Activation and desensitization of neuronal nicotinic receptors modulate glutamatergic transmission on neonatal rat hypoglossal motoneurons.

In the neonate the muscles of the tongue, which are exclusively innervated by the XII cranial nerves originating from the brainstem nucleus hypoglossus, must contract rhythmically in coincidence with breathing, suckling and swallowing. These motor commands are generated by hypoglossal motoneurons excited by glutamatergic inputs. Because in forebrain areas the efficiency of glutamatergic transmission is modulated by neuronal nicotinic receptors (nAChRs), the role and identity of nAChRs within the nucleus hypoglossus of the neonatal rat were explored using an in vitro brainstem slice preparation

Involvement of purinergic nerves in the NANC inhibitory junction potentials in pigeon oesophageal smooth muscle.

1. Electrical field stimulation (EFS) (0.5 ms in train of 2-32 Hz for 300 ms) in smooth muscle of pigeon oesophagus, in the presence of atropine (1 microm) and guanethidine (1 microm), elicited an inhibitory response consisting of a transient hyperpolarization (inhibitory junction potential, IJP) associated with muscle relaxation. 2. Sodium nitroprusside (SNP, 100 microm) induced hyperpolarization correlated to mechanical relaxation. 3. The nitric oxide (NO) synthase inhibitor N(omega)-nitro-l-arginine (from 0.1 to 100 microm) caused a concentration-dependent reduction of electromechanical res

Nicotinic cholinergic receptor expression in the human nasal mucosa.

Twenty-four nasal mucosa specimens were obtained from the inferior or middle turbinates of 6 normal subjects and 18 patients with chronic sinusitis, inflammatory polyp formation, or sinus allergies. Reverse transcription-polymerase chain reaction analysis was used to identify the non-neuronal nicotinic cholinergic receptor (nAChR) subunits that were expressed in the nasal mucosa. Collectively, transcripts for alpha (alpha1, alpha2, alpha3, alpha4, alpha6, alpha7) and beta (beta2, beta3, beta4) nAChR subunit genes were detected in the respiratory mucosa. The alpha3, alpha7, and beta2 subunits w

[The effects of the antagonists of muscarinic acetylcholine receptor subtypes in rat brain on urinary bladder contraction].

BACKGROUND: Pontine micuturition center and storage center are controlled by upper brain. Pharmacological and molecular biological data indicate the distribution of muscarinic receptor subtypes in rat brain, however, the effect of central muscarinic cholinergic mechanisms on the bladder activity has not been evaluated. We investigated the diversity of effect of each subtype in rat brain. METHODS: The muscarinic agents such as muscarine (non-specific agonist), atropine (non-specific antagonist), pirenzepine (M1 receptor antagonist), AF-DX116 (M2 receptor antagonist) and 4-DAMP (M3 receptor ant

In vivo and in vitro studies of glycine- and glutamate-evoked acetylcholinesterase release from spinal motor neurones: implications for amyotrophic lateral sclerosis/motor neurone disease pathogenesis.

To investigate the spinal cellular structures and molecular mechanisms involved in acetylcholinesterase (AChE) release evoked by both glycine (GLY) and glutamate (GLU)--responses that might play a role in chronic neurotoxicity--we analysed AChE histochemistry and histology upon systemic administration of aspartate (ASP), and conducted in vitro experiments in synaptosomes and slices prepared from mouse spinal ventral horns. Upon superfusion and incubation exposure of these preparations to GLY- and GLU-receptor agonists, we assayed both tissue content and release of AChE, butyrylcholinesterase a

Physiological and morphometric studies into the pathophysiology of detrusor hyperreflexia in neuropathic patients.

The in vitro responses of isolated detrusor strips obtained from neuropathic patients with neuropathic bladder dysfunction were found to be supersensitive to both carbachol [ED50 1.2 x 10(-6) M. versus 2.5 x 10(-6) M. in control strips; p < 0.005] and potassium chloride [ED50 41.8 mM. versus 49.3 mM. in control strips, p < 0.05]. There were no significant differences in the frequency-response curves obtained with intramural nerve stimulation. However, expressed as a percentage of the maximal response with carbachol, the maximal responses to intramural nerve stimulation in neuropathic strips we

Neurotransmitter-induced hypothalamic-pituitary-adrenal axis responsiveness is defective in inflammatory disease-susceptible Lewis rats: in vivo and in vitro studies suggesting globally defective hypothalamic secretion of corticotropin-releasing hormone.

The susceptibility of female Lewis (LEW/N) rats to the development of streptococcal cell wall (SCW)-induced arthritis and other autoimmune phenomena is associated with the inability of their hypothalamic-pituitary-adrenal (HPA) axis to adequately respond to inflammatory stimuli. In contrast, resistance to the development of SCW-induced arthritis and other inflammatory autoimmune manifestations in histocompatible female Fischer rats (F344/N) is related to their intact HPA axis response to inflammatory mediators. To evaluate the mechanism and the specificity of the HPA axis defect in LEW/N rats,

Senile plaques, amyloid beta-protein, and acetylcholinesterase fibres: laminar distributions in Alzheimer's disease striate cortex.

The laminar distributions of senile plaques and amyloid beta-protein (A beta P) within the striate cortex of patients with Alzheimer's disease (AD) were studied with enhanced Bielschowsky (roughly equivalent to the Campbell technique) and immunohistochemical methods. The laminar distribution of acetylcholinesterase (AChE) fibres within the striate cortex of both AD patients and control patients was studied with an enzyme histochemical method. Quantification of Bielschowsky-stained plaque numbers along intersect lines drawn parallel to laminar boundaries revealed a significant aggregation of pl

[The significance of vasoactive intestinal polypeptide (VIP) in diagnosis of sleep apnea syndrome].

Vasoactive intestinal polypeptide (VIP) is a neurotransmitter of the non-adrenergic and non-cholinergic system (NANC). It has been detected in many organs, including the airways, central and peripheral nervous system, the blood, and CSF. As a result of neuronal overflow, vagal stimulation leads to an elevation of VIP levels in the plasma. VIP leads to the activation of adenyl-cyclase, and thus to elevated cAMP in cells bearing VIP receptors. In 25 patients (24 males, 1 female) with a mean age of 50.3 +/- 9.9 years, and a Broca Index of 134.5 +/- 27.0%, plasma VIP was measured in the morning un

Apolipoprotein E and Alzheimer's disease. A role in amyloid catabolism.

It has been shown over the past few years that apolipoprotein E (apoE) plays a central role in the brain response to injury and neurodegeneration in mammalian species. The coordinated expression of apoE and its different receptors, the so-called LDL receptor family, appears to regulate the transport of cholesterol and phospholipids during the early and middle phases of the reinnervation in the adult mammalian brain. As neurons undergo dendritic remodelling and synaptogenesis using cholesterol internalization through the apoE/LDL receptor pathway, they progressively shut down 3,3-hydroxymethylg

Pineal gland hormone and idiopathic scoliosis: possible effect of melatonin on sleep-related postural mechanisms.

Experimental and clinical evidences indicate that endocrine mechanisms, particularly involving the pineal gland, exert a role in the development of postural deficits leading to the occurrence of idiopatic scoliosis (IS). In particular, experiments performed in bipedal animals have shown that removal of the pineal gland, which secretes melatonin (M), induced a scoliosis, and that in such preparations, administration of this hormone prevented the development of this deformity (cf. 131). It appears also that adolescents with IS showed a reduced level of serum M with respect to age-related control

Neuroprotective Effects of Food-Derived Cyanidin Glycosides in an Alzheimer's Disease-Like Zebrafish Model Through Modulation of PI3K/Akt/GSK3β Signaling.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder lacking effective and safe interventions. Food-derived anthocyanins have gained increasing attention due to their neuroprotective potential; however, the specific mechanisms of individual cyanidin glycosides remain insufficiently understood. Notably, the comparative efficacy and mechanistic differences among structurally distinct cyanidin glycosides have not been systematically evaluated in vivo. In the present study, three cyanidin glycosides, cyanidin-3-O-rutinoside (C3R), cyanidin-3-O-sophoroside (C3S), and cyanidin-3-O-xy

A compound pulsed magnetic field achieves superior cognitive benefits against Alzheimer's disease progression via multi-level restoration of neural oscillations and cerebral perfusion.

The link between impaired gamma oscillations and Alzheimer's disease (AD) has inspired therapies using rhythmic physical stimuli. However, given that cognition requires cross-frequency interactions like theta-gamma coupling, single-frequency stimulation may yield limited benefits. This study therefore applied a compound pulsed magnetic field (cPMF) with theta rhythm-modulated gamma frequency to evaluate its efficacy and mechanisms against AD pathology compared with single gamma-frequency pulsed magnetic field (sPMF). Local field potential results showed that cPMF outperformed sPMF by significa

Benzimidazole as a Versatile Scaffold for Developing Neurotherapeutics Against Neurodegenerative Diseases.

Neurodegenerative diseases (NDs) such as Alzheimer's disease (AD), Parkinson's disease (PD), Huntington's disease (HD), and amyotrophic lateral sclerosis (ALS) are characterized by progressive neuronal loss, leading to severe cognitive and motor dysfunction. Benzimidazole, a privileged heterocyclic scaffold, has emerged as a promising pharmacophore in modulating key pathological targets across these disorders. In AD, benzimidazole derivatives inhibit cholinesterases, glycogen synthase kinase-3β (GSK-3β), and glutaminyl cyclase (QC), thereby addressing cholinergic dysfunction, tau phosphorylati

Defective efferocytosis links autonomic dysfunction and atrial fibrillation: multi-omics integration and in vivo validation.

INTRODUCTION: Atrial fibrillation (AF) is the most common and clinically significant arrhythmia requiring treatment. Although an imbalance between the sympathetic and vagal branches of autonomic nervous system (ANS) is known to trigger and sustain AF, the underlying mechanisms remain incompletely understood. Furthermore, there are yet no clinically authorized antiarrhythmic drugs currently targeting the modulation of ANS. METHODS: Targeted LC-MS/MS profiling of plasma 39 neurotransmitters was conducted in a clinical cohort. To identify shared mechanisms, atrial transcriptomes and a sympatheti

Neural state as a determinant of tumor immunity in lung cancer.

Immune checkpoint blockade has reshaped lung cancer therapy, yet durable benefit remains confined to a minority of patients, and resistance often emerges even in initially responsive disease. An increasingly coherent explanation is that neural state-encoded through hierarchical neuroimmune interactions-can profoundly recalibrate anti-tumor immunity and influence treatment responsiveness. In this Review, we synthesize evidence across three integrated levels of neuro-immune regulation in lung cancer. First, lung tumors engage in local neural-immune-tumor interactions, including synapse-like coup

Prolonged Light Exposure Induces Long-Lasting Retinal Wave Plasticity via Retrograde Melanopsin-Dopamine Signaling.

The spontaneous activity in the developing retina is necessary for the maturation of the neuronal circuitry in visual-associated brain areas. While previous studies have shown that the activation of intrinsically photosensitive retinal ganglion cells (ipRGCs) contributes to visual development, its effects and mechanisms remain largely unclear. Here, using microelectrode array recordings from both male and female mice, we demonstrated that prolong light exposure reduces the interwave interval and coupling distance of Stage 2 retinal waves, which can persist for at least 1 h after light exposure

Walnut Peptide KG-7 Alleviates Scopolamine-Induced Memory Deficits and Enhances Paracellular Transport via Tight Junction Modulation in a Mouse Model.

Walnut peptide Lys-Gly-His-Leu-Phe-Pro-Asn (KG-7) is a food-derived bioactive peptide with a high antioxidant capacity. We systematically evaluated the ameliorative effects of KG-7 on scopolamine-induced memory deficits in mice and its intestinal absorption mechanisms through integrating motion behavior analysis, molecular biochemistry research, and fluorescence imaging technology. Morris water maze tests revealed that KG-7 significantly improved the behavioral performance of these mice. Further mechanistic investigations demonstrated that KG-7 restored cholinergic function by reducing acetylc

Microinjection of kynurenic acid and lidocaine into the dorsal motor nucleus of vagus evokes airway smooth muscle relaxation in ferrets.

The dorsal motor nucleus of vagus (DMV) provides a critical source of parasympathetic motor output to the airway smooth muscle; however, the central neurotransmitter mechanisms that regulate this activity remain incompletely understood. This study investigated how blockade of excitatory glutamatergic transmission or transient functional suppression of the DMV modulates vagally mediated airway constriction in anaesthetized ferrets (n = 10), divided into two experimental groups (n = 5 per group). Animals were maintained under alpha-chlorose anesthesia and mechanically ventilated. The pressure in

Redox-active copper oxide nanoparticles trigger glial-centred neurotoxicity and developmental instability in Drosophila.

The use of copper oxide nanoparticles (CuO NPs) is increasing in biomedical applications and various industrial sectors. The increased use of these nanoparticles raises concerns regarding their potential neurotoxicological consequences on copper homeostasis. The present study established a CuO NP feeding-based Drosophila melanogaster model to investigate copper-induced toxicity with emphasis on glial-associated neurodegenerative mechanisms. The CuO NP of average size of ∼88 nm was synthesised in-house and exposed to flies through oral mode. The locomotor abnormality was investigated in larvae
