than 20% of normal). In cultured skin fibroblasts, KGDH…

Natural establishment of thiaminase activity in the alimentary tract of newborn lambs and effects on thiamine status and growth rates.

Thiaminase activity was detected in the faeces of lambs at 2 to 5 days of age. Levels of activity increased for 10 days and then declined over the next 3 to 4 weeks. Decreased erythrocyte transketolase activity indicated thiamine insufficiency in lambs with high thiaminase activity. Mean growth rates were 17% less in lambs with high thiaminase activity than in lambs with zero or low thiaminase activity. Bacillus thiaminolyticus was the only organism isolated which produced thiaminase. Treatment of newborn lambs with intramuscular injections of sulphadoxine did not prevent them from excreting t…

[35S-thiamine transport in the rat small intestine in hypovitaminosis B1 and acute alcoholic intoxication].

35S-thiamine absorption was studied on an isolated strip of the small intestine in experiments on male albino rats with hypovitaminosis B1 and acute alcohol poisoning. The degree of hypovitaminosis was determined by the analysis of blood transketolase. Hypovitaminosis B1 provoked increased thiamine absorption by the mucous membrane of the small intestine. Concurrently, no changes in active transport of thiamine through the wall of the small intestine were observed. Single administration of ethanol in a dose of 5 g/kg body weight into the stomach resulted in a substantial inhibition of active t…

Animal models of alcoholic neuropathy: morphologic, electrophysiologic, and biochemical findings.

A chronic high alcohol intake was induced in rats through the use of two procedures: the schedule-induced polydipsia technique and the liquid diet technique. Rats consumed 11-12 g of ethanol per kilogram body weight per day for 16 to 18 weeks. Morphologic evidence of a mild distal axonal neuropathy in the ventral caudal nerve was proposed. The red blood cell transketolase levels were normal, indicating that the rats were not deficient in thiamine and suggesting that the axonal degeneration was due to the direct toxic effect of alcohol. Axonal transport studies demonstrated a significant increa…

Fulminant beriberi heart disease with lactic acidosis: presentation of a case with evaluation of left ventricular function and review of pathophysiologic mechanisms.

Cardiac beriberi is considered a rare disease in western society. A patient with fulminant Shoshin-type beriberi was studied in the acute phase and found to have severe metabolic acidosis, high output biventricular failure, and markedly low systemic vascular resistance. Red blood cell transketolase activity was abnormally low. Following treatment with thiamine, diuretics, digitalis and oxygen, all abnormalities disappeared. The historical background of the disease is reviewed along with a discussion of pathophysiologic mechanisms responsible for the hemodynamic profile and lactic acidosis. Ang…

Thiamine Deficiency Increases Ca2+ Current and CaV1.2 L-type Ca2+ Channel Levels in Cerebellum Granular Neurons.

Thiamine (vitamin B1) is co-factor for three pivotal enzymes for glycolytic metabolism: pyruvate dehydrogenase, α-ketoglutarate dehydrogenase, and transketolase. Thiamine deficiency leads to neurodegeneration of several brain regions, especially the cerebellum. In addition, several neurodegenerative diseases are associated with impairments of glycolytic metabolism, including Alzheimer's disease. Therefore, understanding the link between dysfunction of the glycolytic pathway and neuronal death will be an important step to comprehend the mechanism and progression of neuronal degeneration as well…

Thiamine, riboflavin and pyridoxine deficiency in psychiatric in-patients.

One hundred and seventy-two successive admissions to a district general hospital psychiatric unit were examined. Routine psychiatric, drug and dietary histories were taken and signs of avitaminosis B specifically noted. Red cell transketolase (for thiamine deficiency), glutathione reductase (for riboflavin deficiency) and aspartate transaminase (for pyridoxine deficiency) were measured. Of the patients, 53 per cent were deficient in at least one vitamin, 12 per cent in more than one (30 per cent in thiamine, 27 per cent in riboflavin and 9 per cent in pyridoxine). Schizophrenics and alcoholics…

Thiamin status of the elderly: dietary intake and thiamin pyrophosphate response.

OBJECTIVE: The present study was undertaken to investigate the relationship between thiamin intake and its biochemical status in 60 free-living (30 male, 30 female) elderly subjects (> or = 65 years). DESIGN: Dietary intake was estimated using a 3-nonconsecutive-day food record. Biochemical status was assessed by measuring the stimulating effect of thiamin pyrophosphate (TPP) on transketolase activity (TPP effect) in hemolyzed erythrocytes, which is a functional test indicative of metabolic availability of thiamin. RESULTS: Average daily thiamin intake was above the recommended requirement (…

Wernicke's encephalopathy in patients on peritoneal dialysis or hemodialysis.

Although the occurrence of Wernicke's encephalopathy (WE) in patients on dialysis is frequently alluded to, review of the literature reveals … ty of WE in patients on dialysis may in part be explained by studies indicating a genetic defect in transketolase activity. Patients on dialysis are also potentially at risk for thiamine deficiency because of anorexia, vomiting, and intravenous alimentation. Other factors altering thiamine requirements, such as glucose load or infections, may also contribute. P …

[Thiamine, riboflavine and pyridoxine supply in chronic alcoholism (author's transl)].

Tests on 50 chronic alcoholics demonstrated that there was a reduced supply of thiamine (measured with the transketolase activation test) and riboflavine (glutathione reductase test) compared with results obtained in 1152 healthy controls. A high risk of thiamine deficiency was present in 45.4% of male and 50% of female alcoholics; risk of riboflavine deficiency was 11.1% and 16.7%, respectively. Corresponding results in the normal controls were 2% for thiamine and 0.4% for riboflavine deficiency. There was a significant relationship between enzymatic values of thiamine deficiency, on the one …

[A nutritional survey of 38 single elderly persons with free choice of food].

The nutritional habits of 38 randomly selected single aged people with free choice of food have been investigated by the precise weighing method. Blood serum levels of the vitamins A, C, and E and of beta-carotene and biotin were analyzed, and the nutritional status with respect to the vitamins B1, B2, and B6 was examined by the transketolase-test, the glutathione-reductase-test, and the glutamic-acid-oxalo-acetic-transaminase-test. In relation to the desirable daily supply of nutrients and other dietary components the food was deficient in the vitamins B1, and B6 and in magnesium. For women a…

Thiamine deficiency in fulminant hepatic failure and effects of supplementation.

Nine out of 24 patients with acute hepatocellular necrosis leading to fulminant hepatic failure showed biochemical evidence of thiamine deficiency early in the course of their illness, probably as a result of inadequate intake of the vitamin. This was corrected by twice daily administration of intravenous vitamin supplements containing thiamine hydrochloride (100 mg b.d.). These studies indicate that conversion of thiamine hydrochloride to its biologically active co-enzyme form, thiamine pyrophosphate, is possible even in the presence of severe acute hepatocellular necrosis, and it is suggeste

TKT maintains intestinal ATP production and inhibits apoptosis-induced colitis.

Inflammatory bowel disease (IBD) has a close association with transketolase (TKT) that links glycolysis and the pentose phosphate pathway (PPP). However, how TKT functions in the intestinal epithelium remains to be elucidated. To address this question, we specifically delete TKT in intestinal epithelial cells (IECs). IEC TKT-deficient mice are growth retarded and suffer from spontaneous colitis. TKT ablation brings about striking alterations of the intestine, including extensive mucosal erosion, aberrant tight junctions, impaired barrier function, and increased inflammatory cell infiltration. …

Thiamin deficiency and uncomplicated falciparum malaria in Laos.

OBJECTIVE: Thiamin deficiency complicates severe Plasmodium falciparum malaria in Thailand and may contribute to acidosis. We therefore estimated the frequency of biochemical thiamin deficiency in patients presenting with uncomplicated falciparum malaria in southern Laos. METHODS: Red cell transketolase activation coefficients (alpha) were measured in 310 patients presenting with uncomplicated falciparum malaria and 42 days after starting treatment. RESULTS: Twelve per cent of patients had biochemical evidence of severe deficiency (alpha values >1.31) at presentation, declining to 3% 42 days…
