Plasma metabolomics combined with machine learning for postoperative prognostic stratification in hormone receptor-positive/human epidermal growth factor receptor 2-negative early breast cancer.

BACKGROUND AND AIMS: Hormone receptor-positive/human epidermal growth factor receptor 2-negative (HR+/HER2-) breast cancer (BC) accounts for the majority of BC cases. Although early-stage patients generally have favorable outcomes, recurrence and metastasis substantially worsen prognosis. We aimed to evaluate whether plasma metabolomics combined with machine learning could predict postoperative outcomes in HR+/HER2- early BC. METHODS: A total of 178 patients with HR+/HER2- BC were prospectively enrolled. Preoperative plasma samples were analyzed using untargeted metabolomics based on liquid c

Glycemia Range and Offspring Weight and adiposity in response To Human milk (GROWTH) study: protocol for an observational cohort designed to study lactational programming.

INTRODUCTION: Maternal human milk feedings continue an offspring's exposure to the programming stimuli of maternal metabolism during the postnatal period. While considerable research focuses on associations between in utero environments and offspring metabolic disease, few studies have been able to specifically measure how human milk composition modifies programming of children's growth in conjunction with comprehensive measures of maternal glycaemia during pregnancy. METHODS AND ANALYSIS: The Glycemia Range and Offspring Weight and adiposity in response To Human milk (GROWTH) Study is a long

Metabolomics integrated with network pharmacology and serum-urine pharmacochemistry unveils the antidiabetic mechanism of Anemarrhenae Rhizoma.

OBJECTIVE: Anemarrhenae Rhizoma (AR) is a traditional Chinese medicine widely used for the treatment of type 2 diabetes mellitus (T2DM). However, the specific bioactive constituents responsible for its in vivo effects and their underlying mechanisms of action remain unclear. We hypothesise that serum-absorbed and metabolised AR components modulate key metabolic and inflammatory pathways in T2DM. To test this hypothesis, this study employs an integrated strategy combining metabolomics with serum-urine pharmacochemistry and network pharmacology to systematically identify AR's active constituents

Low-Carbon Fabrication of Fluorine-Free Superhydrophobic Copper-Based Materials for Acquiring Freshwater.

By utilizing the etching reaction between a hydrophobic copper mesh (CuM) and an alkali solution to obtain a superhydrophilic copper mesh (AT CuM), and then only depending on the modification of linoleic acid (LA) at room temperature, a fluorine-free superhydrophobic copper-based material (CuM@Cu(OH)2@LA) was fabricated. Subsequently, a funnel-shaped fog collector was constructed by simple folding for fog collection, which collected 10 400 mg of water with a water-harvesting rate (WHR) of 13.9 mg min-1 cm-2 after 30 min. Meanwhile, the AT CuM was deposited with polydopamine to obtain a phototh

Therapeutic Effect of Brucea Javanica Oil Emulsion in Mice with Irinotecan-Induced Delayed Diarrhea.

BACKGROUND: Chemotherapy-induced diarrhea (CID), particularly delayed diarrhea, often limits clinical use. Brucea javanica oil emulsion (BJOE), an adjuvant chemotherapy agent, has been shown to reduce irinotecan-related gastrointestinal side effects. However, its underlying molecular mechanism remains unclear. The cGAS-STING pathway, composed of the cytosolic DNA sensor cyclic GMP-AMP synthase (cGAS) and the adaptor protein stimulator of interferon genes (STING), plays an essential role in delayed diarrhea. This work aimed to investigate the therapeutic potential and underlying mechanism of BJ

Modulation of lipid metabolism through multiple pathways during oocyte maturation and embryo culture in bovine.

Lipid accumulation occurs in cultured embryos and is associated with reduced cryotolerance. Here we report the use of a multiple pathway lipid modulator cocktail (l-carnitine, linoleic acid and forskolin) to improve cryosurvival. First, we stained oocytes and embryos with Oil Red to examine the time course of lipid accumulation during in vitro fertilization (IVF) and embryo culture. Then we evaluated the effects of the lipid modulators cocktail on lipid content, developmental rates and survival after vitrification. In our conditions, lipid accumulation was detected (P < 0.05) at the end of in

Emamectin benzoate induced enzymatic and transcriptional alternation in detoxification mechanism of predatory beetle Paederus fuscipes (Coleoptera: Staphylinidae) at the sublethal concentration.

In this study, the detoxification enzyme activity and the transcriptional profile changes in the second instar through RNA-sequencing technology due to emamectin benzoate (EB) were assessed. The cytochrome P450 monooxygenases (P450) enzyme activity was not altered by EB due to the change in concentration and exposure time in all treatments. The glutathione S-transferase (GST) enzyme was not considerably varying in all treatments, while exposure time significantly changed the enzyme activity. Results showed that the esterase (Ests) activity was elevated with the increasing concentrations and ex

Nitrite elicits divergent NO-dependent signaling that associates with outcome in out of hospital cardiac arrest.

Brain and heart injury cause most out-of-hospital cardiac arrest deaths but limited pharmacotherapy exists to protect these tissues. Nitrite is a nitric oxide precursor that is protective in pre-clinical models of ischemic injury and safe in Phase I testing. Protection may occur by cGMP generation via the sGC pathway or through S-nitrosothiol and nitrated conjugated linoleic acid (NO2-CLA) formation. We hypothesized that nitrite provided during CPR signals through multiple pathways and that activation of signals is associated with OHCA outcome. To this end, we performed a secondary analysis of

Meconium fatty acid ethyl esters as biomarkers of late gestational ethanol exposure and indicator of ethanol-induced multi-organ injury in fetal sheep.

BACKGROUND: Meconium fatty acid ethyl esters (FAEE) constitute a biomarker of heavy fetal ethanol exposure. Our objective was to measure meconium FAEE in fetal sheep following daily, relatively moderate-dose ethanol exposure in late gestation, and to evaluate their utility in identifying fetal organ-system injury. METHODS: Pregnant ewes received ethanol (0.75 g/kg; n = 14) or saline (n = 8) via 1-h i.v. infusion daily during the third trimester equivalent, while additional pregnant sheep served as untreated controls (n = 6). The daily ethanol regimen produced similar maximal maternal and feta

Prevalence of fetal ethanol exposure in a regional population-based sample by meconium analysis of fatty acid ethyl esters.

Challenges in identifying children exposed prenatally to ethanol necessitate the development of a biomarker for neonates at risk for fetal alcohol spectrum disorder. Meconium fatty acid ethyl esters (FAEE), products of nonoxidative ethanol metabolism, have been established as a novel biomarker of fetal ethanol exposure. We present the first application of this biomarker to a population-based sample in Canada. Six-hundred eighty-two meconium specimens were anonymously collected in the region of Grey Bruce, Ontario, Canada. Meconium FAEE were extracted by liquid-liquid and solid-phase extraction

Association between gestational diabetes mellitus and long-term all-cause and cause-specific mortality risks: a prospective cohort study based on metabolomics.

BACKGROUND: Gestational diabetes mellitus (GDM) is one of the most common metabolic disorders during pregnancy. Women with GDM face an increased risk of type 2 diabetes and cardiovascular disease (CVD) after childbirth, even after accounting for obesity, lifestyle, and socioeconomic factors. However, whether GDM raises long-term all-cause and cause-specific mortality remains uncertain. This study aimed to clarify the long-term links between GDM-related metabolic profiles and mortality using nuclear magnetic resonance (NMR) metabolomics. METHODS: Using data from the UK Biobank (UKB) prospective
