Astrocytes Resist HIV-1 Fusion but Engulf Infected Macrophage Material.

HIV-1 disseminates to diverse tissues and establishes long-lived viral reservoirs. These reservoirs include the CNS, in which macrophage-lineage cells, and as suggested by many studies, astrocytes, may be infected. Here, we have investigated astrocyte infection by HIV-1. We confirm that astrocytes trap and internalize HIV-1 particles for subsequent release but find no evidence that these particles infect the cell. Astrocyte infection was not observed by cell-free or cell-to-cell routes using diverse approaches, including luciferase and GFP reporter viruses, fixed and live-cell fusion assays, m

Potential protective effects of autophagy activated in MPP+ treated astrocytes.

Astrocytes, which have various important functions, have previously been associated with Parkinsons disease (PD), particularly in 1-methyl-4-phenylpyridinium (MPP+) and 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) models of PD. MPP+ is the toxic metabolite of MPTP and is generated by the enzymatic activity of monoamine oxidase B, which is predominantly located in astrocytes. MPP+ acts as a mitochondrial complex I inhibitor. Autophagy is an evolutionarily conserved self-digestion pathway in eukaryotic cells, which occurs in response to various types of stress, including starvation and ox

Effects of Ranolazine on Astrocytes and Neurons in Primary Culture.

Ranolazine (Rn) is an antianginal agent used for the treatment of chronic angina pectoris when angina is not adequately controlled by other drugs. Rn also acts in the central nervous system and it has been proposed for the treatment of pain and epileptic disorders. Under the hypothesis that ranolazine could act as a neuroprotective drug, we studied its effects on astrocytes and neurons in primary culture. We incubated rat astrocytes and neurons in primary cultures for 24 hours with Rn (10-7, 10-6 and 10-5 M). Cell viability and proliferation were measured using trypan blue exclusion assay, MTT

Cytokine regulation by modulation of the NMDA receptor on astrocytes.

The N-methyl-d-aspartate receptor (NMDA-R) is crucial for synaptic transmission and plasticity. Over-activation, as well as complete blockade, of receptor function can lead to severe impairment. However, modest modulation of the receptor has been reported to be neuroprotective via endogenous regulation of the receptor and its subunit composition in response to pathophysiological conditions. As an important model for de- and remyelination in the central nervous system (CNS) we examined NMDA-R regulation in the mouse cuprizone model. We were able to show an upregulation of the NR2 subunit on hip

Myelin phagocytosis by astrocytes after myelin damage promotes lesion pathology.

Astrocytes are key players in the pathology of multiple sclerosis and can assume beneficial and detrimental roles during lesion development. The triggers and timing of the different astroglial responses in acute lesions remain unclear. Astrocytes in acute multiple sclerosis lesions have been shown previously to contain myelin debris, although its significance has not been examined. We hypothesized that myelin phagocytosis by astrocytes is an early event during lesion formation and leads to astroglial immune responses. We examined multiple sclerosis lesions and other central nervous system path

Epigenetic Features Induced by Ischemia-Hypoxia in Cultured Rat Astrocytes.

Epigenetic mechanisms are involved in the maintenance of long-term hypoxia-adapted cellular functions. Astrocytes as the dividing neuroglia cell in the nervous tissue can be activated under hypoxia condition. In the present study, the epigenetic characteristics such as DNA methylation and histone acetylation, as well as S-adenosylmethionine (SAM) role were explored in cultured astrocytes under ischemia-hypoxia (IH) condition. IH could induce the hypermethylation of global DNA and hypoacetylation of histone H3/H4 in an enzyme-dependent manner. c-Jun amino terminal kinase (JNK) signal pathway wa

Dual role of astrocytes in perinatal asphyxia injury and neuroprotection.

Perinatal asphyxia represents an important cause of severe neurological deficits including delayed mental and motor development, epilepsy, major cognitive deficits and blindness. However, at the moment, most of the therapeutic strategies were not well targeted toward the processes that induced the brain injury during perinatal asphyxia. Traditionally, experimental research focused on neurons, whereas astrocytes have been more related with the damage mechanisms of perinatal asphyxia. In this work, we propose to review possible protective as well as deleterious roles of astrocytes in the asphyct

Differentiation capacity of native pituitary folliculostellate cells and brain astrocytes.

Pituitary folliculostellate (FS) cells are characterized by producing S100B protein, as do brain astrocytes. FS cells have some functions in the pituitary gland, i.e. scavenger functions, sustentacular cell activity through cytokines, and intercellular communication through gap junctions. However, the biological significances of FS cells, especially their differentiation capacities in the anterior pituitary gland, are still under discussion. To understand FS cells with new approaches, we generated a transgenic rat expressing GFP under S100b gene promoter, which regulates tissue-specific expres

Effect of oxidative stress on UDP-glucuronosyltransferases in rat astrocytes.

The present work reports data regarding effects of an induced oxidative stress on the mainly expressed isoforms of UDP-glucuronosyltransferases (UGTs) in the brain. UGT1A6 and UGT1A7 expression and enzymatic activities toward the 1-naphthol were analyzed in rat cultured astrocytes following the exposure for 48 h to redox-cycling xenobiotic compounds such as quinones and bipyridinium ions. The expression of NADPH:cytochrome P450 reductase and NAD(P)H:quinone oxidoreductase 1 (NQO1) was also investigated. Oxidative stress induced significant deleterious changes in astrocyte morphology, decreased

Four mood stabilizers commonly induce FEZ1 expression in human astrocytes.

OBJECTIVES: Mood stabilizers influence the morphology, chemotaxis, and survival of neurons, which are considered to be related to the mood-stabilizing effects of these drugs. Although previous studies suggest glial abnormalities in patients with bipolar disorder and an effect of mood stabilizers on certain genes in astrocytes, less is known about the effects of mood stabilizers in astrocytes than in neurons. The present study identifies a common underlying response to mood stabilizers in astrocytes. METHODS: Human astrocyte-derived cells (U-87 MG) were treated with the four most commonly used

Induction of IP-10 (CXCL10) in astrocytes following Japanese encephalitis.

Chemokines and their receptors are important elements for the selective attraction and activation of various subsets of leukocytes. Interferon-gamma inducible protein (IP-10 or CXCL-10) is a potent chemoattractant and has been suggested to enhance the severity of virus infection and neuronal injury. In order to assess functional importance of this chemokine in viral encephalitis, we have exploited an experimental model of Japanese encephalitis. We report for the first time that in Japanese encephalitis, astrocytes are the predominant source of IP-10. A progressive increase in IP-10 induction f

TNF-alpha down-regulates CXCR4 expression in primary murine astrocytes.

CXC chemokine receptor 4 (CXCR4) is a co-receptor for human immunodeficiency virus (HIV) infection and is believed to be involved in the pathogenesis of AIDS-associated neurologic disorders and brain tumors. The physiological roles of CXCR4 in developmental patterning of the nervous and hematopoietic system; gastrointestinal angiogenesis; and cardiac organogenesis were established by studies in gene-targeted mice. Studies on CXCR4 expression and regulation in neuroepithelial cells are fundamental for understanding its physiopathologic roles in the central nervous system (CNS). We show here tha

[Astrocytes--anonymous cells in the brain, participating in neurasthenic symptoms].

During rehabilitation after brain damage due to stroke or trauma, or after meningitis, patients often suffer from mental fatigue and decreased capacity for concentration and learning. We hypothesize that one pathogenic cause of these symptoms could be a failure on the part of astroglia to remove glutamate and K+ from synaptic regions during glutamatergic transmission. As astroglial support of neuronal glutamate transmission has been shown to be of utmost importance, such an astroglial failing might lead to a decreased signal-to-noise ratio in glutamatergic neurotransmission, especially when th

Heterogeneity of expression of neuropeptide genes by astrocytes: functional implications.

A specific subset of neuropeptides are expressed in astrocytes, with specificity in terms of both brain region and gene, as well as with specific developmental patterns: the overall pattern of expression is thus heterogeneous. These neuropeptides have been demonstrated to have a number of trophic effects, including regulation of cell number, of length of axons and dendrites, and of neuronal phenotype. The developmental regulation of expression of neuropeptides in astrocytes further supports functional roles for the neuropeptides as trophic modeling factors. Not all the trophic effects have to

Sudanophilic lipid accumulation in astrocytes in periventricular leukomalacia in monkeys.

Sudanophilic lipid accumulation is a characteristic feature of periventircular leukomalacia (PVL) of infants. At least two types of lipid-containing cells have been identified, one being the macrophage, the other the "pre-myelin glial cell". A third type of lipid-containing cell has been seen in two monkeys with spontaneous PVL. Electron microscopically this cell appears to be an astrocyte. This probably represents a reaction of the astrocyte to hypoxia and may be the equivalent of the "hypertrophic" astrocytes found in human infants.

Preface to the Special Issue "Astrocytes in Cognition and Behavior".

Astrocytes, once viewed as passive support cells in the central nervous system, are now recognized as active and dynamic regulators of neural function. Across multiple domains of central nervous system function, astrocytes interact closely with neurons to influence synaptic transmission, plasticity, and behavior. These include key brain and spinal cord functions related to information processing that impact cognition and animal behavior. This Special Issue features 12 comprehensive reviews and original data articles that highlight key advances and ongoing challenges in this field. These works

Novel Characterization of Prox-1 Expression on Optic Nerve Astrocytes.

Prox-1 (prospero homeobox protein 1) is a master control gene for lymphatic vessel development and it has been widely used as a marker to detect lymphatic vessels inside and outside the eye. We herein report a novel finding of Prox-1 expression on the astrocytes of the optic nerve, a critical structure that conveys visual information from the retina to the brain. We identified that Prox-1 expressing cells were widely distributed inside the optic nerve. These cells co-expressed both GFAP (glial fibrillary acidic protein) and AQP4 (aquaporin-4) which are typical markers for astrocytes. Interesti

BAG3 modulates clathrin-mediated endocytosis and tau uptake in astrocytes.

Astrocytes maintain neuronal homeostasis by removing extracellular disease-relevant proteins, such as tau, to prevent their uptake by neurons. In Alzheimer's disease (AD), this astrocytic function is impaired, contributing to pathological tau accumulation. Many AD-associated risk genes are linked to endocytosis pathways, suggesting their role in AD pathogenesis. Although astrocytes can internalize, degrade, and release tau, the mechanisms governing these processes remain unclear. Bcl2-associated athanogene 3 (BAG3), a multifunctional protein regulating vacuolar processes, interacts with compon
