Normalizing Body Weight with a Dietary Strategy Mitigates Obesity-Accelerated Pancreatic Carcinogenesis in Mice.

BACKGROUND: Obesity is a modifiable risk factor for pancreatic cancer, but the impact of dietary changes leading to weight loss in pancreatic carcinogenesis remains unknown. OBJECTIVES: This study aims to determine the effects of weight normalization via dietary switch on pancreatic carcinogenesis and associated mechanisms. METHODS: Five-wk-old male and female LSL-KrasG12D/+; p48Cre/+ (KC) mice (8-12/diet group/sex) were fed a high-fat, diet-induced obesity diet (DIO; 60% kcal from fat) or a low-fat, control diet (CD; 11% kcal from fat) for 21 wk. A subset of mice was fed the DIO for 8 wk, t

Transcriptome analysis reveals the pathogenesis of spontaneous tibial dyschondroplasia in broilers.

Tibial dyschondroplasia (TD) is a severe bone disease that affects fast-growing broiler chickens and causes economic loss. Despite previous studies, the regulatory mechanism of TD remains unclear and is thought to be primarily based on thiram induction, which may differ from that of naturally occurring diseases. To better understand TD, a digital X-ray machine was used in the present study to determine its incidence in four hundred yellow-feathered broiler chickens. The results showed that the incidence of TD was 22% after 6 weeks and gradually decreased after 8 and 10 weeks. The body weight o

Plasma-targeted proteomic and lipidomic profiling of MASLD, MASH, and hepatitis C virus infection.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) and metabolic dysfunction-associated steatohepatitis (MASH) are chronic liver diseases characterized by lipid accumulation and persistent inflammation, often progressing to fibrosis or hepatocellular carcinoma (HCC). Hepatitis C virus (HCV) infection shares overlapping pathological features, including chronic inflammation and fibrogenesis. Despite their prevalence, reproducible plasma-level molecular data that capture disease-associated systemic alterations remain limited. METHODS: We conducted targeted proteomic and

Spatiotemporally programmed hybrid nano-prodrugs orchestrate multidimensionally synergistic cascade lethality in NSCLC.

RATIONALE: Non-small cell lung cancer (NSCLC) develops a high GSH/GPX4 antioxidant phenotype under persistent oxidative pressure, which suppresses membrane lipid peroxidation and ferroptosis, constituting a core mechanism underlying chemotherapy resistance and suboptimal therapeutic efficacy. Breaking this resistance barrier demands not simply attacking the tumor but disarming the antioxidant defense to intensify oxidative damage and awaken durable antitumor immunity. METHODS: A self-assembled nano-prodrug, E-R@ISSL, was engineered by co-assembling a carbamate-linked RGD-modified etoposide (E

Paeoniflorin improves atherosclerosis by regulating the gut microbiota and fecal metabolites.

Atherosclerosis (AS) is a chronic inflammatory disease and a key factor in cardiovascular disease. Paeoniflorin (PF), a monomer component extracted from red and white paeony roots, has been used for the prevention and treatment of cardiovascular diseases, but the specific mechanism remains unclear. ApoE-/- mice fed a high-fat diet were randomly divided into a model group, a PF treatment group, a positive control group, and a control group fed a non-high-fat diet. The mice in the PF group were treated with 100 mg/kg (PF-100), 50 mg/kg (PF-50), and 25 mg/kg (PF-25) of PF through gavage. The posi

The metabolomics analysis of cecal contents elucidates significant metabolites involved in the therapeutic effects of total flavonoids derived from Sonchus arvensis L. in male C57BL/6 mice with ulcerative colitis.

Ulcerative colitis (UC), an inflammatory disease affecting the colon and rectal mucosa, is characterized by chronic and heterogeneous behavior of unknown origin. The primary cause of UC is chronic inflammation, which is closely linked to the development of colorectal cancer. Sonchus arvensis L. (SAL), a plant consumed worldwide for its nutritional and medicinal properties, holds significance in this context. In this study, we employed the total flavone in SAL as a treatment for male C57BL/6 mice with UC. The cecal contents metabolic profile of C57BL/6 mice in different groups, including UC (gr

Phospholipase A2 regulates autophagy in gouty arthritis: proteomic and metabolomic studies.

BACKGROUND: Acute gouty arthritis is inflammatory joint arthritis. Gouty arthritis (GA) involves multiple pathological processes. Deposition of joints by monosodium urate (MSU) crystals has been shown to play a critical role in the injury process. Due to the different effects of MSU stimulation on the joints, the exact changes in the synovial fluid are unknown. We want to explore the changes in proteins and metabolites in the joints of gouty arthritis. Regulating various functional substances in the joint can reduce inflammation and pain symptoms. METHODS: 10 patients with gouty knee arthriti

Can antibody conjugated nanomicelles alter the prospect of antibody targeted therapy against schistosomiasis mansoni?

BACKGROUND: CLA (conjugated linoleic acid)-mediated activation of the schistosome tegument-associated sphingomyelinase and consequent disruption of the outer membrane might allow host antibodies to access the apical membrane antigens. Here, we investigated a novel approach to enhance specific antibody delivery to concealed surface membrane antigens of Schistosoma mansoni utilising antibody-conjugated-CLA nanomicelle technology. METHODOLOGY/PRINCIPAL FINDINGS: We invented and characterised an amphiphilic CLA-loaded whey protein co-polymer (CLA-W) as an IV injectable protein nanocarrier. Rabbit

Role of hormonal and other factors in human prostate cancer.

American men have a lifetime risk of about 18% for prostate cancer diagnosis. Large international variations in prostate cancer risks and increased risks among migrants from low- to high-risk countries indicate important roles for environmental factors. Major known risk factors include age, family history, and country/ethnicity. Type 2 diabetes appears to reduce risk, while high birth weight and adult height are linked to increased risk of aggressive prostate cancer. Limited evidence supports an association with a history of sexually transmitted infections. A previous meta-analysis of eight co

Effect of complexation with randomly methylated beta-cyclodextrin on the aqueous solubility, photostability and antioxidant activity of an indolinonic nitroxide radical.

The interaction between the hydrophobic indolinonic nitroxide radical, 1,2-dihydro-2-methyl-2-phenyl-3H-indole-3-one-1-oxyl and hydrophilic alpha-, beta- and gamma-cyclodextrin derivatives was investigated in water by phase-solubility analysis. Among the studied cyclodextrins, random methyl-beta-cyclodextrin (RM-beta-CD) had the greatest solubilizing activity (1312-fold increase in. the intrinsic aqueous solubility). Solid complexes were prepared by the freeze-drying method and characterized by powder X-ray diffractometry and thermal analysis. Complexation of the nitroxide with RM-beta-CD was

Dual roles for lipolysis and oxidation in peroxisome proliferation-activator receptor responses to electronegative low density lipoprotein.

Low density lipoprotein (LDL) exists in various forms that possess unique characteristics, including particle content and metabolism. One circulating subfraction, electronegative LDL (LDL(-)), which is increased in familial hypercholesterolemia and diabetes, is implicated in accelerated atherosclerosis. Cellular responses to LDL(-) remain poorly described. Here we demonstrate that LDL(-) increases tumor necrosis factor alpha (TNFalpha)-induced inflammatory responses through NF kappa B and AP-1 activation with corresponding increases in vascular cell adhesion molecule-1 (VCAM1) expression. LDL

Increased toll-like receptors and p53 levels regulate apoptosis and angiogenesis in non-muscle invasive bladder cancer: mechanism of action of P-MAPA biological response modifier.

BACKGROUND: The new modalities for treating patients with non-muscle invasive bladder cancer (NMIBC) for whom BCG (Bacillus Calmette-Guerin) has failed or is contraindicated are recently increasing due to the development of new drugs. Although agents like mitomycin C and BCG are routinely used, there is a need for more potent and/or less-toxic agents. In this scenario, a new perspective is represented by P-MAPA (Protein Aggregate Magnesium-Ammonium Phospholinoleate-Palmitoleate Anhydride), developed by Farmabrasilis (non-profit research network). This study detailed and characterized the mecha

Malnutrition and cachexia in ovarian cancer patients: pathophysiology and management.

In ovarian cancer patients the poor nutritional status and cachexia are caused by the metabolic effects of the enlarging tumor masses and bowel obstruction. These patients may have a high resting energy expenditure due to increase in Cori cycle activity, glucose and triglyceride-fatty acid cycling and gluconeogenesis. Biochemical mediators of cachexia include cytokines, such as tumor necrosis factor and interleukin-6, and tumor-produced catabolic factors, such as lipid-mobilizing factor, proteolysis-inducing factor, and anemia-inducing factor. Mechanisms involved in the pathogenesis of obstruc

Time-course transcriptome and WGCNA reveal tissue-specific genes in 'White BK-1' rye during early cold acclimation stage.

INTRODUCTION: The early stage of cold acclimation (CA) is a critical period during which plants gradually transition from cold sensing to the establishment of acquired freezing tolerance. Winter rye (Secale cereale L.) is a cereal crop with superior cold hardiness. However, the temporal transcriptional dynamics during early CA and the tissue-specific expression patterns between leaves and roots remain poorly understood. METHODS: We evaluated the growth and freezing survival of 'White BK-1' during long-term CA and profiled leaf and root transcriptomes at multiple time points during the first 4
