Design and development of novel therapeutics for brucellosis treatment based on carbonic anhydrase inhibition.

Carbonic anhydrase is a metalloprotein, an enzyme with strong inhibition in antibacterial treatment. This study presents QSAR modeling for a series of 41 chemical compounds, 40 sulfonamides and one sulfamate, including 13 clinically tested drugs as carbonic anhydrase inhibitors based on the Monte Carlo optimization with molecular descriptors based on the SMILES notation and local invariants of the molecular graph, and field 3D based methods. Conformation independent QSAR models were developed for three random splits and a 3D QSAR model for one random split into the training and test sets. The

Understanding the within-host dynamics of influenza A virus: from theory to clinical implications.

Mathematical models have provided important insights into acute viral dynamics within individual patients. In this paper, we study the simpl … load, the time to peak viral load and the level of cell death due to infection. Using Markov chain Monte Carlo methods, we fitted the models to data collected from 12 untreated volunteers who participated in two clinical studies that tested the antiviral drugs oseltamivir and zanamivir. Based on the results, we also discuss various difficulties in deriving precise …

Estimation of the carcinogenic risk of radiotherapy of benign diseases from shoulder to heel.

BACKGROUND AND PURPOSE: To estimate risk on fatal tumour induction in patients by radiotherapy of benign diseases at various body sites, including heterotopic ossification, omarthritis, gonarthrosis, heel spurs and hidradenitis suppurativa. MATERIAL AND METHODS: The carcinogenic risk is estimated by applying the effective dose concept from the ICRP with the average risk factor of 10% per Sv for high dose and high dose rate. Although, the concept of effective dose for the present study has limitations, its use is considered acceptable for a fairly rough risk estimate. The organ doses are calcu

A Hybrid Frequency Estimation Framework for Long-Range FMCW LiDAR Under Low-SNR Conditions.

Frequency-modulated continuous-wave (FMCW) light detection and ranging (LiDAR) has become increasingly significant for long-distance ranging fields. However, the beat signal is vulnerable to noise interference during detection, resulting in a low signal-to-noise ratio (SNR) and inaccurate ranging results. Although traditional methods have improved accuracy through various denoising and spectral analysis algorithms, they often struggle to maintain robust performance in long-range scenarios under low-SNR conditions. To address this challenge, this paper proposes the HOIGL, a hybrid frequency est

Machine learning approaches reveal methylation signatures associated with pediatric acute myeloid leukemia recurrence.

Acute myeloid leukemia (AML) is a severe hematological malignancy characterized by high recurrence rates, especially in pediatric patients, highlighting the need for reliable prognostic markers. This study proposes methylation signatures associated with AML recurrence using computational methods. DNA methylation data from 696 newly diagnosed and 194 relapsed pediatric AML patients were analyzed. Feature selection algorithms, including Boruta, least absolute shrinkage and selection operator, light gradient boosting machine, and Monte Carlo feature selection, were employed to screen and rank met

Fast and trustworthy nowcasting of dengue fever: A case study using attention-based probabilistic neural networks in São Paulo, Brazil.

Nowcasting methods are crucial in infectious disease surveillance, as reporting delays often lead to underestimation of recent incidence and can impair timely public health decision-making. Accurate real-time estimates of case counts are essential for resource allocation, policy responses, and communication with the public. In this paper, we propose a novel probabilistic neural network (PNN) architecture, named NowcastPNN, to estimate occurred-but-not-yet-reported cases of infectious diseases, demonstrated here using dengue fever incidence in São Paulo, Brazil. The proposed model combines stat

ESTABLISHMENT OF RADIATION DOSES FOR PEDIATRIC X-RAY EXAMINATIONS IN A LARGE PEDIATRIC HOSPITAL IN TURKEY.

Pediatric patients are more sensitive to ionizing radiation when compared with adults. The aim of this study was to evaluate the radiation doses for some common pediatric x-ray examinations performed with various digital radiography systems. Quality control tests of the digital radiography systems were carried out according to international published protocols before the pediatric dose measurements. Radiation dose measurement was performed by using the x-ray tube outputs and thermoluminescent dosimeter dose measurement methods. In the present study, radiation doses were assessed for 247 chest,

A pilot study of patient-specific cardiovascular MDCT dose maps and their utility in estimating patient-specific organ and effective doses in obese patients.

BACKGROUND: Estimates of effective dose (E) for cardiovascular CT are obtained from a scanner-provided dose metric, the dose-length product (DLP), and a conversion factor. These estimates may not adequately represent the risk of a specific scan to obese adults. OBJECTIVE: Our objective was to create dose maps sensitive to patient size and anatomy in the irradiated region from a patient's own CT images and compare measured E (EDoseMap) to doses determined from standard DLP conversion (EDLP) in obese adults. METHODS: 21 obese patients (mean body mass index, 39 kg/m(2)) underwent CT of the pulm

A systematic procedure to optimise dose and image quality for the measurement of inter-vertebral angles from lateral spinal projections using Cobb and superimposition methods.

BACKGROUND: Patients with vertebral column deformations are exposed to high risks associated with ionising radiation exposure. Risks are further increased due to the serial X-ray images that are needed to measure and asses their spinal deformation using Cobb or superimposition methods. Therefore, optimising such X-ray practice, via reducing dose whilst maintaining image quality, is a necessity. OBJECTIVES: With a specific focus on lateral thoraco-lumbar images for Cobb and superimposition measurements, this paper outlines a systematic procedure to the optimisation of X-ray practice. METHODS:

BACKGROUND: Closed-incision negative pressure therapy (ciNPT) decreases the rate of wound complications in oncoplastic breast surgery (OBS) but at a fiscal cost. Our aim was to examine the cost-utility of ciNPT in OBS. METHODS: A literature review was performed to obtain the probabilities and outcomes for the treatment of unilateral breast cancer with OBS with ciNPT versus without. Reported utility scores in the literature were used to calculate quality-adjusted life years (QALYs) for each health state. A decision analysis tree was constructed with rollback analysis to determine the more cost

Performance evaluation of DEWMA3 in phase-II for capturing changes in simple linear profiles based on run rule mechanism.

In Statistical Process Control, many techniques exist for monitoring the stability of a process over time. In this work, we study the relationship of the response variable with explanatory variables in the form of linear profiles for detecting changes in slope and intercept of the linear quality profiles. We used the transformation of explanatory variables approach used for make the regression estimates independent of each other to have zero average. A comparative study of three phase-II methods using DEWMA statistics in monitoring and capturing undesirable deviations in the slope, intercept,

Early Surgery Versus Biologic Therapy in Limited Nonstricturing Ileocecal Crohn's Disease-A Decision-making Analysis.

BACKGROUND: Surgery is the preferred option for patients with symptomatic localized fibrostenotic ileocecal Crohn's disease (CD) but not for those with predominantly active inflammation without obstruction. The benefit of early surgery in patients with a limited nonstricturing ileocecal CD over biologic treatment is still a debate. OBJECTIVE: Our objective is to formulate a decision analysis model based on recently published data to explore whether early surgery in patients with limited nonstricturing CD is preferred over biologic treatment. METHODS: We constructed a Markov model comparing 2

Nanohybrid-based immunosensor prepared for Helicobacter pylori BabA antigen detection through immobilized antibody assembly with @ Pdnano/rGO/PEDOT sensing platform.

The gastric colonization of human hosts by Helicobacter pylori (H. pylori) increases the risk of developing gastritis, ulcers and gastric cancer. To detect H. pylori, a nanohybrid-based BabA immunosensor is developed herein. BabA is an outer membrane protein and one of the major virulence factors of H. pylori. To design the immunosensor, an Au electrode is loaded with palladium nanoparticles (Pdnano) by electrodeposition to generate reduced graphene oxide (rGO)/poly(3,4-ethylenedioxythiophene) (PEDOT). The immobilization of these nanostructured materials imparts a large surface area and electr

Quantitative risk assessment of Listeria monocytogenes in bulk cooked meat from production to consumption in China: a Bayesian approach.

BACKGROUND: To estimate the public health risk related to cooked meat in bulk products contaminated with Listeria monocytogenes, a generic Bayesian network (BN) risk-assessment model was developed to simulate influencing factors and processes of products from the industry level to the consumer level. To quantify the model, parameter values of prior distributions were acquired from the literature, websites, and expert opinions. Using the Markov chain Monte Carlo (MCMC) simulation approach, posterior probability distributions were calculated according to the incorporated evidence, which allowed
