In the anterior pituitary of the rat, target cells of 1,25 (OH)2 vitamin D3 are identified as those that secrete thyroid stimulating hormone by means of a combined technique of thaw-mount autoradiography and immunohistochemistry. The results for the first time provide evidence that suggests a central effect of 1,25 (OH)2 vitamin D3 on the modulation of thyrotropin secretion in a manner similar to that of other steroid hormones at the level of the pituitary.

Cerebral blood flow and glucose utilisation were measured in rats using [14C]iodoantipyrine and [14C]2-deoxyglucose quantitative autoradiography respectively following repeated exposure to the serotonergic neurotoxin methylenedioxyamphetamine. In both control and treated animals blood flow and glucose use were similarly correlated but the ratio was increased following lesion. Some focal increases in flow were greatly in excess of metabolic demand. These results are consistent with a cerebrovascular vasoconstrictor role for serotonin.

The effects of vibration on the neurosecretory system of the brain were studied using the method of autoradiography. Both control and exposed to vibration (for 6 or 12 days) guinea pigs were injected with tritiated methionine and the uptake of the isotope was determined after 1, 6, 12, 24 h. Some differences were found in the number of silver grains in the brain neurosecretory system between the controls and both experimental groups. These differences, were however, not statistically significant.

The tissue culture and autoradiography techniques were used to study the effect of infiltration by inflammatory cells and of HP infection on the synthesis of glycoproteins by the gastric antrum. The results showed that in active phase of gastritis, the synthesis of glycoprotein was increased but no correlation between HP infection and synthesis of glycoprotein was noted. The mechanism of increased synthesis in chronic gastritis was discussed. It is suggested that the inflammatory cells and media released by them may play important role.

A peroxidase staining method for von Willebrand factor (vWF) multimer analysis was modified for comparison with an autoradiographic method. This method consists of SDS-agarose electrophoresis followed by blotting to a nitrocellulose membrane and a sensitive peroxidase staining method. The resolution of vWF multimers on the nitrocellulose membrane is comparable to that on the conventional autoradiography. Results can be obtained in 3 days. This nonradioisotopic method will be useful for the determination of the type of von Willebrand's disease in clinical laboratories.

Anatomical and functional aspects of the circadian timekeeping system containing the suprachiasmatic nucleus (SCN) were compared in normal and genetically narcoleptic dogs. The retinohypothalamic tract was delineated by tritiated amino acid autoradiography, the SCN was identified and examined by morphological techniques, and the circadian rhythm of melatonin concentrations in cerebrospinal fluid was measured by radioimmunoassay. Results suggest that the retinal input, cytoarchitecture, and essential timekeeping function of the SCN are intact in narcoleptic dogs.

Fresh rat neural lobe slices were incubated in the presence of [125I] alpha-scorpion toxin (ScTX), a specific marker of Na+ channels. Quantitative electron microscope autoradiography revealed preferential, irregularly spaced labeling of the axolemma of neurosecretory axons, with a significantly higher crude specific activity than any other neuronal or non-neuronal compartment. The number of specific binding sites at the neural lobe surface was calculated to be about 23 per microns2 of axolemma.
