α1-acid Glycoprotein mitigates MASLD progression by modulating liver and adipose tissue function.

Dysregulation of hepatokines has been observed in patients with metabolic dysfunction-associated steatotic liver disease (MASLD), suggesting their involvement in disease progression. In this study, we aimed to investigate the role of α1-acid glycoprotein (AGP), a hepatokine, in the progression of MASLD and to evaluate its potential therapeutic utility. Reanalysis of hepatic RNA sequencing datasets from patients with fatty liver disease showed that expression of the hepatic AGP gene decreased with disease progression. A high-fat diet (HFD)-induced mouse model of MASLD also showed decreased plas

Integrin beta-like 1 protects against liver injury by inhibiting ferroptosis in metabolic dysfunction-associated steatohepatitis.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatohepatitis (MASH) is characterized by hepatic lipid accumulation, inflammation, and oxidative injury. However, the regulatory role of integrin beta-like 1 (ITGBL1) in MASH remains unknown. METHODS: ITGBL1 knockout (KO) and liver-specific overexpressing mice were fed an MCD diet for 4 weeks. In vitro, AML‑12 hepatocytes were treated with palmitic acid (PA), with ITGBL1 overexpression (OE) or knockdown (sgITGBL1). Liver injury, steatosis, inflammation, and ferroptosis markers were assessed by biochemical assays, histology, RNA‑seq, west

Liraglutide Modulates Zinc Release and Improves Mitochondrial Function in Insulin-Resistant Senescent Cardiomyocytes.

Aging and insulin resistance are intertwined factors in the development of metabolic diseases such as type 2 diabetes and cardiovascular disorders. Liraglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, has shown promising cardioprotective effects in preclinical and clinical studies of metabolic diseases. Yet, its action on insulin-resistant aged subjects is not clearly defined. This study aimed to investigate the effects of liraglutide on intracellular zinc levels, including its modulation of oxidative stress, mitochondrial function, and Endoplasmic Reticulum (ER) stress in a novel

Piperine inhibits biofilm formation and efflux activity and dysregulates lipid metabolism in Mycobacterium abscessus.

The nontuberculous Mycobacterium abscessus is a human pathogen that causes chronic lung infections and soft tissue infections. The bacterium forms biofilms and efflux pumps contribute to its tolerance of antibiotics. Efflux pumps also transport lipids and other molecules to the bacterial outer cell surface for biofilm formation. The effects of piperine, an alkaloid derived from black pepper, on biofilm formation, efflux activity and lipid biosynthesis in M. abscessus have not been reported. We report that, at sub-minimum inhibitory concentration levels, piperine inhibits biofilm formation in M

Melatonin impedes NCOA4/ACSL4-dependent ferroptosis by targeting STING to ameliorate diabetic cognitive dysfunction.

Melatonin (MLT) is a potentially effective therapeutic agent for mitigating brain injury under various pathological conditions. However, the molecular mechanisms of its ability to ameliorate diabetic cognitive dysfunction (DCD) require further elucidation. In this study, we demonstrated that MLT administration improved the learning and memory deficits in type 2 diabetes mellitus (T2DM) mice, concurrently attenuated acyl-CoA synthetase long-chain family member 4 (ACSL4) - dependent ferroptosis in diabetic brain tissue and neuronal cells exposed to high glucose/palmitic acid (HG + PA), as indica

Sappanone A, a potential natural inhibitor of PI3K, alleviates metabolic dysfunction-associated steatohepatitis in experimental models.

Metabolic dysfunction-associated steatohepatitis (MASH) affects a large proportion of the global population and is widely regarded as the fastest growing cause of hepatocellular carcinoma. Currently, approved therapeutic strategies for MASH are limited. Therefore, this study used the Connectivity Map (CMap) database to identify a candidate compound for MASH, evaluate its efficacy in experimental models, and explore its mechanism of action. Based on the gene expression profile of GSE126484, Sappanone A (SA) was screened using the CMap. In a palmitic acid-induced cell model, SA notably reduced t

Ethanol causes rapid decreases in the hepatic retinoid levels shaping the early steps of alcohol-associated liver disease.

BACKGROUND: Chronic alcohol drinking causes hepatic vitamin A (retinoids and derivatives) decreases, which correlate with the progression and severity of alcohol-associated liver disease (ALD). However, the effects of short-term ethanol (EtOH) intake on liver retinoids and ALD are still undefined. METHODS: Using high-performance liquid chromatography and high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC, HPLC-MS/MS), and molecular biology techniques in mice and cultured human hepatocytes, we investigated the temporal EtOH effects on retinoids and ALD. RESULTS

Pirin knockdown alleviates the progression of metabolic dysfunction-associated steatotic liver disease by inhibiting hepatic lipid deposition, inflammation, and fibrosis.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global disease with a broad spectrum of symptoms. Pirin (PIR) is involved in a variety of biological and molecular processes. However, the role of PIR in MASLD remains unclear. Rats were fed with high-fat and high-cholesterol (HFC) diet to induce MASLD. Time-course results showed that liver in HFC-fed rats exhibited a progressive increase in hepatic tissue damage over time (3, 6, and 9 weeks), and developed in obvious hepatic fibrosis and steatosis at week 9. Meanwhile, HFC feeding also gradually increased PIR expression in

Targeting ferroptosis and ferritinophagy improves tooth extraction socket healing in type 2 diabetes mellitus via human bone marrow mesenchymal stem cells modulation.

Delayed tooth extraction socket (TES) healing is a significant complication in patients with poorly controlled type 2 diabetes mellitus (T2DM). Ferroptosis, a regulated form of cell death driven by iron-dependent lipid peroxidation, has been implicated in various T2DM-induced pathologies; however, its role in TES healing remains unclear. This study aimed to investigate the mechanisms of ferroptosis induced by a high-glucose environment in T2DM, focusing on its effects on human bone marrow mesenchymal stem cells (HBMSCs), iron content, and ferritinophagy. Additionally, we evaluate the therapeut

A high-fat diet suppresses growth hormone synthesis and secretion by influencing the Vit D receptor and Pit1.

BACKGROUND: A long-term high-fat diet (HFD) leads to excessive lipid deposition, which may cause many diseases, including NAFLD, diabetes, and thyroid dysfunction. In addition, HFD leads to a decrease in serum growth hormone (GH) levels to further increase lipid deposition and obesity. However, the mechanism of such reduction of GH has not been fully elucidated. METHODS: Male Sprague-Dawley rats were fed a regular diet (CD) or a high-fat diet (HFD) for 29 weeks. GH synthesis and secretion were evaluated in pituitary and blood samples, respectively. An in vitro model was constructed by treatin

Effectiveness and safety of the SREBP1/2 inhibitor, fatostatin, in a preclinical model of metabolic dysfunction-associated steatotic liver disease progression.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a prevalent liver disorder that can progress to metabolic dysfunction-associated steatohepatitis (MASH), potentially leading to liver fibrosis, cirrhosis, and cancer. As there is no sufficient specific pharmacological treatment for the progression of MASLD to MASH, we focused on the study of master transcriptional regulators involved in energy, cholesterol and lipid metabolism and adipogenesis. Therefore, our experimental work aimed to investigate the possible hepatoprotective effects and safety of fatostatin, which is the inh

Puerarin reduces susceptibility to ventricular arrhythmias and inhibits ferroptosis via Sirt1/Nrf2 signaling in high-fat-diet rats.

Obesity is a significant risk factor for cardiac arrhythmias, and the ferroptosis is closely related to cardiac arrhythmias. This study aimed to investigate whether puerarin (Pue), a natural isoflavone, could reduce the susceptibility to ventricular arrhythmias (VAs) associated with obesity and inhibit ferroptosis, with a particular focus on the Sirt1/Nrf2 signaling pathway. Male rats were randomly divided into three groups: normal chow diet (NC), high-fat diet (HFD), and HFD with Pue treatment (100mg/kg, HFD + Pue). After 16 weeks, electrophysiological, structural, and molecular analysis were

Targeting ASK1 by CS17919 alleviates kidney- and liver-related diseases in murine models.

BACKGROUND: Apoptosis signal-regulating kinase 1 (ASK1) is a MAP3K kinase in the MAPK signaling pathway activated by stressors and triggers downstream biological effects such as inflammation and apoptosis; therefore, inhibition of ASK1 kinase activity can protect cells from pathological injury. In this study, we designed and synthesized a novel selective ASK1 inhibitor, CS17919, and investigated its pharmacological effects in various animal models of metabolic injury. METHODS: First, we validated the ability of CS17919 to inhibit ASK1 in vitro and then tested the safety profile of CS17919 in

Increased hepatic putrescine levels as a new potential factor related to the progression of metabolic dysfunction-associated steatotic liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic liver condition that often progresses to more advanced stages, such as metabolic dysfunction-associated steatohepatitis (MASH). MASH is characterized by inflammation and hepatocellular ballooning, in addition to hepatic steatosis. Despite the relatively high incidence of MASH in the population and its potential detrimental effects on human health, this liver disease is still not fully understood from a pathophysiological perspective. Deregulation of polyamine levels has been detected in various pathological condition

OTUD7B inhibited hepatic injury from NAFLD by inhibiting K48-linked ubiquitination and degradation of β-catenin.

Non-alcoholic fatty liver disease (NAFLD) is the prevalent liver disease. Ovarian tumor domain-containing 7B (OTUD7B) is a deubiquitinating enzyme and its role in NAFLD remains unclear. In high-fat diet (HFD)-induced NAFLD mouse model and palmitic acid (PA)-treated HepG2 cells, OTUD7B expression was decreased. Adenoviral overexpression of OTUD7B in mice resulted in reduced body weight and liver injury, with decreased serum aminotransferase (ALT) and aspartate aminotransferase (AST) levels. OTUD7B overexpression attenuated hepatic lipid deposition (serum TG, TC, LDL-C, HDLC, and FFA levels), wh
