Genome-wide analysis of the HDAC gene family in Pyrus betulifolia and functional validation of PcHDAC5 and PcHDT1.2 in Pyrus calleryana seed dormancy release.

Histone deacetylation, catalyzed by histone deacetylases (HDACs), is a core epigenetic regulatory mechanism in plants. However, no systematic characterization of the HDAC gene family has been reported in pear (Pyrus spp.), a globally important temperate fruit crop. In this study, we performed genome-wide identification and functional characterization of HDAC genes in pear, with a specific focus on their regulatory roles in seed dormancy release. We identified 22 PbHDAC genes from the genome of Pyrus betulifolia Bunge, which were classified into three conserved subfamilies. Evolutionary analysi

Effects of a 10-week creative dance intervention on social-emotional competence in preschool children: A preliminary cluster-randomized trial.

BACKGROUND: Effective interventions to enhance social-emotional competence in typically developing preschoolers remain scarce, particularly those aligning with young children's natural, embodied modes of learning. OBJECTIVE: To examine the effects of a 10-week Creative Dance intervention on social-emotional competence in preschool children in a preliminary cluster-randomized trial design. METHODS: Four kindergarten classes were randomly allocated to a Creative Dance group (n = 62) receiving two 30-40 min sessions weekly for 10 weeks, or a control group (n = 59) continuing usual activities. Smart Sensors, Smarter Hearts: The Wearable Revolution in Cardiology.

Cardiovascular disease (CVD) continues to claim more lives than any other condition worldwide. Traditional clinic visits capture only brief snapshots of health, leaving many opportunities for prevention and early intervention unmet. Wearable technologies now extend cardiovascular care into daily life, delivering continuous physiologic and behavioral data that could transform how we detect, treat, and ultimately prevent CVD. This review highlights how wearable devices, ranging from consumer-grade wristbands to advanced sensor-embedded textiles, are reshaping cardiovascular medicine. We discuss

Predicting the Potential Distribution of Amyelois transitella (Walker) in China Under Climate Change Using a Biomod2-Based Ensemble Model.

The Navel Orangeworm (Amyelois transitella Walker, 1863), a primary pest of nut crops native to North America, poses a significant potential threat to China's agricultural biosecurity, yet its potential distribution dynamics under climate change remain unquantified. This study utilized the Biomod2 ensemble model platform to predict habitat suitability under current and future climate scenarios (SSP1-2.6 and SSP5-8.5). We evaluated the prediction accuracy of the ensemble model using calibration data, with TSS = 0.898 and AUC = 0.978, while spatially stratified cross-validation confirmed moderat

Acute and Seasonal Effects of Temperature on Bite Performance of Alligator Snapping Turtles (Macrochelys temminckii).

Environmental temperatures influence most aspects of ectotherm biology, especially when fluctuating on daily and seasonal scales. The Alligator Snapping Turtle (Macrochelys temminckii) is a species of conservation concern inhabiting temperate latitudes in the southeastern United States. To study the effect of temperature and season on the bite performance of M. temminckii, we used a force transducer and high-speed videography to measure bite force and kinematics, including bite duration, jaw velocity and acceleration, and lunge velocity and acceleration, for 21 captive subadult M. temminckii.

Different Temperature Regimes Affect Gonadal Development and Energy Budget in Apostichopus japonicus.

Apostichopus japonicus is a temperate marine echinoderm whose growth and reproduction are strongly influenced by temperature. We compared adult sea cucumbers exposed to stepwise warming (10-12, 12-14, and 14-16 °C across three successive 20-day stages) with animals maintained at 15 °C. Growth indices, gonadosomatic index (GSI), gonadal histology, and energy-budget components were evaluated in females and males. The 60 d experiment used three replicate tanks for each treatment-by-sex combination. Compared with the constant 15 °C treatment, the stepwise-warming regime showed a higher female GSI

A New Challenge to Species Delimitation: Remarkable Genomic and Ecological Diversity in the Butterfly Melitaea diamina.

Evolution relentlessly challenges any concept of species, and human efforts to fit a taxonomic hypothesis to patterns in nature often seem unsatisfactory. Genomics show that a solution does not always come with more data: a fine-scaled view may reveal populations with a more gradual array of differentiation, infer cases of hybrid origin and bring to light the existence of ghost populations. The false heath fritillary, Melitaea diamina, is a Palearctic butterfly with patchy, localized populations highly dependent on humid temperate habitats. Despite its extensive geographic range, populations a

Dominance and toxicity without lethality: Exploring biomass, cyanometabolites, and Daphnia responses across Cyanobacterial strains.

Cyanobacteria are cosmopolitan, important components of biomass with a remarkable ability to synthesize a diverse array of bioactive compounds, some of which may have toxic effects on other organisms and ecosystems. In this study, filamentous cyanobacterial strains isolated predominantly from temperate European freshwaters (Aphanizomenon, Chrysosporum, Cuspidothrix, Dolichospermum, Planktothrix, Raphidiopsis), with a subset from Arctic waters (Microcoleus, Phormidesmis), were qualitatively screened for commonly studied cyanometabolites, using immunoassays and chromatographic techniques. We als

The larch DAL1 interacts with PEBP family genes to regulate the annual growth cycle.

Boreal and temperate perennial woody plants coordinate annual growth cycle with seasonal environmental changes with age-dependent phenological differences. However, the underlying molecular mechanisms remain limited. In this study, we demonstrated that the MADS-box transcription factor DAL1 in Larix kaempferi (Japanese larch) directly regulates the expression of PEBP family genes LkFT1 and LkMFT to govern the annual growth cycle. LkDAL1 binds to the LkFT1 promoter to repress its expression, and to the LkMFT promoter to activate its expression. LkFT1 is highly expressed in the dormant stage, wi

Spatiotemporal Dynamics of Human Metapneumovirus and Potential Impact of Respiratory Syncytial Virus Interventions in the United States.

Human metapneumovirus (HMPV) causes acute lower respiratory infections, primarily affecting young children and older adults, with seasonal outbreaks peaking annually in March or April in the United States and other temperate regions in the Northern hemisphere. However, the factors driving HMPV seasonality in the United States remain poorly understood. We analyzed laboratory-confirmed HMPV cases and age-specific emergency department visits across 10 US regions, fitting an age-stratified dynamic transmission model to assess spatiotemporal patterns and investigate the influence of environmental v

Pathophysiology, assessment, and treatment of pain in patients with pancreatic ductal adenocarcinoma.

Pain is one of the most frequent and debilitating symptoms associated with pancreatic ductal adenocarcinoma (PDAC). More than 60% of patients suffer from significant pain at diagnosis. The prevalence increases during the progression of the disease and is associated with anorexia, weight loss, and impaired social interactions. The pathophysiology of pain includes the combined effects of tumor growth and spread, perineural invasion, neuroimmune interactions, peripheral nerve remodeling, and central nervous system sensitization. Pain is additionally modulated by comorbid conditions, such as anxie

Adaptations of the cardiovascular system in hibernating mammals.

Winter in temperate and high latitudes imposes severe challenges for animal survival, associated with low ambient temperature and limited food availability. In some mammalian species, these challenges are met by hibernation, a state characterized by marked reduction in core temperature and metabolic activity. Despite these extreme conditions, hibernating mammals preserve cardiac function and avoid life-threatening arrhythmias and vascular failure. This review summarizes the current evidence on cardiovascular adaptations supporting survival during torpor, focusing on cardiac electrophysiology,

Sirolimus and Mimetics: Modulator Sine Qua Non for Cardiac Xenotransplantation?

The last decade has seen Promethean advances in cardiac xenotransplantation, led by genetic editing of porcine xenografts, such that they lack the most immunogenic antigens. Nonetheless, cardiac xenotransplants have reached an apparently insuperable survival limit of around 50 days. Immunity is not only provoked by alien antigens but equally by the absence of self-antigens. This is the frequently overlooked "missing self" rejection. It is amenable to inhibition by sirolimus and other mammalian target of rapamycin (mTOR) signaling suppressors. In this hypothesis, it is posited the next step in

Effect of Manual Tungsten Inert Gas Welding on the Microstructure and Mechanical Properties of Welded Joints in Thick SIMP Steel Plates.

The novel martensitic heat-resistant SIMP steel is considered a primary candidate structural material for Accelerator Driven Sub-critical Systems (ADSs) due to its excellent comprehensive properties. However, welding difficulties arising from its high alloy content represent a major bottleneck for its engineering application. This study explored and established optimized manual Tungsten Inert Gas (TIG) welding process parameters suitable for 20 mm thick SIMP steel plates, involving 24 layers and 110 passes, with an average heat input of approximately 4 kJ/cm, successfully achieving well-formed

Semaphorin 3E regulates the response of regulatory T cells to lipopolysaccharide-induced systemic inflammation.

Sepsis is a life-threatening systemic inflammation marked by an initial hyperinflammatory phase. Tregs are pivotal in tempering this early immune response. Semaphorins, originally neuronal guidance cues, have emerged as immune modulators. Notably, Sema3E influences dendritic cell and T cell responses, but its role in Treg-mediated regulation during endotoxemia is unclear. WT and Sema3e-/- mice were subjected to LPS-induced endotoxemia. Treg frequencies, proliferation and migration were assessed by flow cytometry and Transwell assays, respectively. Serum cytokines (eg IL-10) were quantified usi

Systemic Therapy for Salivary Gland Cancers: A Review of Targeted and Chemotherapeutic Approaches.

Systemic therapy for recurrent and metastatic salivary gland malignancies (SGMs) remains a major therapeutic challenge. Traditional chemotherapy offers modest response rates with limited durability, and its role is largely palliative. The most meaningful advances have occurred in biomarker-selected subgroups: androgen receptor blockade for AR-positive salivary duct carcinoma and HER2-directed therapy for HER2-positive tumors have demonstrated superior response rates and survival (often exceeding 50%), compared with the single digit response rates seen with unselected chemotherapy or tyrosine k

Rethinking grapevine downy mildew management: opportunities to disrupt the sexual cycle of the pathogen as a preventive disease control strategy.

Plant diseases caused by fungal and oomycete pathogens with mixed reproduction systems include some of the most damaging crop diseases. These pathogens reproduce asexually under low-stress conditions during the growing season in order to propagate rapidly and efficiently. They then shift to sexual reproduction when conditions become harsher, so as to survive and adapt. For those that are obligate biotroph pathogens with a deciduous host, sexual reproduction is a necessary step in their life cycle. Plasmopara viticola, the oomycete causing grapevine downy mildew (GDM), is a representative examp

[Acupuncture and biological resilience].

This paper systematically elaborates the theories of stress, biological resilience and hormesis. Mild low-dose stress induces organismic overcompensation via preconditioning, activates nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated antioxidant, autophagy, DNA repair and other pathways, and presents an inverted U-shaped biphasic dose-response relationship. Preconditioning is categorized into local and remote types, which exerts long-term protective effects on systemic organs via metabolic reprogramming as well as neuro-humoral and immune pathways, with 2 distinct protective time wi
