Illuminating the therapeutic potential of Schisandrin a against rheumatoid arthritis by targeting ferroptosis: An integrated bioinformatics and experimental study.

Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic inflammation and hyperplasia of the synovial membrane. This study aimed to elucidate the anti-inflammatory mechanisms of Schisandrin A (SCHA), a bioactive component from Schisandra chinensis, on RA fibroblast-like synoviocytes (FLSs). Integrated bioinformatics analysis was performed on transcriptomic datasets from RA and normal FLSs as well as SCHA-treated RA FLSs. Enrichment analysis revealed that differentially expressed genes between groups were significantly enriched in the ferroptosis pathway, suggesting SCHA may

Impact of transcription factors AP-1 and NF-kappaB on the outcome of experimental Staphylococcus aureus arthritis and sepsis.

Staphylococcus aureus infection is, despite adequate antibiotic treatment, a disease characterized by high mortality. The bacterium triggers an exaggerated immune response in the host, which on the one hand acts as an efficient defense, but on the other hand gives rise to tissue damage. In this study we have modulated the hosts response to S. aureus by inhibition of nuclear factor kappaB (NF-kappaB) and activator protein-1 (AP-1)-triggered release of pro-inflammatory cytokines and tissue-destructive proteins, respectively. Mice were administered with antisense oligonucleotides (ODN) to the p65

Studies on type II collagen induced arthritis in rats: an experimental model of peripheral and axial ossifying enthesopathy.

It was suggested that type II collagen induced arthritis in rats may be considered an experimental model for rheumatoid arthritis (RA) because of clinical, histological, and immunological similarities. However, some features separate it from RA. We studied 40 inbred female Wistar Furth rats. Two weeks after immunization with native human type II collagen, they had polyarthritis which progressed to ankylosis associated with ossifying enthesopathy and periosteal new bone formation. Inflammatory nodules of the tail appeared after 2 months, with radiological and histopathological aspects of multis

Comparison of inoculation regimes for the experimental production of swine erysipelas arthritis. I. Clinical, pathological and bacteriological findings.

The arthritic form of swine erysipelas was induced in pigs by multiple intravenous inoculation of 2 strains of Erysipelothrix rhusiopathiae. The strains differed significantly in their arthritogenicity but not in the number of cases of lameness induced. The use of 3 intravenous inoculations instead of 5 did not significantly affect the outcome. In a second trial, the more arthritogenic strain was injected in ten-fold dilutions from 5 x 10(9) to 5 x 10(4) organisms. Pigs receiving the lower doses showed high variability in their arthritic responses that precluded sensitive analysis of the dose

Network Pharmacology and Experimental Validation Reveal Wu Miao Pill's Therapeutic Effects on Acute Gouty Arthritis Through P2X7R Regulation.

Acute gouty arthritis (AGA) is a common inflammatory joint disease with limited therapeutic options and notable side effects from conventional treatments. This study investigated the anti-inflammatory effects and underlying mechanisms of the traditional Chinese herbal formula Wu Miao Pill (WMP) in a rat model of AGA induced by monosodium urate crystals. WMP administration markedly alleviated joint inflammation and swelling, improved synovial histopathology, and reduced the expression of P2X7 receptor (P2X7R) and pro-inflammatory cytokines IL-1β, IL-18, and TNF-α. P2X7R inhibition and knockdown

Systemic overexpression of interleukin-22 induces the negative immune-regulator SOCS3 and potently reduces experimental arthritis in mice.

OBJECTIVE: High levels of IL-22 are present in serum and synovial fluid of patients with RA. As both pro- and anti-inflammatory roles for IL-22 have been described in studies using animal models of RA, its exact function in arthritis remains poorly defined. With this study we aimed to further unravel the mechanism by which IL-22 exerts its effects and to decipher its therapeutic potential by overexpression of IL-22 either locally or systemically during experimental arthritis. METHODS: CIA was induced in DBA-1 mice by immunization and booster injection with type II collagen (col II). Before ar

11β-Hydroxysteroid dehydrogenase type 1, but not type 2, deficiency worsens acute inflammation and experimental arthritis in mice.

Glucocorticoids profoundly influence immune responses, and synthetic glucocorticoids are widely used clinically for their potent antiinflammatory effects. Endogenous glucocorticoid action is modulated by the two isozymes of 11β-hydroxysteroid dehydrogenase (11β-HSD). In vivo, 11β-HSD1 catalyzes the reduction of inactive cortisone or 11-dehydrocorticosterone into active cortisol or corticosterone, respectively, thereby increasing intracellular glucocorticoid levels. 11β-HSD2 catalyzes the reverse reaction, inactivating intracellular glucocorticoids. Both enzymes have been postulated to modulate

Whole-body deletion of LPS-induced TNF-α factor (LITAF) markedly improves experimental endotoxic shock and inflammatory arthritis.

LPS-induced TNF-α factor (LITAF) mediates cytokine expression in response to endotoxin challenge. Previously, we reported that macrophage-specific LITAF-deficient (macLITAF-/-) mice exposed to LPS have a delayed onset in the serum levels of proinflammatory cytokines and prolonged persistence of anti-inflammatory cytokines, but only partial protection from endotoxic shock. We postulated that greater protection might be achieved if LITAF were deleted from all LITAF-producing cells, including macrophages. Using a Cre-loxP system, we engineered a tamoxifen-induced recombination mouse [tamLITAF(i)-

Association between condylar morphology and changes in bony microstructure and sub-synovial inflammation in experimental temporomandibular joint arthritis.

BACKGROUND: In juvenile idiopathic arthritis involvement of the temporomandibular joints (TMJs) is often associated with mandibular growth deviations. The relation between the growth deviations and severity of the inflammation, condylar shape, the micro-architecture, and the quality of the bone has not previously been investigated. This paper studies the effect on the bony structures in mandibular condylar development in rabbits with antigen-induced arthritis. METHODS: Included were 42 juvenile rabbits with ovalbumin-induced arthritis of the TMJs treated with intraarticular saline, intraartic

Condylar lesions in relation to mandibular growth in untreated and intra-articular corticosteroid-treated experimental temporomandibular joint arthritis.

BACKGROUND: Many attempts have been made to improve the treatment success rate of chronic bacterial prostatitis (CBP). However, no treatment modality has achieved complete cure. The growth and development of the prostate is under direct hormonal control, and it is possible that prostatitis may be directly influenced by its hormonal milieu in a similar fashion to benign prostatic hyperplasia and prostate cancer. Therefore, the effects of androgen deprivation on the treatment of CBP were investigated in rats. METHODS: Experimental CBP was induced in one hundred, male Wistar rats by instillation

Addition of bisphosphonate to antibiotic and anti-inflammatory treatment reduces bone resorption in experimental Staphylococcus aureus-induced arthritis.

Bacterial arthritis is a disease with high morbidity leading to rapidly progressive bone resorption. We have shown earlier that treatment with antibiotics in combination with corticosteroids decreases joint inflammation and mortality but does not significantly affect bone/cartilage destruction of the joints. This study was performed to assess the effect of treatment with bisphosphonate [zoledronic acid (ZA)] in combination with antibiotics and corticosteroids, on the course and outcome of Staphlococcus aureus-induced arthritis. Three days after intravenous inoculation with S. aureus, mice were

Dual anti-inflammatory and immunomodulatory effects of clove essential oil and eugenol: targeting neutrophil functions and experimental arthritis.

Polymorphonuclear neutrophils (PMNs) contribute to the pathogenesis of arthritis by releasing excessive reactive oxygen species (ROS) and proteolytic enzymes, leading to joint damage. Targeting PMNs activation represents a promising therapeutic approach. This study aimed to evaluate the immunomodulatory and anti-arthritic effects of clove essential oil (CEO) and its main constituent, eugenol, through in vitro, in vivo, and in silico approaches. CEO chemical composition was determined by GC-MS. In vitro assays assessed PMNs degranulation (lysozyme release) and oxidative burst (NBT reduction), a

Sanmiao pill ameliorates rheumatoid arthritis via PPARγ-mediated lipid accumulation in macrophage: A multi-omics and experimental validation.

BACKGROUND: The enhanced infiltration of M1-polarized macrophages into synovium acts as a central driver of chronic inflammation characteristics of rheumatoid arthritis (RA). Sanmiao pill (SMP), a classic traditional formulation, has long been employed in the treatment of arthralgia syndromes. Despite its recognized therapeutic efficacy, the underlying mechanism remains unclear. PURPOSE: To explore the therapeutic potential and protective mechanisms of SMP against RA. METHODS: A collagen-induced arthritis (CIA) rat model was established, the therapeutic effects were evaluated by arthritis se

Role of inter-alpha-trypsin inhibitor heavy chain 4 and its citrullinated form in experimental arthritis murine models.

Inter-α-trypsin inhibitor heavy chain 4 (ITIH4) is a major protein in serum and reported to be upregulated at the onset of rheumatoid arthritis (RA). Its citrullinated form, cit-ITIH4, is specifically found in the serum and synovial fluid of patients with RA. However, the detailed function of ITIH4 in arthritis remains unknown. The aim of this study was to clarify the role of ITIH4 and cit-ITIH4 using experimental arthritis models. ITIH4 and cit-ITIH4 expression was examined in steady-state mice and two different arthritis models, and their pathological effects were examined in Itih4-deficient

Biomaterial-based combinatorial approach of aescin-comprised zein-coated gelatin nanoparticles alleviates synovial inflammation in experimental inflammatory arthritis.

Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disease that mostly affects joints. Although RA therapy has made significant progress, difficulties including extensive medication metabolism and its quick clearance result in its inadequate bioavailability. The anti-inflammatory effect of zein was reported with other medications, but it has certain limitations. There are reports on the anti-oxidant and anti-inflammatory effect of aescin, which exhibits low bioavailability for the treatment of rheumatoid arthritis. Also, the combinatorial effect of zein with other effective drug de

Sorafenib suppresses proliferation rate of fibroblast-like synoviocytes through the arrest of cell cycle in experimental adjuvant arthritis.

OBJECTIVES: Rheumatoid arthritis, a recurrent incendiary autoimmune joint syndrome, features by prominent synovial hyperplasia. Fibroblast-like synoviocytes are the executive components in the pathogenesis of rheumatoid arthritis. It is generally accepted that excessive proliferation and reduced apoptosis of fibroblast-like synoviocytes lead to synovial hyperplasia. Our previously studies found that sorafenib could inhibit adjuvant arthritis in rats and induced adjuvant arthritis fibroblast-like synoviocytes apoptosis. Presently, we aim to investigate the inhibitory effect with mechanisms of a
