The global introduction of inactivated polio vaccine can circumvent the oral polio vaccine paradox.

This literature review identifies the factors that influence the decision to introduce inactivated polio vaccine (IPV) in developing countries as opposed to the policy of vaccine cessation. Attenuated viruses in the oral polio vaccine (OPV) can replicate, revert to neurovirulence and become transmissible circulating vaccine-derived polioviruses (cVDPVs), preventing use of the vaccine in the post-eradication era. This literature review identifies (1) risks of complete cessation of vaccination, (2) barriers and (3) solutions for the introduction of IPV in developing countries. The reviewed liter

Randomized comparison of early and late vaccination with inactivated poliovirus vaccine after allogeneic BMT.

Forty-five adult HLA-matched sibling BMT recipients were randomized to receive inactivated poliovirus vaccine (IPV) at 6, 8 and 14 months (early group, n = 23) or at 18, 20 and 26 months after BMT (late group, n = 22). Ninety-five percent of the early group patients had protective antibody titres of > or = 4 to poliovirus type 1 (PV1), poliovirus type 2 (PV2) and poliovirus type 3 (PV3) by a microneutralization assay prior to the first vaccination, at 6 months after BMT. The corresponding proportion for the late group patients was only 67% at 18 months. The antibody responses 1 month after eac

Current issues in evaluating the efficacy of oral poliovirus vaccine and inactivated poliovirus vaccine immunization.

Although epidemic poliomyelitis in the United States has been eliminated, there are a number of current issues that concern the use of oral attenuated poliovirus vaccine in comparison with the new enhanced-potency inactivated vaccine. Although wild-type poliovirus is almost eradicated in developed nations, vaccine-induced polio exists, at a low but persistent rate. This article reviews the current issues and suggests a return to the Institute of Medicine's 1988 recommendations concerning combined immunization with inactivated poliovirus vaccine and oral poliovirus vaccine.

The Effectiveness of Four Quadrivalent, Inactivated Influenza Vaccines Administered Alone or in Combination with Pneumococcal and/or SARS-CoV-2 Vaccines: A Population-Wide Cohort Study.

Background: Several influenza vaccine formulations are available, including adjuvanted, high-dose, trivalent, and quadrivalent vaccines, and direct, comparative evidence on the relative effectiveness is limited. Real-life data on the potential impact of the co-administration of pneumococcal and/or SARS-CoV-2 vaccinations are also very scarce. During the 2023-2024 influenza season, we carried out a retrospective cohort study on the entire elderly population of the Pescara province, Italy, in order to evaluate the effectiveness of the quadrivalent influenza vaccine, offered alone or in combinati

Immunological response in horses following West Nile virus vaccination with inactivated or recombinant vaccine.

To evaluate the immunological response following vaccination, 40 WNV serologically negative horses were selected and divided in two groups of 20 animals. One group was vaccinated (booster after 28 days) with a whole inactivated viral strain and the second group with a live recombinant canarypox virus expressing the genes coding for the WNV preM/E viral proteins. IgM, IgG and neutralizing antibodies were monitored by class specific ELISAs and serum neutralization assay for 360 days. In both groups, IgM antibodies were first detected 7 days post vaccination (dpv). However, in the group vaccinate

Lymphocyte blast transformation and antibody responses after vaccination with inactivated mumps virus vaccine.

Mumps antibody response and the development of cell-mediated immunity (CMI) to mumps antigen as assayed by the lymphocyte blast transformation method were studied after immunization with an inactivated mumps vaccine in seven sero-negative and ten sero-positive subjects. The transformation response of peripheral blood lymphocytes to mumps antigen generally appeared by 20 weeks post vaccination and reached levels found in sero-positive subjects years after mumps infection. There was no clear-cut correlation between the magnitude of blast transformation and antibody responses.

Rift Valley fever vaccine--antibody and immune response in cattle to a live and an inactivated vaccine.

In a study of the response in cattle to a live and an inactivated Rift Valley fever (RVF) vaccine prepared from cell cultures infected with RVF virus, the effects of varying doses and combinations of these vaccines were compared. The antibody response to a primary injection of either vaccines was poor when measured by the serum virus neutralization test (SVN) and the haemagglutination-inhibition test (HI) but a booster dose of inactivated vaccine evoked a good anamnestic response in cattle previously injected with either of these vaccines. Cattle vaccinated with the live vaccine but negative t

Identification of metabolic signatures of immune response following mRNA and inactivated vaccines against COVID-19: a systematic review.

BACKGROUND: Metabolomic profiling offers insights into immune responses, yet a synthesis of systemic metabolic changes after COVID-19 vaccination is lacking. This review aims to characterize vaccination-induced metabolomic alterations and identify correlative biomarkers of responsiveness. METHODS: Following PRISMA 2020 guidelines (PROSPERO 1181037), four databases (PubMed, Embase, Scopus, Web of Science) were searched for studies using LC-MS, GC-MS, or NMR to analyse venous blood after COVID-19 vaccination. Inclusion criteria focused on original human studies. Risk of bias was assessed using

Dynamics of Circulating Follicular Helper and Regulatory T-Cell Memory Induced by mRNA, Inactivated, and Live-Attenuated Vaccines.

OX40 signaling promotes the differentiation of memory CD4+ T cells into T follicular helper (Tfh) cells that support germinal center (GC) B-cell responses. Using an extended activation-induced marker (AIM) assay based on OX40 (CD134) and CD25 coexpression, we quantified antigen-specific circulating memory Tfh and T follicular regulatory (Tfr) cells following in vitro stimulation with influenza, SARS-CoV-2, or measles antigens. Responses were assessed longitudinally in recipients of inactivated influenza (n = 20) and mRNA COVID-19 (n = 13) vaccines and cross-sectionally in individuals vaccinate

Orally Dissolving Film-Based Influenza Vaccines Confer Superior Protection Compared to the Oral Administration of Inactivated Influenza Virus.

Background: Self-administered orally dissolving films (ODFs) encapsulating inactivated influenza vaccines represent an effective strategy for stimulating mucosal immunity. While this vaccination method offers several advantages over conventional influenza vaccines, a comparative efficacy study remains lacking. Methods: Female BALB/c mice were immunized with inactivated A/PR/8/34 (H1N1) either via orogastric inoculation or through the oral mucosal delivery using pullulan and trehalose-based ODF vaccines. Each group received equivalent antigen doses across three immunizations. Humoral responses

Effectiveness of Enterovirus 71 inactivated vaccines against hand, foot, and mouth disease: A test-negative case-control study.

The Enterovirus A71 (EV-A71) vaccine was introduced in China in December 2015 as a preventive measure against hand, foot, and mouth disease (HFMD) caused by EV-A71. However, the effectiveness of the vaccine (VE) in real-world settings needs to be evaluated. We conducted a test-negative case-control study to assess the effectiveness of EV-A71 vaccines in preventing EV-A71-associated HFMD. Children aged 6-71 months with HFMD were enrolled as participants. The case group comprised those who tested positive for EV-A71, while the control group comprised those who tested negative for EV-A71. To esti

Hesitancy, reactogenicity and immunogenicity of the mRNA and whole-virus inactivated Covid-19 vaccines in pediatric neuromuscular diseases.

The mRNA-based BNT162b2 and inactivated whole-virus CoronaVac are two widely used COVID-19 vaccines that confer immune protection to healthy individuals. However, hesitancy toward COVID-19 vaccination appeared to be common for patients with neuromuscular diseases (NMDs) due to the paucity of data on the safety and efficacy in this high-risk patient population. Therefore, we examined the underlying factors associated with vaccine hesitancy across time for NMDs and assessed the reactogenicity and immunogenicity of these two vaccines. Patients aged 8-18 years with no cognitive delay were invited
