Background/Objectives: The RET (Rearranged during Transfection) gene encodes a receptor tyrosine kinase. RET plays a critical role in embryonic development and postnatal physiology. This review provides a comprehensive overview of RET-associated disorders, focusing on the molecular mechanisms of RET activation, associated clinical phenotypes and therapeutic implications. In addition, we present an updated RET mutation database. Methods:RET mutation database is built through the integration and curation of data from two major RET mutation repositories: the Leiden Open Variation Database (LOVD)

The growth plate plays a crucial role in long bone development and elongation, housing stem cells that contribute to bone formation. This study investigates the role of primary cilia, specialized organelles that regulate stem cell fate, in the development and repair of long bones. Here, we report the presence of primary cilia in all zones of the growth plate, particularly during embryonic development. Spatial transcriptomics and analysis of conditional knockout (CKO) mice identified that primary cilia mediate critical developmental signaling pathways within the growth plate. Disruption of prim

Serotonin (5-HT) signaling plays essential roles in vertebrate development beyond neurotransmission, including in ocular morphogenesis. Here, we investigated the effects of Ritanserin, a non-selective 5-HT2 receptor antagonist, on zebrafish embryonic development with a focus on eye formation. While Ritanserin exposure (0.1-10 μM) did not affect mortality or hatching rates up to 96 h post-fertilization (hpf), higher concentrations (5-10 μM) induced developmental abnormalities, such as microphthalmia, pericardial edema, blood congestion, and craniofacial malformations by 5 days post-fertilizat

Bone morphogenetic proteins (BMPs) are critical for craniofacial development. We previously reported that cranial neural crest cell (CNCC)-specific enhanced BMP signaling through the ALK2 receptor causes ectopic cartilage formation in the face during mouse embryonic development. However, the downstream effectors triggering this ectopic chondrogenesis remain unclear. Here, we investigated the targets of BMP signaling responsible for ectopic cartilage formation. A microarray analysis using CNC-derived ectomesenchymal cells from the first branchial arches identified T-box transcription factor 20

BACKGROUND: Persistent primitive hypoglossal artery (PPHA) is a rare embryological variant of the cerebral circulation in which the posterior circulation is supplied through a persistent embryonic carotid-basilar anastomosis. While often discovered incidentally, PPHA may have important clinical consequences when associated with atherosclerotic disease, aneurysms, or other cerebrovascular abnormalities.
OBJECTIVE: To introduce a structured clinical classification system for PPHA that captures its anatomical spectrum, pathological associations, and symptomatic presentations, with the aim of imp

Prenatal hypoxia, often accompanied by maternal glucocorticoid stress, can predispose offspring to neurological disorders in adulthood. If placental ischemia (PI) primarily reduces fetal oxygen supply, the maternal hypoxia (MH) model also elicits a pronounced fetal glucocorticoid exposure. Here, we compared MH and PI in rats to distinguish their unique and overlapping effects on embryonic and newborn brain development. We analyzed glucocorticoid transport into the developing brain, glucocorticoid receptor (GR) expression, and GR-dependent transcription, along with key enzymes regulating glucoc

Nicotinamide adenine dinucleotide (NAD) is essential for embryonic development. To date, biallelic loss-of-function variants in 3 genes encoding nonredundant enzymes of the NAD de novo synthesis pathway - KYNU, HAAO, and NADSYN1 - have been identified in humans with congenital malformations defined as congenital NAD deficiency disorder (CNDD). Here, we identified 13 further individuals with biallelic NADSYN1 variants predicted to be damaging, and phenotypes ranging from multiple severe malformations to the complete absence of malformation. Enzymatic assessment of variant deleteriousness in vit

BACKGROUND: The incidence of spontaneous abortion (SA), which affects approximately 15-20% of pregnancies, is the most common complication of early pregnancy. Pathogenic copy number variations (CNVs) are recognized as potential genetic causes of SA. However, CNVs of variants of uncertain significance (VOUS) have been identified in products of conceptions (POCs), and their correlation with SA remains uncertain.
RESULTS: Of 189 spontaneous abortion cases, trisomy 16 was the most common numerical chromosome abnormality, followed by monosomy X. CNVs most often occurred on chromosomes 4 and 8. Gen

Sn(IV) complex of N-Confused Porphyrin (Sn(IV)-NCP) has been prepared and characterized by several spectroscopic techniques to verify its structure and purity. Sn(IV)-NCP shows a red shift in both the Soret and Q bands compared to the free base NCTPP. The last Q band appears in the NIR region. Based on these characteristics, we investigated the antiproliferative properties and antimicrobial photodynamic therapy (a-PDT) efficiency of Sn(IV)-NCP against the bacteria Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). Further, we investigated the photodynamic activity of Sn (IV)-NCP

BACKGROUND: Cerebral autosomal-dominant arteriopathy with subcortical infarction and leukoencephalopathy (CADASIL) is an inherited small-vessel disease that affects the white matter of the brain. Recent studies have confirmed that the deposition of NOTCH3ECD is the main pathological basis of CADASIL; however, whether different mutations present the same pathological characteristics remains to be further studied. Some studies have found that mitochondrial dysfunction is related to CADASIL; however, the specific effects of NOTCH3ECD on mitochondrial remain to be determined.

Antiepileptic drugs affect embryonic development when administered during pregnancy, generating severe alterations, such as as cleft lip, spina bifida, heart abnormalities, or neuronal alterations. The compound DL-4-hydroxy-4-phenylhexanamide (DL-HEPB), a phenyl alcohol amide structurally different from known anticonvulsants, has shown good anticonvulsant effects in previous studies. However, its effects on intrauterine development are unknown. So, the purpose of this study was to determine the potential of DL-HEPB to produce alterations in conceptus. Pregnant Wistar rats were orally exposed t

Chlorophenols represent one of the most abundant families of toxic pollutants emerging from various industrial manufacturing units. The toxicity of these chloroderivatives is proportional to the number and position of chlorine atoms on the benzene ring. In the aquatic environment, these pollutants accumulate in the tissues of living organisms, primarily in fishes, inducing mortality at an early embryonic stage. Contemplating the behaviour of such xenobiotics and their prevalence in different environmental components, it is crucial to understand the methods used to remove/degrade the chlorophen

Antipsychotic drugs are the mainstay of treatment for schizophrenia and provide adjunct therapies for other prevalent psychiatric conditions, including bipolar disorder and major depressive disorder. However, they also induce debilitating extrapyramidal syndromes (EPS), such as Parkinsonism, in a significant minority of patients. The majority of antipsychotic drugs function as dopamine receptor antagonists in the brain while the most recent 'third'-generation, such as aripiprazole, act as partial agonists. Despite showing good clinical efficacy, these newer agents are still associated with EPS

Sphingolipids are a diverse family of lipids with critical structural and signalling functions in the mammalian nervous system, where they are abundant in myelin membranes. Serine palmitoyltransferase, the enzyme that catalyses the rate-limiting reaction of sphingolipid synthesis, is composed of multiple subunits including an activating subunit, SPTSSA. Sphingolipids are both essential and cytotoxic and their synthesis must therefore be tightly regulated. Key to the homeostatic regulation are the ORMDL proteins that are bound to serine palmitoyltransferase and mediate feedback inhibition of en

BACKGROUND: Persistent sciatic artery (PSA) is a congenital malformation due to incomplete involution during the embryonic period. Its etiology is unknown, with an estimated incidence of 0.02 - 0.04% in the whole population and a mean age of 60-65 years. Its presentation can be bilateral. It is asymptomatic in most cases and is usually detected accidentally; however, some symptoms may appear, such as claudication, sciatic neuralgia, and pain in the affected limb. It can also manifest as an aneurysmal dilatation or thrombosis that can generate distal embolism with ischemia.

Protein kinase C and casein kinase substrate in neurons (PACSINs), or syndapins (synaptic dynamin-associated proteins), are a family of proteins involved in the regulation of cell cytoskeleton, intracellular trafficking and signalling. Over the last twenty years, PACSINs have been mostly studied in the in vitro and ex vivo settings, and only in the last decade reports on their function in vivo have emerged. We first summarize the identification, structure and cellular functions of PACSINs, and then focus on the relevance of PACSINs in vivo. During development in various model organisms, PACSIN

Reactive oxygen species (ROS) are oxygen-containing molecules which are reactive in nature and are capable of independent existence in the body. ROS comprise mostly of free radicals that contain at least one unpaired electron. Endogenous sources are the foremost birthplaces of ROS, which include mitochondrial electron transport chain, endoplasmic reticulum and peroxisome. Conversely, numerous enzymatic pathways such as xanthine oxidase and cyclooxygenase systems are among the prominent generators of cellular ROS. Major sources of ROS in the female reproductive tract include Graafian follicles,

Cisplatin is a commonly used chemotherapeutic for the treatment of many solid organ cancers; however, its effectiveness is limited by the development of acute kidney injury (AKI) in 30% of patients. AKI is driven by proximal tubule cell death, leading to rapid decline in renal function. It has previously been shown that sphingolipid metabolism plays a role in regulating many of the biological processes involved in cisplatin-induced AKI. For example, neutral ceramidase (nCDase) is an enzyme responsible for converting ceramide into sphingosine, which is then phosphorylated to become sphingosine-

Inositol is an essential metabolite that serves as a precursor for structural and signaling molecules. Although perturbation of inositol homeostasis has been implicated in numerous human disorders, surprisingly little is known about how inositol levels are regulated in mammalian cells. A recent study in mouse embryonic fibroblasts demonstrated that nuclear translocation of inositol hexakisphosphate kinase 1 (IP6K1) mediates repression of myo-inositol-3-P synthase (MIPS), the rate-limiting inositol biosynthetic enzyme. Binding of IP6K1 to phosphatidic acid (PA) is required for this repression.

In this study, it was aimed to determine the effect of destruction of lyophilized and frozen-thawed ram sperm plasma and acrosomal membrane on development of embryos produced by intracytoplasmic sperm injection (ICSI). Semen samples were divided into two groups for lyophilization (L) and freezing (F). For the removal of the plasma membrane, L and F groups were incubated with Triton X-100 (LTX-100 and FTX-100, respectively). Integrities of the plasma membrane, acrosome and chromatin structure were evaluated. Oocytes were injected with these sperm groups. Although no plasma membrane and acrosome
