Polycystin 2: A calcium channel, channel partner, and regulator of calcium homeostasis in ADPKD.

Polycystin 2 (PC2) is one of two main protein types responsible for the underlying etiology of autosomal dominant polycystic kidney disease … tential (TRP) channel family of proteins. As a TRP protein, PC2 is a nonselective, cation-permeant, calcium-sensitive channel expressed in all tissue types, where it localizes primarily on the endoplasmic reticulum (ER), primary cilia, and plasma membrane. In addition to its channel function, PC2 interacts with and acts as a regulator of a number of other …

Insights into calcium-sensing receptor trafficking and biased signalling by studies of calcium homeostasis.

The calcium-sensing receptor (CASR) is a class C G-protein-coupled receptor (GPCR) that detects extracellular calcium concentrations, and modulates parathyroid hormone secretion and urinary calcium excretion to maintain calcium homeostasis. The CASR utilises multiple heterotrimeric G-proteins to mediate signalling effects including activation of intracellular calcium release; mitogen-activated protein kinase (MAPK) pathways; membrane ruffling; and inhibition of cAMP production. By studying germline mutations in the CASR and proteins within its signalling pathway that cause hyper- and hypocalca…

[Bone and calcium metabolism associated with malignancy. Bone and calcium metabolism associated with malignancy.].

The bones are the most common organ where estrogen-receptor positive breast cancer tends to metastasize. Thus, it is important to treat and prevent bone metastases by studying pathogenesis of developing metastasis to the bones. As advancing treatments have significantly improved the quality of life in patients with bone metastases of breast cancer. Among them, bone-modifying agents have been playing the most crucial roles by reducing the rate of skeletal related events and also having a possibility of favorable effects on prognosis. Zoledronic-acid and denosumab are currently available and den

The role of the calcium-sensing receptor in disorders of abnormal calcium handling and cardiovascular disease.

PURPOSE OF REVIEW: The calcium-sensing receptor (CaSR) has a central role in parathyroid gland function. Genetic alterations in CaSR are well known to cause inherited forms of abnormal calcium homeostasis. This review focuses on studies investigating the role of CaSR in common disorders of abnormal calcium handling and in cardiovascular calcification. RECENT FINDINGS: Genetic population studies tested the association of common allelic CASR variants with serum and urine calcium levels, kidney stone disease, primary hyperparathyroidism and bone mineral density. The results of these association …

Control of renal calcium, phosphate, electrolyte, and water excretion by the calcium-sensing receptor.

Through regulation of excretion, the kidney shares responsibility for the metabolic balance of calcium (Ca(2+)) with several other tissues including the GI tract and bone. The balances of Ca(2+) and phosphate (PO4), magnesium (Mg(2+)), sodium (Na(+)), potassium (K(+)), chloride (Cl(-)), and water (H2O) are linked via regulatory systems with overlapping effects and are also controlled by systems specific to each of them. Cloning of the calcium-sensing receptor (CaSR) along with the recognition that mutations in the CaSR gene are responsible for two familial syndromes characterized by abnormalit…

The importance of measuring ionized calcium in characterizing calcium status and diagnosing primary hyperparathyroidism.

CONTEXT: Serum total calcium (tCa) is routinely measured for diagnosing calcium disorders but may not reflect levels of biologically active ionized calcium (iCa) in disease or detect all cases of primary hyperparathyroidism. OBJECTIVE: We investigated the utility of measuring iCa and tCa for diagnosing primary hyperparathyroidism. DESIGN: This was an observational, retrospective, cross-sectional study. PATIENTS: We studied a biochemistry cohort of consecutive ambulatory outpatients with suspected bone or calcium metabolism disorders referred for calcium metabolism biochemistry panels and a …

Is the fasting calcium/creatinine a bone resorption marker in patients with calcium renal stones?

Osteoporosis and osteopenia are an important endocrine-metabolic disease that affect women and men from a certain age and it has a high risk and health cost. The aim of this short communication is to show that fasting calcium/creatinine ratio in patients with calcium stones is a marker of bone resorption. We studied 180 patients with renal stones with calcium composition and the relationship of them between the calcium/creatinine in urine after 8 h of fasting with bone densitometry (T-score) and values of bone resorption marker β-crosslaps (ng/ml). The Pearson correlation test was applied for t…

Sodium-calcium exchange is essential for effective triggering of calcium release in mouse heart.

In cardiac myocytes, excitation-contraction coupling depends upon sarcoplasmic reticular Ca2+ release triggered by Ca2+ influx through L-type Ca2+ channels. Although Na+-Ca2+ exchange (NCX) is essential for Ca2+ extrusion, its participation in the trigger process of excitation-contraction coupling is controversial. To investigate the role of NCX in triggering, we examined Ca2+ sparks in ventricular cardiomyocytes isolated from wild-type (WT) and cardiac-specific NCX knockout (KO) mice. Myocytes from young NCX KO mice are known to exhibit normal resting cytosolic Ca2+ and normal Ca2+ transients

Cch1 mediates calcium entry in Cryptococcus neoformans and is essential in low-calcium environments.

The ability of Cryptococcus neoformans to grow at the mammalian body temperature (37 degrees C to 39 degrees C) is a well-established virulence factor. Growth of C. neoformans at this physiological temperature requires calcineurin, a Ca(2+)/calmodulin-dependent protein phosphatase. When cytosolic calcium concentrations are low ( approximately 50 to 100 nM), calcineurin is inactive and becomes active only when cytosolic calcium concentrations rise ( approximately 1 to 10 microM) through the activation of calcium channels. In this study we analyzed the function of Cch1 in C. neoformans and found…

Evidences for calcium-dependent inactivation of calcium current at the frog motor nerve terminal.

Assessment of calcium-dependent inactivation of calcium current in nerve terminals is not feasible due to technical reasons. Perineural measurement of calcium-flow, however, might be utilized as indirect means to evaluate synaptic currents. Using perineural recording from frog neuromuscular junction, supra-threshold stimuli applied to motor nerve in paired-pulse manner with varying inter-pulse intervals (5-50 ms) are demonstrated in this study to cause paired-pulse depression (PPD) of Ca(2+)-current. PPD of Ca(2+)-flow was reduced at lower extracellular Ca(2+) concentrations, in BAPTA-AM and E…

Calcium spikes and calcium currents in neurons from the medial preoptic nucleus of rat.

Ca2+ spikes, their contribution to firing patterns, and the underlying Ca2+ currents in neurons of the medial preoptic nucleus of rat were investigated by tight-seal whole-cell recordings in a slice preparation. Two different types of spikes were recorded: Low-threshold spikes were generated from membrane potentials <-75 mV. High-threshold spikes were recorded when K+ currents were reduced, and were readily evoked from membrane potentials near -40 mV. Both types of spikes were blocked by substitution of Co2+ for Ca2+ in the external medium, but were insensitive to 2.0 microM TTX. Under voltage

Effects of calmodulin antagonists on calcium pump and cytosolic calcium level in human neutrophils.

The concentration of cytosolic free calcium was monitored in suspensions of intact human neutrophils in phosphate-buffered saline by means of the fluorescent indicator Indo 1 trapped in the cytosol. Trifluoperazine and n-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide markedly reduced the amplitude of the transient increase in cytosolic Ca2+ triggered by CaCl2 as well as by N-formyl-methionyl-leucyl-phenylalanine. The effect of the calmodulin antagonists on the calcium burst observed upon cell activation was much more pronounced in the presence of extracellular free calcium than in EGTA-conta…

Duodenal calcium binding protein and active calcium transport in rats: are they functionally related?

The effects of calcitriol and a novel calcitriol analogue, 22-oxacalcitriol (OCT) on duodenal Ca transport, calbindin-D9k mRNA, and calbindin-D9k content were studied in two animal models reflecting common human pathologies, namely arterial hypertension and chronic renal failure, as well as in normal rats. The hormone or its analogue were administered intraperitoneally to vitamin-D-replete rats. Active Ca transport was increased in both spontaneously hypertensive rats (SHR) and in normotensive control WKY rats 5 h after calcitriol dosing of either 60 and 600 ng per rat. In WKY, calbindin-D9k c

Visibility of calcium on MR and CT: can MR show calcium that CT cannot?

PURPOSE: To test the hypothesis that calcium can be visible on MR images without being visible on CT. METHODS: Five different calcium salts ranging in concentration from 0 to 0.45 g/mL were suspended in 2% agarose gel and studied using T2-weighted MR, T1-weighted MR, and CT. MR signal intensity, CT attenuation, and image noise were measured. Relative visibility was determined from these measurements. RESULTS: CT was shown to be more than 10 times as sensitive as T2-weighted MR or T1-weighted MR for the detection of calcium. CONCLUSION: MR cannot show calcium that is occult on CT.

Regulation of calcium current by voltage and cytoplasmic calcium in canine gastric smooth muscle.

The regulation of Ca2+ current by intracellular Ca2+ was studied in isolated myocytes from the circular layer of canine gastric antrum. Ca2+ current was measured with the whole cell patch-clamp technique, and changes in cytoplasmic Ca2+ ([Ca2+]i) were simultaneously measured with indo-1 fluorescence. Ca2+ currents were activated by depolarization and inactivated despite maintained depolarization. Ca2+ current inactivation was fit with a double exponential function. Using Ba2+ or Na+ as charge carriers removed the fast component of inactivation, whereas enhanced intracellular buffering of Ca2+

Calcium metabolism in postmenopausal osteoporotic women is determined by dietary calcium and coffee intake.

Eighty-five patients, age 48 to 77 y with postmenopausal crush fracture osteoporosis, were investigated using a 7-d combined calcium balance and 47Ca tracer-kinetic turnover method taking the dermal calcium loss into account. Individual dietary records were obtained based on a 4-d registration at home before hospital admission and on questioning by a dietitian. The following dietary constituents were estimated: energy content, protein, methionine, cysteine, calcium, phosphate, magnesium, coffee, fiber and vitamin C. All patients were served individual diets based on the dietary records during …

Pathways for the movements of ions during calcium-free perfusion and the induction of the 'calcium paradox'.

The intracellular sodium content of cardiac cells in fish and amphibia, measured with either an isotope technique or with sodium-sensitive micro-electrodes, rises steeply from around 15 mmol/l in calcium-containing solution to as much as 70 mmol/l, during exposure to a Ca2+-free solution. This increase is associated with the development of spontaneous and prolonged action potentials so that the membrane may stabilise around -20 mV. On reperfusion with calcium-containing medium the membrane repolarises before a strong contracture develops. Inhibition of the Na-pump increases both the sodium gai…

Crystallization of calcium-calmodulin-trifluoperazine complex and an attempt at crystallizing calcium-free calmodulin.

Crystals suitable for X-ray structure analysis were obtained for CaM complexed with both calcium ions and a phenothiazine drug, trifluoperazine (TFP). The TFP/CaM binding ratio in crystals was experimentally determined to be nearly 1. An attempt at crystallizing calcium-free calmodulin (CaM) resulted in rigid but non-birefringent solids which exhibited no X-ray reflections.

Specificity of hepatic aldehyde dehydrogenase inhibition by calcium carbimide (calcium cyanamide) in the rat.

Acute, oral administration of 7.0 mg/kg calcium carbimide (calcium cyanamide) to rats, 2 h before sacrifice, produced complete inhibition of hepatic, low-Km (less than 1 microM acetaldehyde) mitochondrial and cytosolic aldehyde dehydrogenase enzymes and significantly inhibited high-Km (approximately 1 mM acetaldehyde) mitochondrial, cytosolic, and microsomal aldehyde dehydrogenase isozymes. Calcium carbimide had no effect on several other hepatic enzyme activities including mitochondrial glutamate dehydrogenase and monoamine oxidase, cytosolic alcohol dehydrogenase, microsomal NADPH-cytochrome…
