IgE Reactivity of Recombinant Pac c 3 from the Asian Needle Ant (Pachycondyla chinensis).

BACKGROUND: Stings from the Asian needle ant are an important cause of anaphylaxis in East Asia. A 23-kDa protein homologous to antigen 5 is the major allergen produced by these ants. In this study, we aimed to produce a recombinant antigen 5 allergen, Pac c 3. METHODS: Recombinant Pac c 3 allergen from the Asian needle ant was expressed in Pichia pastoris and purified by ammonium sulfate precipitation and Ni affinity chromatography. IgE reactivity was demonstrated by ELISA and immunoblotting. RESULTS: The recombinant protein was recognized in 5 of 6 (83.3%) serum samples from patients with

Insecticide resistance of Anopheles sinensis and An. vagus in Hainan Island, a malaria-endemic area of China.

BACKGROUND: Malaria is one of the most important public health problems in Southeast Asia, including Hainan Island, China. Vector control is the main malaria control measure, and insecticide resistance is a major concern for the effectiveness of chemical insecticide control programs. The objective of this study is to determine the resistance status of the main malaria vector species to pyrethroids and other insecticides recommended by the World Health Organization (WHO) for indoor residual sprays. METHODS: The larvae and pupae of Anopheles mosquitoes were sampled from multiple sites in Hainan

[Molecular approaches to the control of onchocerciasis in Mexico].

Onchocerciasis is one of the major causes of blindness in the World, with about 17.7 million infected, particularly in West Africa. In Mexico, onchocerciasis is also present and has been subjected to control since 1923. The standard diagnosis of onchocerciasis is by the detection of microfilariae by skin biopsy and transmission is evaluated by detection of Onchocerca volvulus larvae in the vector. Classically, this was carried out by manual dissection of Simuliumn ochraceun s.l. However, with the use of ivermectin, a drug that kills microfilariae but not the adult worms, the skin biopsy is bec

Biocidal action, characterization, and molecular docking of Mentha piperita (Lamiaceae) leaves extract against Culex quinquefasciatus (Diptera: Culicidae) larvae.

Mosquitoes are found in tropical and subtropical areas and are the carriers of a variety of diseases that are harmful to people's health. e.g., malaria, filariasis, chikungunya, dengue fever, etc. Although several insecticides are available, however, due to insect resistance and environmental hazards, more eco-friendly chemicals are needed for insect control. So, the current research was planned to explore the prospective of Mentha piperita to be used for the formulation of larvicides against mosquito Culex quinquefasciatus. The ethanolic and water extracts of M. piperita leaves were prepared …

The complete mitochondrial genome of Acanthaspis geniculata (Hsiao, 1976) (Hemiptera: Reduviidae) and its phylogenetic analyses.

The genus Acanthaspis (Hemiptera: Reduviidae) comprises predatory insects of ecological importance, yet mitochondrial genomic resources for this group remain limited. Here, we present the complete mitochondrial genome of Acanthaspis geniculata (Hsiao, 1976). The mitogenome is 16,226 bp in length and contains the standard set of 37 mitochondrial genes (13 protein-coding genes, 2 rRNA genes, and 22 tRNA genes), along with a control region (D-loop). The nucleotide composition is A (40.4%), T (29.6%), C (17.9%), and G (12.1%), with an overall AT content of 70.0%. Phylogenetic analyses based on par

Mathematical modelling and release thresholds of transgenic sterile mosquitoes.

We formulate simple differential equation models to study the impact of releases of transgenic sterile mosquitoes carrying a dominant lethal on mosquito control based on the modified sterile insects technique. The early acting bisex, late acting bisex, early acting female-killing, and late acting female-killing lethality strategies are all considered. We determine release thresholds of the transgenic sterile mosquitoes, respectively, for these models by investigating the existence of positive equilibria and their stability. We compare the model dynamics, in particular, the thresholds of the mo

An Overview on the Potential Hazards of Pyrethroid Insecticides in Fish, with Special Emphasis on Cypermethrin Toxicity.

Pesticides are chemicals used to control pests, such as aquatic weeds, insects, aquatic snails, and plant diseases. They are extensively used in forestry, agriculture, veterinary practices, and of great public health importance. Pesticides can be categorized according to their use into three major types (namely insecticides, herbicides, and fungicides). Water contamination by pesticides is known to induce harmful impacts on the production, reproduction, and survivability of living aquatic organisms, such as algae, aquatic plants, and fish (shellfish and finfish species). Note: High-speed optical tracking of a flying insect.

We developed a video recording system with the capability of tracking moving objects and used it to track the flight of an insect. The system consists of two galvano mirrors, which redirect the light coming from the object in two orthogonal directions toward a high-speed camera to capture the image. An additional high-speed camera, which views the same object through a beam splitter placed between one of the galvano mirrors and the observation camera, detects the position of the object. The mirror angle is controlled to maintain the position of the object at the center of the view, allowing th

Functional analysis of genes in Aedes aegypti embryos.

Blood-feeding mosquitoes, including the dengue and yellow fever vector Aedes aegypti, transmit many of the world's deadliest diseases. Such diseases have resurged in developing countries and pose clear threats for epidemic outbreaks in developed countries. Recent mosquito genome projects have stimulated interest in the potential for arthropod-borne disease control by genetic manipulation of vector insects, and genes that regulate development are of particular interest. In recent years, RNA interference (RNAi) has proven to be an effective strategy for inhibiting gene function in many organisms

Plague.

Bubonic plague is an often fulminant systemic zoonosis, caused by Yersinia pestis. Conventional microbiology, bacterial population genetics, and genome sequence data, all suggest that Y pestis is a recently evolved clone of the enteric pathogen Yersinia pseudotuberculosis. The genetic basis of this organism's rapid adaptation to its insect vector (the flea) with transmission between mammalian hosts by novel subcutaneous and pneumonic routes of infection is becoming clearer. This transition provides a paradigm for the way in which new pathogens could emerge. Plague in humans is controlled by su

knot is required for the hypopharyngeal lobe and its derivatives in the Drosophila embryo.

The genetic control of segmentation in the Drosophila larval head has been only partially elucidated. We report that the knot (kn) gene of Drosophila is required for the formation of the hypopharyngeal lobe in the germ-band stage embryo and the ventral arms and lateralgräte of the larval head skeleton. Rearrangement mutations of knot disrupt the previously characterized gene, collier, which encodes a COE-family transcription factor. kn is required for the activation of cnc (cap'n'collar, a bZIP homeotic selector gene) in the progenitors of hypopharyngeal lobe. kn is also required for the activ

An integrated overview of the midgut bacterial flora composition of Phlebotomus perniciosus, a vector of zoonotic visceral leishmaniasis in the Western Mediterranean Basin.

BACKGROUND: The Leishmania developmental life cycle within its sand fly vector occurs exclusively in the lumen of the insect's digestive tract in the presence of symbiotic bacteria. The composition of the gut microbiota and the factors that influence its composition are currently poorly understood. A set of factors, including the host and its environment, may influence this composition. It has been demonstrated that the insect gut microbiota influences the development of several human pathogens, such as Plasmodium falciparum. For sand flies and Leishmania, understanding the interactions betwee

Evaluation of the protective efficacy of a transfluthrin-based spatial repellent product to reduce malaria prevalence in Uganda: study protocol for a cluster-randomised double-blinded control trial-the Mossie-GO trial.

BACKGROUND: Progress towards elimination and eventual eradication of malaria is threatened by challenges such as the rise in insecticide resistance and low coverage of existing vector control tools. Spatial repellents offer personal and household protection against biting mosquitoes by disseminating repellents into a given area. The trial described here aims to evaluate the efficacy of an active transfluthrin-based spatial repellent device (Mossie-GO™) against malaria in Uganda, using a placebo-controlled, double-blinded cluster randomised control trial. The study's primary objective is to dem

Hexamerin a novel protein associated with Bacillus sphaericus resistance in Culex quinquefasciatus.

Bacterial insecticides like, Bacillus sphaericus and Bacillus thuringiensis serovar israelensis, have been used for the control of nuisance and vector mosquitoes for more than two decades. For many years, it was assumed that the use of microbial larvicides based on B. sphaericus would not lead to resistance in mosquitoes. However, recent reports have shown that B. sphaericus toxins are not free from this problem. Therefore, the resistance of mosquito populations to be will seriously threaten the sustainability of current mosquito control programme using these microbial insecticides. In the pre

Resistance of Mexican fruit fly to quarantine treatments of high hydrostatic pressure combined with heat.

High-pressure processing (HPP) has been proposed as an alternative quarantine method against the Mexican fruit fly Anastrepha ludens Loew (Diptera: Tephritidae), which is one of the most important pests infesting mangoes, citrus, and other fruits in Mexico and other Latin-American countries. However, processing conditions used to destroy eggs and larvae also affect the shelf life of fruits. The objective of this study was to assess the biological viability of A. ludens eggs treated with HPP, establishing whether undestroyed eggs were able to develop larvae, pupae, and adults capable of reprodu

Distinct Roles of Transketolase (TktA) and Transaldolase (talB) in Metabolism, Biofilm Formation, and Flea Colonization in Yersinia pestis.

The flea-borne transmission of Yersinia pestis relies on biofilm formation and metabolic adaptation within the insect gut. The pentose phosphate pathway (PPP) is central to these processes, yet the contribution of its non-oxidative branch remains poorly defined. Here, we investigated the roles of transketolase (TktA) and transaldolase (TalB) in plague bacillus physiology, metabolism, and flea colonization. TktA was essential for growth, preventing assessment of its role in biofilm formation and in vivo colonization. In contrast, TalB was dispensable for growth but required for optimal biofilm

A temperature-sensitive CRISPR-Cas12a system for sterile insect technique.

The sterile insect technique (SIT) reduces population numbers by releasing sterile males that produce non-viable progeny. Specifically, CRISPR/Cas9-based precision-guided SIT (pgSIT) generates sterile males through genetic crosses of two transgenic lines: a Cas9 strain and a guide RNA (gRNA) strain targeting male sterility and female viability or infertility. However, pgSIT requires separate maintenance of the two lines and sorting to obtain sterile males, creating possible challenges for scaling. To overcome this, we propose using Cas12a nuclease, which is inoperative at lower temperatures bu

Functional Characterization of an Odorant Receptor in the Brown Planthopper, Nilaparvata lugens, Tuned to 2-Dodecanone.

Insect olfactory systems detect environmental volatiles to guide key behaviors. The specific binding of odorants to olfactory neurons mediated by odorant receptors (ORs) constitutes the fundamental basis of olfaction. The brown planthopper (BPH, Nilaparvata lugens) is a major global rice pest; however, there have been few studies on the function of BPH NlugORs. Here, we conducted transcriptome sequencing of adult and nymph antennae with differential expression analysis and quantitative real-time PCR (qPCR) validation to screen candidate OR genes. We functionally characterized three NlugORs (Nl

Histological investigation of the effects of fenoxycarb on neurosecretory cells in the silkworm, Bombyx mori brain.

Fenoxycarb 0-ethyl N-(2-(4-pheoxyphenoxy)-ethyl) carbamate is the most potent juvenile hormone analogue against a variety of insect species including the silkworm Bombyx mori. In this study, the effects of fenoxycarb on silkworm Bombyx mori brain neurosecretory cells in 5th instar were investigated. Fenoxycarb (1 ng/10 µl) was applied topically along the dorsa-medial line to the animals in the spinning behavior on day 1 of the experimental group. Brains removed by dissection were histologically examined by hematoxylin eosin (hem&eosin) and paraldehyde fuchsin staining. Three types of neurosecr
