The effects of prenatal ethanol exposure on the NMDAR1 protein expression (postnatal days 1 and 7) and on the developmental profile of the NMDAR2A and NMDAR2B subunits in rat forebrain and hippocampus were investigated. Forebrain and hippocampal membrane proteins were isolated from pups of various ages (postnatal days 1 to 21) from prenatally ethanol exposed, pair-fed and ad libitum control groups. A semiquantitative immunoblot procedure was used with antibodies raised against the NMDAR1, NMDAR2A, and the NMDAR2B subunits to assess the NMDA subunit protein expression in the samples. NMDAR1 pro

Alcohol-exposed children display delayed motor development and impaired fine- and gross-motor skills, including deficits in the maintenance of balance. In a recent study, we assessed the contribution of visual, somatosensory, and vestibular information to the ability to maintain balance. Our findings suggested that alcohol-exposed children were overly reliant on somatosensory information and were unable to compensate by using the visual and/or vestibular systems. To understand the nature of these observed balance deficits, corrective postural reactions were examined by exposing standing subjec

BACKGROUND: Fetal Alcohol Syndrome (FAS) represents an important array of abnormalities in the development of offspring born of alcoholic mothers. This investigation used a murine Balb/cJ model to investigate the effects of maternal ethanol intake on craniomandibular and long bone development.
METHODS: Experimental 8-week-old female mice (daughters of alcoholic female mice) drank an ethanol solution increasing from 1-20%, and 6 weeks later were mated with non-alcoholic males. The control group consisted of normal Balb/cJ male and female mice that drank water without ethanol. Alcohol intake wa

Early ethanol exposure depletes neurons in the developing nervous system, however the effects on neuronal precursors are not homogeneous. Some cells are more susceptible to ethanol toxicity than others. Growth factors are important mitogens for neuronal precursors. We tested the hypothesis that the differential sensitivity of neuronal precursors to ethanol is determined by their responses to growth factors using an in vitro model (SH-SY5Y, SK-N-SH, and IMR32 neuroblastoma cells) of neuronal precursors. The three cell lines were raised in a medium containing 10% or 0% fetal calf serum. Cells we

Fetal alcohol syndrome (FAS) is characterized by growth retardation, craniofacial malformations, and heart and neural defects; the cellular and molecular mechanism(s) responsible for ethanol's teratogenicity remains unknown. Although the phenotype suggests that prenatal ethanol exposure perturbs neural crest cell development, direct proof that these cells are an in utero target is still lacking. Previous research suggested that cranial neural crest cells are eliminated by ethanol-induced apoptosis. We tested this hypothesis using a chick embryo model of FAS. A single dose of ethanol, chosen to

The efficacy of alcohol screening questionnaires, the TWEAK, T-ACE, NET, MAST, and CAGE, in detecting periconceptional risk-drinking, > or = 1 oz absolute alcohol/day, was investigated in 4743 African-American women attending an inner-city prenatal clinic who had reported ever drinking. Sensitivity, specificity, positive predictive value, efficiency, follow-up rates, and receiver operating characteristics of the questionnaires were examined to compare the overall effectiveness of the questionnaires and their performance at cut-points defining positive scores ranging from 1 to 3. Prenatal exposure to ethanol (EtOH) alters developing catecholamine (CA) systems and acute sensitivity to the locomotor stimulant effects of EtOH. As an extension of previous work involving CA agents, this study addressed whether prenatal EtOH exposure influences central norepinephrine (NE) systems by examining the motoric effects of the direct alpha 2 adrenoreceptor agonist clonidine given alone and in combination with a low-dose stimulant challenge of EtOH. Standard lab chow or liquid diets containing either 25% EtOH-derived calories (EDC), or 0% EDC (pair-fed group) were given to pregnant C

Ethanol exposure during development can lead to structural damage to the cerebellum with associated behavioral dysfunction. Deficits in cerebellar weight and Purkinje cell number in the cerebellum of the rat are a function of the timing of the exposure and the peak blood alcohol concentration (BAC). Purkinje cell loss is greater following exposure during the third trimester equivalent than during the second trimester equivalent. Interestingly, Purkinje cells switch rapidly from being highly vulnerable on postnatal days 4 and 5 to being much more resistant after postnatal day 7. Furthermore, th

An eight-week old infant with alcohol embryopathy, weighing 3,700 g, was found to have abnormal liver functions (GPT 312 U/l, Quick value 25%) after surgical repair of a stenosis of the left ureter at its origin. The hospital notes indicated that the infant had been given a total of 1.6 g paracetamol over 60 hours for postoperative restlessness and pain. The serum paracetamol level was 60 mg/l 8 hours after the last dose of the drug. Blood exchange transfusion lowered the paracetamol level to 11 mg/l within 14 hours. After the exchange transfusion further signs of poisoning, namely renal impai

The effects of the synthetic dipeptide, L-pyroglutamyl-D-alaninamide (LPDA) were studied in the experiments on offspring of alcoholized during the pregnancy (5 g/kg/day) females. This dipeptide, which revealed the nootropic activity in previous experiments, was injected to the pups in dose of 1 mg/kg from 8 to 19 days of life. LPDA was shown to prevent the delayed disturbances of learning in passive avoidance test, of extrapolatory behaviour in escape test, to attenuate the emotional hyperreactivity. LPDA normalized EEG power spectrum, decreased interhemispheric asymmetry. This substance atten

We examined the developing synaptic junctions in the rat frontal cortex in cases of fetal alcohol syndrome, the objective being to determine the synapse-mental retardation relationship. On day 21 of gestation, the ultrastructural synaptic junction revealed no obvious differences between the ethanol-exposed and control rats; however, the number of synapses in ethanol-exposed rats was one third that of the controls. The possible relationship between synaptic density in the frontal cortex and mental development has to be considered.

The anterograde horseradish peroxidase method was used to determine if prenatal exposure to ethanol affected the development of the characteristic afferent lamination pattern of the commissural projection to the dentate gyrus. Mean ethanol consumption for the ethanol-consuming dams was 12.7 g/kg +/- 0.3 g per day. Adult offspring of rats that consumed a liquid diet containing 35% ethanol-derived calories during days 1-21 of gestation, and both pair-fed and normal controls were examined. Brain weights and volumes of the ethanol and pair-fed control rats did not differ significantly from normal

The effects of chronic ethanol consumption during gestation on the development of layer V pyramidal cells was studied quantitatively in the somatosensory cerebral cortex of the newborn guinea-pig. The spread of the basilar dendritic arborizations and counts of dendritic spines on the apical dendrite of neurons that had been processed with the rapid Golgi method were compared with those found in age-matched controls receiving an isocaloric diet without alcohol. There were significant differences in the number of primary basilar dendrites (P less than 0.05) and dendritic ramifications at a dista

Behavioral changes following pre- and postnatal ethanol administration were examined in rat pups of various ages. A significant increase in locomotor activity occurred at all ages (16, 22 and 30 days) examined. Nose poking activity in 22-day-old pups and standing activity in 30-day-old pups were also significantly higher following perinatal ethanol exposure. No significant change in grooming behavior was observed. The overactivity in rat pups following perinatal ethanol exposure is consistent with the frequently observed hyperactivity in fetal alcohol syndrome children.

As a possible preventative measure for brain dysfunction in the fetal alcohol syndrome, the effect of zinc supplementation together with ethanol on the fetal cerebrum, especially the hippocampus, was investigated. Female rats of an ethanol group and a control group received 30% ethanol and water, respectively. During pregnancy half of both groups received 0.01% zinc together with ethanol or water. Fetuses on gestational day 21 were examined biochemically and ultrastructurally. Administration of zinc with ethanol resulted in an increased cerebral weight and cerebral RNA content compared to admi

Fostered rat offspring whose mothers had been administered 7.5% alcohol solution (4.85 g/kg/day) as drinking fluid during 0-17 days of gestation were compared with equivalent groups whose dams had given ad libitum and restricted volume of tap water in the following behavioral tests: reflex test, swimming behavior test, open-field test, radial-maze learning and two-way avoidance learning. The treatment with alcohol resulted in developmental delay in ear-flaps uncurling, placing reflex, swimming behavior, walking in addition to reduction in body weight at 1 and 7 days of age. The treated group a

Female Sprague-Dawley rats were fed a complete liquid diet containing either 5.5% ethanol (mean daily intake of about 9g of ethanol per kg body weight) or an isocaloric amount of dextrose (control group), with additional water available ad libitum. The diets were fed for four weeks prior to and throughout pregnancy. On day 20 of gestation cardiac output and blood flow to the the placenta, heart, kidneys and uterus were measured and plasma osmolality and muscle dry weight were determined. No significant differences were seen between alcohol and control groups with respect to litter size, fetal

In this retrospective study, the clinical findings of 26 cases borne by alcoholic mothers in Japan are summarized. The information on maternal alcohol drinking and abnormal offspring was gathered by a questionnaire survey. Twenty cases, mostly in institutions for mental retardation, were checked from all areas of Japan. Six children from another group were followed during health examinations of children in a selected area with many heavy drinkers. Growth deficiency was found in half of these cases with an increased rate of low birth weight babies (42% of cases) including small-for-date babies
