New insight for management of blunt splenic trauma: significant differences between young and elderly.

BACKGROUND/AIMS: Although highly successful in children and young patients, the non-operative management of blunt splenic injury in the elderly has yet to be clearly studied. The purpose of this study was to determine whether or not a relationship exists among the mechanism of injury, the grade of splenic injury, the associated injuries, and whether patterns of injury differ between the young group (younger than 60 years old) and the elderly group (60 years and older than 60 years). METHODOLOGY: One hundred and sixty-seven patients (116 young patients including 30 early deaths and 51 elderly

A Systematic Evaluation of Molecule Generation Models for De Novo Drug Design: From Benchmarks to Practical Insights.

Molecule generation has emerged as a powerful computational tool for de novo drug design, enabling the exploration of the chemical space beyond the limits of conventional virtual screening. The field has progressed rapidly, driven by advances in molecular representations, generative architectures, and target-aware modeling strategies. However, existing reviews typically address specific model families or application scenarios in isolation rather than offering an integrated perspective on how these components collectively form a coherent generation workflow. In this review, we present a compreh

Multi-Omics and Liquid Biopsy Integration for Precision Stratification of Breast Cancer.

Breast cancer is a biologically heterogeneous disease that requires accurate diagnosis, risk stratification, treatment selection and longitudinal assessment. Conventional imaging and tissue biopsy remain the mainstays of clinical management. However, tissue sampling represents a specific anatomical site and time point, and may not reflect the molecular heterogeneity or evolution of disease. Liquid biopsy offers a minimally invasive complementary approach, analyzing circulating tumor cells (CTCs), circulating tumor DNA (ctDNA) and extracellular vesicles. Applications include molecular character

Ancient Greek votive epigraphy: Ancient religion between individual choice and cultural trends.

Ancient votive inscriptions, numbering in the tens of thousands across the Mediterranean, offer direct evidence for religious practice but have rarely been studied comprehensively across time and space due to their sheer volume. This paper presents a "distant reading" analysis of approximately 11,000 Greek dedicatory inscriptions (sixth century BCE - seventh century CE) drawn primarily from the GIST/I.PHI digital corpus. By tagging inscriptions for key features (votive/dedicatory terminology, deities, motives, gender) and analyzing their distribution chronologically and geographically while co

Exploring the pharmacological basis of adverse effects in oxazolidinone and antidepressant cotherapy: an in silico study.

Linezolid and related oxazolidinones are frontline antibiotics against multidrug-resistant Gram-positive pathogens, including methicillin-resistant Staphylococcus aureus (MRSA), yet their co-administration with antidepressants has been clinically associated with serotonin syndrome (SS) or hypertensive crisis (HC). While traditionally attributed to monoamine oxidase (MAO) inhibition, clinical observations remain variable and the underlying mechanism is not completely understood. In this exploratory study, we applied an integrated in silico framework combining target prediction, network similari

pH-triggered liposomal delivery of alendronate enables acid-responsive drug release and improves osteoblast viability.

Bisphosphonates, such as sodium alendronate (ALN), are primary treatments for osteoporosis and osteosarcoma; however, their long-term use is limited by causing severe side effects. A pH-sensitive liposomal formulation was developed to enhance ALN delivery, enabling controlled drug release specifically in acidic biological microenvironments and niches. Release efficiency of pH-sensitive liposomes was evaluated and compared with that of standard and PEGylated liposomes. Molecular dynamics simulations demonstrated the stability and structural behavior of liposome formulations under physiological

A Hierarchical-Dynamic integration model for the formation of interpersonal neural synchrony during spoken communication.

Interpersonal neural synchrony (INS) is widely observed during spoken communication, yet the cognitive and neural processes through which it emerges remain incompletely understood. This review synthesizes evidence on the functional significance, representational organization, and neural implementation of INS and proposes a Hierarchical-Dynamic Integration Model (HDIM) of its formation. HDIM conceptualizes INS as the cross-brain manifestation of ongoing coordination between two interdependent processes: multilevel linguistic alignment and contextual-intentional integration. The hierarchical org

Physics-informed neural ordinary differential equations for PBPK-based internal dose prediction in environmental health risk assessment.

Reliable prediction of internal doses, especially systemic blood concentrations, is crucial for exposure science and environmental health risk assessment. While traditional physiologically based pharmacokinetic (PBPK) models provide mechanistic insights, their fixed parameters often fail to capture nonlinear dynamics at high concentrations, as encountered in acute contamination scenarios. This study introduces a hybrid PBPK-constrained neural ODE framework that combines neural ordinary differential equations with PBPK modeling. By integrating a minimal two-neuron network correction at the live

A Novel Peak-Shape Aware Approach for Mass Alignment in Mass Spectrometry.

RATIONALE: In mass spectrometry measurements, mass shifts may be inadvertently introduced due to instrumental drift and calibration inaccuracies, potentially compromising the accuracy of subsequent data analysis. This work presents a novel, label-free algorithm to improve relative mass alignment between mass spectra. The warping function is modeled as a natural cubic spline, a suitable model for gradual, nonlinear mass shifts. METHODS: The algorithm is validated on a dataset generated from human glioblastoma multiforme samples using matrix-assisted laser desorption/ionization time-of-flight m

The transformative role of single-cell analysis in multifactorial disorders research.

Multifactorial inherited disorders (MIDs) arise from complex interactions between polygenic risk and environmental exposures, presenting major challenges for mechanistic discovery, patient stratification, and targeted therapy development. While traditional approaches like genome-wide association studies (GWAS) and bulk omics profiling have identified broad associations, they often struggle to resolve the cellular context in which these interactions drive pathogenesis.Emergingsingle-cell technologies now offer unprecedented resolution to dissect tissue heterogeneity, define rare or transient di

Freezing of Gait in Parkinson's Disease: A Scoping Review on the Path Towards Real-Time Therapies.

BACKGROUND: Freezing of gait (FoG) is a common symptom of Parkinson's disease, especially in its later stages of progression. Characterized by involuntary stopping during normal gait patterns, FoG greatly increases fall risk, reducing quality of life. Given the complex presentation and etiology of FoG, current treatments have proven ineffective in managing episodes. In recent years, machine learning algorithms have been leveraged to derive actionable clinical insights from biomedical datasets. As a manifestation of neuromechanical dysfunction, impending FoG episodes may be characterized throug

Combining Device Information and Molecular Fingerprints into a Global Model for Reliable Prediction of Power Conversion Efficiency of Dye-Sensitized Solar Cells.

Solar cells hold the potential to meet the growing worldwide demand for clean and sustainable energy. Today most solar cells are based on silicon, yet new alternatives are continuously emerging. In particular, dye-sensitized solar cells (DSSCs) constitute low-cost electrochemical energy-harvesting devices. The design of new DSSCs with improved properties could be aided by computational techniques, in particular machine learning (ML) models. In this work, we therefore present the first global ML model based on nearly all entries (4351 out of 4426) in the DSSC Database (DSSCDB). Recognizing that
