Current insights and prospects for the pathogenesis and treatment of clinical manifestations associated with Down syndrome through neurotransmitter, inflammatory, and oxidative stress pathways.

Individuals with Down syndrome exhibit various changes in the human body systems, including alterations in the ocular, neurological, and dermatological systems. Especially, preclinical and clinical studies have determined Down syndrome patients to possess reduced intellectual and cognition abilities, which neurobehavioral effects are associated with altered molecular markers in the brain. For instance, neuroinflammation and increased brain oxidative stress are reported in animals models of Down syndrome, and the reversal of those markers lead to positive effects. Dopaminergic and serotonergic

Interneuron Loss and Microglia Activation by Transcriptome Analyses in the Basal Ganglia of Tourette Disorder.

BACKGROUND: Tourette disorder (TS) is characterized by motor hyperactivity and tics that are believed to originate in the basal ganglia. Postmortem immunocytochemical analyses has revealed decreases in cholinergic (CH), as well as parvalbumin and somatostatin GABA (gamma-aminobutyric acid) interneurons (INs) within the caudate/putamen of individuals with TS. METHODS: We obtained transcriptome and open chromatin datasets by single-nucleus RNA sequencing and single-nucleus ATAC sequencing, respectively, from caudate/putamen postmortem specimens of 6 adults with TS and 6 matched normal control s

Rivastigmine ameliorates botulinum-induced hippocampal damage and spatial memory impairment in male rats.

Botulinum toxin (Botox) is widely used in beauty industry and its long-term consequences can be a matter of concern. The hippocampal cholinergic system plays a significant role in memory and learning that could be affected by Botulinum toxin. However, to date, the effect of Botox on memory system has been controversial. This survey aimed to examine the effects of Botox on spatial memory, and biochemical and histological parameters of the hippocampus in male rats by using Rivastigmine (R) as a cholinesterase inhibitor that is more selective for the central nervous system (CNS). Thirty-five male

A Potential Role of Cholinergic Dysfunction on Impaired Colon Motility in Experimental Intestinal Chagas Disease.

BACKGROUND/AIMS: Chagasic megacolon is caused by Trypanosoma cruzi, which promotes in several cases, irreversible segmental colonic dilation. This alteration is the major anatomic-clinical disorder, characterized by the enteric nervous system and muscle wall structural damage. Herein, we investigate how T. cruzi -induced progressive colonic structural changes modulate the colonic contractile pattern activity. METHODS: We developed a murine model of T. cruzi-infection that reproduced long-term modifications of the enlarged colon. We evaluated colonic and total intestinal transit time in animal

Antipsychotics: Mechanisms underlying clinical response and side-effects and novel treatment approaches based on pathophysiology.

Antipsychotic drugs are central to the treatment of schizophrenia and other psychotic disorders but are ineffective for some patients and associated with side-effects and nonadherence in others. We review the in vitro, pre-clinical, clinical and molecular imaging evidence on the mode of action of antipsychotics and their side-effects. This identifies the key role of striatal dopamine D2 receptor blockade for clinical response, but also for endocrine and motor side-effects, indicating a therapeutic window for D2 blockade. We consider how partial D2/3 receptor agonists fit within this framework,

The splanchnic anti-inflammatory pathway: could it be the efferent arm of the inflammatory reflex?

What is the topic of this review? We review the current literature on the neural reflex termed the 'inflammatory reflex' that inhibits an excessive release of inflammatory mediators in response to an immune challenge. What advances does it highlight? The original model proposed that the inflammatory reflex is a vago-vagal reflex that controls immune function. We posit that, in the endotoxaemic animal model, the vagus nerves do not appear to play a role. The evidence suggests that the efferent motor pathway, termed here the 'splanchnic anti-inflammatory pathway', is purely sympathetic, travelli

Maternal choline supplementation in a mouse model of Down syndrome: Effects on attention and nucleus basalis/substantia innominata neuron morphology in adult offspring.

The Ts65Dn mouse model of Down syndrome (DS) and Alzheimer's disease (AD) exhibits cognitive impairment and degeneration of basal forebrain cholinergic neurons (BFCNs). Our prior studies demonstrated that maternal choline supplementation (MCS) improves attention and spatial cognition in Ts65Dn offspring, normalizes hippocampal neurogenesis, and lessens BFCN degeneration in the medial septal nucleus (MSN). Here we determined whether (i) BFCN degeneration contributes to attentional dysfunction, and (ii) whether the attentional benefits of perinatal MCS are due to changes in BFCN morphology. Ts65…

The hypothalamic peptidergic system, hypocretin/orexin and vigilance control.

Using forward and reverse genetics, the genes (hypocretin/orexin ligand and its receptor) involved in the pathogenesis of the sleep disorder, narcolepsy, in animals, have been identified. Mutations in hypocretin related-genes are extremely rare in humans, but hypocretin-ligand deficiency is found in most narcolepsy-cataplexy cases. Hypocretin deficiency in humans can be clinically detected by CSF hypocretin-1 measures, and undetectably low CSF hypocretin-1 is now included in the revised international diagnostic criteria of narcolepsy. Since hypocretin-ligand deficiency is the major pathophysio

Survival and mitogenesis of neuroepithelial cells are influenced by noradrenergic but not cholinergic innervation in cultured embryonic rat neopallium.

alpha-Adrenoreceptors are present in the ventricular (VZ) and subventricular (SVZ) zones of the mammalian embryonic forebrain, and contribute towards cell cycle controls in the germinal neuroepithelium [Pabbathi et al., Brain Res. 760 (1997) 22-33.]. Since noradrenaline-containing fibres from the locus coeruleus (LC) and other brainstem nuclei innervate many parts of the CNS from an early stage of embryogenesis, we have used heterochronic cocultures to investigate whether noradrenergic innervation may be the source of trophic support. Dorsal neopallium from E13 rat embryo was cultured in proxi

Effects of extracellular choline concentration and K+ depolarization on choline kinase and choline acetyltransferase activities in superior cervical sympathetic ganglia excised from rats.

The activities of choline kinase (CK) and choline acetyltransferase (ChAT) were examined in vitro in superior cervical sympathetic ganglia (SCG) excised from rats following aerobic incubation for 1 h in a medium containing various choline concentrations, with and without application of a high KCl level (70 mM). Ganglionic CK activity was strongly inhibited (by approximately 75%) at low extracellular choline concentrations (1-5 microM) but rose as the choline concentration was raised to 10-50 microM in the incubation medium, then fell and rose again with further increases in choline concentrati

Evidence that growth hormone-releasing factor stimulates somatostatin release in vitro via beta-endorphin.

Previous results indicate that GH-releasing factor (GRF) induces a dose-related stimulation of somatostatin (SRIF) release from median eminence fragments incubated in vitro. In the present investigation we examined whether this action was mediated by other neurotransmitters or neuromodulators. Studies using receptor blockers for dopamine (pimozide), alpha-adrenergic receptors (phentolamine), and muscarinic cholinergic receptors (atropine) revealed that these receptor blockers, at a dose of 10(-6) M, which was capable of blocking the response to the relevant transmitter in previous studies, had

Chitosan-coated cyclodextrin-based niosomes enable enhanced nose-to-brain delivery of rivastigmine and asiaticoside for Alzheimer's disease.

The co-delivery of rivastigmine hydrogen tartrate (RHT) and asiaticoside (AS) offers a promising strategy for enhancing acetylcholinesterase (AChE) inhibition in Alzheimer's disease (AD) by targeting both cholinergic deficits and neurodegeneration. However, formulating a combined delivery system is challenging due to the hydrophilic nature of RHT and poor solubility of AS. Accordingly, we developed a scalable intranasal delivery system using cyclodextrin-enabled niosomes. Inclusion complexes of RHT, AS, and 2-hydroxypropyl-β-cyclodextrin were prepared and encapsulated into niosomes via thin-fi

Small-molecule PCSK9 inhibition enhances BBB amyloid-β clearance and suppresses microglial inflammation in Alzheimer's disease models.

Impaired amyloid-β (Aβ) clearance and chronic neuroinflammation are central drivers of Alzheimer's disease (AD) progression, yet therapeutic strategies targeting these processes remain limited. Proprotein convertase subtilisin/kexin type 9 (PCSK9) negatively regulates low-density lipoprotein receptor family members, including low-density lipoprotein receptor -related protein 1 (LRP1), a key mediator of blood-brain barrier (BBB) Aβ efflux, and has emerging roles in inflammatory signaling. In this study, to define the therapeutic relevance of PCSK9 inhibition in AD-like pathology, we examined SB

Immuno-neural mechanisms in gastrointestinal tumorigenesis: bridging inflammation, neural regulation, and therapeutic innovation.

Gastrointestinal (GI) tumors remain a leading cause of global cancer mortality, with late-stage diagnosis and metastatic dissemination posing major clinical challenges. This review synthesizes current understanding of the intricate interplay between immune regulation, neural signaling, and tumor microenvironment dynamics in GI malignancies. We highlight how chronic inflammation, driven by pathogens like H. pylori or inflammatory bowel disease, establishes a pro-tumorigenic milieu through cytokine networks (IL-1β, TNF-α, IL-6) and Wnt/β-catenin signaling, while neural components (serotonergic,

Identification of Potential Multitarget Directed Ligands for Alzheimer's Disease by Coupling Virtual Screening and Experimental Validation.

Alzheimer's disease (AD) is a neurodegenerative disorder (NDD) associated with the accumulation of beta-amyloid plaques (βA), oxidative stress, and a decrease in cholinergic activity among other pathologies. Given the limitations of current treatments, multitarget strategies present a promising alternative. In this study we prioritized six AD-related protein targets: acetylcholinesterase (AChE), beta-secretase 1 (BACE-1), cannabinoid receptor type 2 (CB2), glycogen synthase kinase 3 beta (GSK-3β), monoamine oxidase A (MAO-A), and the neuronal acetylcholine receptor subunit alpha-7 (nAChR7). Multi-target Triazole-Benzopyrone hybrids modulating cholinergic dysfunction, oxidative stress, and neuroinflammation through GFAP/NF-κB/APOE/NLRP3 axis in Alzheimer's disease.

Alzheimer's disease (AD) is a degenerative neurological disorder characterized by a deterioration in cognitive abilities, especially memory and learning. The main aim of this study is to evaluate the effects of our agents on oxidative stress, neuroinflammation, cognitive function, and behavioral performance in an LPS-induced AD animal model and comprehensive in vitro and in vivo assays. The synthesized compounds, namely 4b, 5b, 6, 8a-d, and 11a-c, revealed acetylcholinesterase inhibitory activity (3.50-5.91 nM) superior to that of the reference drug donepezil (6.33 nM). In silico docking and in vivo assessment of Mondia whitei fruit extract in attenuating CdCl2-Induced neurodegeneration in rat.

Cadmium is a toxic heavy metal found in the atmosphere. Its frequent exposure can lead to neurodegeneration. Medicinal plants are scavengers of free radicals that could result from exposure to heavy metals. In this study, the neuroprotective effect of the aqueous extract of Mondia whitei fruit (AEMW) against cadmium chloride (CdCl2)-induced neurotoxicity was investigated using an in silico and in vivo approach. The biological constituents in the AEMW were identified using high-performance liquid chromatography, and the compounds were docked against a target protein related to neurodegeneration

Therapeutic potential of apigenin in neurodegenerative diseases and brain health.

Neurodegenerative disorders are characterized by progressive neuronal loss and remain a major global health concern, largely due to their complex and multifactorial nature. These conditions, including Alzheimer's disease, Parkinson's disease, and Huntington's disease, involve overlapping mechanisms such as oxidative stress, chronic inflammation, mitochondrial dysfunction, and impaired neurotransmission. In recent years, natural bioactive compounds have gained attention as potential therapeutic candidates. Among them, apigenin, a dietary flavonoid widely found in fruits and vegetables, exerts n
