Amoebiasis is a parasitic infectious disease caused by the enteric protozoan Entamoeba histolytica, a leading basis of deaths accounted to parasites, succeeding malaria and schistosomiasis. Conventional treatment methodologies used to deal with amoebiasis mainly rely on the administration of anti-amoebic compounds and vaccines but are often linked with substantial side-effects on the patient. Besides, cases of development of drug resistance in protozoans have been recorded, contributing further to the reduction in the efficiency of the treatment. Loopholes in the efficacious management of the

Gill parasitic infections challenge farming of rainbow trout (Oncorhynchus mykiss, Walbaum) in freshwater facilities. Apart from flagellates (Ichthyobodo, (Pinto) and ciliates (Ichthyophthirius (Fouquet), Ambiphrya (Raabe), Apiosoma (Blanchard), Trichodinella (Sramek-Husek) and Trichodina (Ehrenberg)), we have shown that amoebae are prevalent in Danish trout farms. Gills were isolated from farmed rainbow trout in six fish farms (conventional and organic earth pond and recirculated systems) and placed on non-nutrient agar (NNA) moistened with modified Neff's amoeba saline (AS) (15°C). Gill amoe

A 74-year-old woman who exhibited drowsiness was referred to our hospital. Enhanced head magnetic resonance imaging (MRI) revealed multiple ring-enhancing lesions and lesions showing partial mild hemorrhaging. The patient gradually progressed to a comatose condition with notable brain deterioration of unknown cause on follow-up MRI. On day nine, the patient inexplicably died, although brain herniation was suspected. Autopsy and histopathology revealed numerous amoebic trophozoites in the perivascular spaces and within the necrotic tissue. Brain immunostaining tested positive for Balamuthia man

A 3-year-old male child presented with moderate-to-high grade fever and non-projectile vomiting, generalised seizures and altered sensorium for 1 month. CT scan revealed a communicating hydrocephalus with no basal exudates. The microbiological tests were negative for Streptococcus pneumoniae, Haemophilus influenzae, Neisseria meningitides, brucellosis, cryptococcosis, HIV and Mycobacterium tuberculosis. Intracranial pressure was relieved by ventriculo-peritoneal shunt, and the child was empirically started on ceftriaxone, and antitubercular therapy with isoniazid, rifampicin, ethambutol

Protozoa can cause severe diseases, including malaria, leishmaniasis, Chagas disease, sleeping sickness and amoebiasis, all being responsible for morbidity and mortality particularly in tropical countries. To date there are no protective vaccines against any of these diseases, and many of the available drugs are old or elicit serious adverse reactions. Moreover, parasite resistance to existing drugs has become a serious problem. Owing to lack of financial returns, research in this field is of limited interest to pharmaceutical companies and largely depends on funding by public authorities. Thi

OBJECTIVE: The purpose of this report is to describe the clinical history, treatment, pathology, and imaging in two cases of rare CNS infection caused by free-living amebas. The Naegleria fowleri and Acanthamoeba species cause primary amebic meningoencephalitis and granulomatous amebic encephalitis, respectively. We describe the neuroimaging findings of a case involving a nonspecific cerebral edema pattern in primary amebic meningoencephalitis and a case involving focal enhancing lesions in granulomatous amebic encephalitis.
CONCLUSION: Primary amebic meningoencephalitis and granulomatous ame

Acanthamoeba culbertsoni is one of the free-living amoebae which are known to be pathogenic to man, causing granulomatous amoebic encephalitis (GAE). In this study, Acanthamoeba culbertsoni was isolated from a water sample of El-Mahmoudia Canal in Alexandria, in July-1993. This was used to infect mice intranasally to study the histopathological picture of the brain and lungs. The neuropathological features consisted of chronic granulomatous encephalitis in which cysts and trophozoites were found. Associated Acanthamoeba pneumonitis with massive consolidation was also observed. Congenital infec

Studies of immunoglobulins pattern in 64 patients with different clinical symptoms of amoebiasis revealed that IgG showed no rise in patients with acute amoebic dysentery and asymptomatic cyst passers, while it was significantly increased in patients with active hepatic amoebiasis. IgM, IgA and IgE were significantly increased in both acute amoebic dysentery and active hepatic amoebiasis, but no significant changes in asymptomatic cyst passers. ELISA (depended on level of IgG) was valid and reliable in diagnosis of active hepatic amoebiasis (100%), IHAT (depended on level of IgM) gave 72.7% an

In xenic culture, isolates of Entamoeba histolytica from asymptomatic carriers are characterized, with rare exception, by possession of a nonpathogenic zymodeme. During the process of axenizing such an isolate, strain CDC:0784:4, a change in the pattern of the isoenzymes from nonpathogenic zymodeme I to pathogenic zymodeme II was observed 40 days after the amebae had been transferred from a medium for xenic cultivation to one used for axenic cultivation, but before axenization of the amebae had actually occurred. Axenization was accomplished by feeding the amebae lethally irradiated bacteria w

We have described a case of primary amebic appendicitis with liver abscess, and we suggest that this diagnosis be considered in patients with appendicitis after returning from endemic areas. Careful histologic preparation will establish the diagnosis and dictate appropriate therapy.

Myelin membranes were purified from normal as well as experimental primary amoebic meningo-encephalitis-afflicted mouse brains. Infected myelin fractions were observed to form a floating fraction in 0.32 M sucrose. Total proteins, total lipids, phospholipids, cholesterol and galactolipids decreased markedly due to amoebic infection. Phosphatidyl choline, phosphatidyl ethanolamine plus phosphatidyl serine and sphingomyelin were decreased with a concomitant increase in lysophosphatidyl choline, lysophosphatidyl ethanolamine plus lysophosphatidyl serine and phosphatidic acid. The differential diagnostic possibilities in colitic changes are presented. In addition, the specific objectives of major examination procedures in the differentiation of the underlying diseases are indicated. In this connection, mention is also made of the etiopathogenic factors-in those conditions in which they are known with certainty.

Primary amebic meningoencephalitis and granulomatous amebic encephalitis are well recognized clinicopathological entities caused by free-living amebas. Associated arteritis and "mycotic aneurysms" with infiltration of intracranial arteries by lymphocytes, amebic trophozoites and cysts have not been previously reported. A 26-month-old girl had a 3-week history of encephalitis, characterized, initially, by vomiting and low-grade fever. Subsequently, she developed ataxia, generalized weakness, lethargy, and esotropia. The first CSF showed 490 RBC/microliters, 705 WBC/microliters with 90% mononucl

A novel tetrahydroquinolinyl ester, quinfamide, administered orally in multiple doses for 3 days had an ED50 of 0.25 mg/kg/day (total dose 0.75 mg/kg) for eradicating Entamoeba criceti in hamsters in several tests. It was significantly more active by direct comparison than 3 commercially available amoebicides and at least as active as 2 other esters of the parent compound, 1-(dichloroacety)-1,2,3,4-tetrahydro-6-quinolinol. After administration of a single dose, ED50 calculations for quinfamide averaged 0.9 mg/kg. Quinfamide was considerably more active than the other tetrahydroquinolinols, dil
