Comparison of adjuvants to optimize influenza neutralizing antibody responses.

Seasonal influenza vaccines represent a positive intervention to limit the spread of the virus and protect public health. Yet continual influenza evolution and its ability to evade immunity pose a constant threat. For these reasons, vaccines with improved potency and breadth of protection remain an important need. We previously developed a next-generation influenza vaccine that displays the trimeric influenza hemagglutinin (HA) on a ferritin nanoparticle (NP) to optimize its presentation. Similar to other vaccines, HA-nanoparticle vaccine efficacy is increased by the inclusion of adjuvants dur

[Acute myelitis associated with anti-neutral glycolipid antibody].

A 48-year-old man with rapid onset of fever elevation developed acute myelitis over a period of a week. MRI of the spinal cord revealed a longitudinal T2-hyperintense intraspinal lesion extending from C6 to Th8 level. Clinical symptoms and findings resolved with immunotherapy. In serological analysis, no antibodies related to various collagen diseases, anti-aquaporin-4 (AQP4) antibody and anti-myelin oligodendrocyte glycoprotein (MOG) antibody were detected. Anti-lactosylceramide (LacCer) antibodies were detected in the acute phase of serum and cerebrospinal fluid, with titers showing decremen

Heritability of vaccine-induced measles neutralizing antibody titers.

Understanding how genetics influences inter-individual variation of antibody titers in response to measles vaccination is vital to understanding possible sources of vaccine failure as well as improved vaccine development. Although it is recognized that both the human leukocyte antigen (HLA) genes and the immunoglobulin allotype genes play significant roles in immune response, there is significant variation in antibody titers that is not explained by these genes. To obtain a more complete estimate of the role of the entire genome, we used a large panel of single nucleotide polymorphisms to esti

Structural basis for antibody-mediated neutralization of Lassa virus.

The arenavirus Lassa causes severe hemorrhagic fever and a significant disease burden in West Africa every year. The glycoprotein, GPC, is the sole antigen expressed on the viral surface and the critical target for antibody-mediated neutralization. Here we present the crystal structure of the trimeric, prefusion ectodomain of Lassa GP bound to a neutralizing antibody from a human survivor at 3.2-angstrom resolution. The antibody extensively anchors two monomers together at the base of the trimer, and biochemical analysis suggests that it neutralizes by inhibiting conformational changes require

Human picornaviruses associated with neurological diseases and their neutralization by antibodies.

Picornaviruses are the most commonly encountered infectious agents in mankind. They typically cause mild infections of the gastrointestinal or respiratory tract, but sometimes also invade the central nervous system. There, they can cause severe diseases with long-term sequelae and even be lethal. The most infamous picornavirus is poliovirus, for which significant epidemics of poliomyelitis were reported from the end of the nineteenth century. A successful vaccination campaign has brought poliovirus close to eradication, but neurological diseases caused by other picornaviruses have increasingly

JC polyomavirus mutants escape antibody-mediated neutralization.

JC polyomavirus (JCV) persistently infects the urinary tract of most adults. Under conditions of immune impairment, JCV causes an opportunistic brain disease, progressive multifocal leukoencephalopathy (PML). JCV strains found in the cerebrospinal fluid of PML patients contain distinctive mutations in surface loops of the major capsid protein, VP1. We hypothesized that VP1 mutations might allow the virus to evade antibody-mediated neutralization. Consistent with this hypothesis, neutralization serology revealed that plasma samples from PML patients neutralized wild-type JCV strains but failed

A requirement for FcγR in antibody-mediated bacterial toxin neutralization.

One important function of humoral immunity is toxin neutralization. The current view posits that neutralization results from antibody-mediated interference with the binding of toxins to their targets, a phenomenon viewed as dependent only on antibody specificity. To investigate the role of antibody constant region function in toxin neutralization, we generated IgG2a and IgG2b variants of the Bacillus anthracis protective antigen-binding IgG1 monoclonal antibody (mAb) 19D9. These antibodies express identical variable regions and display the same specificity. The efficacy of antibody-mediated ne

Development and evaluation of a rapid neutralizing antibody test for rabies.

The level of virus-neutralizing antibody, which plays a crucial role in the prevention of rabies, is determined by rabies virus (RABV) neutralizing test, which are time- and cost-consuming. In order to determine the level of neutralizing antibody in vaccinees, an easy and reliable method is needed. Based on the principle of immunochromatography, we developed a RAPINA (RAPId Neutralizing Antibody) test to determine the presence of neutralizing antibody in serum. In the RAPINA test, if neutralizing antibody equivalent to 0.5IU/ml of serum sample are mixed with an optimal amount of inactivated RA

[Anti-verotoxin-neutralizing antibody in intravenous gammaglobulin preparations].

Intravenously administered immune globulin therapy has been reported to be an effective treatment for serious patients with verotoxin-producing Escherichia coli infections while an efficacy for VT2 are under discussion. We therefore examined in vitro commercially available immune globulin preparations for the presence of anti-VT neutralizing antibodies in expectation of protecting a patient from serious complications as HUS. We examined 47 lots of gamma-globulin prepared by 5 pharmaceutical companies in Japan. Of them 29 lots were prepared from imported blood and 18 lots were from Japanese don

Neutralization of bee venom lethality by immune serum antibodies.

The lethal effects of Africanized honey bee venom depend on the absorption of venom delivered during simultaneous sting attacks by large numbers of bees. The hypothesis that antibodies to whole bee venom and bee venom components could neutralize the lethal effect of bee venom was tested. Antibodies from beekeepers and immunized rabbits were incubated with bee venom and neutralization was studied by survival of intravenously injected mice. Beekeeper serum antibodies were found effective in protecting mice challenged with whole venom, and serum from rabbits immunized with phospholipase A2 (PLA2)

Treatment of toxic shock syndrome with endotoxin-neutralizing antibody.

The enhancement of host susceptibility to endotoxin (lipopolysaccharide, LPS) by toxic shock syndrome toxin-1 (TSST-1) is an important mechanism in the pathogenesis of toxic shock syndrome. In these studies, we sought to determine whether an endotoxin-neutralizing monoclonal antibody could be useful in the treatment of toxic shock syndrome. We isolated a murine monoclonal hybridoma (3-H3) which secreted monoclonal antibody (mAb) specific for Escherichia coli 0111:B4 LPS. Spleen cells cocultured with E. coli 0111:B4 LPS demonstrated up to a 60% decrease in mitogenic activity in the presence of

Neutralizing antibody responses to Japanese encephalitis vaccine in children.

Two shots of the current Japanese encephalitis (JE) vaccine were given to children and their immune responses to the Nakayama strain (the vaccine strain) and two wild strains (JaGAr-01 and E-50) of virus were examined by neutralizing (N) antibody titrations. Seventy vaccinees without antibody to JE virus before the first vaccination were bled one month after the second vaccination. The N antibody responses to the JaGAr-01 and E-50 strains were found to be similar and to be less than that to the Nakayama strain after the second vaccination: the geometric mean titers (GMT) of N antibodies to the

Determination of serum neutralizing antibodies reveals important difference in quality of antibodies against pertussis toxin in children after infection.

OBJECTIVES: To determine neutralizing antibodies to pertussis toxin (PTNAs) in children with suspected pertussis and to compare results of PTNAs and anti-PT IgG antibodies. METHODS: 172 hospitalized children with suspected pertussis were included. Pertussis was confirmed by culture, PCR and/or serology. PTNAs were determined by Chinese hamster ovary (CHO) cell assay. RESULTS: A correlation between titers of PTNAs and anti-PT IgG levels was noticed in 172 patients (Spearman R = 0.68, P < 0.001). Subjects with same concentrations of anti-PT IgG antibodies could have different titers of PTNAs a

Neutralizing antibodies as A therapy for hepatitis E virus: Advances, challenges and opportunities.

Hepatitis E virus (HEV) infection is a leading cause for acute hepatitis worldwide posing a significant public health threat but no FDA-approved therapy is currently available. Studies have shown that HEV-specific antibodies are protective against HEV infection both in vitro and in vivo; therefore, antibodies may be considered a potential therapy for high-risk populations (e.g., pregnant women and immunocompromised individuals). Many potent neutralizing monoclonal antibodies (mAbs) against HEV have been identified and characterized, showing great promise for therapeutic development. Here we pr

Potently neutralizing and protective human monoclonal antibodies against the measles fusion protein trimer.

Measles remains a significant source of morbidity and mortality globally. There are currently no licensed treatments for patients at high risk of severe disease. There is limited knowledge about the human antibody response to a measles infection or vaccination or the structural determinants of antibody-mediated neutralization on the measles virus surface hemagglutinin (H) and fusion (F) glycoproteins. Here, we describe a panel of human monoclonal antibodies isolated from individuals who had been naturally infected with the measles virus. Several antibodies that recognized a prefusion-stabilize

Broadly neutralizing monoclonal antibodies against influenza A viruses: current insights and future directions.

Monoclonal antibodies (mAbs) have become attractive tools for both the treatment and prevention of influenza A viruses due to their ability to target several viral components, which confers broad therapeutic potential. Advances in biotechnology, such as hybridoma technology, phage display technology, B cell immortalization, and artificial intelligence (Al)-driven antibody design, have significantly accelerated the development of effective mAbs. Clinical trials have shown that mAbs can improve clinical outcomes particularly in high-risk and immunocompromised populations by lowering viral loads

Zika virus infection in early pregnancy increases the likelihood of persistent neutralizing antibodies.

BACKGROUND: Neutralizing antibodies (nAbs) play a central role in protection against Zika virus (ZIKV) infection. Higher maternal ZIKV nAb titers during pregnancy have been associated with reduced risk of congenital anomalies. However, limited data exist on the long-term persistence of these antibodies in women infected during pregnancy and in children exposed in utero. METHODOLOGY/PRINCIPAL FINDINGS: We conducted a cross-sectional serological study using stored serum samples from a cohort of mother-child pairs with confirmed maternal ZIKV infection during pregnancy in Campo Grande, Brazil.
