Several authors have suggested that african and north american forms of Blastomyces dermatitidis appeared separate taxa. Morphologic and pathogenic differences not only prevail, but also the failing to mate between them. African forms lose their mycelium-yeast conversion capability in old cultures. So I propose to create the complementary appellation Blastomyces dermatitidis tunisiens nov.subsp. from the african forms, in memory of the first case of systemic blastomycosis in this continent, discovered in Tunisia.

Transplacental infection with Blastomyces dermatitidis is rare in humans and unknown in the dog. A Doberman pinscher bitch was diagnosed with blastomycosis 25 days after whelping. Clinical signs were noted after whelping and were progressive. All 9 pups were free of clinical signs and had negative urine Blastomyces antigen tests at 6 weeks of age and remained free of signs of illness through 11 months of age. The bitch responded to treatment with itraconazole.

The inbred mouse strains C3H/HeJ and DBA/1J have been shown to represent the extremes of susceptibility and resistance, respectively, to pulmonary blastomycosis. This pattern was completely reversed when challenge was performed by the intraperitoneal route, whether a virulent or an attenuated strain of Blastomyces dermatitidis was utilized. By a third route (subcutaneous), the differences were insignificant. Inhibition of replication of blastomyces in vitro by macrophages from both strains, before or after activation by subcutaneous infection, was similar.

We applied a modified immunofluorescence and immunoperoxidase method, utilizing labeled Blastomyces dermatitidis antigens, to look for specific antibody-bearing B/plasma cells in the tissue infiltrates of blastomycosis lesions induced in hamsters. No specific anti-blastomyces antibodies were detectable by this method, although such antibodies were present in blood samples as demonstrated by routine immunodiffusion techniques. These studies suggest that humoral immune reactions do not play a major role in the pathogenesis of lesions of blastomycosis in hamsters.

C57BL/6J mice rendered hypersensitive to Blastomyces dermatitidis were protected from the lethal effects of a blastomyces infection. This protection was observed following a lethal intraperitoneal challenge with viable cells 15 days after subcutaneous inoculation with 3.9 x 10(4) viable cells. Delayed hypersensitivity was induced in C57 mice by two injections of Merthiolate-killed cells in adjuvant or by a single injection of viable cells. Development of hypersensitivity was determined at appropriate intervals by footpad injection with killed yeast cells.

BACKGROUND: Blastomycosis is a rare, potentially fatal fungal infection caused by inhalation of Blastomyces spores, typically acquired outdoors in the midwestern and eastern United States. In 2023, the largest recorded U.S. blastomycosis outbreak occurred among workers at a paper mill in Michigan's Upper Peninsula. Few data exist on occupational risk factors or indoor exposure to Blastomyces, limiting prevention efforts.

A case of localized oral mandibular blastomycosis is described in a five-year-old dog. Complete resolution of clinical signs and oral radiographic changes were seen following itraconazole therapy at 5 mg/kg/day for four and a half months. The patient remained free of Blastomyces at the one year follow up based on the Mira Vista Blastomyces urine antigen test by EIA (Enzyme Immunoassay)a. A literature review of localized blastomycosis cases in humans and dogs was performed, available diagnostic tests evaluated, and treatment comparisons made.
