Stem Cell and Cell-Free Strategies for Osteoarthritis: Toward Durable Regenerative Therapies.

Osteoarthritis (OA) is the most prevalent degenerative joint disease and a leading cause of pain and disability, yet current treatments remain largely palliative and fail to alter disease progression. Stem cell-based therapies offer a promising alternative, with mesenchymal stromal/stem cells (MSCs) representing the most clinically advanced approach. Preclinical and clinical studies demonstrate that MSCs exert anti-inflammatory, immunomodulatory, chondroprotective, and analgesic effects primarily through paracrine signaling rather than durable engraftment. Clinical trials consistently confirm

MGRN1 is linked to recessive heart and laterality defects: the first genotype-phenotype report in humans.

Recurrent idiopathic severe fetal structural anomalies present major challenges for reproductive decision-making and genetic counselling. A non-consanguineous healthy Estonian couple had experienced two electively terminated pregnancies at 12-13 weeks' gestation due to unexplained major fetal malformations and one early miscarriage. Their three pregnancies had resulted in unaffected newborns. Exome sequencing of fetal tissues from both terminated pregnancies identified a homozygous rare missense variant in MGRN1 (mahogunin ring finger 1), c.881G>A, p.(Arg294His) (Genome Aggregation Database V.

MFN2-mediated remodelling of ER-mitochondria communication alleviates ochratoxin A-induced hepatocyte ferroptosis by enhancing mitophagy.

Mycotoxins are ubiquitous environmental contaminants that widely occur throughout ecosystems and the food chain, posing persistent threats to environmental safety and public health. Ochratoxin A (OTA) is a widespread and chemically stable mycotoxin with recognized hepatotoxicity, yet the mechanisms linking OTA exposure to liver injury remain poorly defined. This study investigated the role of mitofusin 2 (MFN2)-mediated ER-mitochondria communication in OTA-induced hepatotoxicity, with particular emphasis on its regulation of mitophagy and ferroptosis. An OTA-exposed chicken model and primary c

Prenatal NO₂ exposure and cardiac developmental dysfunction: CD4⁺ T cell imbalance drives cardiac fibrosis through placenta-heart axis immune transmission.

Epidemiological studies show maternal nitrogen dioxide (NO2) exposure associates with congenital heart disease (CHD) risk, but subtypes and mechanisms unclear. Here, we first conducted a systematic review and meta-analysis, which confirmed that prenatal NO₂ exposure significantly increased the risk of coarctation of the aorta (COA) (OR = 1.15, 95% CI: 1.04-1.26). Using a mouse model of prenatal NO₂ exposure, we demonstrated that prenatal NO2 exposure adversely affected fetal cardiac structure and function at embryonic day 18.5, characterized by ventricular dilation, myocardial wall thinning, s

Herpesviruses hijack ISGylation to promote viral capsid assembly.

Herpesviruses are common pathogens of the oral cavity, yet how they interact with other oral microbes are poorly understood. Using murine gamma-herpesvirus 68 (MHV68) as a model for human gamma-herpesviruses, we find that ISGylation is hijacked to facilitate viral capsid assembly and lytic replication. Coinfection with the oral Aggregatibacter actinomycetemcomitans (A.actinomycetemcomitans) and MHV68 synergistically induced interferon-stimulated gene 15 (ISG15) expression and global ISGylation. Proteomic profiling revealed viral structural proteins as the dominant targets of ISGylation in MHV6

Tmem216 deficiency induces hydrocephalus via impaired ciliogenesis and disrupted ependymal planar polarity in mice.

Defects in multiciliated ependymal cells (ECs), which line the cerebral ventricular walls and generate unidirectional cerebrospinal fluid (CSF) flow through coordinated ciliary beating, can lead to CSF accumulation and hydrocephalus. TMEM216, a ciliopathy-associated gene mutated in Joubert, Meckel, and related syndromes, encodes an essential component of the protein complexes that form the transition zone (TZ) at the base of primary cilia. Here, we demonstrate that central nervous system-specific inactivation of Tmem216 results in severe postnatal hydrocephalus. TMEM216 localizes to the TZ of

Effects of the ecpA gene on the biological characteristics and pathogenicity of avian pathogenic Escherichia coli strain FJLY68.

Avian pathogenic Escherichia coli (APEC) is a major cause of colibacillosis in poultry, yet the role of the ecpA gene, which encodes the major structural subunit of the Escherichia coli common pilus (ECP), remains incompletely defined in APEC pathogenesis. To investigate the role of ecpA in the biological characteristics and pathogenicity of Avian Pathogenic Escherichia coli (APEC) strain FJLY68, an ecpA deletion mutant (ΔecpA) and its corresponding complemented strain (CΔecpA) were constructed using the CRISPR/Cas9 system and verified by PCR and Sanger sequencing. Phenotypic analyses revealed

Clinical Spectrum of Nodoventricular Accessory Pathways: An Asian Collaborative Study.

BACKGROUND: Nodoventricular accessory pathways (NV-APs) are rare embryonic remnants connecting the atrioventricular node to the ventricle. We describe their electrophysiological characteristics and ablation outcomes across a multicenter Asian cohort, highlighting a developmental link to Ebstein anomaly. CASE SERIES: Five patients presented with supraventricular tachycardia due to NV-APs (4 concealed, 1 manifest). Programmed stimulation confirmed ventricular participation and excluded the atrium from the circuit. Two patients had Ebstein anomaly, clinically extending histological data regardin

Laminin 332 regulates glial fibrillary acidic protein expression and astrocyte maturation during spinal cord development.

The white matter of the spinal cord is essential for sensory and motor signaling, and its proper development is crucial for establishing functional neuronal circuits. However, the mechanisms underlying white matter formation remain incompletely understood. We hypothesized that the extracellular matrix, particularly laminins, plays a key role in this process. The spatiotemporal expression patterns of laminins during spinal cord white matter development have not been fully characterized. Here, we examined the distribution and function of laminins during spinal cord development. Laminin 332 local

Fibroblast heterogeneity and its role in generating protective immunity in the secondary lymphoid organs.

Fibroblasts are cells of mesenchymal origin with a range of phenotypic diversity and heterogeneity. One of the major functions of fibroblasts is the formation and turnover of the extracellular matrix and establishing a tissue structure by forming a matrisome from embryonic development to the adult stage. It plays an indispensable role in extracellular matrix remodeling during injury, repair, and infection, providing a scaffold for cell-to-cell interaction. Despite their important pathophysiological roles, molecular markers for tissue-resident fibroblasts are only now being identified. Fibrobla

[Transcription Factor PAX4: Role in Differentiation of Insulin- Producing β Cells during Pancreas Development and Association with Diabetes].

PAX4 (Paired Box 4) is a transcription factor that is expressed mainly in the pancreas and plays a key role in the development of insulin-producing β cells at the embryonic stage. In mature cells, PAX4 acts as a main regulator of their adaptation under pathological conditions. The importance of PAX4 to the proper function of pancreatic islets has been demonstrated in studies of the relationship between mutations of the PAX4 gene and various forms of diabetes mellitus (DM). PAX4 overexpression in adult islets stimulates β-cell proliferation and resistance to apoptosis. Taken together, the data

Iron Deficiency Impairs Mitochondrial Energetics and Early Axonal Growth and Branching in Developing Hippocampal Neurons.

Each stage of neuronal development (i.e., proliferation, differentiation, migration, neurite outgrowth and synapse formation) requires functional and highly coordinated metabolic activity to ultimately ensure proper sculpting of complex neural networks. Energy deficits underlie many neurodevelopmental, neuropsychiatric and neurodegenerative diseases implicating mitochondria as a potential therapeutic target. Iron is necessary for neuronal energy output through its direct role in mitochondrial oxidative phosphorylation. Iron deficiency (ID) reduces mitochondrial respiratory and energy capacity

Hemodynamic homeostasis disequilibrium in critical illness.

Over millions of years, the circulatory system evolved from primitive forms into a highly specialized network capable of overcoming time-distance constraints and enhancing diffusion efficiency. This structural advancement laid the physiological foundation for the regulation of hemodynamics and systemic homeostasis. Hemodynamic homeostasis is a fundamental biological process that ensures the continuous delivery of oxygen and substrates while facilitating the removal of carbon dioxide and metabolic waste. Such balance is essential for sustaining cellular metabolism and maintaining the function o

The Role of Prenatal Microglial Activation and Its Sex Differences in the Development of Neuropsychiatric Disorders and Neurodegenerative Diseases.

Maternal Immune Activation (MIA) is a phenomenon of pathophysiological stimulation of the maternal immune system during gestation which potentially leads to functional and structural disturbances of fetal neurogenesis. It occurs due to the alteration of paracrine signals between the maternal organism and the developing nervous system of the fetus. Any disturbances in the brain at embryonic and early postnatal stages might compromise its natural developmental trajectory, which could potentially increase the risk of developing neuropsychiatric disorders, such as schizophrenia, autistic spectrum

Splicing and frameshift variants in QSER1 may be involved in developmental phenotypes.

Human development is a complex process that requires precise control of gene expression through regulatory proteins. Recently, heterozygous variants in PRR12, encoding a proline-rich regulatory protein, were found to cause a variable phenotype involving developmental delay/cognitive impairment, neuropsychiatric diagnoses, structural eye anomalies, congenital heart and kidney defects, and poor growth. QSER1, encoding glutamine- and serine-rich protein 1, represents a paralog of PRR12 that shares 28% overall identity at the protein level and stronger conservation (43%) in the C-terminal region.

How is rosette formation in brain tumours linked with cerebrospinal fluid spread?

Rosette formation, a characteristic histopathological feature of various paediatric brain tumours, appears to be linked with cerebrospinal fluid (CSF) dissemination. Tumours like medulloblastoma, ependymoma, retinoblastoma, pineal region, and embryonal tumours, known for their rosette formations, also exhibit a propensity for CSF spread, which can manifest as drop metastases and leptomeningeal involvement. CSF dissemination is detected early in the disease course and contributes to significant diagnostic and management challenges. The structure of rosettes, consisting of tumour cells arranged
