Identification and neutralization of biological activities in the venoms of Loxosceles spiders.

The biological activities of the venom of three species of spiders of the genus Loxosceles were studied (L. gaucho, L. laeta and L. intermedia). The dermonecrotic and lethal activities are shared by all three Loxosceles venoms. Only low levels of proteolytic, myotoxic and phospholipase A2 activities were demonstrable even when a large amount of venom was used. No direct hemolytic activity was detected. L. intermedia venom was the most lethal (LD50 0.48 mg/kg), the L. laeta venom was the least lethal (LD50 1.45 mg/kg) whereas L. gaucho venom showed an intermediate value (LD50 0.74 mg/kg). The a…

Muscle extract of hedgehog, Erinaceus europaeus, inhibits hemorrhagic activity of snake venoms.

The antihemorrhagic activity of muscle extract of hedgehog, Erinaceus europaeus, was tested on various snake venoms with hemorrhagic activity. The extract inhibited strongly hemorrhagic activity of venoms from Bitis arietans, Bothrops jararaca and Vipera latastei gaditana, and remarkably that of venoms from Agkistrodon halys blomhoffi, Bitis gabonica rhinoceros, Bitis nasicornis, Bothrops atrox asper, Crotalus horridus horridus and Vipera berus. The antihemorrhagic activity against eight other snake venoms was below the detection level.

A comparative study of the biological properties of some sea snake venoms.

1. The protease, phosphodiesterase, alkaline phosphomonoesterase, L-amino acid oxidase, acetylcholinesterase, phospholipase A, 5'-nucleotidase, hyaluronidase, arginine ester hydrolase, procoagulant, anticoagulant and hemorrhagic activities of ten samples of venoms from seven taxa of sea snakes were examined. 2. The results show that venoms of sea snakes of both subfamilies of Hydrophiinae and Laticaudinae are characterized by a very low level of enzymatic activities, except phospholipase A activity and, for some species, hyaluronidase activity. 3. Because of the low levels of enzymatic activit…

Venomic and F(ab')₂ antivenomic studies of Bothrops alternatus venom proteins.

In this study, using a proteomic approach, we performed the venomic characterization of B. alternatus venom proteins. The venom comprises toxins from eleven protein families and the main identified included PIII-SVMP (27.32%), PII-SVMP (12.33%), Disintegrin-SVMP (10.26%), SVSP (8.80%), PLA2 (6.85%), BBP/C-NP (4.85%), CRVP (4.87%), CTL (6.80%), LAAO (4.14%), SVGF (1.90%), VPA (1.64%), PDE (0.07%), and an unidentified percentage (11.64%). Antivenomic analysis showed that both F(ab')₂ antivenoms recognized all venom proteins but had low affinity for small peptides like BPP. The BIOL antivenom dem…

Beyond venomous fangs: Uloboridae spiders have lost their venom but not their toxicity.

BACKGROUND: Venom, one of nature's most potent secretions, has played a crucial role in the evolutionary success of many animal groups, including spiders. However, Uloboridae spiders appear to lack venom and capture their prey, unlike venomous spiders, by extensive silk-wrapping and regurgitation of digestive fluids onto the entire prey package. A prevailing hypothesis posits that toxins may have been reallocated from the venom to alternative secretions, like silk or digestive fluids. Yet, whether uloborids have retained venom toxins and the mechanisms underlying prey immobilisation remain unr

Preliminary Biochemical and Venomic Characterization of the Venom of Phalotris lemniscatus (Serpentes, Colubridae).

BACKGROUND: For many decades, research on snake venom toxinology focused mainly on the venoms of Viperidae and Elapidae species, which were traditionally the only ones considered as venomous. However, much less interest has been given to the venom produced by opisthoglyphous colubrid snakes, since they were typically considered of no clinical relevance. OBJECTIVE: The aim of this work is to perform a preliminary biochemical and venomic characterization of the venom of the colubrid snake Phalotris lemniscatus, a species that has been responsible for two relevant cases of envenomation in Urugua…

Venom gland transcriptomics for identifying, cataloging, and characterizing venom proteins in snakes.

Snake venoms are cocktails of protein toxins that play important roles in capture and digestion of prey. Significant qualitative and quantitative variation in snake venom composition has been observed among and within species. Understanding these variations in protein components is instrumental in interpreting clinical symptoms during human envenomation and in searching for novel venom proteins with potential therapeutic applications. In the last decade, transcriptomic analyses of venom glands have helped in understanding the composition of various snake venoms in great detail. Here we review …

Combined snake venomics and venom gland transcriptomic analysis of Bothropoides pauloensis.

Unraveling the repertoire of venom toxins of Bothropoides pauloensis was assessed by snake venomics and venom gland transcriptomic surveys. Both approaches yielded converging overall figures, pointing to metalloproteinases (~37%), PLA(2)s (26-32%), and vasoactive (bradykinin-potentiating) peptides (12-17%) as the major toxin classes. The high occurrence of SVMPs, PLA(2) molecules, vasoactive peptides, along with serine proteinases, explains the local and systemic effects observed in envenomations by B. pauloensis. Minor (<3%) C-type lectin, serine proteinase, L-amino acid oxidase, nerve growth

Functional structure of Agelena labyrinthica's (Araneae:Agelenidae) venom gland and electrophoresis of venom.

The funnel-web spider, Agelena labyrinthica, is widely distributed throughout Turkey. The objective of the present study was to describe the histological and functional fine structure of A. labyrinthica's venom gland by using light microscope, scanning (SEM) and transmission electron microscope (TEM). We have also preliminarily analyzed venom components by SDS-PAGE. Each venom gland has surrounded by a thin adventitia and gross striated muscular bundles. Basal lamina underlies between muscular bundles and the inner glandular epithelium, and ties up them each other. The striated muscular bundle

[Hymenoptera venom allergy].

This review finds that diagnostics should only be performed in patients who might be candidates for venom immunotherapy (VIT). VIT is recommended for children and adults with documented sensitisation and systemic anaphylactic reactions (respiratory and/or circulatory symptoms) or reduced quality of life due to less severe reactions. VIT is not recommended for children with monosymptomatic urticaria. VIT should be continued for five years. Patients should carry an adrenaline autoinjector during insect season until a maintenance dose is reached.

Cobra venom keratoconjunctivitis.

We present a case of cobra venom keratoconjunctivitis, an ocular injury caused by a snake's defensive mechanism of spitting venom into a predator's eyes. A male in his 50s reported to the outpatient department of a regional hospital in Songea, Tanzania, with 24 hours of eye pain, photophobia and decreased vision after being sprayed in the face by a cobra. The affected eye was anaesthetised with topical tetracaine hydrochloride 0.5% and irrigated with 1 L of lactated Ringer's solution. Tetracycline hydrochloride 1% ophthalmic ointment was placed in the inferior sulcus, and the lid was taped shu

Addiction to Snake Venom.

The nature of addiction depends on various factors. The tendency to have already used several addictive substances and to seek high sensation experiences as a result of specific personality traits may lead to extreme and peculiar forms of addictions. Even belonging to specific social and cultural background may lead to such forms of addiction such as intentional snake bite and willful envenomation. In this article, we have discussed the peculiarities and practical insight of such addiction to snake venom. The possible molecular mechanism behind such venom-mediated reinforcement has also been h

Snake venom metalloproteinases.

Recent proteomic analyses of snake venoms show that metalloproteinases represent major components in most of the Crotalid and Viperid venoms. In this chapter we discuss the multiple activities of the SVMPs. In addition to hemorrhagic activity, members of the SVMP family also have fibrin(ogen)olytic activity, act as prothrombin activators, activate blood coagulation factor X, possess apoptotic activity, inhibit platelet aggregation, are pro-inflammatory and inactivate blood serine proteinase inhibitors. Clearly the SVMPs have multiple functions in addition to their well-known hemorrhagic activi…
