Hepatic Branch Vagotomy Modulates the Gut-Liver-Brain Axis in Murine Cirrhosis.

BACKGROUND: Cirrhosis and hepatic encephalopathy (HE) are linked with an altered gut-liver-brain axis, however, the relative contribution of hepatic vagal innervation is unclear. We aimed to determine the impact of hepatic vagotomy on the gut microbiome, brain, and liver in murine cirrhosis. METHODS: 10-15-week-old male C57BL/6 mice with and without hepatic vagotomy underwent carbon tetrachloride (CCl4) gavage for 8 weeks. Frontal cortex [inflammation, glial/microglial activation, BDNF (brain-derived neurotrophic factor)], liver [histology including inflammation and steatosis, fatty acid synt

Tryptophan-kynurenine metabolic reprogramming along the gut-brain axis alleviates Alzheimer's pathology.

The gut–brain axis influences neuroinflammation and metabolic homeostasis in Alzheimer’s disease (AD). Disruption of gut microbiota and barrier function promotes amyloid and tau pathology via immune and metabolic dysregulation. In this study, Limosilactobacillus fermentum SRK414 (SRK414) was orally administered to ADLPAPT mice, resulting in reduced Aβ and tau pathology and improved cognition. Multi-omics analysis revealed that SRK414 altered gut microbial composition and increased hippocampal kynurenic acid (KYNA), a metabolite linked to neuroimmune regulation. Increased hippocampal KYNA was a

Selenium nanoparticles modulate gut-brain axis via NRF2 to attenuate Parkinsonian neurotoxicity.

Parkinson's disease (PD) is characterized by progressive dopaminergic neurodegeneration driven by mitochondrial dysfunction, oxidative stress, neuroinflammation, and impaired gut-brain communication. Here, we report a biogenic selenium nanoparticle (Se-NP) platform derived from mussel tissue and demonstrate its nano-enabled neuroprotective efficacy in a rotenone-induced zebrafish model of Parkinsonian neurotoxicity. Selenium was extracted from the tissue of Perna viridis (mussel) and used for the biogenic synthesis of Se-NPs through a green reduction approach under controlled conditions. The m

Astragalus polysaccharide ameliorates ischemic stroke via modulating the microbiota-gut-brain axis.

Ischemic stroke (IS) remains a major contributor to global disability and mortality. Astragalus polysaccharide (ASP), a naturally active component derived from Astragalus membranaceus, exhibits therapeutic potential against IS. However, their mechanism against IS via the microbiota-gut-brain axis remains unclear. Our study aimed to evaluate the mechanism of ASP against IS by middle cerebral artery occlusion (MCAO)-induced animal models combined with antibiotics (ABX) and fecal microbiota transplantation (FMT) experiments. In MCAO mice, our results showed that ASP significantly attenuated brain

Persistent molecular scars and gut-brain axis remodeling after acute DSS colitis.

The acute inflammatory phase of dextran sodium sulfate (DSS)-induced colitis has been extensively characterized, often neglecting the long-term systemic consequences that emerge during recovery. Here, we mapped the recovery landscape following acute DSS-induced colitis over a 28-day period, enabling to differentiate transient inflammation from enduring pathophysiological alterations both at peripheral and central level. Colonic recovery showed a transition to an altered steady state, characterized by an increased immune reactivity and vulnerability, rather than a return to baseline. While over

Targeting the Microbiota-Gut-Brain Axis: Emerging Nanomedicine Approaches for Neurodegenerative Diseases.

The microbiota-gut-brain axis (MGBA) is a bidirectional relationship between the gut microbiota (GM) and the brain, where the GM affects the gastrointestinal tract (GIT) and the central nervous system (CNS), and vice versa. Microbiotas are important for several vital body processes, including metabolism, immunity, and homeostasis. The MGBA has three main pathways: the vagal nerve mechanism, the immune-related mechanism, and the neuroendocrine mechanism. GM imbalance, known as dysbiosis, affects the GIT, the brain, and the CNS. Furthermore, dysbiosis is linked to several neurological disorders

Modulating metabolism and reproductive health through microbiome driven gut-brain axis therapies.

The gut microbiome plays a crucial role in regulating metabolic and reproductive health through the gut-brain axis. This review underscores the therapeutic potential of modulating gut microbiota to alleviate conditions such as obesity and polycystic ovarian syndrome (PCOS). Gut dysbiosis as an imbalance microbiome, has direct effect on insulin resistance, hormonal imbalances, and systemic inflammation. It contributes to metabolic and reproductive disparities. This study reveals the coordination between dysbiosis and obesity, and PCOS. Alterations in gut-microbiota is the major contributor of i

Gut-Brain Axis: Unveiling the Interplay Between Diabetes Mellitus and Alzheimer's Disease.

The gut-brain axis (GBA) represents a complex bidirectional communication system linking the gastrointestinal tract with the CNS, influencing various physiological processes, including cognition. Emerging research suggests a significant interplay between diabetes mellitus (DM) and Alzheimer's disease (AD) mediated through this axis. DM, characterized by impaired insulin signaling and chronic inflammation, appears to exacerbate the pathology of AD. Key mechanisms include insulin resistance affecting neuronal function and promoting amyloid-beta accumulation and tau phosphorylation, hallmark feat

COVID-19-associated encephalopathy: connection between neuroinflammation and microbiota-gut-brain axis.

While neurological complications of COVID-19, such as encephalopathy, are relatively rare, their potential significant impact on long-term morbidity is substantial, especially given the large number of infected patients. Two proposed hypotheses for the pathogenesis of this condition are hypoxia and the uncontrolled release of proinflammatory cytokines. The gut microbiota plays an important role in regulating immune homeostasis and overall gut health, including its effects on brain health through various pathways collectively termed the gut-brain axis. Recent studies have shown that COVID-19 pa

HIV-associated neurocognitive disorder: key implications of the microbiota-gut-brain axis.

HIV-associated neurocognitive disorder (HAND) is now recognized to be relatively common in people living with HIV (PLWH), and remains a common cause of cognitive impairment. Unfortunately, the fundamental pathogenic processes underlying this specific outcome of HIV infection have not as yet been fully elucidated. With increased interest in research related to the microbiota-gut-brain axis, the gut-brain axis has been shown to play critical roles in regulating central nervous system disorders such as Alzheimer's disease and Parkinson's disease. PLWH are characterized by a particular affliction,

Fructooligosaccharides and galactooligosaccharides improve hepatic steatosis via gut microbiota-brain axis modulation.

Studies have shown that gut dysbiosis is associated with the steatotic liver disease associated with metabolic dysfunction (MALSD) and its severity. This study evaluated the effects of two commercially available prebiotics fructooligosaccharides (FOS) and galactooligosaccharides(GOS) on hepatic adipogenesis, inflammation, and gut microbiota in high-fat diet-induced MALSD. The results indicated that FOS and GOS effectively reduced insulin resistance, hyperglycaemia, triglyceridemia, cholesterolaemia, and IL-1β serum levels. Moreover, FOS and GOS modulated the lipogenic (SREBP-1c, ACC, and FAS)

Gut-brain axis serotonergic responses to acute stress exposure are microbiome-dependent.

BACKGROUND: Understanding the mechanisms underpinning the response to acute stress is critical for determining how this can be modulated in both health and disease and across sexes. Stress can markedly alter the microbiome and gut-brain axis signaling with the serotonergic system being particularly sensitive to acute stress. As the impact of acute stress on regional serotonergic dynamics in the gut-brain axis and the contribution of the microbiome to this are poorly appreciated, we used microbiota-deficient mice to assess whether the serotonergic response to acute stress exposure is microbiome

Functional gastrointestinal disorders and gut-brain axis: What does the future hold?

Despite their high prevalence, lack of understanding of the exact pathophysiology of the functional gastrointestinal disorders has restricted us to symptomatic diagnostic tools and therapies. Complex mechanisms underlying the disturbances in the bidirectional communication between the gastrointestinal tract and the brain have a vital role in the pathogenesis and are key to our understanding of the disease phenomenon. Although we have come a long way in our understanding of these complex disorders with the help of studies on animals especially rodents, there need to be more studies in humans, e

Diabesity and mood disorders: Multiple links through the microbiota-gut-brain axis.

The global prevalence of diabesity is on the rise, and the clinical, social and economic health burden arising from this epidemic is aggravated by a significant co-morbidity of diabesity with neuropsychiatric disease, particularly depression. Importantly, not only is the prevalence of mood disorders elevated in patients with type 2 diabetes, depressed patients are also more prone to develop diabetes. This reciprocal relationship calls for a molecular and systemic analysis of diabesity-brain interactions to guide preventive and therapeutic strategies. The analysis we are presenting in this revi

Gut commensal Parabacteroides distasonis exerts neuroprotective effects in acute ischemic stroke with hyperuricemia via regulating gut microbiota-gut-brain axis.

BACKGROUND: Hyperuricemia is considered as an independent risk factor for acute ischemic stroke (AIS), and some AIS patients are accompanied by an increase in serum uric acid. Recent studies have highlighted the important role of gut microbiota in both hyperuricemia and AIS, but there is little available data on the relationship between gut microbiota and the pathogenesis of AIS with hyperuricemia (HAS). METHODS: Here we profiled the gut microbiota composition in 63 HAS patients and 269 non-HAS patients through 16s rRNA sequencing. Male rat with hyperuricemia were subjected to middle cerebral

Gut microbiota and Parkinson's disease: exploring pathogenesis and potential therapeutic strategies from a gut-brain axis perspective.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder with a global prevalence exceeding 1‰, posing a significant public health challenge. Although the pathogenesis of PD is not yet fully elucidated, accumulating evidence suggests that it results from the interplay between genetic and environmental factors, highlighting its multifactorial nature. With advances in translational medicine, the gut has emerged as a critical participant in PD onset and progression. This review systematically summarizes the role of the gut in PD, particularly emphasizing potential mechanisms involving n

Never fear, the gut bacteria are here: Estrogen and gut microbiome-brain axis interactions in fear extinction.

Sex differences in the prevalence, symptomatology, severity, and other aspects of various neuropsychiatric diseases have been consistently reported. Stress and fear-related psychopathologies, such as anxiety disorders, depression, and post-traumatic stress disorder, are more prevalent in women. Investigations of the mechanisms underlying this sex disparity have described the influence of gonadal hormones in both humans and animal models. However, gut microbial communities are also likely to play a role, as these communities differ between the sexes, are involved in a bidirectional cycling of s

Heat exposure remodels the gut microbiota to promote heat acclimation through a serotonin-mediated gut-brain axis.

Rising global temperatures pose an increasing challenge to the survival of endotherms. In this context, heat acclimation is a critical adaptive process that enables long-term survival under warming conditions. However, the mechanisms underlying this adaptive process remain poorly understood in birds, especially heat-sensitive domestic poultry. Here, we investigated how heat acclimation enhances adaptation to high-temperature environments in broiler chickens and examined the role of the gut microbiota in this process. Heat acclimation gradually induced a hypometabolic phenotype in broilers, cha

Dynamic dual roles of gut microbial metabolites in Alzheimer's disease: Translational insights for gut-brain axis interventions.

Alzheimer's disease (AD) is a complex neurodegenerative disorder in which the microbiome-gut-brain axis (MGBA) plays a crucial regulatory role through microbial metabolites. This review analyzes the concentration trajectories of representative metabolites across different disease stages and compartments, and synthesizes current preclinical evidence on how these metabolites influence AD pathogenesis, with particular attention to the mechanisms underlying their dual roles. We integrate evidence for bidirectional pathological crosstalk: gut dysbiosis accelerates central neurodegeneration through
