Bioactivation of leukotoxins to their toxic diols by epoxide hydrolase.

Leukotoxin is a linoleic acic oxide produced by leukocytes and has been associated with the multiple organ failure and adult respiratory distress syndrome seen in some severe burn patients. Leukotoxin has been reported to be toxic when injected into animals intravenously. Herein, we report that this lipid is not directly cytotoxic in at least two in vitro systems. Using a baculovirus expression system we demonstrate that leukotoxin is only cytotoxic in the presence of epoxide hydrolases. In addition, it is the diol metabolite that proves toxic to pulmonary alveolar epithelial cells, suggesting

Metabolic reprogramming in differentiated thyroid cancer progression: A tumor metabolomics analysis.

BACKGROUND: Differentiated thyroid cancers exhibit biologic and clinical heterogeneity not fully explained by conventional staging. Metabolic reprogramming is a hallmark of cancer, and its contribution to differentiated thyroid cancer progression is an ongoing area of study. Metabolomics-the comprehensive analysis of metabolites within biologic systems-may identify stage-specific metabolic signatures. METHODS: Fifty-five differentiated thyroid cancer specimens underwent untargeted metabolomic profiling using liquid chromatography-mass spectrometry. Stage was assigned per American Joint Commit

Tobacco intercropping with Allium crops reshapes soil microbiome-metabolome interactions and improves plant health and tobacco leaf quality.

BACKGROUND: Intercropping systems play important roles in improving soil conditions, enhancing crop performance and suppressing soil-borne diseases. However, the mechanisms by which tobacco-Allium intercropping regulates soil microecology, tobacco growth and leaf quality remain insufficiently understood. In this study, tobacco monoculture and tobacco intercropping systems with onion, garlic or Chinese chive were established to evaluate tobacco agronomic traits, disease occurrence, cured leaf chemical composition and defence-related enzyme activities. Soil physicochemical properties, microbial

Histopathological effects of the fruit extract of Citrullus colocynthis on the integument of the female tick Hyalomma dromedarii.

Hyalomma dromedarii is a hard tick species parasitizing domestic animals, particularly camels. Heavy infestation results in huge economic losses through severe blood loss and transmission of pathogens, in addition to crucial problems for camel production. Worldwide control of ticks is mainly based on acaricides, which have led to environmental pollution, resistance development, and an increase in the cost of control. To reduce the drawbacks of chemical acaricides, new tick control methods are therefore required, such as the application of natural plant extracts. Citrullus colocynthis, commonly

Diversity and functions of fungal VOCs with special reference to the multiple bioactivities of the mushroom alcohol.

Different species of fungi usually share many common pathways but they also have some unique metabolic pathways (e.g. those for specialised metabolites). It is not clear how gene expression patterns significantly contribute to the creation of diverse volatile compounds. Based on the research of most VOCs, the functions of different fungal volatile compounds are mainly as follows: inhibitory or synergistic effects on other microorganisms, promoting growth in plants or inducing a defensive response in crops, and participating in the material cycle or affecting the interactions between organisms

Microbiome-Metabolomics Analysis of the Impacts of Balantidium Coli Infection in Rhesus Monkeys (Macaca mulatta).

Balantidium coli (B. coli) is a prevalent intestinal parasite in monkeys, significantly impacting their health. Previous studies have demonstrated that B. coli infection in pigs leads to severe dysregulation of the gut microbiota. However, there has been no report on the alterations in fecal microbiota and metabolites in rhesus monkeys infected with B. coli. In order to investigate the differences in gut microbiota and metabolites between healthy rhesus monkeys and those infected with B. coli, we conducted gene sequencing and gas chromatography-mass spectrometry (GC-MS) analysis of fecal sampl

A multi-omics investigation of the lung injury induced by PM2.5 at environmental levels via the lung-gut axis.

Long-term exposure to fine particulate matter (PM2.5) posed injury for gastrointestinal and respiratory systems, ascribing with the lung-gut axis. However, the cross-talk mechanisms remain unclear. Here, we attempted to establish the response networks of lung-gut axis in mice exposed to PM2.5 at environmental levels. Male Balb/c mice were exposed to PM2.5 (dose of 0.1, 0.5, and 1.0 mg/kg) collected from Chengdu, China for 10 weeks, through intratracheally instillation, and examined the effect of PM2.5 on lung functions of mice. The changes of lung and gut microbiota and metabolic profiles of m

Failure to withstand oxidative stress induced by phospholipid hydroperoxides as a possible cause of the lens opacities in systemic diseases and ageing.

Lipid peroxidation (LPO) is a causative factor of cataract. The increased concentrations of primary molecular LPO products (diene conjugates, lipid hydroperoxides) and end fluorescent LPO products, were detected in the lipid moieties of the aqueous humor samples obtained from patients with senile and complicated cataracts as compared to normal donors. The degrees of lens clouding were assessed quantitatively by measuring the optical density indices and areas of equidensities using digital image analysis. Human cataractous lenses showed decreased activity of glutathione peroxidase (GPX, catalyz

The involvement of cytochrome P-450 in hepatic microsomal steroid hydroxylation reactions supported by sodium periodate, sodium chlorite, and organic hydroperoxides.

The mechanism of steroid hydroxylation in rat liver microsomes has been investigated by employing NaIO4, NaClO2, and various organic hydroperoxides as hydroxylating agents and comparing the reaction rates and steroid products formed with those of the NADPH-dependent reaction. Androstenedione, testosterone, progesterone, and 17beta-estradiol were found to act as good substrates. NaIO4 was by far the most effective hydroxylating agent followed by cumene hydroperoxide, NADPH, NaClO2, pregnenolone 17alpha-hydroperoxide, tert-butyl hydroperoxide, and linoleic acid hydroperoxide. Androstenedione was

Cryo-EM structure of locked spike glycoprotein from bat SARS-like coronavirus WIV1, molecular dynamics and biophysics across host range.

As made evident by severe acute respiratory syndrome coronavirus 1 (SARS-CoV-1) and SARS-CoV-2 pandemics, the possibility of a SARS-like coronavirus (SL-CoV) emerging again in humankind after zoonotic spillover represents a significant global health threat. Given the role of spike (S) glycoprotein in mediating SL-CoV cell entry and cross-species transmission, there remains urgent need of structural information on SL-CoV S to guide therapeutic countermeasure development. Among SL-CoVs, bat-derived WIV1 is capable of using as receptor the angiotensin converting enzyme 2 (ACE2) from a variety of

Olive Leaf Extract Suppresses Sebogenesis and Inflammation via AKT/ERK and SREBP-1/PPAR-γ Signaling in Human Sebocytes.

This study evaluated olive leaf extract (OLE) as a multifunctional dermocosmetic candidate for sebum-related and inflammatory responses relevant to oily and acne-prone skin using an axis-aligned in vitro panel: (i) sebocyte lipogenesis, (ii) inflammatory mediator production in keratinocytes, and (iii) fibroblast-mediated collagen gel contraction. In addition, supportive mechanistic evidence for the sebum-related effects of OLE was obtained by examining signaling proteins associated with sebocyte lipogenesis, including PPAR-γ and SREBP-1. As a result, OLE significantly inhibited linoleic acid-i
