Determining Intermediate Hosts for Opecoelidae and Microphallidae Species (Platyhelminthes: Trematoda) in the Southeastern Pacific Coast, Using Molecular Markers.

Metacercarial stages of digeneans were collected from decapod crustaceans inhabiting intertidal rocky zones in central Chile. The digeneans were identified through a molecular analysis based on the V4 region of the 18S ribosomal RNA gene. We analyzed 356 crustaceans belonging to 10 species, 115 intertidal fish belonging to 6 species, and 4 specimens of 1 coastal bird species. In total, 74.1% of crustaceans were parasitized with metacercariae. We found 1 species of Opecoelidae. This species showed low genetic divergence (0% and 0.1%) with adult digeneans found in intertidal fish and with the sp

Bovine and ovine rumen fluke in Ireland-Prevalence, risk factors and species identity based on passive veterinary surveillance and abattoir findings.

The prevalence of rumen fluke, the incidence of clinical paramphistomosis and the trematode's species identity were studied in cattle and sheep in the Republic of Ireland using passive veterinary surveillance (faecal examination and necropsy results; 2010-2013) and abattoir data. Based on faecal examination, the prevalence of rumen fluke was higher in cattle than in sheep. Rumen fluke prevalence in cattle and sheep fluctuated over the year and in most years (2011-2013), prevalence was higher in winter (December-February) than in summer (June-August). For 3 of 4 years studied, there was no corr

Active chi-like sequences are present in the ITS1 region of polyembryonic adult Collyriclum faba trematodes encysted in pairs.

Collyriclum faba (Plagiochiida: Collyriclidae) adults occur in pairs within subcutaneous cysts. Here, we tested the extensive C. faba infrapopulation for five DNA loci known to display variability among Central European C. faba individuals. The infrapopulation tested shared 100% similarity in four of the five mitochondrial and nuclear DNA loci tested. Contrariwise, the internal transcribed spacer 1 (ITS1) loci in all but one individual differed from each other. We found only 0.0-1.5 base substitutions per 1,000 sites within the cysts, while we found 0.7-9.0 substitutions between the cysts of t

Persistence of a recurrent group of intestinal helminth species in a coyote population from southern Texas.

The persistence and interrelationships of a recurrent group of intestinal helminth species in a coyote population from southern Texas were determined; data were collected at 4-yr intervals (1979, 1983, and 1987). The a priori expectation was that a defined recurrent group of helminth species persisted over time and that the respective helminth species were stable in terms of their abundances. The Fager index and subsequent recurrent group analysis indicated a small group of taxonomically unrelated recurring species of helminths that always demonstrated affinities with each other and occasional

Human fascioliasis emergence in southern Asia: Complete nuclear rDNA spacer and mtDNA gene sequences prove Indian patient infection related to fluke hybridization in northeastern India and Bangladesh.

Fascioliasis is a snail-borne zoonotic disease with impact on the development of human subjects and communities. It is caused by two liver-infecting fasciolid trematode species, the globally-distributed Fasciola hepatica and the Africa/Asia-restricted but more pathogenic, larger F. gigantica. Fasciola gigantica is the cause of endemicity in livestock throughout the warm lowlands from Pakistan to southeastern Asia since old times. Human fascioliasis is emerging in this region at present, with an increase of patient reports. Complete sequences of rDNA ITS-1 and ITS-2 spacers and mtDNA nad1 and c

PHILOPHTHALMUS HECHINGERI N. SP. (DIGENEA: PHILOPHTHALMIDAE), A HUMAN-INFECTING EYE FLUKE FROM THE ASIAN MUD SNAIL, BATILLARIA ATTRAMENTARIA.

Two cases of human philophthalmosis have been reported in Japan. Gravid flukes removed from the eyes of the patients were broken, but their morphological characteristics suggest that an unknown species of the genus Philophthalmus is involved as a pathogen for humans. The mitochondrial DNA barcode of the human eye fluke enabled us to discover its larval stage from the Japanese mud snail, Batillaria attramentaria. The discovered cercaria had previously been temporarily described as "Philophthalmid sp. I." In this study, we examined the infection status of B. attramentaria with Philophthalmid sp.

Integrative morphological and molecular characterization of Proterodiplostominae metacercariae (Digenea: Diplostomidae) infecting siluriform fishes in the Peruvian Amazon Basin.

Proterodiplostominae Dubois, 1936 is a small but evolutionarily significant lineage of diplostomid trematodes that primarily parasitize crocodilians, with fishes acting as second intermediate or paratenic hosts. In Peru, knowledge of this group has been limited to the record of adult specimens of Proterodiplostomum longum (Brandes, 1888) recovered from Caiman crocodilus Linnaeus, 1758 (Sauropsida: Alligatoridae) in the Loreto region, and molecular evidence linking larval and adult stages has been absent. The present study provides the first integrative morphological and molecular characterizat

Innovating stomach fluke identification: an integrative approach combining Micro-CT imaging and molecular tools.

The rapid loss of biodiversity driven by anthropogenic pressures highlights the urgent need for improved species identification methods. Parasites, vital ecosystem regulators, are being lost at disproportionate rates, with amphistomes-a broadly distributed group of trematode parasites, infecting all major vertebrate groups-facing significant challenges. Many amphistome species remain undescribed, and reference sequences for known species are scarce, partly due to the reliance on labour-intensive identification methods, such as Scanning Electron Microscopy (SEM) and median sagittal sections. Human schistosomiasis in Nigeria: present status, diagnosis, chemotherapy, and herbal medicines.

Schistosomiasis is a neglected tropical disease caused by a parasitic, trematode blood fluke of the genus Schistosoma. With 20 million people infected, mostly due to Schistosoma haematobium, Nigeria has the highest burden of schistosomiasis in the world. We review the status of human schistosomiasis in Nigeria regarding its distribution, prevalence, diagnosis, prevention, orthodox and traditional treatments, as well as snail control strategies. Of the country's 36 states, the highest disease prevalence is found in Lagos State, but at a geo-political zonal level, the northwest is the most endem

Genome of the ramshorn snail Biomphalaria straminea-an obligate intermediate host of schistosomiasis.

BACKGROUND: Schistosomiasis, or bilharzia, is a parasitic disease caused by trematode flatworms of the genus Schistosoma. Infection by Schistosoma mansoni in humans results when cercariae emerge into water from freshwater snails in the genus Biomphalaria and seek out and penetrate human skin. The snail Biomphalaria straminea is native to South America and is now also present in Central America and China, and represents a potential vector host for spreading schistosomiasis. To date, genomic information for the genus is restricted to the neotropical species Biomphalaria glabrata. This limits und

First morphogenetic analysis of parasite eggs from Schistosomiasis haematobium infected sub-Saharan migrants in Spain and proposal for a new standardised study methodology.

Schistosomiasis is a Neglected Tropical Disease caused by trematode species of the genus Schistosoma. Both, autochthonous and imported cases of urogenital schistosomiasis have been described in Europe. The present study focuses on eggs, considered pure S. haematobium by genetic characterisation (intergenic ITS region of the rDNA and cox1 mtDNA). A phenotypic characterisation of S. haematobium eggs was made by morphometric comparison with experimental populations of S. bovis and S. mansoni, to help in the diagnosis of S. haematobium populations infecting sub-Saharan migrants in Spain. Analyses

Host-parasite life-histories of the diurnal vs. nocturnal chronotypes of Schistosoma mansoni: adaptive significance.

OBJECTIVES: To optimise host-to-host transmission, digenean trematodes (parasites) synchronise their cercarial emission patterns with the aquatic activities of their vertebrate hosts. Schistosoma mansoni has two different circadian chronotypes: a diurnal shedding pattern with a mean peak occurring at 11:00 h, and a nocturnal shedding pattern with a mean peak occurring at 20:00 h. We analysed the life-history variations between these two chronotypes at the levels of the parasite and its hosts. METHODS: For each chronotype, we quantified three life-history traits related to the parasite (prepat

Sequence analysis and characterization of pyruvate kinase from Clonorchis sinensis, a 53.1-kDa homopentamer, implicated immune protective efficacy against clonorchiasis.

BACKGROUND: Clonorchis sinensis, the causative agent of clonorchiasis, is classified as one of the most neglected tropical diseases and affects more than 15 million people globally. This hepatobiliary disease is highly associated with cholangiocarcinoma. As key molecules in the infectivity and subsistence of trematodes, glycolytic enzymes have been targets for drug and vaccine development. Clonorchis sinensis pyruvate kinase (CsPK), a crucial glycolytic enzyme, was characterized in this research. RESULTS: Differences were observed in the sequences and spatial structures of CsPK and PKs from h

Schistosomal-derived lysophosphatidylcholine triggers M2 polarization of macrophages through PPARγ dependent mechanisms.

Mansonic schistosomiasis is a disease caused by the trematode Schistosoma mansoni, endemic to tropical countries. S. mansoni infection induces the formation of granulomas and potent polarization of Th2-type immune response. There is great interest in understanding the mechanisms used by this parasite that causes a modulation of the immune system. Recent studies from our group demonstrated that lipids of S. mansoni, including lysophosphatidylcholine (LPC) have immunomodulatory activity. In the present study, our aim was to investigate the role of lipids derived from S. mansoni in the activation

Insights about echinostomiasis by paleomolecular diagnosis.

Echinostomiasis is a zoonosis caused by intestinal trematodes and transmitted by the ingestion of mollusks, crustaceans, fish, amphibians, and reptiles, either raw or poorly cooked. Today human infection is endemic in Southeast Asia and the Far East, but has been reported more recently in other regions of the world. Interestingly eggs identified as Echinostoma sp. were found in coprolites from a mummified body human in Brazil, dated 560 ± 40 BP (before present). However, the specific diagnosis based on morphology of the eggs has not been resolved at the species level. As a follow-up to the pre

Relationships between eosinophilia, anti-Fasciola IgG, and IgM rheumatoid factors, in urban and rural areas of north-western Spain.

A study was carried out, among adult patients attending a hospital in Lugo, in north-western Spain, to investigate possible relationships between eosinophilia, IgG antibodies against the parasitic helminth Fasciola hepatica, and IgM rheumatoid factors (IgM-RF). Blood samples were collected from 1264 individuals and divided into three groups according to eosinophil count: normal (<0.5 x 10(9) eosinophils/litre), eosinophilic (0.5 x 10(9)-3 x 10(9)/litre) or hyper-eosinophilic (>3 x 10(9)/litre). Each sample was checked for IgG against F. hepatica, in an ELISA in which the excretory/secretory pr
