OBJECTIVES: In this study, we investigated the clinical features, etiology, and also predictive factors of secondary erythema nodosum (EN) in patients with EN.
METHODS: A total of 100 patients (mean age: 37 years) diagnosed with EN between 1993 and 2004 in our clinic were included in the study prospectively. A skin biopsy was performed in 46 of the patients. Patients were considered to have secondary EN when an underlying condition was found, and to have primary EN when no such condition was found. For the diagnosis of the underlying diseases, the pertinent diagnostic criteria and/or diagnost

Accurate and species-specific diagnosis of Plasmodium vivax malaria remains a significant challenge in malaria management due to the insuffi … abling clear, instrument-free interpretation. The assay exhibited no cross-reactivity with P. falciparum LDH or human LDH, thereby ensuring robust species-specific discrimination. Given its high sensitivity, portability, and strong performance in complex biological matrices, this CNB-based LFIA represents a promising next-generation point-of-care diagn …

BACKGROUND: Artemisinin-based combination therapies are the cornerstone of Plasmodium falciparum malaria treatment; however, delayed parasite clearance and treatment failure occur even in the absence of established parasite genetic resistance. While parasite-intrinsic resistance mechanisms have been extensively studied, the contribution of host erythroid heterogeneity to antimalarial drug response remains poorly defined.
METHODS: Highly purified human CD71+ve reticulocytes and mature erythrocytes were used to assess parasite invasion, intraerythrocytic development, and drug susceptibility. In…

Malaria caused by Plasmodium falciparum remains a health burden worldwide due to drug resistance and limited treatment options. Calcium-dependent protein kinase 1 (CDPK1) plays a central role in parasite development and invasion, but the downstream molecular alterations that occur upon its disruption remain poorly understood. We present a proteogenomic-based data analysis pipeline for the reanalysis of the publicly available P. falciparum CDPK1 mutant dataset (PRIDE: PXD005207), integrating proteomic and phosphoproteomic data with six-frame genome translation. This led to the discovery of 24 n…

BACKGROUND: In sub-Saharan Africa, including Ethiopia, malaria continues to pose a significant public health risk. Efforts to control the disease are complicated by emerging resistance to current drugs and concerns about the sustained effectiveness of antimalarial medications. The purpose of this systematic review and meta-analysis is to determine the nationwide prevalence of the Pfcrt K76T and Pfmdr1 N86Y genetic mutations in Ethiopia.
METHODS: We queried multiple sources for literature, including the Google Scholar, Cochrane Library, Scopus, Web of Science, and PubMed/MEDLINE databases. Fin

BACKGROUND AND OBJECTIVES: Infection with multiple genetically distinct Plasmodium falciparum parasites is common in human malaria cases. Within an infected patient, a particular genotype may thrive, influencing its chance of transmission. Competitive growth assays provide a quantitative index to assess relative dynamic fitness disparities among co-infecting parasites across experimentally controlled, physiologically relevant conditions to test specific hypotheses. The ability to assess the relative competitive fitness of parasite genotypes present in patient-derived samples will be particular…

Artemisinin-based combination therapies (ACTs) remain the cornerstone of malaria treatment, yet the emergence of artemisinin partial resistance (ART-R) in Africa threatens their efficacy. ART-R is primarily associated with mutations in Plasmodium falciparum kelch13 (K13), notably R561H, which has been linked to delayed parasite clearance in East Africa. We genotyped 2,866 P. falciparum isolates from seven districts in Tanzania's Kagera region (2021-2023) using 121 molecular inversion probes (MIP) targeting key resistance loci to characterize trends in ART-R and other resistance markers. The WH…

BACKGROUND: Plasmodium falciparum malaria remains a major cause of morbidity and mortality in endemic regions, with outcomes influenced by host genetic factors. The ABO blood group system has been linked to clinical severity, though findings differ regarding blood groups and severe malaria susceptibility. This analysis aims to synthesise evidence on ABO blood group's role in severe P. falciparum malaria (SM) through meta-analysis and trial sequential analysis.
MATERIALS AND METHODS: A search was conducted across PubMed, Scopus, Embase, Web of Science and Science Direct databases on August 31,…

Plasmodium falciparum is the most dangerous malaria parasite affecting humans. Compounds such as mukonal, 7-hydroxymukonal, and 7-methoxymukonal have shown potential for the treatment of this disease. Density Functional Theory (DFT), molecular docking, and pharmacokinetic analyses are valuable computational approaches for predicting potential drug candidates against P. falciparum. In this study, three compounds mukonal, 7-hydroxymukonal, and 7-methoxymukonal were optimized and evaluated through molecular docking and pharmacokinetic analysis against the target protein PF3D7_1441900 (transcripti…

DNA N6-methyladenine (6mA) has emerged as a significant epigenetic modification across a broad range of eukaryotes, from unicellular protists to metazoa. However, its role in unicellular eukaryotic parasites with highly AT-rich genomes, such as malaria-causing Plasmodium falciparum , remains unclear. Using mass spectrometry, South-western blotting, and Single Molecule Real-Time sequencing (Pacific Biosciences) across four stages of P. falciparum intra-erythrocytic development (IED), we confirmed that 0.02-0.04% of genomic adenines are modified to 6mA, with over 60% of the sites being stably ma…

The Plasmodium falciparum circumsporozoite protein ( Pf CSP) is the major surface antigen on Pf sporozoites. WHO-recommended vaccines RTS,S/AS01 E and R21/Matrix-M target the Pf CSP major repeat region and C-terminal domain (ctCSP). Although multiple studies associated protection with antibody responses to ctCSP, only a few ctCSP-specific monoclonal antibodies (mAbs) have been characterized. Here, crystal structures of 11 Fab-ctCSP complexes reveal how mAbs against the conserved β-epitope region achieve diverse modes of strain-transcending recognition, in contrast to mAbs to the hypervariable …

BACKGROUND: Although both preventable and treatable, malaria remains a significant burden on health and economic stability, particularly in regions of high transmission such as Africa and parts of Asia. The parasite's genetic diversity enhances its ability to evade the host immune system and adapt, posing challenges to effective treatment strategies.
OBJECTIVE: The primary objective of this study was to assess the genetic diversity of Plasmodium falciparum by analyzing polymorphisms in the merozoite surface protein genes among clinical isolates collected from public health facilities in Arba …

INTRODUCTION: Malaria is a fatal disease affecting millions of people worldwide, primarily due to infection by Plasmodium falciparum. The emergence of multidrug-resistant parasite strains has necessitated the exploration of novel therapeutic targets. Plasmodium falciparum DNA gyrase A (pfDNA gyrase A), an essential topoisomerase II that is not present in humans, is a promising antimalarial drug target. This study explores the inhibitory potential of three Withania somnifera withanolide derivatives, D, E, and O, against pfDNA gyrase A.
METHODS: An integrated structure-based computational analy…

Because P. falciparum and P. vivax coexist in the Gedeo Zone, accurate malaria diagnosis is essential for effective case management. Low parasitemia and pfhrp2/3 gene deletions limit the performance of conventional HRP2-based RDTs. The new Bioline Malaria multi-Ag Pf/Pf/Pv RDT, detecting HRP2 and dual pLDH for both species, addresses this limitation. This study evaluates its field performance in detecting low-density infections during peak transmission season using expert microscopy and qPCR as references. A health facility-based cross-sectional study enrolled 579 febrile patients from health …
