Monitoring virus strain variation following infection with VZV: is there a need and what are the implications of introducing the Oka vaccine?

Varicella zoster virus (VZV) is a stable virus showing relatively little variation. Nevertheless, recent data have shown there to be at least four distinct viral strains. For the most part these are geographically segregated, but in areas of the world such as the UK, where mixed populations live, there is evidence for spread of all the genotypes. Little is known about the biological differences, if any, between these strains, yet recent data have shown that even a single nucleic acid change can affect the biological behaviour of the virus. The Oka vaccine has been licensed for mass vaccination

Continuous OFT-Based Behavioral Phenotyping Reveals Neural Stem Cell-Associated Shifts in Depression-like Mice.

Background: Depression-like phenotypes in animal models often show graded behavioral variation rather than uniform group-wise changes, yet treatment effects are still commonly evaluated using categorical comparisons. In particular, it remains difficult to determine whether treated animals retain model-like behavioral features or shift toward a more control-like state. Here, we developed a continuous behavioral phenotyping framework based on multidimensional open-field-test (OFT) features to quantify model-like behavioral states and treatment-associated behavioral shifts. Methods: In the presen

Single-cell computational machine learning approaches to immune-mediated inflammatory disease: New tools uncover novel fibroblast and macrophage interactions driving pathogenesis.

Recent advances in single-cell sequencing technologies call for greater computational scalability and sensitivity to analytically decompose diseased tissues and expose meaningful biological relevance in individual cells with high resolution. And while fibroblasts, one of the most abundant cell types in tissues, were long thought to display relative homogeneity, recent analytical and technical advances in single-cell sequencing have exposed wide variation and sub-phenotypes of fibroblasts of potential and apparent clinical significance to inflammatory diseases. Alongside anticipated improvement

Scientific and Clinical Studies on Variation in Individual Response to Ionising Radiation: A Scoping Review.

Ionising radiation, defined as radiation with sufficient energy to remove electrons from atoms and molecules, can produce significant biological effects in living tissue. Individual response to ionising radiation varies considerably and underpins both deterministic tissue reactions and stochastic effects such as cancer. The concepts of radiosensitivity and radiosusceptibility are central to this variability; however, their definitions and applications remain inconsistent across the literature. This scoping review aims to systematically map and analyse how radiosensitivity and radiosusceptibili

Biochemical monitoring of treatment for galactosaemia: biological variability in metabolite concentrations.

Red cell galactose 1-phosphate (Gal-1-P) concentrations and urinary galactitol excretion have been suggested as biochemical indices of dietary compliance in classical transferase-deficient galactosaemia. We report our experience of measuring both in 32 patients over 0-10.9 years (median 3.45). A total of 438 blood specimens for Gal-1-P and 383 urine specimens for galacitol assay were received; 317 pairs of specimens were collected at the same time. Concentrations of both analytes fell rapidly over the first 2-3 months following dietary intervention, to mean (geometric SD) levels of 225 (1.60)

A practical guide to biologic treatments for psoriasis in resource-limited areas.

Biologic therapies have transformed the management of moderate to severe psoriasis, providing targeted and effective interventions against key inflammatory pathways. Due to their action specifications compared to conventional therapies, they generally provide better efficacy-safety profiles, especially the second-generation of biologics for psoriasis. Furthermore, the broader aspects of patients' quality of life can improve dramatically. This review overviews the efficacy, safety, and real-world application of biologic agents, including tumor necrosis factor-alpha inhibitors, interleukin (IL)-

Human anthrax vaccines: Current status and future prospects.

Bacillus anthracis is a harmful pathogen that can pose a severe hazard to the public when deployed as a biological weapon. Human anthrax vaccines, as an effective pre-exposure prevention, can significantly reduce morbidity and mitigate public panic. However, there are still concerns toward current vaccines, such as suboptimal immune efficacy, complex vaccination procedures, adverse reactions and variations in individual immune responses. Researchers have devoted great efforts to addressing these issues and deficiencies, which have yielded many novel findings. This review mainly covers the biol

Effect of Glycerol Administration on ECochG and VEMP Findings in Individual with Meniere's Disease.

The activity of several components of the vascular system appears to be diurnally regulated. Endothelial cell activation, leukocyte and platelet interactions and lipoprotein metabolism have all been shown to vary with time of day, but whether these variations are due to the endogenous circadian clock, exogenous factors, such as the light-dark cycle, or an interaction between the two remains to be determined. Endothelium-dependent vasodilation also varies diurnally. This rhythmicity is lost in individuals with established coronary disease has been shown to occur in the early stages of atheroscl

Biological Variation of Natriuretic Peptides and Other Cardiovascular Disease-Related Biomarkers: A Systematic Review and Meta-analysis.

BACKGROUND: The clinical application of cardiovascular disease (CVD) biomarkers requires robust biological variation (BV) data to enable both interpretation of serial results and definition of appropriate analytical quality. A critical review and meta-analysis of eligible BV studies of natriuretic peptides and other CVD-related biomarkers was therefore undertaken, with a focus on differences observed in different states of health and the impact of sampling intervals on BV. METHODS: BV studies of N-terminal pro-B-type natriuretic peptide (NT-proBNP), brain natriuretic peptide, N-terminal pro-A

Genetic susceptibility to hypertensive renal disease.

Hypertensive renal disease occurs at increased frequency among the relatives of patients with this disease compared to individuals who lack a family history of disease. This suggests a heritable risk in which genetic variation may play a role. These observations have motivated a search for genetic variation contributing to this risk in both experimental animal models and in human populations. Studies of animal models indicate the capacity of natural genetic variants to contribute to disease risk and have produced a few insights into the disease mechanism. In its current phase, human population

Some statistical issues in HIV vaccine trials.

Efficacy trials of prophylactic HIV vaccines will be among the most difficult clinical trials ever attempted. Not only will there be challenges with the recruitment and retention of high-risk uninfected individuals, there will be many statistical challenges to the design, conduct, analysis, and interpretation of these trials. General features of an efficacy trial are described, including choice for the primary endpoint and testing for and estimating vaccine efficacy. Secondary objectives of trials are also discussed. These include determining the correlates of protective immunity, assessing th

Epigenetic mechnisms involved in posttraumatic stress disorder.

Posttraumatic stress disorder (PTSD) is a complex mental health condition influenced by various factors, including the environment, biology, and genetic variations. This chapter delves into how these factors interact, focusing on trauma exposure, brain mechanisms, and genetic expression changes that contribute to the development of PTSD. The chapter provides an overview of PTSD prevalence, diagnostic criteria, key patterns in its occurrence, interindividual symptoms variations, and factors that increase the risk of developing PTSD, such as inherited trauma effects passed down from one generati

Precision of a phage susceptibility spot assay assessed with 154 clinical Staphylococcus aureus isolates.

Lytic phages are a promising consideration to combat recalcitrant bacterial infections. To advance the possibility of using phage therapy in the clinical routine, standardized phage susceptibility testing methods are needed. Here, staphylococcal phage K was tested against 154 periprosthetic joint infection-associated Staphylococcus aureus isolates using a double overlay spot assay in technical and biological triplicates. Individual plaques were assessed using a morphology scoring system ranging from 0 (no plaques) to 4 (large, clear plaques), with scores ≥2 considered indicative of infectivity

Effects of CYP2C19 and CYP2C9 genotypes on pharmacokinetic variability of valproic acid in Chinese epileptic patients: nonlinear mixed-effect modeling.

PURPOSE: To evaluate the effects of CYP2C19 and CYP2C9 genotypes on the pharmacokinetic variability of valproic acid (VPA) in epileptic patients using a population pharmacokinetic (PPK) approach. METHODS: VPA concentrations were measured in 287 epileptic patients, who were genotyped for CYP2C19*2/*3 and CYP2C9*3. Patients who were on monotherapy with VPA were divided into two groups, a PPK-model group (n = 177) and a PPK-valid group (n = 110). The PPK parameter values for VPA were calculated in the PPK-model group by using the NONMEM software. Ultimately, a biological model and a final model

Individual-dependent molecular heterogeneity in human endometrial stromal cells: a potential determinant of female fertility.

BACKGROUND: Human endometrial stromal cells (hEnSCs) are central to female fertility. This study investigated the cellular and molecular landscape of hEnSCs and identified inter-individual variations that may influence reproductive potential. METHODS AND RESULTS: Endometrial biopsies were obtained from 16 women undergoing gynecologic evaluation. Primary hEnSCs were isolated, expanded, and characterized for mesenchymal stem cell properties. Stemness, receptivity, and decidualization capacity were assessed using RT-qPCR and RNA sequencing, followed by differential gene expression and pathway enr

Indirect Effects of the Coping Power Intervention on Latent Suicidal Thoughts and Behaviors: an Integrative Data Analysis.

There has been growing interest in preventive intervention "crossover" effects on suicidal thoughts and behaviors (STBs), in which targeting early risk factors may mitigate distal risk for STBs without STBs having been the targeted outcomes of the primary study. The present study extends an 11-study integrative data analysis of the Coping Power (CP) intervention (N = 3182) to assess indirect effects of different forms of CP on teacher- and parent-reported STBs as transmitted through different subdimensions of internalizing and externalizing problems. Compared to school-as-usual, all forms of C

Identifying Genomic Variations in Monozygotic Twins Discordant for Autism Spectrum Disorder Using Whole-Genome Sequencing.

Autism spectrum disorder (ASD) presents a set of childhood neurodevelopmental disorders with impairments in social communication and restricted, repetitive, and stereotyped patterns of behavior. Here, based on the whole-genome sequencing (WGS) data of three monozygotic twins discordant for ASD, we explored multiple patient-specific genetic variations and prioritized a list of ASD risk genes. Our results identified DVMT (discordant variation in monozygotic twin) observed in at least two twin pairs, including 14,310 SNPs, 2,425 indels, and 16,735 CNVs, referring to a total of 2,174 genes, and 37

Is there a biological component in gender identity?

INTRODUCTION: Gender identity is each person's internal sense of being a woman, a man, both, neither, or anywhere along the gender spectrum, which may (cisgender) or may not (transgender) coincide with the sex assigned at birth. The multiple difficulties experienced by transgender individuals constitutes a risk factor for mood disorders and self-harming behaviors. However, knowledge about biological influences on gender identity development has the potential to reduce the stigmatization of gender minorities. MATERIALS AND METHODS: We conducted a literature review of the available literature o

Complex interactions between dietary and genetic factors impact lycopene metabolism and distribution.

Intake of lycopene, a red, tetraterpene carotenoid found in tomatoes is epidemiologically associated with a decreased risk of chronic disease processes, and lycopene has demonstrated bioactivity in numerous in vitro and animal models. However, our understanding of absorption, tissue distribution, and biological impact in humans remains very limited. Lycopene absorption is strongly impacted by dietary composition, especially the amount of fat. Concentrations of circulating lycopene in lipoproteins may be further influenced by a number of variations in genes related to lipid absorption and metab
