Stability and Activity Collaborative Improvement of Carbonyl Reductase Based on the Modification Strategy for Transition Zone of the Flexible and Rigid Regions: An Application for Vibegron Chiral Intermediate Synthesis in High Efficiency.

Carbonyl reductases (EC 1.1.1.148, CRs) make up a group of oxidoreductases that catalyze the asymmetric reduction of prochiral ketones or aldehydes to produce the corresponding chiral alcohols, which are widely used in pharmaceutical and fine chemical industries. However, challenges in improving the enzymatic activity and stability continue to hinder the broader industrial application of the biocatalysts. In this study, a novel strategy was developed to target the transition region between the rigid and flexible domains of EaSDR6 from Exiguobacterium sp. s126. B-factor analysis was used to gui

SOX4 promotes triple-negative breast cancer progression by promoting SRPX2 transcription to regulate AKR1C1.

Triple-negative breast cancer (TNBC) is a specific subtype of breast cancer that poses a serious threat to women's health. The aldo-keto reductase type 1 C (AKR1C) family serves as a crucial ferroptosis defense system, catalyzing the conversion of aldehydes and ketones into their corresponding alcohols. SRY-Box4 (SOX4), a transcription factor of the SOX (sry-related HMG-box) family, is important in regulating tumor progression. This study aims to investigate the mechanism through which AKR1C1 mediates the growth, invasion, and ferroptosis of triple-negative breast cancer cells. SRPX2 was highl

Characterization of the microbiome and volatile compounds in anal gland secretions from domestic cats (Felis catus) using metagenomics and metabolomics.

Many mammals rely on volatile organic chemical compounds (VOCs) produced by bacteria for their communication and behavior, though little is known about the exact molecular mechanisms or bacterial species that are responsible. We used metagenomic sequencing, mass-spectrometry based metabolomics, and culturing to profile the microbial and volatile chemical constituents of anal gland secretions in twenty-three domestic cats (Felis catus), in attempts to identify organisms potentially involved in host odor production. We found that the anal gland microbiome was dominated by bacteria in the genera

Proteomic analysis of oxidative stress-resistant cells: a specific role for aldose reductase overexpression in cytoprotection.

We are using a proteomic approach that combines two-dimensional electrophoresis and tandem mass spectrometry to detect and identify proteins that are differentially expressed in a cell line that is resistant to oxidative stress. The resistant cell line (OC14 cells) was developed previously through chronic exposure of a parent cell line (HA1 cells) to increasing hydrogen peroxide concentrations. Biochemical analyses of this system by other investigators have identified elevated content and activity of several classical antioxidant proteins that have established roles in oxidative stress resista

Aldose reductase (AKR1B3) regulates the accumulation of advanced glycosylation end products (AGEs) and the expression of AGE receptor (RAGE).

Diabetes results in enhanced chemical modification of proteins by advanced lipoxidation end products (ALEs) and advanced glycation end products (AGEs) precursors. These modifications have been linked to the development of several secondary diabetic complications. Our previous studies showed that aldose reductase (AR; AKR1B3) catalyzes the reduction of ALEs and AGEs precursors; however, the in vivo significance of this metabolic pathway during diabetes and obesity has not been fully assessed. Therefore we examined the role of AR in regulating ALEs and AGEs formation in murine models of diet-ind

Design, Synthesis, In Silico ADME, Toxicity Prediction and Molecular Docking Studies of Benzimidazole-Oxadiazole Derivatives for α-Glucosidase and Aldose Reductase Pathways as Potent Anti-Diabetic Agents.

In this study, due to the side effect profiles and low efficacy of currently used inhibitors, novel benzimidazole-oxadiazole derivatives (6a-6e, 7a-7e) were synthesized as dual inhibitors of α-GLY and AR. Their structures were elucidated using 13C-NMR and 1H-NMR techniques. Their binding properties were investigated by molecular docking studies, and their ADME properties were screened in silico. AR and α-GLY inhibitory effects of the synthesized compounds were examined. The compounds were observed to exhibit partially similar inhibitory effects to the reference drug epalrestate (IC50: 0.78 nM;

Design, synthesis, and aldose reductase inhibition assessment of novel Quinazolin-4(3H)-one derivatives with 4-Bromo-2-Fluorobenzene functionality.

Aldose reductase (ALR2) inhibition is a promising therapeutic strategy for managing diabetes-related complications, including neuropathy, retinopathy, and nephropathy. This study reports the design, synthesis, and biological evaluation of eighteen novel quinazolin-4(3H)-one derivatives incorporating a 4-bromo-2-fluorobenzylidene moiety as ALR2 inhibitors. Among the synthesized compounds, the cyclohexyl-substituted derivative (compound 9) exhibited the highest potency as a competitive ALR2 inhibitor, with a Ki of 0.064 μM-15 times more effective than the standard inhibitor epalrestat (EPR) (Ki

Aldose reductase inhibition: emerging drug target for the treatment of cardiovascular complications.

Diabetes is a major metabolic disorder and leading cause of morbidity and mortality in the developed world. Indeed, vascular complications are the major cause of mortality in diabetics. Impaired glucose metabolism has been implicated as a cause for the observed cardiac dysfunction in diabetics. Dramatic increase in polyol pathway flux during hyperglycemia could cause damage to blood vessels that leads to heart failure. Recent investigations and patent applications suggest the interesting and important role of polyol pathway enzyme, aldose reductase (AR) in mediating oxidative stress-induced in

Schwann cell changes induced as early as one week after galactose intoxication.

In galactose neuropathy, aldose reductase inhibitor (ARI)-preventable Schwann cell injury has been reported in studies in which galactose feeding continued over a period of months. Given the link between these morphologic changes and polyol pathway flux, polyol accumulation after just days of galactose feeding points to the possibility that structural changes occur much earlier than previously reported. The aim of this study was to examine rat sciatic nerve after 7 days of galactose feeding for evidence of myelinated fiber injury and establish whether it is related to polyol accumulation. Comp

X-ray affinity and photoaffinity inhibitor of aldose reductase.

A reversible affinity inhibitor of rat lens aldose reductase (RLAR), that can be transformed into an irreversible inhibitor by low doses of X-rays or by exposure to light, has been synthesized and characterized. The inhibitor consists of quercitrin, a reversible receptor binding moiety, that is connected through a hexanediamine spacer to an azido containing moiety which becomes chemically reactive when irradiated. This reagent, N-(2-nitro-4-azidophenyl)-hexamethylenediamine-N'-(3'-O-quercit rin), (NAP-HEX-Q), represents a new class of affinity agents which have potential applications as target

Hyperglycemia as a cause of diabetic retinopathy.

Diabetes mellitus is marked by hyperglycemia and a variety of other metabolic disorders. The significance of hyperglycemia in the pathogenesis of diabetic retinopathy has proven difficult to evaluate in patients. Diabetic dogs are known to develop retinal lesions morphologically identical to those typical of diabetes in man, provided hyperglycemia in the animal is allowed to persist at least for many months and usually for 3 to 5 years. The development of retinopathy in this animal model can be inhibited by careful improvement of diabetic (glycemic) control. Comparable retinopathy has recently

The purification and properties of formate dehydrogenase and nitrate reductase from Escherichia coli.

The membrane-bound formate dehydrogenase of Escherichia coli grown anaerobically in the presence of nitrate was solubilized with deoxycholate and purified to near homogeneity. The purification procedure included ammonium sulfate fractionation and chromatography on Bio-Gel A-1.5m and DEAE Bio-Gel A in the presence of the nonionic detergent, Triton X-100. This detergent caused a significant decrease in the molecular weight of the soluble formate dehydrogenase complex and allowed the enzyme then to be resolved from other membrane components. Anaerobic conditions were required throughout due to th

Parasitoid wasp venom elevates sorbitol and alters expression of metabolic genes in human kidney cells.

Venom from the parasitoid wasp Nasonia vitripennis dramatically elevates sorbitol levels in its natural fly hosts. In humans, sorbitol elevation is associated with complications of diabetes. Here we demonstrate that venom also induces this disease-relevant phenotype in human cells, and investigate possible pathways involved. Key findings are that (a) low doses of Nasonia venom elevate sorbitol levels in human renal mesangial cells (HRMCs) without changing glucose or fructose levels; (b) venom is a much more potent inducer of sorbitol elevation than glucose; (c) low venom doses significantly al

Inhibiting wild-type and C299S mutant AKR1B10; a homologue of aldose reductase upregulated in cancers.

AKR1B10 is an aldose reductase (AR) homologue overexpressed in liver cancer and various forms of that enzyme in carcinomas catalyze the reduction of anticancer drugs, potential cytostatic drug, and dl-glyceraldehyde but do not catalyze the reduction of glucose. Kinetic parameters for wild-type and C299S mutant AKR1B10 indicate that substitution of serine for cysteine at position 299 reduces the affinity of this protein for dl-glyceraldehyde and enhances its catalytic activity. Fibrates suppress peroxisome proliferation and the development of liver cancer in human. Here we report the potency of

[New aspects of hexobarbital metabolism: stereoselective metabolism, new metabolic pathway via GSH conjugation, and 3-hydroxyhexobarbital dehydrogenases].

Hexobarbital, a short-acting hypnotic, is metabolized to 3'-hydroxyhexobarbital by cytochrome P450, and then to 3'-oxohexobarbital by liver cytosolic dehydrogenase. New methods of separation for hexobarbital and its metabolites by TLC have been developed and applied to study the metabolism of hexobarbital enantiomers and stereoselective metabolism of hexobarbital. (+)-Hexobarbital preferentially was transformed into beta-3'-hydroxyhexobarbital and the (-)-enantiomer preferentially transformed into alpha-3'-hydroxyhexobarbital by rat liver microsomes. Identification of a common ketohexokinase-dependent link driving alcohol intake and alcohol-associated liver disease in mice.

Alcohol and sugar share reinforcing properties and both contribute to liver disease progression, ultimately leading to cirrhosis. Emerging evidence suggests that ethanol activates the aldose reductase pathway, resulting in endogenous fructose production. Here we investigated whether alcohol preference and alcohol-associated liver disease (ALD) are mediated through fructose metabolism by ketohexokinase (KHK)-A/C. Using global, conditional and tissue-specific KHK-A/C knockout mice, we assessed ethanol intake, reinforcement behaviours and liver injury. Ethanol consumption increased portal vein os

Installing the neurospora carotenoid pathway in plants enables cytosolic formation of provitamin A and its sequestration in lipid droplets.

Vitamin A deficiency remains a severe global health issue, which creates a need to biofortify crops with provitamin A carotenoids (PACs). Expanding plant cell capacity for synthesis and storing of PACs outside the plastids is a promising biofortification strategy that has been little explored. Here, we engineered PAC formation and sequestration in the cytosol of Nicotiana benthamiana leaves, Arabidopsis seeds, and citrus callus cells, using a fungal (Neurospora crassa) carotenoid pathway that consists of only three enzymes converting C5 isopentenyl building blocks formed from mevalonic acid in

Vascular biotransformation of organic nitrates is independent of cytochrome P450 monooxygenases.

BACKGROUND AND PURPOSE: Organic nitrates such as nitroglycerin (NTG) or pentaerythritol tetranitrate (PETN) have been used for over a century in the treatment of angina or ischaemic heart disease. These compounds are prodrugs which release their nitrovasodilators upon enzymic bioactivation by aldehyde dehydrogenase (ALDH2) or cytochromes P450 (CYP). Whereas ALDH2 is known to directly activate organic nitrates in vessels, the contribution of vascular CYPs is unknown and was studied here. EXPERIMENTAL APPROACH: As all CYPs depend on cytochrome P450 reductase (POR) as electron donor, we generate
