Short-term magnesium deficiency downregulates telomerase, upregulates neutral sphingomyelinase and induces oxidative DNA damage in cardiovascular tissues: relevance to atherogenesis, cardiovascular diseases and aging.

1) short-term dietary deficiency of magnesium (Mg; 21 days) in rats (MgD) would result in a downregulation of telomerase in cardiac and aortic smooth muscle cells, 2) low levels of Mg(2+) added to drinking water (DW) would either prevent or greatly reduce the downregulation of telomerase in MgD, 3) MgD in rats would cause an upregulation of neutral-sphingomyelinase (N-SMAse) and p53, 4) short-term MgD would result in oxidation of DNA in diverse cardiac muscle and aortic smooth muscle cells as exemplified by measurement of 8-hydroxydeoxyguanosine (8-OH-dG), and 5) cross-talk between telomerase,

The role of Rhox homeobox factors in tumorigenesis.

Emollients and moisturizing creams are used to break the dry skin cycle and to maintain the smoothness of the skin. The term 'moisturizer' is often used synonymously with emollient, but moisturizers often contain humectants in order to hydrate the stratum corneum. Dryness is frequently linked to an impaired barrier function observed, for example, in atopic skin, psoriasis, ichthyosis, and contact dermatitis. Dryness and skin barrier disorders are not a single entity, but are characterized by differences in chemistry and morphology in the epidermis. Large differences also exist between moisturi

Podocyte-specific acid sphingomyelinase overexpression promotes gasdermin D dependent pyroptosis by impairing autophagic flux during obesity.

Recent studies suggest that gasdermin D (GSDMD) pore formation contributes to inflammasome-mediated cytokine release and pyroptosis in podocytes under pathological conditions. However, the molecular mechanisms regulating GSDMD pore formation in these cells remain unclear. Given the established role of the lysosomal acid sphingomyelinase (ASM)-ceramide pathway in obesity-related glomerulopathy (ORG), we investigated whether ASM regulates obesity-induced GSDMD pore formation and pyroptosis in podocytes, thereby influencing the progression of ORG. We found that podocyte-specific Smpd1 (the gene e

Identification and functional characterization of plasma exosomal miR-618 as a novel biomarker in recurrent depressive disorder.

BACKGROUND: Given the substantial clinical and financial burden of Recurrent Depressive Disorder (RDD) and the critical lack of reliable biomarkers, this study investigated plasma exosomal microRNA (miRNA) as candidate biomarkers and therapeutic targets to overcome reliance on subjective diagnosis. The pathogenesis of RDD remains incompletely understood. To this end, we focused on miR-618 and conducted in vivo functional experiments to reveal its molecular mechanism in RDD. METHODS: Differentially expressed miRNAs (DEmiRNAs) in plasma exosomes between RDD patients and healthy controls (HCs) w

Severe Dermatologic Toxicity During Amivantamab-Lazertinib Treatment in Epidermal Growth Factor Receptor (EGFR)-Mutated Non-Small Cell Lung Cancer.

Amivantamab is an epidermal growth factor receptor (EGFR) and mesenchymal-epithelial transition (MET) antibody. Lazertinib is an EGFR tyrosine kinase inhibitor with central nervous system penetration. Used in combination, this targeted therapy is effective in treating non-small cell lung cancer (NSCLC). However, it has been associated with severe dermatologic adverse reactions. A 61-year-old woman with a history of EGFR-mutated NSCLC with brain metastases on amivantamab-lazertinib therapy presented for follow-up with drainage from her scalp incision after undergoing a right parietal craniotomy

Attenuation of colitis-related metabolic dysregulation via consumption of wholemeal sourdough bread but not wholemeal bread: a molecular discovery.

Epidemiological studies reveal that the majority of adult patients with inflammatory bowel disease (IBD) are obese and/or overweight. Whole wheat sourdough produced by lactic acid bacterial (LAB) fermentation can provide numerous benefits. In this study, the potential therapeutic effects of consuming whole wheat sourdough bread (singly fermented by two high-metabolite-producing LAB strains, Lactobacillus crustorum (LC) or Lactobacillus fermentum (LF)) on colon and liver metabolic function in an obese (high-fat diet) and ulcerative colitis (5% dextran sulfate sodium, DSS) induced male rat model

Regulation of cell cycle by the novel GATA1/TAL1/Sphingomyelin Synthase 1 ( SGMS1 ) transcriptional axis. Implications for anti-leukemic strategies.

UNLABELLED: Sphingomyelin Synthase 1 (gene name: SGMS1 ) participates in regulation of sphingolipid levels by synthesizing sphingomyelin from ceramide and phosphatidylcholine. Evidence have supported SGMS1's functions in regulating proliferation, cell cycle, cell death and migration. While its functions have begun to be explored, very little is known about upstream regulators. Here, we demonstrate that SGMS1 is a direct gene target of the GATA1-TAL1 transcriptional complex in K562 erythroleukemic cells. A predicted GATA1 consensus DNA binding sequence was identified with in a newly characteriz

Investigating the Pathological Relevance of N-acylsphingosine Amidohydrolase 2 (ASAH2) and Related Proteins in Alzheimer's Disease.

Background Alzheimer's disease (AD) is a progressive neurodegenerative disorder mainly characterized by progressive cognitive decline and memory loss. Identifying candidate biomarkers before the clinical onset of AD is crucial for enabling earlier diagnosis and timely therapeutic intervention. Among different molecular targets, N-acylsphingosine amidohydrolase 2 (ASAH2), a key enzyme in ceramide metabolism, has been linked to many neurodegenerative diseases, including AD. This study investigates ASAH2 expression in human serum and AD mouse models to explore its potential as an early biomarker

A specific serum lipid signature characterizes patients with glycogen storage disease type Ia.

Glycogen storage disease type Ia (GSDIa) is a rare, inherited glucose-6-phosphatase-α (G6Pase-α) deficiency-induced carbohydrate metabolism disorder. Although hyperlipidemia is a hallmark of GSDI, the extent of lipid metabolism disruption remains incompletely understood. Lipidomic analysis was performed to characterize the serum lipidome in patients with GSDIa, by including age- and sex-matched healthy controls and age-matched hypercholesterolemic controls. Metabolic control and dietary information biochemical markers were obtained from patients with GSDIa. Patients with GSDIa showed higher to

Coordinated Regulation of Membrane Homeostasis and Drug Accumulation by Novel Kinase STK-17 in Response to Antifungal Azole Treatment.

The emergence of antifungal resistance, especially to the most widely used azole class of ergosterol biosynthesis inhibitors, makes fungal infections difficult to treat in clinics and agriculture. When exposed to azoles, fungi can make adaptive responses to alleviate azole toxicity and produce azole tolerance. However, except for azole efflux pumps and ergosterol biosynthesis genes, the role of most azole responsive genes in azole resistance is unknown. In this study, STK-17, whose transcription is upregulated by azoles, was characterized as a novel kinase that is required for azole resistance

A Drosophila Model of Neuronopathic Gaucher Disease Demonstrates Lysosomal-Autophagic Defects and Altered mTOR Signalling and Is Functionally Rescued by Rapamycin.

Glucocerebrosidase (GBA1) mutations are associated with Gaucher disease (GD), an autosomal recessive disorder caused by functional deficiency of glucocerebrosidase (GBA), a lysosomal enzyme that hydrolyzes glucosylceramide to ceramide and glucose. Neuronopathic forms of GD can be associated with rapid neurological decline (Type II) or manifest as a chronic form (Type III) with a wide spectrum of neurological signs. Furthermore, there is now a well-established link between GBA1 mutations and Parkinson's disease (PD), with heterozygote mutations in GBA1 considered the commonest genetic defect in

Structural requirements of anti-GD1a antibodies determine their target specificity.

The acute motor axonal neuropathy (AMAN) variant of Guillain-Barré syndrome (GBS) is associated with anti-GD1a and anti-GM1 IgG antibodies. The basis of preferential motor nerve injury in this disease is not clear, however, because biochemical studies demonstrate that sensory and motor nerves express similar quantities of GD1a and GM1 gangliosides. To elucidate the pathophysiology of AMAN, we have developed several monoclonal antibodies (mAbs) with GD1a reactivity and reported that one mAb, GD1a-1, preferentially stained motor axons in human and rodent nerves. To understand the basis of this p

Imiglucerase low-dose therapy for paediatric Gaucher disease--a long-term cohort study.

BACKGROUND: Gaucher disease is the most common lysosomal storage disorder caused by the insufficiency of the lysosomal enzyme, glucocerebrosidase. This deficiency results in absent or inefficient conversion of glucocerebroside (a membrane lipid) to ceramide and glucose. Accumulation of glucocerebroside occurs primarily in macrophage lysosomes (i.e. monocytes and macrophages) during phagocytic degradation of red blood cells. Clinical symptoms arise due to the displacement of normal cells by lipid-engorged Gaucher cells. Enzyme replacement therapy (ERT) targets the macrophage system and has been

Glycosphingolipids and insulin resistance.

Glycosphingolipids are structural membrane components, residing largely in the plasma membrane with their sugar-moieties exposed at the cell's surface. In recent times a crucial role for glycosphingolipids in insulin resistance has been proposed. A chronic state of insulin resistance is a rapidly increasing disease condition in Western and developing countries. It is considered to be the major underlying cause of the metabolic syndrome, a combination of metabolic abnormalities that increases the risk for an individual to develop Type 2 diabetes, obesity, cardiovascular disease, polycystic ovar

Integrated gut microbiota and metabolome signatures revealed by deep metagenomic sequencing in post-stroke cognitive impairment with type 2 diabetes.

UNLABELLED: Post-stroke cognitive impairment (PSCI) is significantly exacerbated in individuals with type 2 diabetes mellitus (T2DM), yet the underlying gut microbial and metabolic mechanisms remain unclear. In this study, baseline fecal samples from 28 diabetic PSCI (PSCI-DM) patients and 29 matched non-PSCI non-diabetic controls were subjected to deep metagenomic sequencing and untargeted metabolomics. Although alpha diversity was preserved, subtle but meaningful shifts were observed in bacterial and fungal composition. The PSCI-DM group exhibited depletion of beneficial butyrate-producing t
