The impact of chlorophyllin on deoxynivalenol transport across jejunum mucosa explants obtained from adult pigs.

Regardless of the efforts put into preventing or reducing fungal growth, extensive mycotoxin contamination has been reported in animal feeds. In the case of pigs, one of the mycotoxins of major concern is deoxynivalenol (DON). The use of adsorbents as feed additives represents one of the strategies to control mycotoxins' contamination in feedstuff. Therefore, the aim of the study was to verify the ability of chlorophyllin (CHL) to reduce the absorption rate of DON in swine mucosa explants. Intestine was obtained from routinely slaughtered adult pigs. The mucosa explants were studied by means o

Exploring Ecological Alternatives for Crop Protection Using Coriandrum sativum Essential Oil.

Essential oils (EOs) are a natural source of active compounds with antifungal, antimycotoxigenic, and herbicidal potential, and have been successfully used in organic agriculture, instead of chemical compounds obtained by synthesis, due to their high bioactivity and the absence of toxicity. The aim of this study was to highlight the importance of Coriandrum sativum essential oil (CEO) as a potential source of bioactive constituents and its applications as an antifungal and bioherbicidal agent. The CEO was obtained by steam distillation of coriander seeds and GC-MS technique was used to determi

Co-occurrence of aflatoxins B₁, B₂, G₁ and G₂, ochratoxin A, zearalenone, deoxynivalenol, and citreoviridin in rice in Brazil.

A total of 230 samples of processed rice and its sub-products or derived products were analysed to establish the co-occurrence of several mycotoxins. Samples were analysed in the period 2007-2009 due to the outbreak of beriberi associated with the consumption of rice stored in inappropriate conditions in Brazil. According to data from the Ministry of Health, 323 cases of disease were registered in 2006, of which at least 47 cases resulted in death. The occurrence of total aflatoxin (AFT) (aflatoxin B(1) + B(2) + G(1) + G(2)), ochratoxin A (OTA), zearalenone (ZON), deoxynivalenol (DON), and cit

Diacetoxyscirpenol toxicity in pigs.

Purified diacetoxyscirpenol (DAS) mycotoxin fed to growing pigs at 0, 2, 4, 8, and 9 ppm for as long as nine weeks caused several lesions at all levels. The pigs developed multifocal, proliferative, gingival, buccal and lingual lesions. The small intestine had both glandular and mucosal cell hyperplasia. No other lesions were seen at necropsy or in any of the 19 tissues examined microscopically using haematoxylin and eosin stain. A one-way analysis of variance revealed statistically significant (P less than 0.05) decreased ration consumption and weight gain at all levels of DAS. Total ration r

[Effect of mycotoxins on the rate of fermentation in Saccharomyces cerevisiae].

The rate of fermentation of Saccharomyces cerevisiae is partially inhibited by different mycotoxins. This effect is remarkable with T2-toxin and diacetoxyscirpenol, slight with aflatoxin-B1, penicillic acid and patulin. On the contrary, the butenolide appears as a stimulator of the alcoholic fermentation.

Recent advances in the mechanism of deoxynivalenol-induced hepatotoxicity and protective strategies.

Deoxynivalenol (DON) is a type B trichothecene mycotoxin produced by fusarium species, which widely contaminates grains and feed, posing a serious threat to human and animal health, with the liver as one of its key target organs. This review summarises in vivo and in vitro studies indicating that DON induces liver injury through multiple synergistic mechanisms, among which ferroptosis has attracted increasing attention; other core mechanisms include ribosomal stress response triggered by ribosomal function inhibition, reactive oxygen species (ROS) burst and lipid peroxidation (LPO), inflammato

Mechanistic insights into deoxynivalenol-Induced hepatic cholestasis via IRE1α/HNF1α/FXR signaling dysregulation in mice.

Deoxynivalenol (DON), a trichothecene mycotoxin ubiquitously contaminating agricultural commodities, foodstuffs, and water systems, poses significant health risks to humans and livestock. As the primary detoxification organ, the liver exhibits marked susceptibility to DON-induced toxicity. Our study demonstrated that DON triggers hepatocellular injury by disrupting bile acid (BA) homeostasis and activating pro-inflammatory cascades. In murine models, DON exposure significantly elevated systemic and intrahepatic total bile acid (TBA) levels while upregulating pro-inflammatory cytokine expressio

Quercetin alleviates Deoxynivalenol-induced myocardial fibrosis in chicken by regulating MCU-mediated mitochondrial calcium overload.

Deoxynivalenol (DON), a trichothecene mycotoxin commonly found in contaminated cereals, poses a major threat to animals and human health. While DON-induced damage to the intestines, liver, and kidneys in chickens is well-documented, its cardiac toxicity remains insufficiently characterized. Quercetin (QUE), a plant-derived flavonoid with antioxidant and cytoprotective properties, has been shown to protect against cardiac damage. However, it is unclear whether QUE has a protective effect against DON-induced cardiac damage in chicken. Therefore, this study combined an in vivo chicken model of DO

Effects of toxic exposure to molds and mycotoxins in building-related illnesses.

The authors studied 100 patients who had been exposed to toxic molds in their homes. The predominant molds identified were Alternaria, Cladosporium, Aspergillus, Penicillium, Stachybotrys, Curvularia, Basidiomycetes, Myxomycetes, smuts, Epicoccus, Fusarium, Bipolaris, and Rhizopus. A variety of tests were performed on all, or on subgroups of, these patients. Sensitivities and exposures were confirmed in all patients by intradermal skin testing for individual molds (44-98% positive), and by measurement of serum antibodies. Abnormalities in T and B cells, and subsets, were found in more than 80%

Mitochondrial dysfunction via oxidative stress and calcium imbalance mediates antifungal activity of calcium propionate against Trichothecium roseum.

Trichothecium roseum is an important postharvest pathogenic fungus, which can cause postharvest disease of diverse fruit. In addition to causing decay and quality deterioration, T. roseum infection can also metabolize trichothecenes that pose potential health risks to consumers. Calcium propionate (CP) is widely recognized as a food-grade antifungal agent. However, limited information is available regarding its antifungal mechanism against T. roseum and its potential application in controlling postharvest diseases of fruit. In this study, the antifungal efficacy of CP. against T. roseum was sy…

Synergistic effects of fumonisin B1 and ochratoxin A: are in vitro cytotoxicity data predictive of in vivo acute toxicity?

Contamination of food and feeds by mycotoxins is a major problem of human and animals health concern which is also extremely detrimental to economy. Mycotoxins producing moulds may produce a diversity of toxins such as aflatoxins, ochratoxins, trichothecenes, zearalenone, fumonisins, tremorgenic toxins and ergot alkaloids. Although toxicological, environmental and epidemiological studies have addressed the problem of these toxins one by one, more than one mycotoxin are found usually in the same contaminated commodities. That rises the incommensurable problem of multi-toxicosis in which the res…

Ultra-sensitive deoxynivalenol detection via SPR-enhanced ellipsometry on an electrochemically deposited nanofilm.

A highly sensitive SPRe-TIRE (Surface Plasmon Resonance-enhanced Total Internal Reflection Ellipsometry) aptasensor was developed for label-free detection of deoxynivalenol using the DN-2 aptamer as the specific recognition element. A mercaptophenyl monolayer was electrochemically grafted onto an SF-10/Au substrate via diazonium reduction, and characterized by ellipsometry and X-ray photoelectron spectroscopy. Gold nanoparticles were then self-assembled to enhance plasmonic coupling, followed by thiol-modified DN-2 aptamer immobilization. The aptasensor exhibited a wide semi-log linear range f

A Dual-Focus Workflow for Simultaneously Engineering High Thermostability of Aldo-Keto Reductase for the Degradation of 3-Keto-Deoxynivalenol.

Aldo-keto reductase AKR13B3 catalyzes the reduction of 3-keto-deoxynivalenol (3-keto-DON) to nontoxic 3-epi-DON, thereby alleviating mycotoxin contamination in food and feed. However, the previously reported AKR13B3 mutant AKR-V3 exhibited high catalytic activity but poor thermostability, restricting its industrial application. Here, guided by molecular dynamics flexibility analysis and consensus sequence design, we engineered the variant M5 with markedly improved thermostability. M5 showed a 38.3-fold longer half-life at 60 °C and retained slightly enhanced catalytic efficiency. Quantum mecha

The Influence of Non-Starch Polysaccharide Enzymes in Diets Contaminated with Deoxynivalenol and Its Modified Form, Deoxynivalenol-3-Glucoside, on Growth Performance and Toxicokinetics in Broiler Chickens.

The interaction between modified forms of deoxynivalenol (DON), such as DON-3-glucoside (DON-3G), and feed enzymes is underestimated. We assessed the interaction between DON and non-starch polysaccharide enzymes (NSPases) on growth performance, and excreta levels of DON and its metabolites (Experiment 1), as well as DON toxicokinetics (Experiment 2). Broiler chickens were fed maize-based diets naturally contaminated with low (LD; 516-792 μg DON/kg diet) or moderate (MD; 2395-3020 μg DON/kg diet) levels of DON. The LD and MD diets were supplemented with or without an NSPase blend. The MD diet i

Dietary β-sitosterol alleviates reproductive toxicity induced by combined exposure of low-dose deoxynivalenol and high-fat diet in male C57BL/6J mice.

Deoxynivalenol (DON), a mycotoxin commonly found in grains, is difficult to remove during processing, posing a risk of long-term low-dose exposure in both humans and animals. Meanwhile, there is a trend toward the increasing prevalence of high-fat diets (HFD) in contemporary human populations. Therefore, combined exposure to DON and HFD will exacerbate their health threats to humans and animals, especially potentially impairing male reproductive health and reducing fertility. This study aims to investigate the potential male reproductive toxicity induced by combined exposure to long-term low-d

Solvent Regulation Enhanced Aggregation-Induced Electrochemiluminescence of Piperazine-Functionalized Carbon Dot Aggregates for Ultrasensitive Detection of Deoxynivalenol.

The environmental contamination caused by deoxynivalenol (DON) has become an emerging public health issue, and its sensitive and accurate analysis is crucial as its trace amounts and microchange in environmental monitoring and management. Here, piperazine-functionalized carbon dot aggregates (P-CDAs) with obvious aggregation-induced electrochemiluminescence (AIECL) properties strengthened by solvent regulation were synthesized, based on which an ECL aptasensor was constructed for the efficient detection of DON. In addition to the surface states optimizing originated from piperazine modificatio
