Homocysteine and related B-vitamins throughout our International Conferences: a historical perspective.

In this invited keynote review, I reflect on biochemical, clinical, and epidemiological evidence on the role of folate and vitamin B12 in one-carbon metabolism and their relationship with homocysteine, integrating over five decades of research. One-carbon metabolism represents a central biochemical network linking folate-dependent one-carbon transfer reactions with vitamin B12-dependent remethylation of homocysteine to methionine. Disruptions in this pathway result in elevated homocysteine concentrations and impaired methylation capacity, providing a mechanistic link between micronutrient stat

Iron deficiency anemia in pregnancy and related complications with specific insight in Rivers State, Nigeria: a narrative review.

Iron deficiency anemia (IDA) is one of the most common nutritional disorders affecting pregnant women worldwide, with significant implications for maternal and neonatal health. In developing regions like Rivers State, Nigeria, the prevalence of IDA in pregnancy remains alarmingly high, exacerbated by factors such as poverty, inadequate healthcare access, suboptimal nutrition, and endemic parasitic infections like malaria. This narrative review provides an updated overview of IDA in pregnancy, emphasizing its prevalence, risk factors, and complications, with a specific focus on Rivers State. Th

The role of B vitamins in stroke prevention.

Elevated plasma levels of homocysteine (Hcy) are a recognized risk factor for stroke. This relationship represents one aspect of the debated `Hcy hypothesis'. Elevated Hcy may be an independent and treatable cause of atherosclerosis and thrombotic vascular diseases. Further observations indicate that proper dietary supplementation with B-vitamins decreases total plasma Hcy concentrations and may be an effective intervention for stroke prevention. Metabolic vitamin B12 deficiency is a nutritional determinant of total Hcy and stroke risk. Genetic factors may link B vitamins with stroke severity

Clinical and molecular characteristics of imerslund-gräsbeck syndrome: First report of a novel Frameshift variant in Exon 11 of AMN gene.

INTRODUCTION: Imerslund-Gräsbeck syndrome (IGS) is a rare autosomal-recessive disorder characterized by selective vitamin B12 malabsorption, megaloblastic anemia, and proteinuria. The precise incidence of this disorder is unknown in the Middle East and Arab countries. The disease is caused by a homozygous variant in either AMN or CUBN genes. In addition, some compound heterozygous variants are reported. METHODS: Clinical and laboratory data of patients diagnosed with IGS in Oman were retrospectively collected. Mutation analysis for all genes involved in vitamin B12/folic acid metabolism and m

Benzbromarone, Quercetin, and Folic Acid Inhibit Amylin Aggregation.

Human Amylin, or islet amyloid polypeptide (hIAPP), is a small hormone secreted by pancreatic β-cells that forms aggregates under insulin deficiency metabolic conditions, and it constitutes a pathological hallmark of type II diabetes mellitus. In type II diabetes patients, amylin is abnormally increased, self-assembled into amyloid aggregates, and ultimately contributes to the apoptotic death of β-cells by mechanisms that are not completely understood. We have screened a library of approved drugs in order to identify inhibitors of amylin aggregation that could be used as tools to investigate t

Folates from metabolically engineered rice: a long-term study in rats.

SCOPE: The biological impact of folates from folate rice, a metabolically engineered (biofortified) rice line, rich in folates, was investigated. Its consumption may be helpful to fight folate deficiency. Our objective was to investigate the potential of folate rice to supply the organism with folates and evaluate its biological effectiveness using a rat model. METHODS AND RESULTS: Five groups of 12 Wistar rats were monitored during a 7/12-wk depletion/repletion trial. Animals receiving folate-free diet (0 μg/rat/day) and those additionally receiving wild-type rice (on average 0.11 μg/rat/day

Nutritional aspects of Helicobacter pylori infection.

H. pylori is a gram-negative pathogen, etiologically associated with atrophic and non-atrophic gastritis, peptic ulcer, primary gastric B-cell lymphoma and gastric carcinoma. Several observations demonstrated a correlation between H. pylori and malabsorption of essential nutrients; epidemiological studies have shown an association between H. pylori infection and iron deficiency anemia, while the absorption of some vitamins such as vitamin B12, vitamin A, vitamin C, folic acid and Vitamin E may be affected by the infection. The main mechanism related to malabsorption of this components is the m

C677T polymorphism of the methylenetetrahydrofolate reductase gene does not affect folic acid, vitamin B12, and homocysteine serum levels in Turkish children with neural tube defects.

Association between neural tube defects (NTDs) and C677T polymorphism of the methylenetetrahydrofolate reductase (MTHFR) gene was suspected, because the MTHFR gene codes for a key enzyme in folate metabolism. Its deficiency usually leads to significant reductions in plasma concentrations of folate, vitamin B(12) and methionine, whereas homocysteine levels are increased. We examined folate, vitamin B(12) and homocysteine serum concentrations and polymorphism of the C677T MTHFR gene in Turkish children with neural tube defects. Thirty-three children with NTDs, 26 mothers and 48 healthy individua

Extreme consumption of Beta vulgaris var. rubra can cause metal ion accumulation in the liver.

Redox homeostasis can be considered as the cumulative action of all free radical reactions and antioxidant defences in different tissues, which provide suitable conditions for life. Transition metal ions are ubiquitous in biological systems. Beta vulgaris var. rubra (table beet root) contains several bioactive agents (e.g. betain, betanin, vulgaxanthine, polyphenols, folic acid) and different metal elements (e.g. Al, B, Ba, Ca, Cu, Fe, K, Mg, Mn, Na, Zn), which act on the various physiological routes. Therefore we studied the effect of this metal rich vegetable on element content of the liver

The number of dichorionic twin pregnancies is reduced by the common MTHFR 677C-->T mutation.

In multiple pregnancies, demands for folic acid are considerably increased. The most common inborn error of folate metabolism is mild methylenetetrahydrofolate reductase (MTHFR) deficiency due to the synthesis of a thermolabile variant of the enzyme with impaired catalytic activity which leads to reduced 5-methyltetrahydrofolate (5-methyl-THF) and mildly elevated homocysteine plasma concentrations when folate status is inadequate. To investigate whether the number of offspring is influenced by this mutation, we determined the frequency of the 677C-->T substitution in 156 singleton and 40 twin

Haematological nutrition of schoolchildren in the far northwestern cape.

The prevalence of anaemia and deficiencies of iron, folate and vitamin B12 were investigated in primary schoolchildren of mixed ethnic origin living in four towns in a remote semi-desert area of southern Africa, the Richtersveld. Anaemia was uncommon (6.3%); iron and vitamin B12 deficiencies were not observed. Sub-normal erythrocyte folate concentrations were present in 14.5%, but significant folate deficiency was rare (less than 2%). There were negligible differences in the haematological parameters of the children in the four towns, even though children living in Kuboes received milk at scho

On reducing the frequency of severe abruptio placentae.

At Parkland Memorial Hospital the frequency of abruptio placentae so severe as to kill the fetus has decreased from 1 in 420 deliveries during 1956 through 1969 to 1 in 830 during 1974 through 1989. Major factors in this reduction were elimination of very high parity and a marked increase in the percentage of Latin American women, in whom the risk was 1 in 1473 deliveries compared with 1 in 595 for black women and 1 in 876 for white women. Abdominal trauma was encountered rarely, as was fetoplacental-to-maternal hemorrhage sufficient to impair fetal perfusion seriously. Abnormal development of
