The application of nanoparticles in cancer immunotherapy: Targeting tumor microenvironment.

The tumor development and metastasis are closely related to the structure and function of the tumor microenvironment (TME). Recently, TME modulation strategies have attracted much attention in cancer immunotherapy. Despite the preliminary success of immunotherapeutic agents, their therapeutic effects have been restricted by the limited retention time of drugs in TME. Compared with traditional delivery systems, nanoparticles with unique physical properties and elaborate design can efficiently penetrate TME and specifically deliver to the major components in TME. In this review, we briefly intro…

Stage-related PD-L1 expression in Kaposi sarcoma tumor microenvironment.

BACKGROUND: The immune checkpoint molecule PD-L1 represents an important target in oncological immune therapy. The aim of our study was to evaluate PD-L1 expression and the composition of the tumor microenvironment (TME) in Kaposi sarcoma. METHODS: Immunohistochemical stains were performed for PD-L1, CD3, CD33, CD68, and CD168 in 24 Kaposi sarcoma samples. In PD-L1-positive cases, the double stains for PD-L1, CD31, podoplanin, and HHV8 were added. RESULTS: PD-L1 was observed in 71% of the samples and was predominantly located in the TME. PD-L1 expression was significantly higher in nodular s…

Lipids in the tumor microenvironment: From cancer progression to treatment.

Over the past decade, the study of metabolic abnormalities in cancer cells has risen dramatically. Cancer cells can thrive in challenging environments, be it the hypoxic and nutrient-deplete tumor microenvironment or a distant tissue following metastasis. The ways in which cancer cells utilize lipids are often influenced by the complex interactions within the tumor microenvironment and adjacent stroma. Adipocytes can be activated by cancer cells to lipolyze their triglyceride stores, delivering secreted fatty acids to cancer cells for uptake through numerous fatty acid transporters. Cancer-ass…

The tumor microenvironment shapes hallmarks of mature B-cell malignancies.

B-cell tumorigenesis results from a host of known and unknown genetic anomalies, including non-random translocations of genes that normally function as determinants of cell proliferation or cell survival to regions juxtaposed to active immunoglobulin heavy chain enhancer elements, chromosomal aneuploidy, somatic mutations that further affect oncogenic signaling and loss of heterozygosity of tumor-suppressor genes. However, it is critical to recognize that even in the setting of a genetic disease, the B-cell/plasma cell tumor microenvironment (TME) contributes significantly to malignant transfo…

Metastasis: cell-autonomous mechanisms versus contributions by the tumor microenvironment.

The fatality of cancer predominantly results from the dissemination of primary tumor cells to distant sites and the subsequent formation of metastases. During tumor progression, some of the primary tumor cells as well as the tumor microenvironment undergo characteristic molecular changes, which are essential for the metastatic dissemination of tumor cells. In this review, we will discuss recent insights into pro-metastatic events occurring in tumor cells themselves and in the tumor stroma. Tumor cell-intrinsic alterations include the loss of cell polarity and alterations in cell-cell and cell-…

Oncolytic reovirus: tumor-microenvironment remodeling, engineering constraints, and translational strategies.

Oncolytic reovirus (ReoV) has emerged as a promising cancer immunotherapy platform because of its natural tumor tropism and immunostimulator … ralizing antibodies, heterogeneous receptor expression, stromal barriers, and the immunosuppressive tumor microenvironment. Current strategies therefore combine reovirus with chemotherapy, radiotherapy, immune checkpoint blockade, and targeted agents, although treatment outcomes remain dependent on tumor type, dose, and scheduling. Unlike previous reviews focused primar …

3D nanoscaffolds: Bridging the gap between tumor microenvironment and therapy.

The tumor microenvironment (TME) comprises both cellular components-including cancer cells and non-malignant immune and stromal cells-and non-cellular components, such as the extracellular matrix. Through intricate signaling interactions, these elements collectively regulate tumor initiation, progression, metastasis, and resistance to anticancer therapies. Consequently, targeting and modulating the TME offers novel and valuable insights into the complex mechanisms driving tumor heterogeneity and disease progression. Given recent advances in nanomedicine, a variety of sophisticated 3D scaffolds…

Enzyme-based nanomedicine for tumor microenvironment modulation in cancer therapy.

The tumor microenvironment (TME) is a major driver of therapeutic resistance, shaped by abnormal vasculature, metabolic rewiring, redox imbalance, dense extracellular matrix deposition, and immunosuppressive signaling. These biochemical and biophysical barriers restrict drug delivery, promote tumor progression, and limit the efficacy of cancer therapy. Enzyme-based nanomedicine offers a catalytic strategy to modulate TME features through localized enzymatic reactions. This Review discusses two major platforms: enzyme-loaded nanoparticles, which protect and deliver natural enzymes, and enzyme-m…

Tumor microenvironment shapes the spatial organization of glioblastoma cell states.

Glioblastoma is characterized by heterogeneous and plastic cellular populations that adopt transcriptional programs shaped by genetic alterations and microenvironmental cues. Recent studies have identified at least 4 partially inconvertible cell states-astrocytic-like, neural progenitor-like, oligodendrocyte progenitor-like, and mesenchymal-like-that represent aberrant developmental programs. Expanded analysis further reveals hybrid and intermediate states that form continuous transcriptional and metabolic gradients. These states exhibit spatial organization, assembling into 3 distinct microan

Distinct tumor microenvironment profiles characterize regressive versus progressive anal precancers.

Oncogenesis of anal high-grade squamous intraepithelial lesions (HSIL) is highly variable. Therefore, all HSIL are ablated, leading to overtreatment and associated burden because not all lesions will progress to cancer. This exploratory study investigated differences in the tumor immune microenvironment between regressive and progressive HSIL in people living with HIV to enable a more tailored approach. Multiplex imaging mass cytometry showed more inflammation in HSIL that progressed to cancer and cancer samples, compared with HSIL that spontaneously regressed and controls. Densities of HLA-DR

Solute carrier transporters as metabolic gatekeepers in tumor-microenvironment crosstalk.

The tumor microenvironment (TME) is a metabolically dynamic niche characterized by active metabolic crosstalk between tumor cells and various stromal and immune cell subsets, which critically influences tumor progression and therapeutic outcomes. Within this ecosystem, solute carrier (SLC) transporters are gatekeepers that mediate the transport of a wide range of metabolites, including nutrients, ions, and signaling molecules, across cellular membranes. SLC transporters have emerged as active regulators of TME composition and intercellular communication, representing promising yet underexplore…

Immunomodulation of the Prostate Tumor Microenvironment Following Inorganic Arsenic Exposure.

The tumor microenvironment (TME) influences prostate cancer (PCa) progression through stromal and immune interactions. Adipose-derived mesenchymal stromal cells (ASCs) modulate immune tone, while inorganic arsenic (iAs), a widespread toxicant, is linked to immune suppression and carcinogenesis. Their combined impact on PCa immunity has remained unclear. Using a Ras-driven murine PCa model (TC2Ras, which mimics aggressive, immune-interactive PCa through constitutive Ras signaling), we assessed ASC and chronic iAs exposure effects on tumor growth, immune infiltration, and transcriptomic remodeli…

Atg5/Autophagy inactivation in mouse bone microenvironment promotes tumor development.

Bone is an attractive site for cancer colonization, both for primary tumors such as osteosarcoma and for metastases of various malignancies. Preventing bone metastasis, which is associated with a poor prognosis, is a major challenge and identifying the factors involved in skeletal tumoral development is crucial to improve survival. In the present work, we showed that inactivation of the macroautophagy/autophagy-essential gene Atg5 in osteoblasts, the cells in charge of bone formation, stimulates osteosarcoma and breastbone metastasis growth as well as metastatic dissemination. We determined th

Mathematical modeling of T cell exhaustion in the tumor microenvironment.

T cell exhaustion (TCE), a hallmark of chronic infections and cancer, is characterized by progressive loss of effector function, sustained expression of inhibitory receptors, and stable transcriptional/epigenetic reprogramming. Within the tumor microenvironment (TME), exhausted CD8+ T cells fail to eliminate malignant cells, contributing to immune evasion and resistance to immunotherapy. Although checkpoint blockade has provided clinical benefit, outcomes remain variable, underscoring the need to better understand the temporal and mechanistic basis of exhaustion. Current modeling efforts have …

Quiescent tumor cells shape the immunosuppressive microenvironment in pancreatic cancer.

Immunotherapy, including chimeric antigen receptor (CAR) T-cell therapy, has limited activity in pancreatic ductal adenocarcinoma (PDAC). Using orthotopic PDAC mouse models, we identified a rare population of quiescent PDAC cells that increases after CAR-T cell therapy and exhibits relatively higher clonogenic growth and self-renewal potential than bulk tumor cells. These quiescent cells express high levels of Epiregulin (EREG), a secreted ligand for EGFR and ErbB4, and induce an immunosuppressive tumor microenvironment by increasing the frequency of ErbB4-expressing tumor-associated macrophag…
