RocA truncation underpins hyper-encapsulation, carriage longevity and transmissibility of serotype M18 group A streptococci.

Group A streptococcal isolates of serotype M18 are historically associated with epidemic waves of pharyngitis and the non-suppurative immune sequela rheumatic fever. The serotype is defined by a unique, highly encapsulated phenotype, yet the molecular basis for this unusual colony morphology is unknown. Here we identify a truncation in the regulatory protein RocA, unique to and conserved within our serotype M18 GAS collection, and demonstrate that it underlies the characteristic M18 capsule phenotype. Reciprocal allelic exchange mutagenesis of rocA between M18 GAS and M89 GAS demonstrated that

Prevalence of a septicemia disease in the crested ibis (Nipponia nippon) in China.

This study investigated six cases of septicemia in young crested ibises (Nipponia nippon). These birds all died with similar clinical signs, including sudden death, anorexia, diarrhea, and lameness. Immediately after death, the birds were necropsied; a blood sample was taken from heart and tissues were sampled from liver, lung, spleen, peritoneal mucus, and feces for bacteriologic examination. Anatomic observation showed that the main findings common to the sick birds were arthrocele, associated with congestion in the femur, tibiotarsus, and ventral side; swelling in the liver; hemorrhagic per

A platform for glycoengineering a polyvalent pneumococcal bioconjugate vaccine using E. coli as a host.

Chemical synthesis of conjugate vaccines, consisting of a polysaccharide linked to a protein, can be technically challenging, and in vivo bacterial conjugations (bioconjugations) have emerged as manufacturing alternatives. Bioconjugation relies upon an oligosaccharyltransferase to attach polysaccharides to proteins, but currently employed enzymes are not suitable for the generation of conjugate vaccines when the polysaccharides contain glucose at the reducing end, which is the case for ~75% of Streptococcus pneumoniae capsules. Here, we use an O-linking oligosaccharyltransferase to generate a

Monotherapy or polychemotherapy in the treatment of Campylobacter pylori-related gastroduodenal disease.

Three pilot studies were performed to evaluate the efficacy of bismuth subsalicylate (BSS) and nitrofurantoin to eradicate Campylobacter pylori colonization in man. Nitrofurantoin 3 x 100 mg capsules for 10 days did not clear C. pylori in any of 13 patients, and neither did the combination of BSS and nitrofurantoin suspension (0/6 patients). Immediately after high dose BSS therapy 3 x 900 mg for 28 days, 8/17 patients (47%) had negative cultures and rapid urease tests. There was recrudescence in 5 out of 6 patients so far submitted to follow-up investigations, giving a best possible outcome es

Histological progression and bacterial load dynamics of Renibacterium salmoninarum in Chinook salmon Oncorhynchus tshawytscha.

Renibacterium salmoninarum, the cause of bacterial kidney disease (BKD), severely impacts salmonid populations. Much of our understanding of the BKD pathology in salmonids comes from evaluating fatal infections in wild populations or spawning Pacific Northwest salmonids. Our study investigated the histological progression and bacterial load dynamics of R. salmoninarum infection in Chinook salmon following intraperitoneal injection to enhance understanding of the disease's dynamics. Seventy presmolt salmon were injected with the ATCC-33209 isolate of R. salmoninarum and monitored over 10 wk. Hi

A covR promoter mutation enhances Group B Streptococcus virulence by upregulating pgaC to reinforce capsular surface anchoring.

Group B Streptococcus (GBS) is a major zoonotic pathogen responsible for neonatal sepsis and cerebrospinal meningitis. A single adenine (T) insertion in the covR promoter, identified in a clinical isolate, has been shown to enhance pathogenicity. However, the mechanistic basis remains unclear. In this study, we demonstrated that this insertion downregulates covR expression and enhances hemolysin production. Unexpectedly, we also found that it leads to a thicker and more compact capsule based on dot blot and flow cytometry analyses. Transcriptome analysis performed on the covR promoter mutant r

Decoding fibrotic encapsulation in plastic surgery implants: From biological mechanisms to precision interventions frontiers.

Mitigating fibrotic encapsulation around implants is critical to ensuring the long-term success of implant-based procedures in plastic surgery. Fibrous capsule formation is a pathological foreign body reaction (FBR) that not only causes capsular contracture and implant failure but also creates a permissive microenvironment for bacterial colonization, thereby increasing the risk of infection. These adverse outcomes may ultimately necessitate implant removal or replacement, significantly compromising surgical results and patient satisfaction. Therefore, a thorough understanding of the mechanisms

[Molecular aspects of anthrax pathogenesis].

A model of anthrax infection with the role determined for main pathogenicity factors of Bacillus anthracis exotoxin and capsule is presented. After spore phagocytosis by macrophages, synthesis of the main exotoxin component begins - a protective antigen that in oligomeric form disrupts phagosome membrane. This accelerates the transition of the pathogen from phagosome into the macrophage cytoplasm. Poly-D-glutamine capsule synthesized by the pathogen triggers the exit (exocytosis) of vegetative cells from macrophages and protects them from re-phagocytosis in lymphatic node lumen. The vegetative

Plague in Guinea pigs and its prevention by subunit vaccines.

Human pneumonic plague is a devastating and transmissible disease for which a Food and Drug Administration-approved vaccine is not available. Suitable animal models may be adopted as a surrogate for human plague to fulfill regulatory requirements for vaccine efficacy testing. To develop an alternative to pneumonic plague in nonhuman primates, we explored guinea pigs as a model system. On intranasal instillation of a fully virulent strain, Yersinia pestis CO92, guinea pigs developed lethal lung infections with hemorrhagic necrosis, massive bacterial replication in the respiratory system, and bl

A link between STK signalling and capsular polysaccharide synthesis in Streptococcus suis.

Synthesis of capsular polysaccharide (CPS), an important virulence factor of pathogenic bacteria, is modulated by the CpsBCD phosphoregulatory system in Streptococcus. Serine/threonine kinases (STKs, e.g. Stk1) can also regulate CPS synthesis, but the underlying mechanisms are unclear. Here, we identify a protein (CcpS) that is phosphorylated by Stk1 and modulates the activity of phosphatase CpsB in Streptococcus suis, thus linking Stk1 to CPS synthesis. The crystal structure of CcpS shows an intrinsically disordered region at its N-terminus, including two threonine residues that are phosphory

Modeling the conformations of Neisseria meningitidis serogroup a CPS and a carba-analogue: Implications for vaccine development.

Neisseria meningitidis is a major cause of bacterial meningitis worldwide, especially in Africa. The capsular polysaccharide is the main virulence factor and the target antigen for polysaccharide- and conjugate vaccines. Three tetravalent conjugate vaccines against serogroups A, C, Y and W have been licensed and the monovalent MenAfriVac® was introduced to address the high burden of serogroup A disease in the Meningitis Belt of sub-Saharan Africa. Three of these four vaccines are lyophilized due to the instability of the serogroup A antigen (MenA) in aqueous solution, resulting in a two vial p

A Salmonella enterica serovar Typhi vaccine expressing Yersinia pestis F1 antigen on its surface provides protection against plague in mice.

A recombinant strain of attenuated Salmonella enterica serovar Typhi surface-expressing Yersinia pestis F1 antigen was generated by transforming strain BRD1116 (aroA aroC htrA) with plasmid pAH34L encoding the Y. pestis caf operon. BRD1116/pAH34L was stable in vitro and in vivo. An immunisation regimen of two intranasal doses of 1 x 10(8) cfu of BRD1116/pAH34L given intranasally to mice 7 days apart induced the strongest immune response compared to other regimens and protected 13 out of 20 mice from lethal challenge with Y. pestis. Intranasal immunisation of mice constitutes a model for oral i

A bacteriophage genus infects carbapenem resistant Acinetobacter baumannii via a non-capsular receptor and provides protection in vivo.

Extensive drug resistance (XDR) in Acinetobacter baumannii and other pathogens has revitalized bacteriophage as a therapeutic consideration. Six phages (AB1I1L, AB1I1M, AB1I1P, AB1I1T, AB2I2, and AB2I3) targeting A. baumannii were isolated from wastewater. These represent a previously undescribed phage genus with rapid adsorption and potent lysis. 18/40 A. baumannii clinical isolates, including 11/27 carbapenem-resistant isolates, were susceptible to one of the isolated phages. Importantly, in vitro-derived, phage resistant bacteria were killed in human ascites, demonstrating decreased biofitn

Unlocking data in Klebsiella lysogens to predict capsular type-specificity of phage depolymerases.

Viral entry is a critical step in the infection process. Klebsiella spp. and other clinically relevant bacteria often express complex polysaccharide capsules that act as a barrier to phage entry. In turn, most lytic phages targeting Klebsiella encode depolymerases for capsule removal. This virus-host arms race leads to extensive genetic diversity in both capsules and depolymerases, complicating our ability to understand their interaction. This study exploits the genetic information encoded in Klebsiella prophages to model the interplay between the bacteria, the prophages, and their depolymeras

Impact of Hyperglycemia on Streptococcus pneumoniae Virulence: Insights into Capsular Dynamics and Clinical Implications.

Hyperglycemia, a diabetic condition, profoundly affects the pathogenicity of Streptococcus pneumoniae ( S. pneumoniae ), a leading etiologic pathogen of pneumonia. This review addresses a systematic understanding of how hyperglycemia affects the pathogenicity of S. pneumoniae , especially by immune dysfunction, cytokine dysregulation, and oxidative stress. Although the length of the capsular polysaccharide (CPS) chain plays a role in immune evasion and survival of bacteria, hyperglycemia also affects host immune responses, leading to increased severity of infection in diabetic patients. CPS le

Phenotypic switching of Cryptococcus neoformans and Cryptococcus gattii.

Microorganisms that live in fluctuating environments must constantly adapt their behavior to survive. The host constitutes an important microenvironment in opportunistic and primary fungal pathogens like Cryptococcus neoformans (C. neoformans) and Cryptococcus gattii (C. gattii). In clonal populations, adaptation may be achieved through the generation of diversity. For fungi phenotype switching constitutes a mechanism that allows them to change rapidly. Both C. neoformans and C. gattii undergo phenotypic switching, which allows them to be successful pathogens and cause persistent disease. Simi
