Externalization of host cell protein kinase C during enteropathogenic Escherichia coli infection.

Enteropathogenic Escherichia coli (EPEC) is a common cause of diarrhea in children in developing countries. Protein kinase C (PKC), a serine- and threonine-directed protein kinase, is rapidly activated following EPEC infection and this is accompanied by its translocation to a membrane-bound location where it is tightly bound to phosphatidylserine (PS). EPEC infection causes host cell death, one of whose features is externalization of PS. We hypothesized that externalization of PS would be accompanied by externalization of PKC as well. We report that EPEC infection triggers the externalization

Methyl mercury increases intracellular Ca2+ and inositol phosphate levels in cultured cerebellar granule neurons.

In an effort to explain the previously observed methyl mercury (MeHg)-induced stimulation of protein phosphorylation in cerebellar granule neuron cultures, the effect of MeHg on protein kinase activities in cell-free assays and on second messenger systems in cultured neurons has been examined. Using cell-free assays for several protein kinases, no stimulation of enzyme activity was found at any concentration of MeHg tested. After 24 h exposure, 1-5 microM MeHg was found to have no significant effect on neuronal cyclic AMP levels. In contrast, intracellular levels of Ca2+ and rates of 45Ca2+ up

Tilapia-derived oligopeptide LSGYG inhibits tumor metastasis via electrostatic-lock-like retention at the catalytic region of MMP-2 and suppression of the HIF-1α-mediated EMT-like cascade.

Numerous studies have reported the biological activities of tilapia skin oligopeptides, which exhibit diverse physiological effects. However, research on the relationship between peptide structural characteristics and their activities remains relatively scarce. In this study, we combined computational simulations with biological experiments to investigate the anti-metastatic mechanism of the tilapia skin oligopeptide LSGYG. Computational analyses supported recurrent Zn-neighboring and pocket-associated recognition modes of LSGYG in the catalytic region of matrix metalloproteinase-2 (MMP-2) and

Parathyroid hormone synergizes with non-cyclic AMP pathways to activate the cyclic AMP response element.

Parathyroid hormone (PTH) activates multiple signaling pathways following binding to the PTH1 receptor in osteoblasts. Previous work revealed a discrepancy between cAMP stimulation and CRE reporter activation of truncated PTH peptides, suggesting that additional signaling pathways contribute to activation of the CRE. Using a CRE-Luciferase reporter containing multiple copies of the CRE stably transfected into the osteoblastic cell line Saos-2, we tested the ability of modulators of alternative pathways to activate the CRE or block the PTH-induced activation of the CRE. Activators of non-cyclic

The calcium-dependent chloride conductance mediator pCLCA1.

The regulatory behavior, inhibitor sensitivity, and properties of the whole cell chloride conductance observed in cells expressing the cDNA coding for a chloride conductance mediator isoform of the CLCA gene family, pCLCA1, have been studied. Common C-kinase consensus phosphorylation sites between pCLCA1 and the closely related human isoform hCLCA1 are consistent with a role for calcium in channel activation. Both channels are activated rapidly on exposure to the calcium ionophore ionomycin. Direct involvement of calcium in the activation of pCLCA1 was supported by the finding that treatment w

Mononuclear phagocyte activation: activation-associated antigens.

Mononuclear phagocyte activation is characterized by alterations in cellular metabolism and plasma membrane composition. In rodent and human systems, antibodies (conventional heteroantibodies or monoclonal reagents) that identify plasma membrane antigens selectively expressed by activated macrophages and monocytes have been generated. Among these activation-associated determinants is Mo3e (p50,80), a protease-sensitive antigen that is expressed by human monocytes activated in culture by exposure to bacterial lipopolysaccharide, muramyl dipeptide, or phorbol myristate acetate (PMA) (as well as

Role of activation of 5'-adenosine monophosphate-activated protein kinase in gastric ulcer healing in diabetic rats.

BACKGROUND: The potential utility of 5'-adenosine monophosphate-activated protein kinase (AMPK)-activating agents, such as metformin, in inducing angiogenesis, could be a promising approach to promote healing of gastric ulcers complicated by diabetes mellitus. The aim of the present study was to assess the effect of a drug that activates AMPK, namely metformin, in gastric ulcer healing in streptozotocin-induced diabetic rats. METHODS: Forty male Wistar albino rats were made diabetic by intraperitoneal (i.p.) streptozotocin injection and 10 rats were injected i.p. by a single dose of physiolog

Luteinizing hormone release from chicken pituitary cells: synergism between calcium and protein kinase C and its inhibition by calmodulin antagonists.

Continuous stimulation of cultured chicken pituitary cells with a native chicken hypothalamic GnRH, Gln8-GnRH, caused LH release, followed by rapid desensitization. Continuous exposure to calcium ionophore A23187 also produced a short-lived LH response, followed by desensitization. In contrast, continuous stimulation with a phorbol ester, 12-O-tetradecanoylphorbol-13-acetate (TPA), caused prolonged release of LH, reaching maximum amplitude after 1 h and slowly decreasing thereafter. Cells desensitized to GnRH remained fully responsive to A23187 and TPA, indicating that GnRH-mediated desensitiz

A-Kinase Anchoring Protein 13 (AKAP13) Augments Progesterone Signaling in Uterine Fibroid Cells.

CONTEXT: Uterine leiomyomata (fibroids) are prevalent sex hormone‒dependent tumors with an altered response to mechanical stress. Ulipristal acetate, a selective progesterone receptor (PR) modulator, significantly reduces fibroid size in patients. However, PR signaling in fibroids and its relationship to mechanical signaling are incompletely understood. OBJECTIVE: Our prior studies revealed that A-kinase anchoring protein 13 (AKAP13) was overexpressed in fibroids and contributed to altered mechanotransduction in fibroids. Because AKAP13 augmented nuclear receptor signaling in other tissues, w

ACSS2 Suppresses Ferroptosis to Drive Breast Cancer Brain Metastasis.

UNLABELLED: Brain metastasis in patients with breast cancer represents a terminal disease stage, with a median survival typically measured in months. Tumors that colonize the brain must adapt to its unique microenvironment, such as high acetate levels. Primary brain tumor cells enhance acetate conversion to acetyl-CoA through phosphorylation of acetyl-CoA synthetase 2 (ACSS2) by cyclin-dependent kinase 5 (CDK5), a process regulated by the nutrient sensor O-GlcNAc transferase (OGT). In this study, we showed that brain-metastatic breast cancer cells exhibited elevated O-GlcNAc, OGT, and phosphor

AMPK activation by SC4 inhibits noradrenaline-induced lipolysis and insulin-stimulated lipogenesis in white adipose tissue.

The effects of small-molecule AMP-activated protein kinase (AMPK) activators in rat epididymal adipocytes were compared. SC4 was the most effective and submaximal doses of SC4 and 5-amino-4-imidazolecarboxamide (AICA) riboside were combined to study the effects of AMPK activation in white adipose tissue (WAT). Incubation of rat adipocytes with SC4 + AICA riboside inhibited noradrenaline-induced lipolysis and decreased hormone-sensitive lipase (HSL) Ser563 phosphorylation, without affecting HSL Ser565 phosphorylation. Preincubation of fat pads from wild-type (WT) mice with SC4 + AICA riboside i

Troponin I Tyrosine Phosphorylation Beneficially Accelerates Diastolic Function.

BACKGROUND: A healthy heart is able to modify its function and increase relaxation through post-translational modifications of myofilament proteins. While there are known examples of serine/threonine kinases directly phosphorylating myofilament proteins to modify heart function, the roles of tyrosine (Y) phosphorylation to directly modify heart function have not been demonstrated. The myofilament protein TnI (troponin I) is the inhibitory subunit of the troponin complex and is a key regulator of cardiac contraction and relaxation. We previously demonstrated that TnI-Y26 phosphorylation decreas

Mechanisms underlying the inhibitory actions of the pentacyclic triterpene alpha-amyrin in the mouse skin inflammation induced by phorbol ester 12-O-tetradecanoylphorbol-13-acetate.

The present study evaluated some of the mechanisms through which alpha-amyrin, a pentacyclic triterpene isolated from Protium Kleinii and other plants, exerts its effects against 12-O-tetradecanoylphorbol-acetate (TPA)-induced skin inflammation in mice. Topical application of alpha-amyrin (0.1-1 mg/ear) dose-dependently inhibited TPA-induced increase of prostaglandin E2 (PGE2) levels. In contrast with the selective cyclooxygenase (COX)-1 SC560 [5-(4-chlorophenyl)-1-(4-methoxyphenyl)-3-trifluoromethylpyrazole] or COX-2 rofecoxib inhibitors, alpha-amyrin failed to alter either COX-1 or COX-2 act

Autophagy inhibitors block pathogenic NET release in immune-mediated inflammatory disease without impairing host defence.

OBJECTIVES: Activation of neutrophils and release of neutrophil extracellular traps (NETs), proteases and reactive oxygen species (ROS) is pathogenic in immune-mediated inflammatory diseases like rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), driving inflammation and damaging host tissues. The aim of this research was to identify small molecule inhibitors of NET production using a highly curated panel of narrow-spectrum small molecule kinase inhibitors termed the kinase chemogenomic set (KCGS). METHODS: Neutrophils were isolated from healthy controls (HC) and people with RA

Effects of sphingosine, staurosporine, and phorbol ester on neurites of rat sympathetic neurons growing in compartmented cultures.

Local application of sphingosine (1-10 microM), an inhibitor of protein kinase C, to NGF-supplied, distal neurites of rat sympathetic neurons in compartmented cultures caused their retraction and/or degeneration within 24 hr. This effect was specific for distal neurites because sphingosine (even at 100 microM) applied to cell bodies and/or proximal neurites did not destroy these regions of the cells, and their distal neurites continued to elongate. However, effects of other agents suggest that the retraction/degeneration observed in distal neurites directly exposed to sphingosine is not mediat

A chemical genomics-aggrephagy integrated method studying functional analysis of autophagy inducers.

Macroautophagy/autophagy plays a critical role in the pathogenesis of various human diseases including neurodegenerative disorders such as Parkinson disease (PD) and Huntington disease (HD). Chemical autophagy inducers are expected to serve as disease-modifying agents by eliminating cytotoxic/damaged proteins. Although many autophagy inducers have been identified, their precise molecular mechanisms are not fully understood because of the complicated crosstalk among signaling pathways. To address this issue, we performed several chemical genomic analyses enabling us to comprehend the dominancy

Effects of G-protein activator fluorides, protein kinase C activator phorbol ester and protein kinase inhibitor on insulin binding and hormonal imprinting of Tetrahymena.

AlCl3, BeCl2 and NaF do not influence the insulin binding of Tetrahymena immediately after treatment, but 24 h later insulin binding is decreased or increased by NaF in a dose-dependent manner, AlCl3 barely influences the binding, and BeCl2 increases it. The effect of all the three fluorides is dose-dependent. While NaF and AlF4 decrease binding at low doses and increase the binding at higher doses, BeF3 increases the insulin binding enormously. NaF does not permit insulin imprinting to be developed, AlF4 inhibits or amplifies the imprinting in a dose-dependent manner, while BeF3 allows imprin

Modulation of nicotinic acetylcholine receptor activity in submucous neurons by intracellular messengers.

The effects on acetylcholine-induced membrane currents (ACh currents), produced by agents known to modify the activity of intracellular messengers, were studied in the neurons of the guinea-pig ileum submucous plexus (SMP) using a whole-cell patch clamp recording method. The ACh currents were not affected by forskolin, the adenylate cyclase activator, regardless of whether or not ATP and GTP were present in the intracellular solution, and by phorbol 12-myristate 13-acetate, the protein kinase C activator. The ACh currents were strongly suppressed by thapsigargin, the microsomal calcium ATPase

Clopidogrel-Induced Gastric Injury in Rats is Attenuated by Stable Gastric Pentadecapeptide BPC 157.

AIM: Although Clopidogrel is safe in healthy volunteers, it can induce recurrence of gastric ulcers in high-risk patients. Here, we investigated the protective effect of the natural product, stable gastric pentadecapeptide 157 (BPC 157) on Clopidogrel-induced gastric injury. METHODS: We used acetic acid to induce gastric ulcer in Sprague Dawley rats. Clopidogrel alone or in combination with BPC 157 or L-NAME (nitric oxide system blockade) were administered after healing of acetic acid-induced ulcer. One percent methylcellulose solution was used as control. Ulcer recurrence rate and the ulcer

Signal transduction in lemon seedlings in the hypersensitive response against Alternaria alternata: participation of calmodulin, G-protein and protein kinases.

The development of an effective hypersensitive response (HR) in any plant system relies, not only in their gene composition and expression, but also on an effective and rapid signal transduction system. Lemon seedlings induce the phenylpropanoid pathway, which results in the de novo biosynthesis of the phytoalexin scoparone, as part of the hypersensitive response against Alternaria alternata. In order to elucidate some of the signaling elements that participate in the development of HR in lemon seedlings, we used several compounds that are known as activators or inhibitors of signal transducti
