Enhanced biocontrol of postharvest blue mold in apple by phytopathogen targeted Bacillus subtilis 507.

Postharvest blue mold in apples caused by Penicillium expansum poses a great risk to food safety. Here, we report on a phytopathogen targeted enrichment strategy that led to the isolation of an endophytic biocontrol agent (BCA), Bacillus subtilis 507, with strong antagonism against P. expansum. Using a unique dual application approach that combines both live BCA and its crude metabolites (Bac + CM), our result showed an enhanced efficacy. Relative to the control, the disease incidence of blue mold in apples was reduced by 45.95% and lesion diameter was limited to 19.18 mm. Untargeted metabolom

Discovery of novel viruses related to the Potyviridae family reveals expanded diversity, a broad host range, and evidence of fungal and oomycete infections.

The family Potyviridae represents the largest and most economically important group of plant-infecting RNA viruses. Despite extensive study of crop-associated members, the full diversity, host range, and evolutionary history of potyvirids remain poorly understood. Here, we conducted a large-scale mining of publicly available RNA-seq datasets to systematically search for novel potyvirid sequences. This approach enabled the identification and assembly of 47 previously undescribed members of the family, distributed across 8 recognized genera and, importantly, putative new groups. Beyond expanding

Discovery and Characterization of Conserved N-Oxide-Forming Function of FAD-Dependent Monooxygenases for Fungal Prenylated Indole Alkaloid Biosynthesis.

N-oxides are ubiquitous functional groups in natural products, conferring critical biological properties such as enhanced solubility and modulated membrane permeability. However, the biosynthetic pathways for N-oxide formation in complex prenylated indole alkaloids (PIAs), especially the penicimutamides, remain poorly understood. Here, we describe the discovery and biochemical characterization of new activities of FAD-dependent monooxygenase (FMO) PldC, which sequentially catalyzes the indole 2,3-epoxidation-rearrangement of penicimutamide E, and the regioselective N13-oxidation of penicimutam

Influence of environmental and nutritional conditions on Nakaseomyces glabratus biofilm development in vitro.

Biofilm formation is a major virulence trait in Nakaseomyces glabratus, contributing to antifungal tolerance and persistent infections. Environmental and nutritional parameters influence fungal biofilm development, but standardized experimental frameworks for evaluating N. glabratus biofilm modulation remain limited. This study investigated how defined environmental conditions, including incubation time, agitation, and carbon source supplementation modulate biofilm development under controlled in vitro conditions. Biofilms were formed for 24-72 h under static (0 rpm) and agitated (120 rpm) con

Ecological Management of Crown Gall-Affected Soils: Pathogen Reservoirs, Disease-Suppressive Microbiomes, and Helper Consortia.

Crown gall remains difficult to manage because pathogenic agrobacteria can persist in soil, rhizosphere niches, infected roots, plant residues, and propagative materials, while repeated wounding creates new infection sites. This review examines how these reservoirs interact with soil and plant-associated microbiota to shape disease recurrence. Crown gall-specific studies show that gall and graft-union communities are spatially structured and identify pathogen competition, opine use, and virulence-related functions as processes that may shape disease-associated microbial assemblages. Evidence f

Living co-culture fabrication for biologically crosslinked mycelium-cellulose hydrogels and films.

Living materials, owing to their inherent capacities for growth, self-healing, sensing, and adaptation, have attracted widespread attention in recent years. However, controlling growth of living matter to achieve robust and tunable material properties remains a challenge. Here, we report a co-culture strategy that integrates mycelial microfibers with bacterial cellulose nanofibers into hierarchical hydrogels and transparent films. The biological crosslinking between fungal cell wall mannans and cellulose chains yields an interpenetrating micro-nano network with enhanced interfacial hydrogen bo

Synthesis, Characterization, and Evaluation of the Antifungal Properties of 3-Indolyl-3-Hydroxy Oxindole Derivatives Against Plant Pathogenic Fungi.

To discover novel fungicides with good inhibitory effects on plant fungal diseases, twenty-five 3-indolyl-3-hydroxy oxindole derivatives (3a-3y) were synthesized. These newly derivatives were characterized by NMR and HRMS. Their antifungal activities against five plant pathogenic fungi were assessed in vitro. Most of the compounds exhibited moderate to excellent antifungal activities against the five pathogenic fungi. Notably, compounds 3t, 3u, 3v, and 3w displayed remarkable and broad-spectrum antifungal activities comparable to or superior to those of the fungicides carvacrol (CA) and phenaz

Inhibitors of Type II NADH Dehydrogenase Enzyme: A Review.

Mitochondria are organelles in eukaryotic organisms with an electron transport chain consisting of four complexes (i.e., CI, CII, CIII, and CIV) on the inner membrane, which have functions such as providing energy, electron transport, and generating proton gradients. NADH dehydrogenase type 2 (NDH-2), widely found in bacterial, plant, fungal and protist mitochondria, is a nonproton-pumping single-subunit enzyme bound to the surface of the inner mitochondrial membrane that partially replaces NDH-1. NDH-2 has a crucial role in the energy metabolism of pathogenic microorganisms, and the lack of N

Antifungal activity of ruthenium (II) complex combined with fluconazole against drug-resistant Candida albicans in vitro and its anti-invasive infection in vivo.

With the abuse of antibiotics and azoles, drug-resistant Candida albicans infections have increased sharply and are spreading rapidly, thereby significantly reducing the antifungal efficacy of existing therapeutics. Several patients die of fungal infections every year. Therefore, there is an urgent requirement to develop new drugs. Accordingly, we synthesized a series of polypyridyl ruthenium (II) complexes having the formula [Ru (NN)2 (bpm)] (PF6)2 (N-N = 2,2'-bipyridine) (bpy, in Ru1), 1,10-phenanthroline (phen, in Ru2), 4,7-diphenyl-1,10-phenanthroline (DIP, in Ru3) (bpm = 2,2'-bipyrimidine

Evaluation of Antibiotic Biodegradation by a Versatile and Highly Active Recombinant Laccase from the Thermoalkaliphilic Bacterium Bacillus sp. FNT.

Laccases are industrially relevant enzymes that have gained great biotechnological importance. To date, most are of fungal and mesophilic origin; however, enzymes from extremophiles possess an even greater potential to withstand industrial conditions. In this study, we evaluate the potential of a recombinant spore-coat laccase from the thermoalkaliphilic bacterium Bacillus sp. FNT (FNTL) to biodegrade antibiotics from the tetracycline, β-lactams, and fluoroquinolone families. This extremozyme was previously characterized as being thermostable and highly active in a wide range of temperatures (

Is the C-Terminal Domain an Effective and Selective Target for the Design of Hsp90 Inhibitors against Candida Yeast?

Improving the armamentarium to treat invasive candidiasis has become necessary to overcome drug resistance and the lack of alternative therapy. In the pathogenic fungus Candida albicans, the 90-kDa Heat-Shock Protein (Hsp90) has been described as a major regulator of virulence and resistance, offering a promising target. Some human Hsp90 inhibitors have shown activity against Candida spp. in vitro, but host toxicity has limited their use as antifungal drugs. The conservation of Hsp90 across all species leads to selectivity issues. To assess the potential of Hsp90 as a druggable antifungal targ

Natural phytoalexins inspired the discovery of new biphenyls as potent antifungal agents for treatment of invasive fungal infections.

With the aim of discovering novel and effective antifungal agents derived from natural sources, a series of new biphenyls based on natural biphenyl phytoalexins were designed, synthesized and evaluated for their antifungal activities against four invasive fungi. By modifying the two benzene rings of noraucuparin, a well-known biphenyl phytoantitoxin, some promising compounds with remarkable antifungal activity were discovered. Notably, compounds 23a, 23e and 23h exhibited potent activities and a broad antifungal spectrum with low MICs of 0.25-16 μg/mL, which were 8-256-fold more potent than th

PEGylation of a shell over core-shell MOFs-a novel strategy for preventing agglomeration and synergism in terms of physicochemical and biological properties.

We demonstrate a new strategy of PEGylation over core-shell MOFs of HKUST-1 and Cu-MOF-2 by a solvothermal method. The novel synthesized PEGylated core-shell MOFs has synergistic enhancement in terms of physicochemical and biological properties. FTIR spectroscopy and XRD analysis described the bonding characteristics of the double-shelled-core MOFs PEG@HKUST-1@CuMOF-2 and PEG@CuMOF-2@HKUST-1. XPS and EDAX spectroscopy confirmed the structural features of the PEG@core-shell MOFs. The as-synthesized PEG-modified core-shell MOFs showed a readily identifiable morphology with a reduction in particl

Bacterial community dynamics on bats and the implications for pathogen resistance.

The bats skin microbiota plays an important role in reducing pathogen infection, including the deadly fungal pathogen Pseudogymnoascus destructans, the causative agent of white-nose syndrome. However, the dynamic of skin bacterial communities response to environmental perturbations remains poorly described. We characterized skin bacterial community over time and space in Rhinolophus ferrumequinum, a species with high resistance to the infection with P. destructans. We collected environmental covariate data to determine what factors influenced changes in community structure. We observed signifi

Natural Dibenzo-α-Pyrones: Friends or Foes?

Natural dibenzo-α-pyrones (DAPs) can be viewed from two opposite angles. From one angle, the gastrointestinal metabolites urolithins are regarded as beneficial, while from the other, the emerging mycotoxin alternariol and related fungal metabolites are evaluated critically with regards to potential hazardous effects. Thus, the important question is: can the structural characteristics of DAP subgroups be held responsible for distinct bioactivity patterns? If not, certain toxicological and/or pharmacological aspects of natural DAPs might yet await elucidation. Thus, this review focuses on compar
