A Vaccine Against Fibroblast Activation Protein Improves Murine Cardiac Fibrosis by Preventing the Accumulation of Myofibroblasts.

BACKGROUND: Myofibroblasts are primary cells involved in chronic response-induced cardiac fibrosis. Fibroblast activation protein (FAP) is a relatively specific marker of activated myofibroblasts and a potential target molecule. This study aimed to clarify whether a vaccine targeting FAP could eliminate myofibroblasts in chronic cardiac stress model mice and reduce cardiac fibrosis. METHODS: We coadministered a FAP peptide vaccine with a cytosine-phosphate-guanine (CpG) K3 oligonucleotide adjuvant to male C57/BL6J mice and confirmed an elevation in the anti-FAP antibody titer. After continuou

PLGA-based nanovaccine delivering chimeric IpaD-StxB protein induces robust immune response against Shigella infection.

BACKGROUND AND AIM: Shigellosis is a major cause of morbidity and mortality worldwide, especially among children under five in low- and middle-income countries. Despite its public health burden, there is no licensed vaccine. Recombinant and nanoparticle-based vaccine approaches offer promising alternatives to conventional methods. This study aimed to develop a chimeric nanovaccine based on the fusion of Shigella flexneri IpaD and StxB, encapsulated in PLGA nanoparticles, and to evaluate its immunogenic potential in a murine model. METHODS: The recombinant IpaD-StxB (IdSb) protein was expresse

A Frameshift Peptide Neoantigen-Based Vaccine for Mismatch Repair-Deficient Cancers: A Phase I/IIa Clinical Trial.

PURPOSE: DNA mismatch repair (MMR) deficiency is a hallmark of Lynch syndrome, the most common inherited cancer syndrome. MMR-deficient cancer cells accumulate numerous insertion/deletion mutations at microsatellites. Mutations of coding microsatellites (cMS) lead to the generation of immunogenic frameshift peptide (FSP) neoantigens. As the evolution of MMR-deficient cancers is triggered by mutations inactivating defined cMS-containing tumor suppressor genes, distinct FSP neoantigens are shared by most MMR-deficient cancers. To evaluate safety and immunogenicity of an FSP-based vaccine, we per

A yeast surface display platform for oral delivery of a SARS-CoV-2 receptor-binding domain dimer inducing mucosal and systemic immunity.

The development of scalable and stable antigen delivery systems remains a central challenge in vaccine biotechnology, particularly for oral immunization targeting mucosal immunity. Here, we report a yeast surface display (YSD)-based platform for the oral delivery of a SARS-CoV-2 receptor-binding domain (RBD) dimer. A head-to-tail tandem RBD was genetically fused to the Aga2p subunit and anchored to the cell wall of Saccharomyces cerevisiae via the Aga1p-Aga2p system, generating the YSD-RBD-dimer construct. Flow cytometry confirmed clear surface expression, with 51.1% dual-positive cells recogn

Mapping Immunological, Host Receptor Binding Determinants, and Cathepsin Cleavage Site of EBOV Glycoprotein Utilizing the Qubevirus Platform.

Ebola virus (EBOV) remains a highly infectious human pathogen that causes a severe and lethal disease known as Ebola virus disease (EVD), despite recent progress in vaccine development based on its only surface glycoprotein (GP). In this study, we modeled and inserted four overlapping fragments (F1-4) of the EBOV GP at the C-terminus of the A1 protein of Qubevirus (Qβ) and used the platform to investigate the tropism and immunological functions of the GP by displaying the peptides with 30 overlapping amino acids. The resulting recombinant phages were used to determine their reactivity with GP-

Parenteral adjuvant potential of recombinant B subunit of Escherichia coli heat-labile enterotoxin.

BACKGROUND: The B subunit of Escherichia coli heat-labile enterotoxin (LTB) is a potent mucosal immune adjuvant. However, there is little information about LTB's potential as a parenteral adjuvant. OBJECTIVES: We aimed at evaluating and better understanding rLTB's potential as a parenteral adjuvant using the fused R1 repeat of Mycoplasma hyopneumoniae P97 adhesin as an antigen to characterise the humoral immune response induced by this construct and comparing it to that generated when aluminium hydroxide is used as adjuvant instead. METHODS: BALB/c mice were immunised intraperitoneally with

Plasmodium sporozoite phospholipid scramblase interacts with mammalian carbamoyl-phosphate synthetase 1 to infect hepatocytes.

After inoculation by the bite of an infected mosquito, Plasmodium sporozoites enter the blood stream and infect the liver, where each infected cell produces thousands of merozoites. These in turn, infect red blood cells and cause malaria symptoms. To initiate a productive infection, sporozoites must exit the circulation by traversing the blood lining of the liver vessels after which they infect hepatocytes with unique specificity. We screened a phage display library for peptides that structurally mimic (mimotope) a sporozoite ligand for hepatocyte recognition. We identified HP1 (hepatocyte-bin

Efficacy, Effectiveness, and Immunogenicity of the Respiratory Syncytial Virus Prefusion F Vaccine Administered During Pregnancy: Protocol for a Living Systematic Review and Meta-Analysis.

BACKGROUND: Respiratory syncytial virus (RSV) is a leading cause of lower respiratory tract disease in infants, with the highest burden in the first 6 months of life. Maternal immunization with the bivalent (RSV-A and RSV-B) prefusion F (RSVPreF) protein subunit vaccine (ABRYSVO) is a recently recommended strategy for preventing RSV-related outcomes in infants. First licensed for use in pregnancy in 2023, the RSVPreF vaccine demonstrated efficacy in a single pivotal randomized trial (Maternal Immunization Study for Safety and Efficacy; MATISSE); postlicensure real-world evidence on vaccine eff

Translational and structural vaccinomics approach to design a multi-epitope vaccine against NOL4 autologous antigen of small cell lung cancer.

Small cell lung cancer (SCLC) is one of the most common cancers and it is the sixth common cause for cancer-related deaths. The high plasticity and metastasis have been a major challenge for humanity to treat the disease. Hence, a vaccine for SCLC has become an urgent need of the hour due to public health concern. Implementation of immunoinformatics technique is one of the best way to find a suitable vaccine candidate. Immunoinformatics tools can be used to overcome the limitations and difficulties of traditional vaccinological techniques. Multi-epitope cancer vaccines have become a next-gener

In silico modeling and structural analysis of asparaginyl endopeptidase of schistosoma mansoni (Sm32): Immunological and drug target implications.

Asparaginyl endopeptidase (AE) of Schistosoma mansoni (Sm32), also known as legumain, is a cysteine protease indirectly involved in the digestion of hemoglobin of Schistosoma sp. in the gastrodermis, being a vaccine candidate against this trematode and a potential drug target. This study presents a model for the three-dimensional structure of Sm32 determined by means of homology modeling and a molecular dynamics simulation with explicit solvent refinement. The structure proved to be consistent with other AEs of known crystal structures described in their proenzyme form, revealing a catalytic d

Design of Multi-Epitope DIVA-Compatible Vaccine Candidate Against Canine Parvovirus 2.

Canine parvovirus type 2 (CPV-2) is a highly contagious and often fatal canine pathogen, with antigenic variants (CPV-2a, 2b, 2c), maternally derived antibody (MDA) interference, and lack of Differentiating Infected from Vaccinated Animals (DIVA) capability limiting current vaccines. Using an immunoinformatics-based reverse vaccinology approach, we identified conserved B-cell, cytotoxic T lymphocyte (CTL), and helper T lymphocyte (HTL) epitopes from CPV-2 structural (VP1, VP2) and nonstructural (NS1, NS2) proteins. These were screened for antigenicity, immunogenicity, safety, and stability. Th

Bioinformatics-driven design and protective efficacy assessment of a multi-epitope vaccine for Toxoplasma gondii.

INTRODUCTION: Toxoplasma gondii can cause toxoplasmosis. It is an important type of pathogen within the broad category of emerging and re-emerging zoonoses. As an infectious disease featuring a complex multi-host transmission cycle, it poses an increasingly severe threat to global public health. No licensed vaccines are currently available for pets and humans, and thus a novel high-efficiency vaccine is urgently required. METHODS: Six antigens (GRA1, MIC17A, OWP2, LEA880, LEA870, and a hypothetical protein LEA530) representing different stages of the parasite lifecycle were selected from Toxo

Adjuvant-driven epitope hierarchy correlates with the protective efficacy of FimA vaccine against Klebsiella pneumoniae.

Hypervirulent Klebsiella pneumoniae (hvKP) variants represent urgent multidrug-resistant threats against which no licensed vaccine exists. FimA is indispensable for KP mucosal colonization and is thus a promising vaccine target; however, its protective immunodominant B-cell epitopes in vaccines with different adjuvants against KP remain undefined. Here, we revealed how adjuvants shape different hierarchies of the immunodominant B-cell epitopes of FimA and studied the humoral anti-FimA response and protective efficacy in a murine model of acute pneumonia. BALB/c mice were intramuscularly immuni

Immunoinformatics design of a novel multiepitope vaccine candidate against non-typhoidal salmonellosis caused by Salmonella Kentucky using outer membrane proteins A, C, and F.

The global public health risk posed by Salmonella Kentucky (S. Kentucky) is rising, particularly due to the dissemination of antimicrobial resistance genes in human and animal populations. This serovar, widespread in Africa, has emerged as a notable cause of non-typhoidal gastroenteritis in humans. In this study, we used a bioinformatics approach to develop a peptide-based vaccine targeting epitopes from the outer membrane proteins A, C, and F of S. Kentucky. Additionally, we employed flagellin protein (fliC) from Salmonella Typhimurium (S. Typhimurium) as an adjuvant to enhance the vaccine's

The human adenovirus E4-ORF1 protein subverts discs large 1 to mediate membrane recruitment and dysregulation of phosphatidylinositol 3-kinase.

Adenoviruses infect epithelial cells lining mucous membranes to cause acute diseases in people. They are also utilized as vectors for vaccination and for gene and cancer therapy, as well as tools to discover mechanisms of cancer due to their tumorigenic potential in experimental animals. The adenovirus E4-ORF1 gene encodes an oncoprotein that promotes viral replication, cell survival, and transformation by activating phosphatidylinositol 3-kinase (PI3K). While the mechanism of activation is not understood, this function depends on a complex formed between E4-ORF1 and the membrane-associated ce

Antibody signature of spontaneous clearance of Chlamydia trachomatis ocular infection and partial resistance against re-challenge in a nonhuman primate trachoma model.

BACKGROUND: Chlamydia trachomatis is the etiological agent of trachoma the world's leading cause of infectious blindness. Here, we investigate whether protracted clearance of a primary infection in nonhuman primates is attributable to antigenic variation or related to the maturation of the anti-chlamydial humoral immune response specific to chlamydial antigens. METHODOLOGY/PRINCIPAL FINDINGS: Genomic sequencing of organisms isolated throughout the protracted primary infection revealed that antigenic variation was not related to the inability of monkeys to efficiently resolve their infection.

Amplifying immune responses via a parenteral prime-intranasal boost strategy using a lipidated chitosan nanoparticle-based multi-epitope COVID-19 vaccine.

As COVID-19 continues to spread and new SARS-CoV-2 variants emerge, the immunity provided by first-generation vaccines has been waning, highlighting the need for new strategies to enhance antigen-specific immune responses and overall vaccine efficacy. Nasal vaccination is a promising approach for inducing mucosal immune responses, offering protection at the primary sites of SARS-CoV-2 infection. In this study, we report the development of a novel vaccine formulation, CSOA-LipoCov, consisting of oleic acid-functionalized chitosan nanoparticles loaded with a lipidated multi-epitope peptide-conju

Cytotoxic and immunomodulatory activity of CD151-LEL-based peptides in breast cancer and THP-1 cells.

CD151, a cell surface oncoprotein belongs to the tetraspanin family, drives metastasis, drug resistance, and immune escape, and is recognized as a tumor-associated antigen (TAA). In the screening phase, a positive correlation between CD151 expression and amplification of CCND1, ERBB2, and MYC genes as well with breast cancer (BC) metastasis was observed using data from different GEO datasets. This study identified three immunogenic peptides within the large extracellular loop (LEL) of CD151 using in silico approaches: one each of CTL and HTL epitope, which bind strongly to class I and II MHC m
