Distribution of surfactant and ventilation in surfactant-treated preterm lambs.

Surfactant-deficient ventilated preterm lambs were treated with 100 mg/kg of surfactant radiolabeled with microspheres at 30 min and 2.5 h of age to evaluate the effect of treatment technique on surfactant distribution. The treatments were four positions with four boluses (bolus 4), two lateral positions with two boluses (bolus 2), or a 30-min infusion (infusion). The bolus groups had uniform surfactant distributions to the > 100 pieces analyzed for each lung. Infusion resulted in a very nonuniform surfactant distribution (P < 0.01). Surfactant was recovered equivalently in all lobes of the bo

Effect of ALA on preventing diabetic peripheral neuropathy in rats through mitochondrial axonal transport.

PURPOSE: To explore the mechanism by which alpha-lipoic acid (ALA) regulates mitochondrial axonal transport to protect the sciatic nerve in diabetic rats. METHODS: Among 55 healthy male Sprague‒Dawley rats, 40 were randomly selected, fed a high-carbohydrate/high-fat diet and intraperitoneally injected with streptozotocin (STZ) to induce diabetes. Diabetic rats were randomly divided into diabetic peripheral neuropathy (DPN) and alpha lipoic acid (ALA) groups, with 15 rats in each group, excluding the rats in which diabetes failed to be induced and the dead rats. The other 15 rats were used as

Long-chain fatty acid utilization in Eisenmenger syndrome.

BACKGROUND: Morbidity and mortality in idiopathic pulmonary arterial hypertension (iPAH) are linked to right ventricular failure (RVF), a condition characterized by a metabolic shift from fatty acid oxidation (FAO) to glycolysis. Eisenmenger Syndrome (ES), a unique form of PAH, is associated with a mortality paradox, yet the mechanism for better survival in this condition remains poorly understood. OBJECTIVES: We postulated that beneficial changes in long chain fatty acid (LCFA) utilization may support right ventricular (RV) adaptations in ES, and therefore ssupport favorable RV function and

Iron status influences non-alcoholic fatty liver disease in obesity through the gut microbiome.

BACKGROUND: The gut microbiome and iron status are known to play a role in the pathophysiology of non-alcoholic fatty liver disease (NAFLD), although their complex interaction remains unclear. RESULTS: Here, we applied an integrative systems medicine approach (faecal metagenomics, plasma and urine metabolomics, hepatic transcriptomics) in 2 well-characterised human cohorts of subjects with obesity (discovery n = 49 and validation n = 628) and an independent cohort formed by both individuals with and without obesity (n = 130), combined with in vitro and animal models. Serum ferritin levels, as

Short-Chain Fatty Acids Outpace Ketone Oxidation in the Failing Heart.

BACKGROUND: The failing heart is energy starved with impaired oxidation of long-chain fatty acids (LCFAs) at the level of reduced CPT1 (carnitine palmitoyltransferase 1) activity at the outer mitochondrial membrane. Recent work shows elevated ketone oxidation in failing hearts as an alternate carbon source for oxidative ATP generation. We hypothesized that another short-chain carbon source, short-chain fatty acids (SCFAs) that bypass carnitine palmitoyltransferase 1, could similarly support energy production in failing hearts. METHODS: Cardiac hypertrophy and dysfunction were induced in rats

Mendelian Randomization Revealed Potential of mTOR Inhibitors for Treatment of Osteoporosis: Evidence From GWAS and Transcriptome Data.

AIMS: Numerous preclinical studies suggested that targeted inhibition of mammalian target of rapamycin (mTOR) may be beneficial for the treatment of osteoporosis. However, the relevance of these findings to human populations remains unclear. We hypothesized that lifelong genetically higher mTOR expression leads to reduced bone mineral density (BMD), and we use Mendelian randomization to estimate its causal effect. BACKGROUND: We aimed to utilize existing genome-wide association studies (GWAS) and expression quantitative trait locus (eQTL) data to comprehensively evaluate whether genetically p

A Glucuronic Acid-Palmitoylethanolamide Conjugate (GLUPEA) Is an Innovative Drug Delivery System and a Potential Bioregulator.

Palmitoylethanolamide (PEA) is an endogenous anti-inflammatory lipid mediator and a widely used nutraceutical. In this study, we designed, realized, and tested a drug-carrier conjugate between PEA (the active drug) and glucuronic acid (the carrier). The conjugate, named GLUPEA, was characterized for its capability of increasing PEA levels and exerting anti-inflammatory activity both in vitro and in vivo. GLUPEA treatment, compared to the same concentration of PEA, resulted in higher cellular amounts of PEA and the endocannabinoid 2-arachidonoyl glycerol (2-AG), and increased 2-AG-induced trans

A defined multi-metabolite formulation derived from Salvia miltiorrhiza alleviates obesity-associated cardiac lipotoxicity via modulating mitochondrial dynamics.

BACKGROUND: Obesity is a major risk factor for lipotoxic cardiomyopathy, a process involving lipid metabolism disorders, inflammatory responses, and mitochondrial dysfunction. Salvia miltiorrhiza (Danshen), a traditional Chinese medicinal plant, contains water-soluble metabolites with multitarget regulatory potential. Although some monomers of S. miltiorrhiza have been proved to be effective in cardiovascular diseases, the combined effects of metabolites and the mechanism remains unclear. This study aimed to investigate the cardioprotective effects and potential mechanisms of SABP, a defined f

Integrated physiological responses of Macrobrachium rosenbergii to NaHCO₃ stress: growth inhibition, energy metabolism activation, oxidative injury, and autophagic response.

BACKGROUND: Saline-alkaline stress, particularly sodium bicarbonate-induced alkaline stress, can cause tissue damage and oxidative stress in aquatic animals, thereby inhibiting their growth and even threatening their survival. This has become a key factor limiting the utilization of saline-alkaline water resources. This study for the first time systematically established acute and chronic time-series gradient NaHCO₃ stress treatments, and integrated histological observation, ultrastructural examination, enzyme activity determination as well as transcriptomic and metabolomic profiling to reveal

UHPLC-Q-TOF-MS/MS profiling of resin glycosides and their lipase inhibitory activity in leaves of selected sweet potato (Ipomoea batatas L.) cultivars.

INTRODUCTION: Resin glycosides (RGs) found in the aerial parts of sweet potato are health promoting agents that can be reclaimed from the by-products of sweet potato plantation as agriceuticals for combating obesity. However, RGs have complex structural variations across various sweet potato cultivars, along with an unclear structure-activity relationship. To address this gap, nine types of sweet potato leaves were evaluated, including "Shangshu 19", "Xuzi 20-1", "Xushu 24", "Xushu 32", "Yanshu 25", "Blackheart", "Blackie", and two "Beniazuma" samples. METHODS: RG-rich extracts were obtained

Bio-distribution and deposition of wildfire smoke chemicals into olfactory bulb and brain of rats after intranasal instillation.

Epidemiological and experimental studies suggest wildfire smoke is a potential contributor to neurological dysfunction and associated with neuroinflammation. Using doses comparable to those encountered during intense wildfire events (200-300 μg/m3), we explore the absorption, distribution, metabolism, and elimination (ADME) and pharmacokinetics of representative members of major chemical classes (acid, phenol, PAH, aldehyde) in inhaled wood smoke condensates. Male Sprague Dawley rats were intranasally instilled with smoldering eucalyptus woodsmoke extract (WSE) reconstituted in saline spiked w

Hinokitiol ameliorates MASH in mice by therapeutic targeting of hepatic Nrf2 and inhibiting hepatocyte ferroptosis.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH), an advanced stage of metabolic dysfunction-associated steatotic liver disease (MASLD), still lacks approved effective clinical drugs. Ferroptosis, a form of regulated cell death driven by excessive iron accumulation and uncontrollable lipid peroxidation, has been proven to be a trigger of inflammation and initiation of steatohepatitis. The pathogenic interplay is modulated by oxidative stress, while the Nrf2-mediated antioxidant response plays a regulatory role in ferroptosis. Phytochemical hinokitiol (Hino) has demonstrated

Enhanced ECCD36 signaling promotes skeletal muscle insulin resistance in female mice.

Consumption of a Western diet (WD) increases CD36 expression in vascular, hepatic, and skeletal muscle tissues promoting lipid metabolic disorders and insulin resistance. We further examined the role of endothelial cell-specific CD36 (ECCD36) signaling in contributing to skeletal muscle lipid metabolic disorders, insulin resistance, and their underlying molecular mechanisms. Female ECCD36 wild-type (ECCD36+/+) and knock-out (ECCD36-/-) mice, aged 6 wk, were provided with either a WD or a standard chow diet for a duration of 16 wk. ECCD36+/+ WD mice were characterized by elevated fasting plasma
