Opioidergic receptors in the arcuate nucleus are not involved in the cardiovascular effects of clonidine.

The arcuate nucleus is the bed nucleus for the pro-opiomelancortin system of the brain with important connections with other nuclei involved in cardiovascular function. Clonidine has been reported to produce its cardiovascular effects through an interaction with opioid and alpha 2-adrenergic receptors. The present study examined the arcuate nucleus as a site of action of clonidine. Male spontaneously hypertensive rats were anesthetized with pentobarbital and were instrumented for the measurement of blood pressure and heart rate. Cannulae were placed either through the cisterna magna (IC) or in…

Effects of clonidine on the calcium content and morphology of rat salivary glands.

These effects were examined with and without pretreatment of animals with reserpine and the adrenergic antagonists prazosin (alpha 1), yohimbine (alpha 2) and propranolol (beta). The effects of clonidine on glandular concentrations of norepinephrine and dopamine also were examined. These effects were compared with those of xylazine, a presynaptic alpha 2-adrenergic agonist. A single, high dose of clonidine followed by an overnight fast caused marked increases in calcium content and acinar secretory granules in the submandibular and sublingual glands, similar to those caused by reserpine. Howev…

Evaluation of clonidine suppression and various provocation tests in the diagnosis of pheochromocytoma.

Recent investigations have shown that the widely used clonidine suppression test is sometimes fallible for the diagnosis of pheochromocytoma. A comparative assessment was made of the following suppression and provocation tests, the clonidine suppression test, and the glucagon, metoclopramide, and naloxone provocation tests. The assessment was performed in 6 patients with pheochromocytoma and in 19 patients without pheochromocytoma who were initially suspected of harboring a tumor. BP response did not predict the presence of pheochromocytoma in any test. Plasma norepinephrine (NE) concentration…

Hemodynamic effects of a combination of clonidine and propranolol in conscious cirrhotic rats.

The hemodynamic effects of the combination of clonidine and propranolol were studied in conscious rats with portal hypertension owing to secondary biliary cirrhosis. Pressure and blood flow measurements (radioactive microsphere method) were performed in three groups of eight rats before and after drug administration. The combined effects of clonidine (2 micrograms/100 g body wt., i.v.) and propranolol (0.2 mg/min for 10 min) were compared with those observed after administration of either clonidine alone or propranolol alone. The association of clonidine and propranolol induced significant dec…

Abolition of clonidine's effects on ventricular refractoriness by naloxone in the conscious dog.

The interaction between opiate and adrenergic receptors on cardiac electrophysiologic function in the conscious dog was addressed in our study. We examined the effects of opiate receptor blockade with naloxone on clonidine-induced changes in refractoriness of the cardiac ventricle. Nine dogs were chronically instrumented for recording mean arterial blood pressure, administration of drugs and for measurement of effective refractory period of the ventricle. Clonidine (10 micrograms/kg, i.v.) significantly (p less than 0.05) decreased heart rate to 72 +/- 5 beats/minute from 108 +/- 8 beats/minut…

Effects of clonidine on alprazolam discontinuation in panic patients: a pilot study.

Effects of adjunctive clonidine (0.15 to 0.7 mg/day) on symptoms experienced during and for 4 weeks after gradual alprazolam discontinuation were observed in panic disorder patients after 6 weeks of successful treatment. Twelve of 14 entered patients were considered to have had a sufficient period of discontinuation (2 weeks) and clonidine administration (1 week) for effects to be assessed adequately. Nine of these 12 patients reached zero dose of alprazolam in 3 to 4 weeks. However, 10 of 12 patients experienced new withdrawal symptoms and 11 of 12 experienced recurrent panic attacks during t…

Clonidine in the treatment of Tourette's syndrome exacerbation due to haloperidol withdrawal.

A patient with long-standing Tourette's syndrome had a dramatic exacerbation of symptoms following the rapid withdrawal of haloperidol. Clonidine administration resulted in a disappearance of tics, which recurred 6 days later. The role of clonidine in blocking withdrawal-induced symptoms in patients with Tourette's syndrome treated with long-term neuroleptics is discussed from a clinical and neurobiological perspective. The implications of this case for the treatment of supersensitivity psychosis and tricyclic withdrawal states with clonidine are also discussed.

Clonidine prevents the sympathetic initiation and aggravation of poststenotic myocardial ischemia.

We studied the effects of clonidine (10 micrograms/kg i.v.) on the positive feedback between poststenotic myocardial ischemia and cardiac sympathetic nerve activation and on the initiation of poststenotic myocardial ischemia during bilateral carotid occlusion in 12 anesthetized, open-chest dogs. During 20 min of acute severe coronary stenosis, cardiac sympathetic nerve activity increased by 24 +/- 6%, poststenotic coronary resistance increased from 0.45 +/- 0.05 to 0.61 +/- 0.08 mm Hg . min . 100 g/ml, and the poststenotic myocardial lactate consumption of 25 +/- 6 mumol/min . 100 g at 1 min o…

Regimen for the control of blood pressure and symptoms during clonidine withdrawal.

Abrupt withdrawal of the centrally-acting antihypertensive agent, clonidine, is associated with a high incidence of rebound hypertension and tachycardia, with symptoms of sympathetic overactivity and increased catecholamine excretion. Gradual clonidine withdrawal has been recommended, but does not always avoid the reaction. A regimen is described comprising high doses of the alpha 1-adrenoceptor antagonist, prazosin, the cardioselective beta-blocker, atenolol, and chlordiazepoxide, specifically designed to counter both central and peripheral effects of sudden withdrawal of a central alpha 2-ad…

beta-Endorphin and essential hypertension: importance of the clonidine-naloxone interaction.

Analysis of the effect of naloxone (0.4 mg iv.) on clonidine hypotension in 80 patients with essential hypertension revealed that two groups could be separated. In the responding group (43 pts) naloxone increased blood pressure and heart rate in clonidine-treated patients while in the non-responding group (37 pts) it has no such effect. Patients in the responding group had higher cardiac output, stroke volume, plasma renin activity, plasma adrenaline and beta-endorphin levels and lower total peripheral resistance, shorter history of hypertension and lesser body weight than those in the non-res…

Treatment of Gilles de la Tourette's syndrome with clonidine and neuroleptics.

In an open clinical trial of clonidine hydrochloride and neuroleptics in patients with Gilles de la Tourette's syndrome, neuroleptics resulted in greater improvement across the range of symptoms in a larger proportion of patients than did clonidine. A small subgroup of patients may have benefited from treatment with clonidine. Carefully controlled, randomized, and double-blind studies are necessary to resolve the divergent views about the effectiveness of clonidine in the treatment of Gilles de la Tourette's syndrome.

Cardiovascular changes during morphine withdrawal in the rat: effects of clonidine.

Arterial blood pressure and heart rate were measured in unrestrained rats as an index of the autonomic component of the morphine withdrawal … ent rats also evoked a dose-related increase in MAP to about 40 mmHg. The antiwithdrawal effects of clonidine were tested in this model by pretreating dependent rats with this agent (6-60 micrograms/kg). Clonidine inhibited the pressor response produced by naloxone by 23-60%. These findings indicate that the increase in MAP during opiate withdrawal provides …

Functional restoration of the traumatically injured spinal cord in cats by clonidine.

The long-term, chronic, paralysis resulting from spinal cord injury in the cat has been reversed by the use of an alpha 2-adrenergic receptor agonist, clonidine. Administration of this drug resulted in "normalization" of sensory-motor and autonomic dysfunctions. Preliminary studies of the clonidine in humans with traumatically injured spinal cord indicate that autonomic dysreflexia can be controlled and spasticity minimized. The data suggest that biochemical and pharmacologic manipulation of receptors may ameliorate paralysis following traumatic injury to the spinal cord as well as to the brai…

[Morphine antagonizes the central hypotensive effect of clonidine in the anesthetized cat].

A dose of 1 microgram/kg of clonidine injected intracerebroventricularly (I.C.V.) in the normotensive anaesthetized Cat elicits a significant and long lasting drop in the mean arterial blood pressure (23 +/- 5 mmHg) and the heart rate (34 +/- 5 beats/min.). A pretreatment with morphine (100 micrograms/kg, I.C.V.) completely prevents the central hypotensive effect of clonidine, whereas the bradycardia is only weakly influenced. Administered after clonidine, morphine reverses the hypotensive effect of clonidine whereas morphine does not affect its bradycardic action. These data demonstrate that …

The use of clonidine for management of opiate abstinence in a chronic pain patient.

The use of clonidine in the management of opiate abstinence is presented in a patient dependent upon levorphanol tartrate given for chronic pain. Use of levorphanol was abruptly discontinued, and the patient was monitored for signs and symptoms of opiate withdrawal. He manifested a significant increase in pulse and blood pressure and had perspiration, agitation, and opiate-seeking behavior. Clonidine effectively abolished these signs and symptoms. The mechanism by which clonidine prevents the opiate abstinence syndrome is discussed. Clonidine is a safe and inexpensive means of achieving rapid …

[Blocking by naloxone of the antihypertensive effect of clonidine in hypertensive and normotensive animals].

Antagonism between cardiovascular effects of clonidin (10-30 micrograms/kg) and naloxon (0.15-1.0 mg/kg) was studied on anesthetized spontaneously hypertensive rats (SHR) as compared to Wistar Kyoto (WKY) rats and on awake cats. Naloxon was shown to completely block the clonidin hypotensive effect in SHR and cats and to partially block it in WKY rats. In some experiments, naloxon was administered after the blockade of peripheral M-cholinoreceptors by metacin (2 mg/kg). In these experiments naloxon evoked elevation of blood pressure, cardiac output and heart rate (in cats). It is concluded that

Influence of analgesic antipyretics on the central cardiovascular effects of clonidine in rats.

The centrally acting antihypertensive drug clonidine has been found to stimulate the synthesis of PGF2 alpha in the brain. Centrally administered PGF2 alpha, in turn, induces rises of blood pressure and heart rate. We therefore studied the influence of inhibitors of prostaglandin (PG) synthesis on the cardiovascular effects of clonidine in urethane-anaesthetised rats. Pretreatment with indomethacin or paracetamol (100 micrograms/rat into the fourth cerebral ventricle) antagonised the central hypotensive effect of clonidine (0.125-16.0 micrograms/rat into the fourth cerebral ventricle). The bra…

Clonidine in the central nervous system: site and mechanism of hypotensive action.

The data presented in this paper must be considered in conjunction with the fact that clonidine is both a partial alpha-adrenergic agonist and a partial alpha-adrenergic antagonist. In the anterior hypothalamus, clonidine, acting as an alpha-agonist, excites a pathway that inhibits excitatory cardiovascular neurons. Thus, the effect of neurons from the nucleus tractus solitarii (NTS) in inhibiting sympathetic outflow from the vasomotor center is effectively increased. In the posterior hypothalmus, clonidine, acting as an alpha-antagonist, decreases excitation of excitatory cardiovascular neuro…
