Cold-sensitive C afferents of the urinary bladder were studied in adult cats anaesthetised with alpha-chloralose. The bladder was catheterised for fluid instillations and bladder pressure recordings. Pelvic nerve branches were stimulated electrically close to the bladder. Evoked afferent activity was recorded from dissected filaments of the ipsilateral S1-S2 dorsal roots. Responsive afferents were identified using the 'marking technique', based on activity-dependent decrease in C fibre conduction velocity. Of 108 examined bladder C afferents, 14 were activated by innocuous cooling of the bladd

We investigated the contributions of forebrain, brain stem, and spinal neural circuits to interleukin (IL)-1beta-induced sympathetic nerve discharge (SND) responses in alpha-chloralose-anesthetized rats. Lumbar and splenic SND responses were determined in spinal cord-transected (first cervical vertebra, C1), midbrain-transected (superior colliculus), and sham-transected rats before and for 60 min after intravenous IL-1beta (285 ng/kg). The observations made were the following: 1) lumbar and splenic SND were significantly increased after IL-1beta in sham C1-transected rats but were unchanged af

Hypertonic solutions act in the central nervous system (CNS) to increase mean arterial blood pressure (MAP) by activation of the sympathoadrenal axis. However, adrenal nerve activity (pre- and postganglionic nerve fibers) has not been determined during central osmotic stimulation. Therefore, these experiments evaluated adrenal (AdSNA) and renal (RSNA) sympathetic nerve activity, MAP, and heart rate (HR) following CNS administration of isotonic, hypertonic, and hypotonic sodium chloride solutions in chloralose-anesthetized rats. Injection of isotonic saline (5 microliters) did not alter MAP, HR

Unitary impulse activity from tension receptors in the abomasal antrum in sheep anaesthetized with chloralose was studied with concurrent measurements of isometric tension in the antral wall, intraluminal pressures and electromyography (EMG). Rhythmical background activity had a close correlation with intraluminal pressure changes and usually a close correlation with local increases in EMG activity and isometric tension. Impulse frequencies increased, but were not sustained, following changes from isotonic to isometric conditions. Tension receptors were excited by transmural compression, by an

We tested the hypothesis that acute lung injury (ALI) isolated to a perfused in situ left lung preparation results in sustained reflex cardiovascular depression. Phorbol myristate acetate (PMA), an agent that activates neutrophils, administered into the isolated lung preparation of chloralose-anesthetized dogs resulted in ALI, as assessed by wet-to-dry weight ratios and histopathology, and significant decreases in heart rate (43%), mean arterial pressure (27%), aortic blood flow (29%) and maximum rate of change in left ventricular pressure (30%). Significant reflex effects occurred by 20 min a

In the present study we analyzed the cardiovascular, respiratory and behavioral responses evoked in rats by KCN. Hypertension, hyperpnea and alerting behavior were characteristic responses in unanesthetized animals. Selective denervation of aortic and carotid bifurcation areas confirmed the carotid body chemoreceptor (CBC) origin of these responses. Sino-aortic denervation, as well as selective carotid sinus denervation abolished the responses, while, after selective aortic denervation, the responses were not different from those of the control rats. The administration of prazosin abolished th

Intra-abdominal surgery causes a loss of plasma into tissues within and around the abdomen, predisposing to a decreased tissue viability and postoperative complications. In a rat model of intra-abdominal trauma, we investigated whether neuropeptides released from thin afferent nerve endings may contribute to this extravasation. Newborn male Wistar rats were pretreated with capsaicin 50 mg/kg s.c., leading to a lifelong degeneration of thin afferents. The same animals, when adult, were subjected to experiments under chloralose anesthesia in which tissue clearances of radiolabelled albumin were

We tested the hypothesis that cocaine-induced impairment of left ventricular function results in cardiogenic pulmonary edema. Mongrel dogs, anesthetized with alpha-chloralose, were injected with two doses of cocaine (5 mg/kg iv) 27 min apart. Cocaine produced transient decreases in aortic and left ventricular systolic pressures that were followed by increases exceeding control. As aortic pressure recovered, left ventricular end-diastolic, left atrial (Pla), pulmonary arterial (Ppa), and central venous pressures rose. Cardiac output and stroke volume were reduced when measured 4-5 min after coc

Microneurographic studies have shown that sympathetic vasomotor nerve activity is observed less frequently in patients with peripheral neuropathy than in controls, but when detected its pattern and the baroreflex latencies appear to be normal. Vasomotor nerve function was examined in chloralose-urethane anaesthetised dogs by comparing the discharge of renal sympathetic nerves in control animals and animals with acrylamide neuropathy. There was a normal pattern of pulse related inhibition of sympathetic nerve activity as well as normal amplitude spontaneous compound nerve action potentials in t

Morphine, an opiate alkaloid with mixed mu- and delta-agonist properties, raises the ventricular fibrillation threshold in anesthetized dogs by altering autonomic tone. To elucidate further underlying structure-activity relationships, the effect of fentanyl, a nonalkaloid, mu-selective agonist in wide clinical use, was studied. Fentanyl (30 micrograms/kg) was given intravenously to 27 chloralose-anesthetized dogs, and ventricular fibrillation threshold was measured by means of the single-stimulus technique. In the baseline state fentanyl raised the ventricular fibrillation threshold by 14%. Wh

The present study was undertaken to evaluate the extent that the lung inflation reflex attenuates vasoconstrictor responses in renal cortex and splanchnic beds during severe arterial hypoxia. Hypoxia was induced by inspiration of a 3-5% oxygen gas mixture in three groups of chloralose-anesthetized dogs: Group I, free breathing; Group II, controlled ventilation; Group III, free breathing with arterial PCO2 held constant. Regional vascular conductances (VC) were calculated from regional blood flows measured with 15-microns radioactive microspheres. In Group I, hypoxia caused marked hyperventilat

We recorded the afferent activity of 11 left ventricular mechanoreceptors in filaments of the vagus nerve in 10 chloralose-anaesthetised cats. The fibres showed low irregular spontaneous activity of 2.9 (0.2 to 8.4) spikes X s-1. During temporary occlusion of the left anterior descending or left main coronary artery they were activated to 19.1 (4.8 to 47.0) spikes X s-1. Intravenous infusion of 0.175 and 0.35 mg X kg-1 X min-1 lignocaine lowered heart rate and blood pressure. The spontaneous nerve fibre activity remained unchanged by the local anaesthetic, whereas the maximum activity evoked b

In dogs anesthetized with chloralose-urethan on right heart bypass, left ventricular (LV) performance was assessed at constant LV stroke work before and for up to 2.5 h after crystalloid hemodilution was established. Lowering the hematocrit from 43.3 +/- 1.3% to 13.6 +/- 1.7% (SE) did not significantly change LV end-diastolic pressure (LVEDP) initially. After 80 min LVEDP increased slightly by 1.7 +/- 0.6 cmH2O (P less than 0.05) at a stroke work of 17.3 +/- 2.3 g.m. The value of dP/dt did not change significantly throughout. When LV function curves were generated by increasing cardiac output,

Coronary vascular and myocardial responses to selective hypoxic and/or hypercapnic carotid chemoreceptor stimulation were investigated in constantly ventilated, pentobarbital or urethan-chloralose anesthetized dogs. Bilaterally isolated carotid chemoreceptors were perfused with autologous blood of varying O2 and CO2 tensions via an extracorporeal lung circuit. Systemic gas tensions were unchanged. Effects of carotid chemoreceptor stimulation on coronary vascular resistance, left ventricular dP/dt, and strain-gauge arch output were studied at natural coronary blood flow with the chest closed an

This study investigated the role of γ-aminobutyric acid subtype B (GABAB) receptors in tibial and pudendal neuromodulation of bladder overactivity induced by intravesical administration of dilute (0.5%) acetic acid (AA) in α-chloralose-anesthetized cats. To inhibit bladder overactivity, tibial or pudendal nerve stimulation (TNS or PNS) was applied at 5 Hz and two or four times threshold (T) intensity for inducing toe or anal sphincter twitch. TNS at 2T or 4T intensity significantly (P < 0.05) increased the bladder capacity to 173.8 ± 16.2 or 198.5 ± 24.1%, respectively, of control capacity. Me

BACKGROUND: Traditional tibial nerve stimulation (TNS) has been used to treat overactive bladder syndrome (OAB), but there are some shortcomings. Thus, a novel alternative is needed for the treatment of OAB. The study investigated the effects of a new type of tibial nerve microstimulator on the micturition reflex in cats.
METHODS: An implantable wireless driver microstimulator was implanted around the tibial nerve in 9 α-chloralose anesthetized cats. Cystometry was performed by infusing 0.9% normal saline (NS) or 0.25% acetic acid (AA) through a urethral catheter. Multiple cystometrograms wer

Respiratory effects of intravenous serotonin and nicotine were investigated prior to and after bilateral neurotomy of the carotid sinus nerves (CSNs) in eight pentobarbitone/chloralose-anaesthetised, bilaterally vagotomised and superior laryngeal nerves-sectioned cats. Injection of 188 nmol kg(-1) serotonin (hydrogen oxalate salt, 50 microg x kg(-1)) prior to and after CSNs section induced an expiratory apnoea of, respectively, 7.9 +/- 1.25 s and 8.3 +/- 1.6 s duration (mean +/- S.E.M.) in, respectively, five and three of those cats. In all cats, the serotonin challenge produced a period of ac

Sacral neurones (S1 and S2 segments) projecting to the cervical cord (C6 segment) and to the contralateral gigantocellular reticular nucleus (coGRN) were electrophysiogically studied on 7 adult cats under alpha-chloralose anaesthesia. Antidromic action potentials were recorded from 48 neurones following stimulation of their axons in both the C6 segment and coGRN. Two groups of investigated cells were distinguished in the gray matter of sacral segments: one distributed in laminae V-VI (12 neurones), the other located in medial part of lamina VII and lamina VIII (36 neurones). All cells from the

This study investigates the effect of angiotensin-converting-enzyme inhibition by intravenous enalaprilat (100 micrograms/kg) on splanchnic vascular capacitance during acute left ventricular failure induced by coronary microembolization in alpha-chloralose/urethan anesthetized dogs. Changes in hepatic and splenic vascular volumes were determined from organ diameters (sonomicrometry) at 15, 30, and 45 min after enalaprilat injection. Changes in vascular capacitance were assessed from organ pressure-diameter curves obtained during transient hepatic outflow occlusion. Thirty minutes after enalapr

OBJECTIVE: Despite extensive knowledge about basic baroreceptor function, there continues to be controversy as to whether there is postnatal maturation of the cardiovascular baroreflex. DESIGN/ANIMALS: Acutely studied postnatal developmental changes in baroreflex sensitivity with a neonatal porcine model in 28 chloralose-anesthetized piglets from birth to six weeks of age.
INTERVENTIONS: Steady-state heart rate versus blood pressure response curves were generated by altering blood pressure with increasing doses of phenylephrine or nitroprusside. Data were fitted to the Hill equation for sigmo
