A Neurocomputational Account of How Inflammation Enhances Sensitivity to Punishments Versus Rewards.

BACKGROUND: Inflammation rapidly impairs mood and cognition and, when severe, can appear indistinguishable from major depression. These sickness responses are characterized by an acute reorientation of motivational state; pleasurable activities are avoided, and sensitivity to negative stimuli is enhanced. However, it remains unclear how these rapid shifts in behavior are mediated within the brain. METHODS: Here, we combined computational modeling of choice behavior, experimentally induced inflammation, and functional brain imaging (functional magnetic resonance imaging) to describe these mech

Symptom profile and short term outcome of catatonia: an exploratory clinical study.

BACKGROUND: Catatonia is a potentially life-threatening but treatable neuropsychiatric condition. Although considered more common in low income countries, data is particularly sparse in these settings. In this study we explore the symptomatology, treatment, and short-term outcome of catatonia in Ethiopia, a low income country. METHODS: The study was a prospective evaluation of patients admitted with a DSM-IV diagnosis of catatonia. Diagnosis of Catatonia and its severity were further assessed with the Bush-Francis Catatonia Rating Scale (BFCRS). RESULTS: Twenty participants, 5 male and 15 fe

In vitro and in vivo characterisation of Lu AF64280, a novel, brain penetrant phosphodiesterase (PDE) 2A inhibitor: potential relevance to cognitive deficits in schizophrenia.

Here, we present the pharmacological characterisation of Lu AF64280, a novel, selective, brain penetrant phosphodiesterase (PDE) 2A inhibitor, in in vitro/in vivo assays indicative of PDE2A inhibition, and in vivo models/assays relevant to cognitive processing or antipsychotic-like activity. The in vitro selectivity of Lu AF64280 was determined against a panel of PDE enzymes and 3',5'-cyclic guanosine monophosphate (cGMP) levels in the hippocampus were determined using in vivo microdialysis. Lu AF64280 potently inhibited hPDE2A (Ki = 20 nM), 50-fold above moderate inhibition of both hPDE9A (Ki

Auditory feedback perturbation in children with developmental speech sound disorders.

BACKGROUND/PURPOSE: Several studies indicate a close relation between auditory and speech motor functions in children with speech sound disorders (SSD). The aim of this study was to investigate the ability to compensate and adapt for perturbed auditory feedback in children with SSD compared to age-matched normally developing children. METHOD: 17 normally developing children aged 4.1-8.7 years (mean=5.5, SD=1.4), and 11 children with SSD aged 3.9-7.5 years (mean=5.1, SD=1.0) participated in the study. Auditory feedback was perturbed by real-time shifting the first and second formant of the vow

Impairments in real-world executive function increase from childhood to adolescence in autism spectrum disorders.

OBJECTIVE: Although several studies have investigated developmental trajectories of executive functioning (EF) in individuals with autism spectrum disorders (ASD) using lab-based tasks, no study to date has directly measured how EF skills in everyday settings vary at different ages. The current study seeks to extend prior work by evaluating age-related differences in parent-reported EF problems during childhood and adolescence in a large cross-sectional cohort of children with ASD. METHOD: Children (N = 185) with an ASD without intellectual disability participated in the study. Participants w

Neural mechanisms of frustration in chronically irritable children.

OBJECTIVE: Irritability is common in children and adolescents and is the cardinal symptom of disruptive mood dysregulation disorder, a new DSM-5 disorder, yet its neural correlates remain largely unexplored. The authors conducted a functional MRI study to examine neural responses to frustration in children with severe mood dysregulation. METHOD: The authors compared emotional responses, behavior, and neural activity between 19 severely irritable children (operationalized using criteria for severe mood dysregulation) and 23 healthy comparison children during a cued-attention task completed und

Principal component based analysis of biomechanical inter-trial variability in individuals with chronic ankle instability.

BACKGROUND: Biomechanical variability during movement may influence joint stability in individuals with chronic ankle instability (CAI). The purpose of this study was to compare the kinematic and the kinetic inter-trial variability between healthy and CAI individuals. METHODS: Eleven individuals with CAI and 11 matched controls performed five repetitions of a single-leg landing task. Biomechanical data were collected from 100 ms before to 200 ms after touchdown, and were used to calculate touchdown angles, peak angles and moments at the ankle joint in the frontal and sagittal planes. Ergonomic basket design to reduce cumulative trauma disorders in tea leaf plucking operation.

Work related musculoskeletal disorders are seen as an indicating parameter of occupational stress among the women workers engaged in tea leaf plucking operation. The hand plucking (no mechanized plucking is practiced in Assam) being a highly repetitive task requires forceful exertions to reach to the distant periphery of the tea bushes and mechanical compression (pressing against hard surfaces). Specifically it aggravates with awkward positions adopted. The current research addresses to minimize the risk factors associated with CTDs and to ascertain the role of ergonomic design development in

A proposal for a five-dimensional patient-oriented epilepsy classification.

The recent proposal by the ILAE Task Force for Epilepsy Classification consists of a multi-axial syndrome-oriented approach. Epilepsy syndromes, as defined by the ILAE, group patients according to various, poorly defined parameters. The resulting syndromes have frequently no biological significance, with overlap among different syndromes and syndromes changing with age. Additionally, only a minority of patients can be classified syndromatically, and the axes of this classification system convey redundant information. We propose a five-dimensional, patient-oriented approach to classifying epile

Perception of static eye gaze direction facilitates subsequent early visual processing.

OBJECTIVE: Using event-related potentials (ERPs), it has been recently shown that a reflexive shift of attention following the observation of a dynamic eye gaze cue enhances and speeds up early visual processing of a target presented at the gazed-at location. Here we investigate whether similar early sensory modulations are also elicited by static gaze cues, or if previously described attentional effects were caused mainly by visual motion cues and not by eye gaze direction per se. Furthermore, we explore if these possible attentional orienting effects reflect facilitation of the processing of

Contrastive adversarial mix-up learning for unsupervised cross-modality image segmentation.

BACKGROUND: Unsupervised domain adaptation (UDA) has been extensively investigated in terms of its ability to address the performance degradation caused by distribution shifts between labeled source-domain images and unlabeled target-domain images. In radiotherapy, this problem is particularly relevant for the transfer of segmentation knowledge from planning computed tomography (CT) to cone-beam computed tomography (CBCT), for which target-domain annotations are limited. This study aimed to develop a contrastive adversarial mix-up learning framework for unsupervised cross-modality image segmen

Validation of an automated AI-based micro-CT organ segmentation workflow against expert annotations and its impact on fluorescence quantification.

OBJECTIVE: Automated, artificial intelligence (AI)-based, organ segmentation has the potential to streamline preclinical imaging workflows, but its suitability must be evaluated not only by geometric accuracy, but also by impact on downstream quantitative analyses. We validated a commercially available AI-based organ segmentation workflow for whole-body micro-computed tomography (micro-CT) data regarding segmentation accuracy, reproducibility, processing time, and effects on fluorescence tomography (FLT) quantification. MATERIALS AND METHODS: Heart, lungs, liver, and kidneys were segmented in

Occupational Reproductive Health Risks Among Women Healthcare Workers: A Narrative Review for Clinical Surveillance, Preconception Counseling, and Prevention.

Background/Objectives: Despite well-documented chemical and physical hazards in healthcare settings, existing reviews of occupational reproductive risks have largely focused on single-agent risk estimation and have rarely translated occupational hygiene evidence into clinical decision-making frameworks for reproductive counseling and surveillance. This narrative review synthesizes evidence across multiple occupational exposure categories-antineoplastic agents, high-level disinfectants (HLDs), sterilants, and work-organization factors-and proposes an integrated, clinically operational framework

Prevention of non-contact anterior cruciate ligament injuries in soccer players. Part 1: Mechanisms of injury and underlying risk factors.

Soccer is the most commonly played sport in the world, with an estimated 265 million active soccer players by 2006. Inherent to this sport is the higher risk of injury to the anterior cruciate ligament (ACL) relative to other sports. ACL injury causes the most time lost from competition in soccer which has influenced a strong research focus to determine the risk factors for injury. This research emphasis has afforded a rapid influx of literature defining potential modifiable and non-modifiable risk factors that increase the risk of injury. The purpose of the current review is to sequence the m

SwinTransRNA: A Swin Transformer-Based Framework for Accurate Prediction of RNA Subcellular Localization.

RNA subcellular localization determines the regulatory context in which microRNAs (miRNAs), circular RNAs (circRNAs), and long noncoding RNAs (lncRNAs) exert their functions, yet these RNA classes differ greatly in sequence length and circularity, making fixed-length raw-sequence representations inconvenient for cross-RNA comparison. Here, we present SwinTransRNA, a unified framework that encodes each RNA as a length-normalized 64 × 64 frequency chaos game representation (FCGR) of 6-mer composition, where every cell carries a fixed 6-mer identity and the matrix entries sum to one, enabling dir

The successor representation in high-risk drinking and alcohol-related contexts.

The successor representation (SR) is a reinforcement learning strategy which has been suggested to underlie nuanced forms of habitual behavior, rendering it a candidate mechanism for mental health conditions thought to involve habitual control, such as addiction. However, paradigms used to assess habits in humans cannot detect SR use. Together with the fact that they are usually conducted in disorder-irrelevant contexts, this may explain why the evidence for a hypothesized shift from goal-directed to habitual behavior in addiction is inconsistent. Here, we tested whether individuals with high-

Alcohol reinforcers attenuate goal-directed control in individuals with and without alcohol use disorder.

BACKGROUND: Impaired control over alcohol consumption is a core feature of alcohol use disorder (AUD), yet its mechanisms remain unclear. Habit theories posit a shift from goal-directed to habitual control with repeated alcohol use, but human evidence is mixed and often lacks translational alignment with animal models. Recent accounts therefore call for paradigms assessing contingency sensitivity with disorder-relevant reinforcers. METHODS: We adapted a free-operant contingency degradation task to compare goal-directed control when responding for a natural (juice) versus disorder-relevant rei

Exploring a Range of Potential Factors Promoting and Constraining EF in South African Preschoolers.

BACKGROUND: Socioeconomic disadvantage is widely associated with poorer cognitive and educational outcomes. However, emerging evidence suggests that children in low-income contexts may demonstrate resilience in executive function (EF) development. This study examined the predictors of EF in South African preschoolers from disadvantaged communities to identify modifiable, context-specific influences. METHODS: 125 Children (aged 3-5 years) and their caregivers were recruited from 15 early childhood centers in Cape Town. Children completed three tablet-based EF tasks from the Early Years Toolbox

Emotional Context Modulates Neural Prediction-Error Encoding.

Emotional experiences are closely intertwined with reinforcement learning, yet the extent to which emotional context influences the neural encoding of learning signals remains unclear. The present study examined whether emotional context modulates trialwise neural prediction error (PE) encoding during reinforcement learning. Fifty-six participants completed a drifting three-armed bandit task while electroencephalography (EEG) was recorded. Choice options were consistently paired with positive, neutral, or negative emotional images that were presented during feedback processing. Trial-by-trial
