Copper Promotes Breast Cancer through Immunometabolic Reprogramming: A Multi-omics Mediation Analysis.

BACKGROUND: Elevated copper (Cu) concentration or copper to zinc (Cu/Zn) ratio have been consistently observed in the blood and tumor tissues of patients with breast cancer, although the underlying mechanisms remain unclear. The objective of this study was to assess the effects of plasma Cu on breast cancer risk and the associated immunometabolic pathways. METHODS: We conducted a nested case-control study (226 matched pairs) within the Taizhou Longitudinal Study. Plasma levels of Cu and Zn were quantified, and large-scale proteomic and metabolomic profiling was performed. Structural equation

The Quartet of Core Oncogenic Drivers in Neuroendocrine Prostate Cancer: Multi-Omics Dataset Integration to Forge a Translational Link Between Biology and Precision Therapy.

Prostate cancer (PC) is the most common cancer among American men and the second leading cause of cancer-related deaths. For advanced or metastatic PC, anti-androgen therapies, including androgen deprivation therapy (ADT), are considered standard treatment options. However, these therapies often result in the development of castration-resistant prostate cancer (CRPC) or neuroendocrine prostate cancer (NEPC), both of which present significant treatment challenge. The molecular mechanisms driving the progression from androgen - sensitive PC to castration-resistant and neuroendocrine phenotypes a

Integrated lipidome and miRNome analyses reveal sex-based differences in circulating extracellular vesicles of alcohol use disorder patients.

Integrated multi-omics and extracellular vesicle (EV) analysis are emerging as powerful, complementary strategies for biomarker discovery. These approaches offer promising tools to enhance early detection, diagnosis, and treatment of alcohol use disorder (AUD). Here we applied an integrated miRNomic and lipidomic approach to analyze plasma EVs from AUD patients and controls of both sexes to gain a comprehensive understanding of the underlying molecular mechanisms. We identified an AUD signature with predictive potential for diagnostic applications, representing an initial step in this directio

Insights into the influencing factors of fecal gut microbiota in hepatocellular carcinoma.

Recently, Feng et al published a significant study. Through multi-omics analysis, their research team revealed associations between fecal metabolome, gut bacteria, and serum indicators in patients with hepatocellular carcinoma (HCC). These results suggest that the composition of the metabolome and gut bacteria in fecal samples hold promise as potential biomarkers for the diagnosis of HCC. After carefully reviewing their work, we offer two points of insight: (1) The influence of different factors on gut microbiota changes in HCC; and (2) The impact of the hepatitis B virus and antiviral drugs o

Multi-omics-based insights into the microbial community composition and quality characteristic differentiation potential between Xuanwei and Iberian dry-cured hams.

To uncover the microbial underpinnings of quality and flavor differences between Xuanwei ham (XWH) and Iberian ham (IRH), this study integrated high-throughput sequencing with metabolomics and volatilomics to establish microbiota-metabolite-flavor linkages. Physicochemical characterization revealed that XWH showed higher moisture, water activity, and salt content, whereas IRH had higher fat and protein. These matrix differences imposed selective pressures on microbial colonization. The IRH surface exhibited higher bacterial diversity, while the XWH interior showed higher bacterial diversity; X

Exploiting virulence determinants for precision management of melioidosis: A multi-omics roadmap from pathogenesis to point-of-care : A perspective review.

Melioidosis, caused by Burkholderia pseudomallei, remains challenging to diagnose promptly, partly due to limited use of virulence factors as diagnostic targets. Most existing biomarkers fail to capture the dynamic expression of bacterial virulence factors during active infection. In vivo‑upregulated virulence determinants, such as Hcp1, POMCR/PPEP, and GroEL, are intrinsically superior diagnostic targets. Hcp1 is a paradigm: it is the most advanced virulence factor translated into a point‑of‑care test, with prospective validation in endemic settings. Mechanistic understanding of virulence reg

Multi-omics reveals circadian regulation of bone homeostasis by gut microbiota metabolites: mechanisms and chronotherapeutic implications.

The gut-bone axis plays a pivotal role in skeletal health, yet the integration of multi-omics approaches to elucidate circadian metabolite-bone interactions remains limited. This review synthesizes evidence from metagenomics, metabolomics, and germ-free models to uncover how microbiota-derived metabolites-including short-chain fatty acids (SCFAs), bile acids, tryptophan derivatives, and gaseous molecules-orchestrate bone remodeling in osteoporosis, osteoarthritis, and bone malignancies. Many studies demonstrate that SCFAs inhibit osteoclastogenesis via GPR43/HDAC signaling and promote osteobla

BACKGROUND: Emerging evidence links macrophage polarization to osteoporosis pathogenesis, yet the specific causal genes and underlying molecular mechanisms remain poorly defined. METHODS: We used a multi-stage, summary-data-based Mendelian randomization (SMR) framework to integrate blood-derived methylation, expression, and protein quantitative trait loci (mQTL, eQTL, and pQTL) with large-scale osteoporosis genome-wide association study (GWAS) datasets. We performed SMR and colocalization analyses across predefined macrophage polarization-related genes, followed by multi-omics integration. To

Multi-Omics Integration Identifies a Six-Gene Diagnostic Signature for Ankylosing Spondylitis via Metabolic-Immune Crosstalk.

Ankylosing spondylitis (AS) is a chronic immune-mediated inflammatory disease affecting the axial skeleton, characterized by progressive structural damage and functional impairment. Although biologic therapies targeting tumor necrosis factor and interleukin-17 have improved clinical outcomes, a substantial proportion of patients fail to achieve sustained disease control. Emerging evidence suggests that metabolic alterations may contribute to AS pathogenesis; however, systematic characterization of metabolism-related biomarkers and their regulatory networks remains limited, and the interplay be

Disentangling Shared and Differential Genetic Architectures Between COVID-19 and Other Respiratory Disorders-A Genome-Wide Multi-Omics Framework.

A bidirectional relationship has been observed between COVID-19 and respiratory disorders, where respiratory comorbidities increase severity, and COVID-19 induces respiratory sequelae. The underlying biological and genetic mechanisms remain unclear. While previous studies have identified overlapping genetic loci, few have systematically disentangled the genetic factors shared between these conditions versus those specific to COVID-19, particularly at a multi-omics level. We developed and applied a unified analytical framework to compare three COVID-19 phenotypes with eight respiratory disorder

Integrated Multi-Omic Analyses Reveal Gender-Specific Molecular Mechanisms Regulating Growth and Muscle Development in Zhongshan Partridge Ducks.

Zhongshan Partridge duck is a unique indigenous dual-purpose waterfowl native to China's Pearl River Delta. This once endangered breed has been restored through conserved breeding. Its slow growth and low breast muscle yield limit industrial application, and obvious sexual dimorphism in pectoral muscle growth and metabolism lacks systematic molecular explanation. Herein, we combined GWAS, pectoral RNA-seq and untargeted metabolome to dissect regulatory networks of growth and carcass traits using 365 standardized ducks. Ten growth and carcass phenotypes were collected, and the Gompertz model fi

Mitochondrial Proteostasis Links Diabetes and Sarcopenia: Cross-Scale Convergence from Experimental Models to Human Multi-Omics.

Sarcopenia in type 2 diabetes mellitus is increasingly recognized as a mechanistic consequence of chronic metabolic stress rather than mere age-related comorbidity. This review synthesizes evidence demonstrating how insulin resistance, hyperglycemia, lipotoxicity, and inflammation converge on skeletal muscle mitochondrial proteostasis to drive progressive decline. We evaluate seven pathway modules-mitochondrial dynamics, mitophagy, biogenesis, oxidative phosphorylation, nicotinamide adenine dinucleotide (NAD+)/sirtuin (SIRT)-linked regulation, protein import, and the mitochondrial unfolded pro
