A fadD mutant of Vibrio cholerae is impaired in the production of virulence factors and membrane localization of the virulence regulatory protein TcpP.

In the enteric pathogen Vibrio cholerae, expression of the major virulence factors is controlled by the hierarchical expression of several regulatory proteins comprising the ToxR regulon. In this study, we demonstrate that disruption of the fadD gene encoding a long-chain fatty acyl coenzyme A ligase has marked effects on expression of the ToxR virulence regulon, motility, and in vivo lethality of V. cholerae. In the V. cholerae fadD mutant, expression of the major virulence genes ctxAB and tcpA, encoding cholera toxin (CT), and the major subunit of the toxin-coregulated pilus (TCP) was drasti

Binding of laminin to Paracoccidioides brasiliensis induces a less severe pulmonary paracoccidioidomycosis caused by virulent and low-virulence isolates.

The pathogenic fungus Paracoccidioides brasiliensis is the causative agent of paracoccidioidomycosis (PCM). This pulmonary mycosis, acquired by inhalation of airborne propagules, may disseminate to several internal organs and tissues, leading to severe disease. Adhesion to host cell components is the first step involved in dissemination of pathogens. Previous studies showed that laminin, the most abundant glycoprotein of the basement membrane, binds to P. brasiliensis yeast cells, enhancing their pathogenicity in the hamster testicle model. As PCM is primarily a pulmonary infection, we studied

Virulence, genomic features, and antimicrobial resistance of a highly virulent Streptococcus iniae ST4 from diseased yellow catfish.

Streptococcosis is an emerging bacterial disease in freshwater aquaculture that poses a substantial threat to animal health and production sustainability. In this study, the etiological agent responsible for a "whirling disease" outbreak in yellow catfish (Pelteobagrus fulvidraco) was isolated and comprehensively characterized. A dominant bacterial strain, designated as HSY24518-1S, was identified as Streptococcus iniae, physiological, and biochemical characteristics, as well as lctO and 16S rRNA gene sequence analyses. Multilocus sequence typing (MLST) assigned the isolate to sequence type 4

Genome sequence of Brucella abortus vaccine strain S19 compared to virulent strains yields candidate virulence genes.

The Brucella abortus strain S19, a spontaneously attenuated strain, has been used as a vaccine strain in vaccination of cattle against brucellosis for six decades. Despite many studies, the physiological and molecular mechanisms causing the attenuation are not known. We have applied pyrosequencing technology together with conventional sequencing to rapidly and comprehensively determine the complete genome sequence of the attenuated Brucella abortus vaccine strain S19. The main goal of this study is to identify candidate virulence genes by systematic comparative analysis of the attenuated strai

Tissue tropism in the chicken embryo of non-virulent and virulent Newcastle diseases strains that express green fluorescence protein.

The tissue tropism of non-virulent and virulent Newcastle disease virus (NDV) was investigated using 8-day-old and 14-day-old embryonating chicken eggs (ECE), inoculated with an infectious clone of the non-virulent La Sota strain (NDFL-GFP) or its virulent derivative (NDFLtag-GFP). Both strains expressed the gene encoding jellyfish green fluorescence protein (GFP) as a marker. The GFP was readily expressed in chicken embryo cells infected with the NDV strains indicating virus replication. Whereas both strains replicated in the chorioallantoic membrane (CAM) and infected the skin of 8-day-old E

Detection of bacterial virulence genes by subtractive hybridization: identification of capsular polysaccharide of Burkholderia pseudomallei as a major virulence determinant.

Burkholderia pseudomallei, the etiologic agent of melioidosis, is responsible for a broad spectrum of illnesses in humans and animals particularly in Southeast Asia and northern Australia, where it is endemic. Burkholderia thailandensis is a nonpathogenic environmental organism closely related to B. pseudomallei. Subtractive hybridization was carried out between these two species to identify genes encoding virulence determinants in B. pseudomallei. Screening of the subtraction library revealed A-T-rich DNA sequences unique to B. pseudomallei, suggesting they may have been acquired by horizonta

Role of cell-associated and secretion-independent virulence factors of Pseudomonas aeruginosa in microbial keratitis and anti-virulence therapeutic strategies.

PURPOSE: Microbial keratitis (MK) caused by Pseudomonas aeruginosa is a rapidly progressive and vision-threatening corneal infection worldwide. Even short delays in diagnosis or inappropriate therapy can lead to irreversible corneal scarring and blindness. This opportunistic pathogen colonizes, invades, and damages ocular tissue by taking advantage of disruptions in the corneal epithelial barrier and using a variety of virulence factors. The aim of this study was to summarize the current understanding of P. aeruginosa’s cell-associated, and secretion system-independent virulence factors involv

Acquisition of a large virulence plasmid (pINV) promoted temperature-dependent virulence and global dispersal of O96:H19 enteroinvasive Escherichia coli.

Enteroinvasive Escherichia coli (EIEC) and Shigella are closely related agents of bacillary dysentery. It is widely viewed that EIEC and Shigella species evolved from E. coli via independent acquisitions of a large virulence plasmid (pINV) encoding a type 3 secretion system (T3SS). Sequence Type (ST)99 O96:H19 E. coli is a novel clone of EIEC responsible for recent outbreaks in Europe and South America. Here, we use 92 whole genome sequences to reconstruct a dated phylogeny of ST99 E. coli, revealing distinct phylogenomic clusters of pINV-positive and -negative isolates. To study the impact of

Hemolytic mechanism and virulence of Antarctic psychrophilic Pseudomonas.

Polar warming is expected to release and redistribute psychrophilic microorganisms, thereby increasing their likelihood of encountering warmer environments and potential hosts. However, it remains uncertain whether psychrophilic polar bacteria maintain pathogenic properties relevant to mammalian hosts during such transitions. Here, we investigated two hemolytic Antarctic Pseudomonas isolates, 3S6 and NS21, assigned to P. antarctica and P. yamanorum, respectively. Both isolates exhibited hemolytic activity, biofilm formation, and siderophore production, although these phenotypes were weaker tha

Carbon and nitrogen sources can fuel fungal virulence.

For all pathogenic microbes, metabolic processes are essential for growth, development, and survival during infection. However, host niches are dynamic environments where nutrient availability can vary, and host defense mechanisms often restrict access to host-derived nutrients. Therefore, microbes must adapt to these microenvironments by regulating nutrient acquisition and metabolism, and by employing mechanisms such as virulence factors that release host-derived nutrients. This metabolic plasticity is a key feature that enables certain microbes to thrive during host-microbe interactions. Hen

Phosphoenolpyruvate Carboxykinase Controls Edwardsiella tarda Virulence via Oxaloacetate.

Understanding the metabolic basis of bacterial virulence is essential for developing anti-infective strategies. Phosphoenolpyruvate carboxykinase (PEPCK), encoded by pckA, is a key enzyme at the metabolic junction between the pyruvate cycle and the TCA cycle, but its role in virulence regulation remains largely unexplored. In this study, we generated a pckA deletion mutant, ΔpckA, in Edwardsiella tarda and investigated its impact on virulence, metabolism, and vaccine potential. The ΔpckA mutant exhibited increased antibiotic sensitivity, faster growth, reduced autoaggregation, and significantl

Buprenorphine enhances Streptococcus mutans virulence and biofilm formation.

BACKGROUND: Buprenorphine is widely prescribed for opioid use disorder (OUD). In 2022, the U.S. FDA issued a safety warning on dental diseases associated with buprenorphine. While reports implicate increased caries risk, the microbial mechanisms remain unclear. OBJECTIVE: To evaluate whether buprenorphine directly modulates Streptococcus mutans (S. mutans) virulence traits relevant to cariogenesis. DESIGN: S. mutans UA159 were exposed to buprenorphine. Planktonic growth, acidogenicity, acid tolerance, aggregation, and carbohydrate utilization were assessed. Biofilm biomass and extracellular

Prophages in Skin Pathogens: From Virulence to Therapy.

Prophages are bacteriophage genomes that are part of bacterial chromosomes. They are not just dormant passengers; they actively shape pathogen biology. For example, in skin-infecting pathogens such as Staphylococcus aureus, Streptococcus pyogenes, and Pseudomonas aeruginosa, prophages carry important virulence factors, cytotoxins, superantigens, immune evasion clusters, and epigenetic regulators that directly affect the course of skin and soft tissue infections. This same prophage biology provides a therapeutic strategy: prophage-derived molecules, including endolysins, holins, spanins, and po

Colonic mucus attenuates Bacteroides fragilis virulence in vitro.

OBJECTIVES: Enterotoxigenic Bacteroides fragilis (ETBF) is a subspecies of B. fragilis that is considered to contribute to colorectal carcinogenesis through the activity of the B. fragilis toxin (BFT). However, colonic carriage of ETBF is also evident in a subset of asymptomatic individuals. In vitro studies using the HT29 cell line have linked BFT to the disruption of cell-cell adhesion and inflammation. This cell line, however, fails to consider the role of the mucus barrier in protecting the colonic epithelium in healthy individuals. This study aimed to investigate the role of a cell-associ

NO: from plant immunity to fungal virulence factor.

Traditionally viewed as a plant defense molecule, nitric oxide (NO) has now been shown to play a key role in fungal pathogenesis. A recent study by Zhang et al. reveals that banana pathogenic isolates of Fusarium oxysporum coordinate NO production with host defense responses through an accessory mitochondrial pathway within its genome.

Trichophyton rubrum Phenotypic Virulence Factors in Mexican Strains.

(1) Background: T. rubrum is the most important agent in tinea pedis, tinea manuum, tinea cruris, tinea corporis, and even in subcutaneous dermatophytosis. T. rubrum must overcome several obstacles to adhere, grow, and invade the host, for which their virulence factors are important. Previous studies have demonstrated the capability of T. rubrum strains to produce proteases, phospholipases, hemolysins, and elastases. The aim of this work was the genotypic identification of clinical isolates of T. rubrum to subsequently determine production of the main phenotypic virulence factors associated wi

Broadly inhibitory antibodies against severe malaria virulence proteins.

Plasmodium falciparum pathology is driven by the accumulation of parasite-infected erythrocytes in microvessels. This process is mediated by the parasite's polymorphic erythrocyte membrane protein 1 (PfEMP1) adhesion proteins. A subset of PfEMP1 variants that bind human endothelial protein C receptor (EPCR) through their CIDRα1 domains is responsible for severe malaria pathogenesis. A longstanding question is whether individual antibodies can recognize the large repertoire of circulating PfEMP1 variants. Here, we describe two broadly reactive and binding-inhibitory human monoclonal antibodies
