The clinical significance of apoptotic cells in peripheral blood smears.

Apoptosis as introduced by Kerr and colleagues describes a distinct set of morphological features that represent programmed cell death. Apoptosis frequently arises owing to genetic programming, cytotoxic drugs and physical stimuli. In order to ascertain whether all of the natural triggers of apoptosis have been identified, we looked for the presence of apoptotic cells in all routine blood films throughout a 5-month period. Patients with known malignant disease or on chemotherapy were excluded from the study. Forty-nine blood films, from 10,000 examined in this period, showed apoptosis. The num

Functional competence of dendritic cells in human umbilical cord blood.

Dendritic cells (DC) are the main stimulators of primary T cell responses. Very little is known about DC in cord blood (CB), and whether they are involved in the low incidence and severity of GVHD following CB transplantation. Here, CBDC were identified as a HLA-DR+/lineage marker (lin; CD3, CD11b, CD14, CD16, CD19, CD34, CD56 and glycophorin A antigens) negative population, representing 0.3 +/- 0.1% (mean +/- s.d.; n = 15) of CB mononuclear cells. CBDC expressed the CD4, CD11a, CD18, CD45RA, CD50 and CD54 antigens but revealed no expression of the CD1a, CD11c, CD40, CD45R0, CD58, CD83, CD86 a

Allogeneic peripheral blood progenitor cells: current status and future directions.

Increasing use is being made of mobilized peripheral blood as a source of hematopoietic stem and progenitor cells for transplantation. Initially, these efforts were focused on autologous applications. However, many centers are now using cells derived from normal donors mobilized with recombinant hematopoietic growth factors for allogeneic transplantation. This article reviews the key issues associated with the use of these cells for hematologic reconstitution and summarizes important recent developments in the field.

Miniature field-flow fractionation system for analysis of blood cells.

Field-flow fractionation is an analytical tool that has been historically used to separate species, ranging from molecules to particles or cells several micrometers in size. This technology can effect separation by size, density, charge, or other physical properties, depending on the configuration of the field-flow system. We have developed a miniature field-flow system to analyze cell populations in a small, 125-microns-deep channel 19 cm long. Gravity is used as the primary field to effect separation, and cell analysis is performed in < 20 min. Erythrocytes elute as a single peak when dilute

Oxidative damage of red blood cells in haemolytic uraemic syndrome.

Changes in red blood cell (RBC) lipid peroxidation [measured by malonyl dialdehyde (MDA) concentration], glutathione (GSH) metabolism, antioxidant enzyme activities (catalase, superoxide dismutase, glutathione peroxidase) and haemoglobin (Hb) metabolites (metHb, carboxy Hb) were studied in six children with post-enteropathic (D+) haemolytic uraemic syndrome (HUS) and ten controls. The in vitro effect of hydrogen peroxide [acetylphenylhydrazine (APH) test] on GSH and Hb metabolism was also investigated. MDA levels were significantly higher and the antioxidant enzyme activities were lower in HUS

Hormonal modulation of ionic permeability in human red blood cells.

It has previously been reported that both exogenous adenosine cAMP analogs and forskolin-induced elevations in intracellular cAMP concentrations selective increase relative ionic chloride permeability in normal human red blood cells (RBC). A similar selectively increase in relative ionic chloride permeability was observed in untreated uremic subjects in whom endogenous RBC cAMP concentrations are chronically elevated. To detect which hormones might modulate RBC cAMP and ionic permeabilities, RBC were exposed to norepinephrine, epinephrine, and parathyroid hormone. Thereafter, RBC cAMP concentr…

Cation transport and volume regulation in sickle red blood cells.

Cellular dehydration is one of several pathological features of the sickle cell. Cation depletion is quite severe in certain populations of sickle cells and contributes to the rheological dysfunction that is the root cause of vascular occlusion in this disease. The mechanism of dehydration of sickle cells in vivo has not been ascertained, but three transport pathways may play important roles in this process. These include the deoxygenation-induced pathway that permits passive K+ loss and entry of Na+ and Ca2+; the K(+)-Cl- cotransport pathway, activated by acidification or cell swelling; and t

Functional alterations of swine peripheral blood mononuclear cells by methadone.

In vitro exposure of the synthetic opiate drug methadone allowed evaluation of putative immunomodulatory activities of swine peripheral blood mononuclear cells. Respiratory burst, an index of microbicidal activity, was suppressed by methadone in a dose-dependent manner following exposure for 48 h. The suppression was blocked by the opiate antagonist naloxone. Another macrophage function phagosome-lysosome fusion was impaired by exposure to methadone. A primary lymphocyte-mediated function natural killer cell activity was also affected. In contrast, the macrophage function antibody-mediated pha

Preimplantation genetics and recovery of fetal cells from maternal blood.

Advances in molecular genetics have made possible advances in two new areas of prenatal diagnosis. Preimplantation genetic diagnosis is being explored through polar body biopsy, biopsy of the single cell from the eight-cell embryo, and trophectoderm biopsy of the blastocyst. There are attractive theoretical advantages to the last approach. However, assessment of pregnancies to be continued has been attempted only in the former two, and has succeeded only in the eight-cell embryo. A major problem with polar body biopsy is recombination for loci near the centromere, which may make analysis of th

Antibodies for trypsinized human red blood cells in human sera.

During studies of agglutination of trypsinized human red blood cells (RBC) by anti-Rh sera, it was noticed that some human sera without Rh antibodies agglutinated these erythrocytes. Of 933 normal and pathological sera subsequently screened, 116 produced such agglutination. The agglutinins were of IgM nature and were directed against an antigen exposed or created by tryptic digestion of RBC. They reacted equally well with autologous as with allogeneic trypsinized RBC and were different from T agglutinins and from Paul-Bunnell antibodies. The mode of formation of these antibodies remains unknow…

[Introductory remarks for symposium on "biological membranes of blood cells"].

Recent progress on studies of structure and functions of biological membranes has been successfully achieved. Various topics on membrane abnormalities in blood cells were selected for this symposium. First of all, in red cell disorders, abnormal red cell morphology is crucial for identification of various kinds of hemolytic anemias. In addition, cytoskeletal protein abnormalities in red cell membrane were reviewed, especially on the pathogenesis of hereditary elliptocytosis and hereditary spherocytosis. Abnormal red cell membrane fluidity was also discussed in relation to insulin receptor on r…

Effect of diamide on polyphosphoinositide metabolism in red blood cells.

In intact red cells diamide had no strong influence on monoester phosphate group turnover of phosphatidylinositol 4,5-bisphosphate (PIP2), phosphatidylinositol 4-phosphate (PIP) and phosphatidic acid (PA) due to the potent GSH protection of these pathways in vivo. Following the complete GSH oxidation diamide impaired the turnover of PIP and PA dramatically. However that of PIP2 did not show any change. Indeed, a drastic irreversible decrease of PIP- and PA turnover was caused by a mild oxidative stress with diamide in G6PD deficient red cells.

Membrane-to-membrane transfer of tocopherol in red blood cells.

The direct cell-to-cell transfer of tocopherol between red blood cells (RBCs) was examined. Two kinds of RBCs were provided; 1) Tocopherol-deficient ones showing complete dialuric acid-hemolysis and 2) tocopherol-supplemented ones showing no dialuric acid-hemolysis. The two kinds of RBCs were mixed and incubated with gentle swirling in a buffer solution. If no tocopherol transfer occurred between the two kinds of RBCs in the cell suspensions, hemolysis should not change during incubation because hemolysis is limited only in the -deficient cells in the suspensions. However, it was actually obse…

Pre-B cells in peripheral blood of multiple myeloma patients.

Although multiple myeloma is a disease of plasma cells, abnormalities have been detected in both B and T lymphocytes in peripheral blood. Although multiple myeloma patients are deficient in surface Ig (sIg)-positive B lymphocytes, analysis of lymphocytes present in blood indicates an abnormally large pool of circulating pre-B cells. These pre-B cells express BA-1, do not bear sIg, and contain cytoplasmic mu chains. High numbers of pre-B cells occur in 88% of individuals with frank myeloma and in 44% of individuals with monoclonal gammopathy of undetermined significance. Pre-B cells bearing BA-

Detection of hepatitis B virus DNA in mononuclear blood cells.

The Southern transfer hybridisation technique was used to test mononuclear blood cells for hepatitis B virus DNA. Viral DNA sequences were detected in mononuclear cells of 10 out of 16 patients with hepatitis B virus infection and in none of 21 normal controls. Blood contamination was excluded by the absence of hepatitis B virus DNA in the corresponding serum samples in all cases. Free monomeric hepatitis B virus DNA was found in three patients positive for hepatitis Be antigen (HBeAg) and one positive for anti-HBe, and integrated hepatitis B virus DNA was present in four patients positive for…

Antibody elution from red blood cells by chloroform and trichloroethylene.

Antibody elution by chloroform and trichloroethylene was compared with the Rubin ether method modified by Hughes-Jones for IgG antibodies and the Landsteiner and Miller heat method for IgM antibodies. After washing the cells, the chloroform and trichloroethylene method was done at 37 degrees C for 10 minutes. With this technique the eluate remained on the upper layer above the red blood cell stroma layer, while with Rubin's method the eluate remained below the cell stroma. The reported technique was as effective as the Landsteiner and Miller heat method for IgM antibodies, and a double volume

Reversible plasma and red blood cells volumes increases in acromegaly.

Plasma volume (PV) and red blood cells volume (RBCV) were measured in 76 normotensive acromegalic patients (42 males and 34 females) and compared with those of 28 controls on the basis of body surface. In untreated males PV and RBCV were significantly higher than in normals (P less than 0.001) and total blood volume (TBV) was above the normal range in 85%. Untreated females also showed an elevated PV but had significantly lower RBCV than the male patients. The only vascular volume of the patients with correlated significantly with the body weight or surface was RBCV in females. One to five stu…

Increase in peripheral blood 'null' cells in extrinsic bronchial asthma.

Lymphocyte subpopulations were measured in the peripheral blood of fifteen patients with extrinsic bronchial asthma and twenty-seven normal control subjects. No significant differences in the absolute numbers of relative proportions of T, B or K lymphocytes were found. Antibody-dependent lymphocyte-mediated cytotoxicity (K-cell activity) was also found to be normal. The proportion and absolute number of 'null' cells were significantly increased in the asthmatic group: this population did not correlate with serum IgE levels, the eosinophil count or the duration of disease. The significance of t
