Insulin and IGF1 receptors are essential for the development and steroidogenic function of adult Leydig cells.

The insulin family of growth factors (insulin, IGF1, and IGF2) are critical in sex determination, adrenal differentiation, and testicular function. Notably, the IGF system has been reported to mediate the proliferation of steroidogenic cells. However, the precise role and contribution of the membrane receptors mediating those effects, namely, insulin receptor (INSR) and type-I insulin-like growth factor receptor (IGF1R), have not, to our knowledge, been investigated. We show here that specific deletion of both Insr and Igf1r in steroidogenic cells in mice leads to severe alterations of adrenoc

To ICSI or Not to ICSI.

Intracytoplasmic sperm injection (ICSI) is the most effective assisted reproductive procedure enabling fertilization in severe forms of male factor indications and male gamete dysfunction. Reliability of ICSI has allowed the expansion of its application to other forms of infertility rendering it the most popular assisted reproduction technology (ART) insemination method worldwide. The concern related to the invasiveness of ICSI together with the arbitrary selection of the inseminating spermatozoon has induced the execution of studies to compare the performance of ICSI in non-male factor infert

Open chromatin mapping identifies transcriptional networks regulating human epididymis epithelial function.

The epithelium lining the epididymis in the male reproductive tract maintains a luminal environment that promotes sperm cell maturation. This process is dependent on the coordinated expression of many genes that encode proteins with a role in epithelial transport. We previously generated genome-wide maps of open chromatin in primary human epididymis epithelial (HEE) cells to identify potential regulatory elements controlling coordinated gene expression in the epididymis epithelium. Subsequent in silico analysis identified transcription factor-binding sites (TFBS) that were over-represented in

Castration induces changes in the cation-dependent mannose-6-phosphate receptor in rat epididymis: possible implications in secretion of lysosomal enzymes.

It is believed that the mammalian epididymis participates in the maturation of the sperm due to its secretory activity. High concentrations of several secreted acid hydrolases are found in the epididymal lumen. Moreover, some of these enzymes are secreted by the epididymal epithelium in an androgen-dependent fashion. In this study, we attempted to discern whether mannose-6-phosphate receptors (MPRs) regulate transport and secretion of lysosomal enzymes in the rat epididymis, and if these events are altered when the animals are subjected to hormonal manipulation. We observed that expression of

Aquaporin 9 expression in the developing rat epididymis is modulated by steroid hormones.

Fluid and solute transport across the epithelium of the male excurrent duct is important for sperm maturation and storage. Aquaporin 9 (AQP9), which allows permeation of water and neutral solutes, is abundant throughout the male reproductive tract, where it is expressed at the apical membrane of rat epididymal principal cells as early as at 1 week of age. We evaluated the effect of neonatal exposure to: 1) a GNRH antagonist (GNRHa); 2) diethylstilbestrol (DES); 3) ethinyl estradiol (EE); 4) DES plus testosterone (DES+TE); and 5) the anti-androgen flutamide on AQP9 expression in the epididymis …

In vitro post-meiotic germ cell development from human embryonic stem cells.

BACKGROUND: Investigating the mechanisms of human primordial germ cell (PGC) and gamete development are important for understanding the causes of infertility and effects of environmental chemicals on reproductive development. However, there are practical and ethical difficulties associated with obtaining human tissue in early development. The aim of this study was to investigate whether human embryonic stem cell-hESC-generated germ cells could provide an in vitro model of gamete development. METHOD: Human ESCs were differentiated as embryoid bodies (EBs) in vitro. Gene and protein marker expr

Macrocephaly in bull spermatozoa is associated with nuclear vacuoles, diploidy and alteration of chromatin condensation.

Spermatozoa from 2 dairy AI (artificial insemination) bulls (A and B), identified by their abnormal spermiogram with cells depicting frequent macrocephaly, double tails and nuclear vacuoles, were case-investigated and compared to normal spermatozoa from a control AI sire (C). Head sizes were measured and morphological abnormalities scored using brightfield and differential interference contrast microscopy. The degree of sperm maturation and of resistance to acid-induced DNA denaturation in situ were determined after uploading of acridine orange using flow cytometry of 5,000 cells/sample. Nucle…

The equatorial subsegment (EqSS) was originally identified by atomic force microscopy as a discrete region within the equatorial segment of … embrane. In all five species, tyrosine phosphorylated proteins became restricted to the EqSS during sperm maturation in the epididymis. The major tyrosine phosphorylated proteins in the EqSS of boar and ram spermatozoa were identified by mass spectrometry as orthologs of human SPACA1 (formerly SAMP32). Epididymis of viscacha (Lagostomus maximus maximus): morphological changes during the annual reproductive cycle.

Little is known about morphological changes in the epididymis in relation to the natural photoperiod or their influence on sperm maturation. The viscacha is a seasonal rodent living in the Southern Hemisphere. The adult males exhibit an annual reproductive cycle with periods of maximum gonadal activity and gonadal regression. In this work, we studied seasonal variations in the morphology and cellular population of the epididymis during both periods, and we compared these results with those recorded at the testicular level. Epididymides were removed and studied by light microscopy. Measurements…

Sexual hormones and epilepsy: threat and opportunities.

PURPOSE OF REVIEW: This article reviews recent developments in our knowledge of the reciprocal interactions between epilepsy and sex hormones and how these interactions may play a role in the pathophysiology and treatment of both. RECENT FINDINGS: Community studies confirm that menstrual disorders are overrepresented among women with epilepsy, especially among women with high seizure frequency and in those on valproate or polytherapy. Reproductive function is not affected in women with epilepsy who discontinued antiepileptic drug therapy during pubertal maturation. While valproate has been im

Use of plant-derived compounds for chilled gamete storage in felids and canids.

Wild felids and canids play important roles in maintaining ecosystem stability and biodiversity, yet many species are threatened by habitat loss, climate change, urbanization, and poaching. Assisted reproductive technologies (ARTs) have therefore become valuable tools for wildlife conservation and genetic resource banking. However, prolonged storage and post-mortem deterioration remain major limitations affecting gamete quality and fertilizing ability. Oxidative stress is a major cause of gamete deterioration during chilled storage. During chilled storage, excessive reactive oxygen species (RO

Metaphase-II spindle disruption in oocytes from anti-centromere antibody-positive patients leads to multi-pronuclear formation.

Anti-centromere antibody (ACA), a specific antinuclear antibody (ANA) subset targeting constitutive kinetochore proteins, is recognized as a potent immunological factor impairing reproductive outcomes. While these antibodies interfere with the kinetochore-microtubule interface essential for chromosome segregation, the relationship between meiotic progression involving spindle formation in human oocytes and subsequent fertilization remains largely unknown. This retrospective cohort study analyzed ovum pick-up cycles, comparing ACA-positive patients with ACA-negative and ANA-positive controls. T

m6A RNA methylation orchestrates spermatogenesis: decoding the synergistic regulatory network of signaling pathways and ubiquitination modifications.

N6-methyladenosine (m6A), the most prevalent epitranscriptomic modification in eukaryotic mRNA, dynamically regulates mRNA stability, splicing, and translational efficiency through a sophisticated "writer-eraser-reader" network. This system comprises methyltransferase writers (e.g., METTL3/14, METTL16, WTAP), demethylase erasers (e.g., ALKBH5, FTO), and recognition readers (e.g., YTHDC2, YTHDF2, hnRNPC). This precise regulatory mechanism governs spermatogonial stem cell proliferation, meiotic initiation, and spermatid maturation by orchestrating cell cycle progression, meiotic timing, and sper

Reproductive toxicity of benzo[α]pyrene: Effects, mechanisms, and perspectives.

Polycyclic aromatic hydrocarbons are pervasive environmental pollutants. Benzo[α]pyrene (B[α]P), a prototypical polycyclic aromatic hydrocarbon, elicits increasing apprehension due to its endocrine-disrupting properties and reproductive toxicity in animals; however, the toxicological effects and underlying mechanisms have not been thoroughly compiled and elucidated. This research delineates B[α]P toxicity in the reproductive systems of multiple species, including mammals, amphibians, fish, and invertebrates, alongside the relevant molecular and epigenetic evidence. In males, exposure to B[α]P
