Afferent renal inputs to paraventricular nucleus vasopressin and oxytocin neurosecretory neurons.

Extracellular single-unit recording experiments were done in pentobarbital sodium-anesthetized rats to investigate the effects of electrical stimulation of afferent renal nerves (ARN) and renal vein (RVO) or artery (RAO) occlusion on the discharge rate of putative arginine vasopressin (AVP) and oxytocin (Oxy) neurons in the paraventricular nucleus of the hypothalamus (PVH). PVH neurons antidromically activated by electrical stimulation of the neurohypophysis were classified as either AVP or Oxy secreting on the basis of their spontaneous discharge patterns and response to activation of arteria

Activation of alpha 2-adrenergic receptors decreases nerve trauma-induced afferent barrage but not autotomy.

Effects of the selective alpha 2-adrenoceptor agonist, medetomidine, on a compound volley of a tibial nerve stimulation-evoked spinal reflex, pain-induced phrenic motor responses and on postoperative neuropathic pain behavior were studied in rats. Medetomidine (0.3 mg/kg) decreased the amplitude of the compound volley recorded from peroneal nerve in response to tibial stimulation in pentobarbital (40 mg/kg) anesthetized rats. Atipamezole, an alpha 2-adrenoceptor antagonist (1.5 mg/kg) fully restored the response when given 60 min after the medetomidine administration. Pain-evoked phrenic motor

The interval between the positive peak of premyoclonus spike and the onset of myoclonus is shorter than the cortical latency in cortical myoclonus.

In 6 patients with cortical myoclonus, the intervals between the positive peak of the premyoclonus cortical spike and the onset of the myoclonic jerk [interval (Sp-Mcl)] were precisely measured by the computer-assisted jerk-locked averaging method. The intervals (Sp-Mcl) were 13.0-15.8 ms for the abductor pollicis brevis, 8.0-10.4 ms for the extensor digitorum communis and 5.5 ms for the biceps brachii, respectively. These were apparently shorter than the normal latencies of the responses elicited by transcranial cortical stimulation. The difference between the interval (Sp-Mcl) and the spinal

Fibers innervating different parts of the lateral line system of an Antarctic notothenioid, Trematomus bernacchii, have similar frequency responses, despite large variation in the peripheral morphology.

Regional differences in the architecture and size of lateral line canals and neuromasts were measured in an Antarctic fish, Trematomus bernacchii, and the data were used in models of canal and cupular mechanics to predict the frequency response of these two peripheral structures. These modeled predictions were then compared to frequency response functions measured with single unit recording techniques from anterior and posterior lateral line fibers innervating different canals on the head and trunk of fish of various sizes. Despite large variations in the peripheral morphology of head and trun

Galanin-like immunoreactivity in autonomic regions of the rat lumbosacral spinal cord is sexually dimorphic and varies with the estrous cycle.

These investigations show that there is a heterogeneous distribution of galanin-like immunoreactivity (GAL-LI) within laminae VII and X of the rat thoraco-sacral spinal cord. In either sex, GAL-LI fibers sparsely outline the position of male and female preganglionic sympathetic neurons in thoracic spinal segments; whereas in lumbosacral segments, far greater numbers of GAL-LI fibers surround autonomic preganglionic neurons. An unusual feature of the GAL-LI fibers in lumbosacral autonomic regions is their sexually dimorphic distribution with males containing greater numbers of GAL-LI fibers tha

Multimodal evoked potentials in progressive external ophthalmoplegia with mitochondrial myopathy.

Multimodal evoked potentials were studied in 13 patients affected by progressive external ophthalmoplegia with histologically proven mitochondrial myopathy. Progressive external ophthalmoplegia occurred with craniosomatic spreading in all the patients and with a varying degree of nervous and/or other system involvement in most of them. In all but one of the subjects, at least one evoked potential modality was abnormal; 11 of them demonstrated an abnormal visual evoked potential, but this finding might have been influenced by concurrent retinal dysfunction. Abnormalities in brainstem auditory e

Responses of cat C1 spinal cord dorsal and ventral horn neurons to noxious and non-noxious stimulation of the head and face.

Previous anatomical studies have shown that trigeminal and cervical afferent nerve fibers project to the upper cervical segments of the spinal cord. To determine the response properties of neurons in the upper cervical spinal cord, we studied the response of C1 dorsal and ventral horn cells to electrical and graded mechanical stimulation of the face, head and neck in anesthetized cats. Neurons were classified as low-threshold-mechanoreceptive (LTM), wide-dynamic-range (WDR), nociceptive-specific (NS) or unresponsive, based on their responsiveness to graded mechanical stimulation. Extracellular

Neurophysiological mechanisms in abnormal reflex activities in cerebral palsy and spinal spasticity.

Dorsal root stimulation, carried out during operation in 80 spastic cerebral palsy patients and in one spastic quadriplegic patient, allowed a study of abnormal monosynaptic and polysynaptic reflexes. Frequency-related depression of the monosynaptic reflex was not present; increased activity through non-suppressed polysynaptic pathways was shown.

Brain control of conjugate horizontal and vertical eye movements: a survey of the structural and functional correlates.

(1) It appears that all oculomotor pathways originating within the cerebrum and mediating stimulations and lesions, project from the two sides of the brain through the diencephalon to the brain-stem. (2) The pathways subserving horizontal movements decussate at the level of the oculomotor and trochlear nuclei, across the midsagittal plane. The direction of vector action within the brain above the 'electroanatomical' oculomotor decussation is predominately contraversive; below this levelit is ipsiversive. (3) The pontine reticular formation, the abducens and oculomotor nuclei and the median lon

Limosilactobacillus reuteri normalizes gut microbiota dysfunction and social deficits of rat offspring associated with prenatal exposure to stress.

Prenatal stress (PS) is a potential risk factor for social behavior impairment in offspring. Here, we demonstrate that PS induces gut microb … oxytocin receptor antagonist or subdiaphragmatic vagotomy. Thus, both oxytocin signaling and vagal afferent pathways play essential roles in the observed benefits of L. reuteri. Our findings indicate that social behavior impairments in offspring exposed to prenatal maternal stress can be explained by a novel mechanism involving the gut microbiota-brain axis: where …

Role of neural and humoral mediators in the gastrocolonic response.

The phenomenon that meal ingestion immediately increases colonic motility was at first referred to as the "gastrocolic reflex", but later changed to the "gastrocolonic response" because of the possible involvement of humoral mediators. This review aims to describe the various mechanisms providing stimuli that alter the gastrocolonic response by dividing it into 'cephalic', 'gastric', and 'intestinal' phases, just as the process of gastric acid secretion after feeding, for a better understanding of the gastrocolonic response. The gastrocolonic response occurs within minutes after feeding and co

Long-term NAD+ supplementation prevents the progression of age-related hearing loss in mice.

Age-related hearing loss (ARHL) is the most common sensory disability associated with human aging. Yet, there are no approved measures for preventing or treating this debilitating condition. With its slow progression, continuous and safe approaches are critical for ARHL treatment. Nicotinamide Riboside (NR), a NAD+ precursor, is well tolerated even for long-term use and is already shown effective in various disease models including Alzheimer's and Parkinson's disease. It has also been beneficial against noise-induced hearing loss and in hearing loss associated with premature aging. However, it

[Understanding chronic ocular pain].

Dry eye disease (DED) is a common chronic condition with multifactorial etiologies that is increasing in prevalence worldwide, up to 20% in the elderly. The economic burden and impact of DED on vision, quality of life, work productivity, psychological and physical impact of pain, are considerable. Chronic ocular pain is the most common symptom of DED and there is currently no topical ocular analgesic therapy available to treat this debilitating disease. Eye pain can be perceived as itch, irritation, dryness, grittiness, burning, aching, and light sensitivity. Ocular pain is triggered by cornea

Mechanisms Underlying Overactive Bladder and Interstitial Cystitis/Painful Bladder Syndrome.

The bladder is innervated by extrinsic afferents that project into the dorsal horn of the spinal cord, providing sensory input to the micturition centers within the central nervous system. Under normal conditions, the continuous activation of these neurons during bladder distension goes mostly unnoticed. However, for patients with chronic urological disorders such as overactive bladder syndrome (OAB) and interstitial cystitis/painful bladder syndrome (IC/PBS), exaggerated bladder sensation and altered bladder function are common debilitating symptoms. Whilst considered to be separate pathologi

Functional activation of cerebral metabolism in mice with mutated thyroid hormone nuclear receptors.

Neonatal hypothyroidism impairs structural maturation in the brain and results in diminished electrical activities and energy metabolism. We recently found that glucose utilization (CMR(glc)) is markedly depressed throughout the brain in mice with targeted mutations in thyroid hormone receptor alpha1 (TR alpha 1), but not TR beta. Previous studies had shown that CMR(glc) increases linearly with spike frequency in the afferent pathways to synapse-rich regions in neuropil, but not in neuronal cell bodies. To determine whether the decreased CMR(glc) in mutant TR alpha 1(PV/+) mice reflected lesse…

Calcium-dependent activation of phospholipase C by mechanical distension in renin-expressing As4.1 cells.

One of the major physiological regulators for the production and release of renin from the kidney is blood pressure. The juxtaglomerular (JG) cells, located primarily at the afferent arterioles leading to the glomerulus, are thought to be the baroreceptor of the kidney and adjust their ability to secrete renin in an inverse relationship to changes in pressure (mechanical force). The characteristics of JG cells that allow them to sense and respond to changes in mechanical force at the cellular level are not clear. By use of a renin-expressing clonal cell line (As4.1) as a model for JG cells, it

Magnetic stimulation over the cerebellum in patients with ataxia.

We studied 20 patients with ataxia caused by various disorders using magnetic stimulation over the cerebellum. Results were compared with normal values found for 12 normal volunteers. In normal subjects, a magnetic stimulus over the cerebellum reduced the size of responses evoked by magnetic cortical stimulation when it preceded cortical stimulus by 5, 6 and 7 ms. The grand average of the ratios of the areas of conditioned responses at intervals of 5, 6 and 7 ms to those of control responses was designated the average area ratio (5-7 ms). Suppression of motor cortical excitability was reduced

Medullary and mesencephalic pathways and connections of lateral line neurons of the spiny dogfish Squalus acanthias.

The neuronal connections of the electrosensory dorsal and the mechanosensory medial octavolateralis nuclei of the spiny dogfish Squalus acanthias were studied by horseradish peroxidase, autoradiographic and axonal degeneration methods. Efferents from each nucleus, in addition to extensive commissural components, give rise to ipsilateral and contralateral lemnisci that ascend to midbrain levels and terminate among the cells of the lateral mesencephalic nucleus (LMN). Within LMN, electrosensory and mechanosensory neurons distribute dorsolateral and ventromedial in position, respectively. Ascendi

Pyramidal actions in identified radial motornuclei of the cat.

This study aimed to establish the projection from the corticospinal tract (CST) to the motoneurones innervating the deep radial (DR) forelimb muscles. In the anaesthetized cat stimulation of the contralateral pyramid and intracellular recording from identified forelimb motoneurones was used. A train of pyramidal stimuli evoked disynaptic EPSPs in DR motoneurones. The effects were very similar in the different nuclei. Pyramidal IPSPs had a slightly longer latency and occurred in most cases together with disynaptic EPSPs. It is suggested that the inhibitory actions to the distal forelimb are pre
