The therapeutic potential of neuronal KCNQ channel modulators.

Neuronal KCNQ (Kv7) channels (KCNQ2-5 or Kv7.2-7.5, disclosed to date) were discovered by virtue of their homology with a known cardiac channel involved in long QT syndrome (KvLQT or KCNQ1, Kv7.1) and first disclosed in 1998. The involvement of KCNQ2 (Kv7.2) and KCNQ3 (Kv7.3) in a benign idiopathic neonatal epilepsy, KCNQ4 (Kv7.4) in a form of congenital deafness, and the discovery that neuronal KCNQ heteromultimers were among the molecular substrates of M-channels, resulted in a high level of interest for potential drug development strategies. A number of small-molecule modulators were quickl

Increased function of voltage-dependent Ca++ channels and Ca(++)-activated K+ channels in resting state of femoral arteries from spontaneously hypertensive rats at prehypertensive stage.

The present study examined the possible role of voltage-dependent Ca++ channels (VDCs) and Ca(++)-activated K+ (KCa) channels in the regulation of resting tone of arteries from spontaneously hypertensive rats (SHR) at a prehypertensive stage. Differences in the effects of agents that interact with these channels were assessed in endothelium-denuded strips of femoral arteries isolated from 4-week-old SHR and age-matched normotensive Wistar-Kyoto rats (WKY). Systolic blood pressures at this age were not significantly different between SHR and WKY. The arterial strips from SHR maintained a myogen

Why are antiepileptic drugs used for nonepileptic conditions?

Antiepileptic drugs (AEDs) are used to treat various nonepileptic central nervous system (CNS) disorders, both in neurology and psychiatry. Most AEDs have multiple mechanisms of action (MOAs), which include modulation of γ-aminobutyric acid (GABA)ergic and glutamatergic neurotransmission, and alteration of voltage-gated ion channels or intracellular signaling pathways. These MOAs may explain the efficacy of AEDs in the treatment of bipolar disorder and neuropathic pain. Bipolar disorder and epilepsy have some common features, such as their episodic nature and associated kindling phenomena, whi

Ca(2+)-activated and voltage-gated K+ currents in smooth muscle cells isolated from human mesenteric arteries.

1. Smooth muscle cells were enzymatically isolated from arteries dissected from mesenteric fat removed from patients undergoing routine surgery. The whole-cell patch clamp technique was used to characterize the potassium (K+) currents and passive electrical properties of these cells, using high-K(+)-containing pipette solutions with either 0.2 mM EGTA or 10 mM EGTA and 10 mM BAPTA. 2. Cell capacitance, which is proportional to membrane surface area, was normally distributed around a value of 46 pF, and independent of artery size between 0.4 and 3.6 mm. The mean membrane potential measured unde

Mechanism of generation of spontaneous miniature outward currents (SMOCs) in retinal amacrine cells.

A subtype of retinal amacrine cells displayed a distinctive array of K(+) currents. Spontaneous miniature outward currents (SMOCs) were observed in the narrow voltage range of -60 to -40 mV. Depolarizations above approximately -40 mV were associated with the disappearance of SMOCs and the appearance of transient (I(to)) and sustained (I(so)) outward K(+) currents. I(to) appeared at about -40 mV and its apparent magnitude was biphasic with voltage, whereas I(so) appeared near -30 mV and increased linearly. SMOCs, I(to), and a component of I(so) were Ca(2+) dependent. SMOCs were spike shaped, oc

Cyclic guanosine monophosphate compartmentation in rat cardiac myocytes.

BACKGROUND: Cyclic guanosine monophosphate (cGMP) is the common second messenger for the cardiovascular effects of nitric oxide (NO) and natriuretic peptides, such as atrial or brain natriuretic peptide, which activate the soluble and particulate forms of guanylyl cyclase, respectively. However, natriuretic peptides and NO donors exert different effects on cardiac and vascular smooth muscle function. We therefore tested whether these differences are due to an intracellular compartmentation of cGMP and evaluated the role of phosphodiesterase (PDE) subtypes in this process. METHODS AND RESULTS:

Unveiling the molecular crosstalk between the purinergic P2X7 receptor and cannabinoid subtype 2 receptor in human microglial HMC3 cells: New insights for multitarget therapies in neuroinflammation.

BACKGROUND: Microglia-mediated neuroinflammation plays a pivotal role in the progression and possible onset of numerous neurodegenerative diseases. A promising therapeutic approach involves shifting microglial polarization from a pro-inflammatory to an anti-inflammatory phenotype. Two receptors central to this process are the purinergic P2X7 receptor, an ATP-gated ion channel that promotes pro-inflammatory signaling, and the cannabinoid receptor subtype 2, a Gi/o protein-coupled receptor associated with anti-inflammatory effects. METHODS: Here, we investigated the therapeutic potential of pha

α7 Nicotinic Acetylcholine Receptor as a Therapeutic Target in Central Nervous System Diseases: Prospects and Limitations.

INTRODUCTION: The α7 nicotinic acetylcholine receptor (α7nAChR), a transmitter-gated ion channel, is normally activated by the endogenous neurotransmitter acetylcholine (ACh). Its activation triggers a distinctive cholinergic anti-inflammatory pathway, leading to anti-inflammatory effects in the central nervous system (CNS). This unique pharmacological property has positioned α7nAChR as a promising therapeutic target for multiple CNS disorders. This review delineates α7nAChR's multifaceted roles in CNS pathophysiology, appraises current clinical trial progress of its modulators, and elucidates

Evolving Roles of Primary Cilia in CNS Development and Neural Circuit Function: From Human Disease to Molecular Underpinnings.

Primary cilia are small organelles acting as cellular antennae that sense and transduce diverse signals, including developmental signaling pathways in the developing central nervous system (CNS). This signaling is essential for normal CNS development, as evidenced by the prevalence of neurodevelopmental phenotypes in conditions arising from primary cilia dysfunction (ciliopathies). Though significant research has focused on the roles of primary cilia during CNS development, the functions of primary cilia in the mature CNS have only recently become a focus of investigation. Primary cilia are pr

Genetic Targeting and Conductance-Based Modeling Reveal Novel Diversity within Mouse Type II Spiral Ganglion Neurons.

Spiral ganglion neurons (SGNs) transmit auditory signals from the cochlea to the brain and are divided into two main types: type I and type II, distinguished by their anatomy and connectivity. However, the function of type II SGNs remains poorly understood due to their scarcity and lack of clear physiological markers. In this study, we use two Cre-dependent fluorescent reporter mouse lines of both sexes to enhance the identification and targeting of type II SGNs for whole-cell patch-clamp recordings. We reveal a set of distinguishing biophysical features, notably, the presence of an inactivati

A PIEZO-Gated Mechanotransduction Timing Window Governs Scar Commitment in Wound Healing.

Cutaneous scarring remains a major clinical challenge, often resulting in impaired function, altered tissue mechanics, and poor aesthetic outcomes. Most current anti-fibrotic therapies target late-stage fibrosis, when extracellular matrix deposition and myofibroblast persistence are already established, limiting regenerative benefit. We propose that scar formation is governed not by gradual accumulation of fibrotic signals but by an early mechanically regulated commitment event, that precedes overt fibrosis. We introduce the concept of a PIEZO-gated mechanotransduction timing window, a finite

P2X7R activation promotes ferroptosis in dopaminergic neurons via NF-κB signaling pathway in vitro and in vivo models of MPP+/MPTP-induced Parkinson's disease.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by the loss of dopaminergic neurons (DNs). The role of purinergic receptor P2X ligand-gated ion channel 7 (P2X7R) in mediating neuroinflammation and cell death in PD has attracted increasing research attention. Therefore, we conducted studies in the MPP+-induced PD cellular model and the MPTP-induced PD animal model to assess P2X7R regulates ferroptosis through the nuclear factor-κB (NF-κB) signaling pathway and thereby affects the PD process. This involved assessing changes in mRNA and protein levels of tyrosine hydroxylas

The Identification of Opioid Receptors and Peptide Precursors in Human DRG Neurons Expressing Pain-Signaling Molecules Confirms Their Potential as Analgesic Targets.

The presence and function of the opioidergic system in sensory dorsal root ganglia (DRG) was demonstrated in various animal models of pain. To endorse recent functional and transcriptional evidence of opioid receptors in human DRG, this study compared morphological and transcriptional evidence in human and rat DRG using immunofluorescence confocal microscopy and mRNA transcript analysis. Specifically, it examined the neuronal expression of mu (MOR), delta (DOR), and kappa (KOR) opioid receptors, opioid peptide precursors (POMC, PENK, and PDYN), and key pain-signaling molecules. The results dem

Adenosine A2A receptor blockade attenuates excitotoxicity in rat striatal medium spiny neurons during an ischemic-like insult.

During brain ischemia, excitotoxicity and peri-infarct depolarization injuries occur and cause cerebral tissue damage. Indeed, anoxic depolarization, consisting of massive neuronal depolarization due to the loss of membrane ion gradients, occurs in vivo or in vitro during an energy failure. The neuromodulator adenosine is released in huge amounts during cerebral ischemia and exerts its effects by activating specific metabotropic receptors, namely: A1, A2A, A2B, and A3. The A2A receptor subtype is highly expressed in striatal medium spiny neurons, which are particularly susceptible to ischemic

Ginsenoside Rd, a natural production for attenuating fibrogenesis and inflammation in hepatic fibrosis by regulating the ERRα-mediated P2X7r pathway.

Ginseng, when used as a food and nutritional supplement, has the ability to regulate human immunity. Here, the potential anti-hepatic fibrosis effect of ginsenoside Rd (Rd), one of the protopanaxadiol types of ginsenoside, was investigated. We established a hepatic fibrosis model using intraperitoneal injection of thioacetamide (TAA) for five weeks in mice. In addition, an in vitro model was established by using TGF-β to activate hepatic stellate cells (HSCs), treated with Rd and an estrogen-related receptor α (ERRα) inhibitor (XCT-790). The ERRα knockdown (shRNA-ERRα) of the primary mouse hep

Pharmacological Profiling of a Brugia malayi Muscarinic Acetylcholine Receptor as a Putative Antiparasitic Target.

The diversification of anthelmintic targets and mechanisms of action will help ensure the sustainable control of nematode infections in response to the growing threat of drug resistance. G protein-coupled receptors (GPCRs) are established drug targets in human medicine but remain unexploited as anthelmintic substrates despite their important roles in nematode neuromuscular and physiological processes. Bottlenecks in exploring the druggability of parasitic nematode GPCRs include a limited helminth genetic toolkit and difficulties establishing functional heterologous expression. In an effort to

De novo GRIN2A variants associated with epilepsy and autism and literature review.

N-methyl-D-aspartate receptors (NMDAR) are di- or tri-heterotetrameric ligand-gated ion channels composed of two obligate glycine-binding GluN1 subunits and two glutamate-binding GluN2 or GluN3 subunits, encoded by GRIN1, GRIN2A-D, and GRIN3A-B receptor genes respectively. Each NMDA receptor subtype has different temporal and spatial expression patterns in the brain and varies in the cell types and subcellular localization resulting in different functions. They play a crucial role in mediating the excitatory neurotransmission, but are also involved in neuronal development and synaptic plastici
