A gene expression panel for estimating age in males and females of the sleeping sickness vector Glossina morsitans.

Many vector-borne diseases are controlled by methods that kill the insect vectors responsible for disease transmission. Recording the age structure of vector populations provides information on mortality rates and vectorial capacity, and should form part of the detailed monitoring that occurs in the wake of control programmes, yet tools for obtaining estimates of individual age remain limited. We investigate the potential of using markers of gene expression to predict age in tsetse flies, which are the vectors of deadly and economically damaging African trypanosomiases. We use RNAseq to identi

Detection of the Wolbachia protein WPIP0282 in mosquito spermathecae: implications for cytoplasmic incompatibility.

Cytoplasmic incompatibility (CI) is a conditional sterility induced by the bacterium Wolbachia pipientis that infects reproductive tissues in many arthropods. Although CI provides a potential tool to control insect vectors of arthropod-borne diseases, the molecular basis for CI induction is unknown. We hypothesized that a Wolbachia-encoded, CI-inducing factor would be enriched in sperm recovered from spermathecae of female mosquitoes. Using SDS-PAGE and mass spectrometry, we detected peptides from the 56 kDa hypothetical protein, encoded by wPip_0282, associated with sperm transferred to femal

Plasmid DNA immunization with Trypanosoma cruzi genes induces cardiac and clinical protection against Chagas disease in the canine model.

The only existing preventive measure against American trypanosomosis, or Chagas disease, is the control of the transmitting insect, which has only been effective in a few South American regions. Currently, there is no vaccine available to prevent this disease. Here, we present the clinical and cardiac levels of protection induced by expression to Trypanosoma cruzi genes encoding the TcSP and TcSSP4 proteins in the canine model. Physical examination, diagnostic chagasic serology, and serial electrocardiograms were performed before and after immunization, as well as after experimental infection.

Biology and host preferences of Cryptorhynchus melastomae (Coleoptera: Curculionidae), a possible biocontrol agent for Miconia calvescens (Melastomataceae) in Hawaii.

The introduced plant Miconia calvescens (Melastomataceae) poses a grave threat to Hawaii's native ecosystems and biodiversity. One potential candidate for classical biological control is Cryptorhynchus melastomae (Coleoptera: Curculionidae: Cryptorhynchinae), a stem-boring weevil from Central and South America. This weevil feeds on M. calvescens in its native Costa Rica and has been successfully reared under greenhouse conditions. Comparison of its environmental conditions in Costa Rica with those in the Miconia infested areas of Hawaii indicates the latter is a suitable habitat for C. melasto

Does isometamidium chloride treatment protect tsetse flies from trypanosome infections during SIT campaigns?

African animal trypanosomosis is a major pathological constraint to cattle breeding across 10 million km2 of sub-Saharan West African countries infested by tsetse flies, their cyclic vectors. The release of sterile males (sterile insect technique [SIT]) is a potentially important control technique aimed at eliminating the vectors. Prior to release, tsetse are generally treated with isometamidium chloride, a trypanocide, to prevent them from transmitting parasites. The present study investigated the preventive action of isometamidium chloride (0.5 mg/L) on the subsequent susceptibility of tsets

Eastern equine encephalitis--New Hampshire and Massachusetts, August-September 2005.

During August-September 2005, the New Hampshire Department of Health and Human Services reported seven cases of human eastern equine encephalitis virus (EEEV) disease, the first laboratory-confirmed, locally acquired cases of human EEEV disease reported from New Hampshire in 41 years of national surveillance. Also during August--September 2005, the Massachusetts Department of Public Health reported four cases of human EEEV disease, five times the annual average of 0.8 cases reported from Massachusetts during the preceding 10 years. Four of the 11 patients from New Hampshire and Massachusetts d

A single multiplex assay to identify major malaria vectors within the African Anopheles funestus and the Oriental An. minimus groups.

The African Anopheles funestus and the Oriental An. minimus groups are closely related and composed of major malaria vectors in Africa and Southeast Asia, respectively. None of the species of either the An. funestus or the An. minimus group can be identified with absolute certainty using the adult morphology. Polymorphisms present on the internal transcribed spacer 2 (ITS2) of ribosomal DNA allowed the development of 10 primers that combined with an universal forward primer lead to a simple and sensitive multiplex allele-specific polymerase chain reaction (AS-PCR). Moreover, the possible addit

An outbreak of acute bartonellosis (Oroya fever) in the Urubamba region of Peru, 1998.

During May 1998, we conducted a case-control study of 357 participants from 60 households during an outbreak of acute bartonellosis in the Urubamba Valley, Peru, a region not previously considered endemic for this disease. Blood and insect specimens were collected and environmental assessments were done. Case-patients (n = 22) were defined by fever, anemia, and intra-erythrocytic coccobacilli seen in thin smears. Most case-patients were children (median age = 6.5 years). Case-patients more frequently reported sand fly bites than individuals of neighboring households (odds ratio [OR] = 5.8, 95%

Glycosylation is not required for the fusion activity of the G protein of vesicular stomatitis virus in insect cells.

The gene encoding the complete glycoprotein of vesicular stomatitis virus (VSV, Indiana serotype G protein) with potential asparagine-linked glycans at amino acid residues 179 and 338 was inserted into a baculovirus transfer vector pAcYM1, derived from the nuclear polyhedrosis virus of Autographa californica (AcNPV). The gene was placed under the control of the AcNPV polyhedrin promotor and expressed by the derived recombinant viruses to high levels in Spodoptera frugiperda cell lines. The principal product was the glycosylated version of the G protein, although some alternative (including pro

Assessing dengue transmission risk and a vector control intervention using entomological and immunological indices in Thailand: study protocol for a cluster-randomized controlled trial.

BACKGROUND: Dengue fever is the most common and widespread mosquito-borne arboviral disease in the world. There is a compelling need for cost-effective approaches and practical tools that can reliably measure real-time dengue transmission dynamics that enable more accurate and useful predictions of incidence and outbreaks. Sensitive surveillance tools do not exist today, and only a small handful of new control strategies are available. Vector control remains at the forefront for combating dengue transmission. However, the effectiveness of many current vector control interventions is fraught wi

High household economic burden caused by hospitalization of patients with severe dengue fever cases in Can Tho province, Vietnam.

During 2006-2007, a cohort of 144 confirmed dengue cases in Can Tho Province, Vietnam were compared with a matching set of 144 households that had no dengue cases. Approximately 6-9 months after sickness, there were no significant differences in terms of knowledge of the etiology of dengue, mosquito breeding habitats, and prevention measures in respondents from both sets of households. There was also no difference in the abundance of Aedes aegypti (Linn.) adults but the average numbers of late instar and pupal Ae. aegypti per household were greater in the negative control houses. Thus, the ris

An epidemiological model for West Nile virus: invasion analysis and control applications.

Infectious diseases present ecological and public health challenges that can be addressed with mathematical models. Certain pathogens, however, including the emerging West Nile virus (WN) in North America, exhibit a complex seasonal ecology that is not readily analysed with standard epidemiological methods. We develop a single-season susceptible-infectious-removed (SIR) model of WN cross-infection between birds and mosquitoes, incorporating specific features unique to WN ecology. We obtain the disease reproduction number, R0, and show that mosquito control decreases, but bird control increases

The situation of sleeping sickness in Angola: a calamity.

Although nearly one-fifth of the Angolan population is at risk of becoming infected with trypanosomiasis, only 6% currently have access to surveillance and treatment because of the war and its resultant destruction of the country's infrastructure. The paper outlines the history of human African trypanosomiasis (HAT) control activities in Angola and sums up what measures need to be taken to re-establish them.

Histone Deacetylases Are Required for Aedes aegypti Female Reproduction and Embryonic Development.

Histone acetylation and deacetylation are essential for chromatin modifications that regulate various cellular functions. This study identified nine histone deacetylase (HDAC) genes in the yellow fever mosquito, Aedes aegypti, and investigated their roles in female reproduction and embryonic development using RNA interference (RNAi). RNAi-mediated knockdown of HDAC1 (Histone deacetylase 1), HDAC3 (Histone deacetylase 3), HDAC4 (Histone deacetylase 4), and SIRT2 (Sirtuin 2) resulted in severe defects in oocyte development and reduced egg-laying compared with control insects injected with double

Pigment-dispersing factor neuropeptides act as multifunctional hormones and modulators in tardigrades.

Pigment-dispersing factors (PDFs) are neuropeptides that play key roles in controlling the circadian rhythms in various insects, whereas their function remains elusive in other protostomes including tardigrades (water bears). Here we show that the three PDFs of the tardigrade Hypsibius exemplaris are co-localized in two pairs of inner lobe cells in the brain, whereas only one PDF occurs in four additional cerebral and two extracerebral cells. The axons of the inner lobe cells pass through the contralateral brain hemisphere, descend to the ventral nerve cord and terminate in two pairs of potent

Isolating and identifying fungi to determine whether their biological properties have the potential to control the population density of mosquitoes.

Mosquitoes transmit diseases such as dengue, chikungunya, Zika, and yellow fever to humans. Biological control methods are required for these insects because they can be environmentally friendlier, safer, and more cost-effective than chemical or physical methods currently available. The aim of this research is to identify fungi found in mosquito breeding containers that have the potential to control the population density of mosquitoes. For the identification, water samples were taken from mosquito breeding containers situated in seven districts of Bangkok to obtain pure cultures. Deoxyribonuc

Inhibition of mouse parenchymal cells proliferation by mosquito larvae extract.

As it is known, some cell populations of mosquito Aedes aegypti larvae do not proliferate. This fact suggests the existence of some mitosis inhibiting factor. We have assayed the effect of mosquito larvae crude extract on 85 C3HS young male mice (25 +/- 1 days old) and the effect of the dialyzed fraction of the same extract on 47 partially hepatectomized C3HS adult male mice. The injection was performed at 16:00 hour (before the raising of the DNAS curve) and the controls were made at 08:00/16, 12:00/20 and 16:00/24 (Time of Day/Time Post Injection). Colchicine arrested metaphases of nephrocyt

Species-specific reduction of survival and predation in ladybirds exposed to prolonged and intensified heat waves.

Climate change exacerbates heat waves, threatening ecosystems by disrupting species survival and interactions. Insects are highly sensitive to temperature extremes, exhibiting species-specific stress which can impact community dynamics. Ecosystem services, such as pest control, can therefore be reduced by heat stress. We studied how five ladybird species responded to heat wave conditions, focusing on survival and aphid consumption. While survival was generally unaffected by exposure to realistic heat waves (compared to normal summer temperatures), extreme heat wave (+5°C) reduced survival in f
