The effect of estrogen on appetite.

Such eating disorders as anorexia nervosa, bulimia nervosa, and binge eating disorder are associated with significant morbidity and mortality in women. The etiology of these disorders and the causes of women's increased vulnerability to them remain obscure. The lack of understanding of the biological bases of normal and abnormal human eating behavior impedes development of effective pharmacologic treatment for eating disorders. A review of basic research, implicating estradiol in the physiologic control of eating in laboratory animals, shows potential heuristic and mechanistic significance for

Neurochemical and behavioral factors in the development of tolerance to anorectics.

Tolerance and cross-tolerance among several anorectic drugs were investigated in four separate tests. The same animals served as subjects in all four tests. In the first test, animals given milk shortly after injection of 3 mg/kg amphetamine (Cont group) developed tolerance to amphetamine anorexia, but animals given milk when amphetamine's anorectic effects had worn off (Noncon group) did not develop tolerance in spite of equal drug exposure. In the second test, Cont animals were tolerant to 2 mg/kg apomorphine, a drug with a neurochemical action related to amphetamine. No tolerance to 2 mg/kg

The clinical pharmacology of appetite suppressant drugs.

One way of gaining a greater understanding of the central mechanisms underlying hunger and the regulation of feeding behaviour in humans is to examine the actions and interactions on hunger and food intake of drugs with known or presumed pharmacological modes of action. To this end we have undertaken a number of studies which fall into three main categories: the mechanisms by which amphetamine anorexia is induced; the possible role of endogenous opioids in feeding; the action of amino acids thought to be involved in the regulation of feeding. In this field the potential for cross-fertilization

C-terminal octapeptide of cholecystokinin decreases food intake in man.

In comparison with a saline infusion, the infusion of the C-terminal octapeptide of cholecystokinin (4 ng/kg/min) decreased food intake by an average of 122 g in a group of 12 lean men without objective evidence of untoward side effects. Shapes of the cumulative intake curves under the two conditions were similar, but subjects ate less and stopped eating sooner when receiving octapeptide than when receiving saline. These results are consistent with the hypothesis that cholecystokinin is an endogenous signal for postprandial satiety. The results offer promise for the possible use of the octapep

CCK-octapeptide injected in CSF decreases meal size and daily food intake in sheep.

Cholecystokinin octapeptide (CCK-OP) is a potent and specific suppressor of feeding when administered as a continuous lateral cerebral ventricular injection in fasted sheep, and we have proposed that endogenous CCK-OP in the brain is released during meals and acts to terminate feeding. In previous studies, however, only relatively short-term effects of CCK-OP on feeding were examined. In the first experiment of the present series sheep were adapted to a 6-hr feeding period per day. CCK-OP injected continuously for 6 hr into the lateral ventricles reduced feeding during the entire feeding perio

Associations of maternal food addiction, dietary restraint, and pre-pregnancy BMI with infant eating behaviors and risk for overweight.

Maternal food addiction, dietary restraint, and pre-pregnancy body mass index (BMI) are associated with high-risk eating behaviors and weight characteristics in children and adolescents. However, little is known about how these maternal factors are associated with individual differences in eating behaviors and risk for overweight in infancy. In a sample of 204 infant-mother dyads, maternal food addiction, dietary restraint and pre-pregnancy BMI were assessed using maternal self-report measures. Infant eating behaviors (as measured by maternal report), objectively measured hedonic response to s

Effects of fasting on the gene expression of appetite regulators in three Characiformes with different feeding habits (Gymnocorymbus ternetzi, Metynnis argenteus and Exodon paradoxus).

In order to assess potential interspecific differences in the endocrine mechanisms regulating feeding in Characiformes, we used three fish species with different feeding habits: two Characidae, the omnivore black widow tetra and the carnivore bucktooth tetra, and one Serrasalmidae, the herbivore silver dollar, as models. cDNAs encoding for appetite-regulating peptides (orexin, cocaine and amphetamine regulated transcript CART, cholecystokinin CCK and leptin) were isolated and their tissue distribution examined. The protein sequences of the three species showed most similarities with those of o

A review of the physiological bases of the anorexic action of phenylpropanolamine (d,1-norephedrine).

Phenylpropanolamine (PPA) is a phenethylamine that induces a variety of effects including anorexia and weight loss. The present review compares the acute anorexic effects of PPA with those of amphetamine in animals and humans, describes the persistent chronic effects of PPA on feeding as well as differences in anorexic potency between the norephedrine enantiomers (d-norephedrine less than 1-norephedrine) and summarizes the generality of PPA anorexia. The review also describes the putative mechanisms by which PPA is thought to reduce feeding behavior including alteration of gastric emptying and

Effect of restraint stress on feeding behavior of rats.

The expression of appetite reflects the complex functioning of a psychobiological system organized in different levels closely related to each other, in which emotional changes can influence feeding behavior. Benzodiazepines are widely used as anxiolytics and can change behaviors caused by stress. The aim of the present study was to verify the feeding behavior of rats, submitted or not to fasting, after acute and chronic restraint stress. We also evaluated the response to the ingestion of sweet food of chronically restrained animals after the administration of diazepam. Male adult Wistar rats

Physiology: does gut hormone PYY3-36 decrease food intake in rodents?

Batterham et al. report that the gut peptide hormone PYY3-36 decreases food intake and body-weight gain in rodents, a discovery that has been heralded as potentially offering a new therapy for obesity. However, we have been unable to replicate their results. Although the reasons for this discrepancy remain undetermined, an effective anti-obesity drug ultimately must produce its effects across a range of situations. The fact that the findings of Batterham et al. cannot easily be replicated calls into question the potential value of an anti-obesity approach that is based on administration of PYY

The impact of early neglect on defensive and appetitive physiology during the pubertal transition: a study of startle and postauricular reflexes.

This study tested the effect of early neglect on defensive and appetitive physiology during puberty. Emotion-modulated reflexes, eye-blink startle (defensive) and postauricular (appetitive), were measured in 12-to-13-year-old internationally adopted youth (from foster care or from institutional settings) and compared to non-adopted US born controls. Startle Reflex: adopted youth displayed lower overall startle amplitude across all valences and startle potentiation to negative images was negatively related to severity of pre-adoption neglect. Postauricular reflex (PAR): adopted youth showed lar

Nonmedical prescription stimulant use for suppressing appetite and controlling body weight is uniquely associated with more severe eating disorder symptomatology.

OBJECTIVE: Nonmedical prescription stimulant use (NPS; i.e., use without a prescription or in ways other than prescribed) to suppress appetite or control weight appears to be associated with eating disorder (ED) symptomatology among college students. However, it remains unknown if this relationship is motive-specific and uniquely related to ED symptomatology. This research examined whether engaging in NPS specifically for appetite/weight-related purposes is associated with ED symptomatology and a unique indicator of more severe symptomatology. METHOD: A nonclinical sample of college students

Amiloride sensitivity of the chorda tympani response to sodium chloride in sodium-depleted Wistar rats.

Peripheral gustatory mechanisms that may contribute to the expression of sodium (Na) appetite have been a focus of interest for many years. Because amiloride-sensitive Na transport is involved in the generation of neural signals in response to NaCl stimulation, the present study assessed whether changes in amiloride sensitivity of the neural response to NaCl accompany the induction of a Na appetite in the rat. Na deprivation was achieved by acute depletion with the diuretic furosemide. The magnitude of the whole-nerve chorda tympani response to 0.5 M NaCl was reduced in Na-depleted, compared w

Appetitive responses to computer-generated visual stimuli by the praying mantis Sphodromantis lineola (Burr.).

Tethered adult female praying mantises, Sphodromantis lineola (Burr.), were presented with various computer-generated visual stimuli that moved against patterned or homogeneous white backgrounds in predetermined patterns and at predetermined speeds. The degrees to which the stimulus configurations elicited appetitive behaviors (attempting to approach and/or striking) indicated the relative degrees to which the stimuli were classified as prey. Mantises readily struck at cartoon "crickets" that subtended visual angles as great as 24.5 deg x 62.5 deg, but response rate was suppressed if the stimu…

Abuse of drugs associated with eating disorders.

Concomitant bulimia nervosa and drug abuse are common in women. Drugs used by this group include diuretics, emetics, laxatives, and diet pills, as well as alcohol, cigarettes, and illicit street drugs. This paper applies principles from behavioral pharmacology to the problem of drug use by women with bulimia nervosa. The prevalence of use, primary effects, toxicity, detection, tolerance, withdrawal, and effects on appetite and weight are discussed for drugs used by bulimic women to reduce appetite or weight or to induce purging (e.g., diuretics, emetics, laxatives, and diet aids). Microbiota-gut-brain axis: relationships among the vagus nerve, gut microbiota, obesity, and diabetes.

AIMS: The purpose of this review is to explore the interconnected pathways of the microbiota-gut-brain axis (MGBA), focusing on the roles of the vagus nerve and glucagon like peptide-1 in appetite control, and in the development of obesity and diabetes. METHODS: Type 2 diabetes mellitus (T2DM) and obesity are metabolic disorders whose prevalence has significantly increased in recent decades and is expected to increase every year, to pandemic proportions. These two pathologies often coexist and have substantial public health implications. The term "diabesity" defines the pathophysiological con

The effects of anorexic drugs on free-fed rats responding under a second-order FI15-min (FR10:S) schedule for high incentive foods.

Many similarities exist between the overconsumption of food, which results in obesity, and drug addiction. The present study investigated the effects of anorectic drugs on responding maintained by high incentive, but nutritionally unnecessary, food reinforcers using an FI15(fixed-ratio 10:S) schedule of reinforcement, similar to that used in studies on the incentive properties of drugs of abuse. Rats were trained to respond on a lever to gain access to two high incentive foods--chocolate chip cookies and cheese. Under the FI15(FR10:S) schedule, every 10th response (fixed-ratio 10) delivered a
