A novel mutation in leukocyte adhesion deficiency type II/CDGIIc.

Leukocyte adhesion deficiencies (LAD) are autosomal recessive immunodeficiency syndromes characterized by severe and recurrent bacterial infections, impaired wound healing and leukocytosis. Block in different steps in the leukocyte adhesion cascade causes different types of leukocyte adhesion deficiencies, LAD type I, II and III. In LAD type II, the rolling phase of the leukocyte adhesion cascade is affected due to mutations in the specific fucose transporter GFTP (GDP fucose transporter), causing defect in the biosynthesis of selectin ligands on leukocytes. Thus this syndrome is also called c

SFB flagellin mediates cell adhesion, endocytosis and immune regulation in germ-free mice.

INTRODUCTION: Segmented filamentous bacteria (SFB) colonization dynamics are crucial for host immune regulation. Given this, the present study specifically examined the functions of SFB flagellin in bacterial adhesion, cellular internalization, and immune modulation. METHODS: Lactococcus lactis and Escherichia coli were engineered to express murine and rat SFB flagellin genes. Subsequent in vitro co-culture experiments with intestinal epithelial cell lines (MODE-K and IEC-18) and germ-free mouse colonization assays were conducted. Bacterial counts, immunohistochemical analysis, the AAM-ISO-G1…

Adhesive Functions or Pseudogenization of Type Va Autotransporters in Brucella Species.

Adhesion to host cells is a key step for successful infection of many bacterial pathogens and may define tropism to different host tissues. To do so, bacteria display adhesins on their surfaces. Brucella is an intracellular pathogen capable of proliferating in a wide variety of cell types. It has been described that BmaC, a large protein that belongs to the classical (type Va) autotransporter family, is required for efficient adhesion of Brucella suis strain 1330 to epithelial cells and fibronectin. Here we show that B. suis 1330 harbors two other type Va autotransporters (BmaA and BmaB), whic

Inhibition of Porphyromonas gingivalis adhesion to Streptococcus gordonii by human submandibular-sublingual saliva.

Porphyromonas gingivalis W50 adheres in vitro to biofilms of Streptococcus gordonii G9B. This phenomenon is believed to facilitate the initial colonization of the oral cavity by P. gingivalis and to contribute to the maturation of dental plaque. In this report, we describe the modulating effects of human submandibular-sublingual saliva (HSMSL) on this in vitro model of intergeneric bacterial adhesion (coaggregation). HSMSL inhibited P. gingivalis adhesion to S. gordonii by 50% at a concentration of 57 micrograms of protein per ml. Maximum inhibitory activity was associated with a 43-kDa protei…

Listeria monocytogenes virulence factors that stimulate endothelial cells.

Listeria monocytogenes infection of endothelial cells upregulates surface expression of adhesion molecules and stimulates neutrophil adhesion to infected cell monolayers. The experiments presented here tested the roles of specific bacterial virulence factors as triggers for this inflammatory phenotype and function. Human umbilical vein endothelial cell (HUVEC) monolayers were infected with wild-type L. monocytogenes or L. monocytogenes mutants; then surface expression of E-selectin and neutrophil adhesion were measured. The results showed that delta hly and prfA mutants were the most crippled,

Probing the adhesion potential of Bifidobacterium animalis subsp. lactis MCM-B-2007 via in vitro and in silico analyses.

Bifidobacterium strains are widely recognized for their probiotic properties, and their ability to adhere to the intestinal epithelium is crucial for effective gut colonization and interaction with the host. This study offers a thorough examination of the adhesion capabilities of Bifidobacterium animalis subsp. lactis, integrating in silico analyses with in vitro adhesion assessments. Genome analysis of MCM-B-2007 identified several adhesion proteins that might act as potential mediators of crosstalk between microbes and their host. Molecular docking studies further assessed these adhesion pro

Vitronectin in bacterial pathogenesis: a host protein used in complement escape and cellular invasion.

The multifunctional human glycoprotein vitronectin (Vn) plays a significant role in cell migration, tissue repair and regulation of membrane attack complex (MAC) formation. It also promotes neutrophil infiltration and, thus, enhances the inflammatory process during infection. In the host, a balanced homeostasis is maintained by Vn due to neutralization of the self-reactivity of the MAC. On the other hand, Vn bound to the bacterial surface protects from MAC-mediated lysis and enhances adhesion. Gram-negative bacterial pathogens including Moraxella catarrhalis, Haemophilus influenzae and Neisser

Implementation of phospholipids as pharmacological modalities for postoperative adhesions prevention.

Adhesions formation is considered a significant clinical entity implicating the healing process following major abdominal surgery, with serious clinical consequences and need for substantial health care expenditures. Several agents and substances applied either locally or systematically could potentially function as inhibitors of the formation of peritoneal adhesions endowed by limiting tissue apposition during the critical stages of mesothelial repair. Phospholipids are identified as surfactant-like substances, acting as a temporary membrane-like coverage of serosal defects. The experimental

Type III Methyltransferase M.NgoAX from Neisseria gonorrhoeae FA1090 Regulates Biofilm Formation and Interactions with Human Cells.

Neisseria gonorrhoeae is the etiological factor of the sexually transmitted gonorrhea disease that may lead, under specific conditions, to systemic infections. The gonococcal genome encodes many restriction modification (RM) systems, which main biological role is to defend the pathogen from potentially harmful foreign DNA. However, RM systems seem also to be involved in several other functions. In this study, we examined the effect of inactivation the N. gonorrhoeae FA1090 ngoAXmod gene encoding M.NgoAX methyltransferase on the global gene expression, biofilm formation, interactions with human
